Kcnj16 (Kir5.1) Gene Ablation Causes Subfertility and Increases the Prevalence of Morphologically Abnormal Spermatozoa.

The ability of spermatozoa to swim towards an oocyte and fertilize it depends on precise K+ permeability changes. Kir5.1 is an inwardly-rectifying potassium (Kir) channel with high sensitivity to intracellular H+ (pHi) and extracellular K+ concentration [K+]o, and hence provides a link between pHi and [K+]o changes and membrane potential. The intrinsic pHi sensitivity of Kir5.1 suggests a possible role for this channel in the pHi-dependent processes that take place during fertilization. However, despite the localization of Kir5.1 in murine spermatozoa, and its increased expression with age and

Orchestration of occludins, claudins, catenins and cadherins as players involved in maintenance of the blood-epididymal barrier in animals and humans.

Although spermatozoa are formed during spermatogenesis in the testis, testicular spermatozoa are immature and cannot swim or fertilize. Thes … regulation of these cellular junctions is thought to represent a key determinant in the process of sperm maturation within the epididymis. Tight junctions between adjacent principal cells permit the formation of a specific microenvironment in the lumen of the epididymis that is essential for sperm maturation. Although we have made significant progress in understa …

WAVE1, an A-kinase anchoring protein, during mammalian spermatogenesis.

BACKGROUND: Proper compartmentalization of signalling cascades is paramount to many intracellular activities during spermatogenesis and sperm function. In the present study we focus on the A-kinase-anchoring protein (AKAP) WAVE1, a member of the Wiskott-Aldrich syndrome (WASP) family of adaptor proteins, to study its localization throughout mammalian spermatogenesis. METHODS: Using transmission electron microscopy, immunocytochemistry and western blotting, we examined the distribution of WAVE1 and putative partners during mammalian spermatogenesis. The localization and association of PKA RII,

Selenium mitigates cyclophosphamide-induced toxicity on sperm quality in mice: An experimental study.

BACKGROUND: Cyclophosphamide (CP), a commonly used chemotherapy drug, induces oxidative stress and damages testicular tissue, leading to impaired spermatogenesis. OBJECTIVE: This study aimed to evaluate the effects of selenium, a potent antioxidant, on sperm parameters in adult mice treated with CP. MATERIALS AND METHODS: In this experimental study, 32 adult male NMRI mice were randomly divided into 4 groups (n = 8/each): A) control, B) CP (100 mg/kg/wk, intraperitoneally), C) selenium (1 mg/kg/day, intraperitoneally), and D) CP + selenium. After 35 days of treatment, spermatozoa were collec

Prepubertal arsenic exposure alters phosphoproteins profile, quality, and fertility of epididymal spermatozoa in sexually mature rats.

Arsenic intoxication affects male reproductive parameters of prepubertal rats. Besides, morphological and functional alterations in their testis and epididymis may remain after withdrawal of arsenic insult, causing potential impairment in male fertility during adulthood. In this study, we aimed at analyzing the effect of prepubertal arsenic exposure on the fecundity of epididymal sperm from sexually mature Wistar rats, assessing fertility indexes, sperm parameters, and sperm phosphoproteins content. Male pups on postnatal day (PND) 21 received filtered water (controls, n = 10) and 10 mg L-1 ar

Synthesis and processing of rat sperm-associated alpha-L-fucosidase.

alpha-L-Fucosidase (alpha-fucosidase) is the most active of a number of glycosidases measured in rat epididymal sperm through use of artificial 4-methylumbelliferyl substrates. In addition, enzyme activity can be detected in purified populations of testicular germ cells; in comparison to round spermatids, caput epididymal sperm show at least a 1.8-fold increase in alpha-fucosidase activity per cell. Metabolic labeling of cultured testicular germ cells, followed by immunoprecipitation and SDS-PAGE, reveals synthesis of alpha-fucosidase as a polypeptide of 54 kDa in pachytene spermatocytes and r

Beta-defensins as marker for male fertility: a comprehensive review†.

Bovine male fertility in animals has a direct impact on the productivity of dairy herds. The epididymal sperm maturations involve extensive sperm surface modifications to gain the fertilizing ability, especially by absorptions of the plethora of biomolecules, including glycoprotein beta-defensins (BDs), enzymes, organic ions, protein, and phospholipids. Defensins are broad-range nonspecific antimicrobial peptides that exhibit strong relations with innate and adaptive immunity, but their roles in male fertility are relatively recently identified. In the course of evolution, BD genes give rise t…

Reproductive seasonality drives structural and functional shifts in spermatozoa of the Neotropical rattlesnake Crotalus durissus (Squamata: Viperidae).

In brief: Reproductive seasonality in snakes reflects evolutionarily conserved adaptive mechanisms that sustain sperm function despite seaso … that the distal segment of the ductus deferens plays a role in sperm maintenance and potentially in sperm maturation. The extended viability of spermatozoa outside the active reproductive phase highlights key physiological adaptations and supports potential applications in reptile reproductive management and conservation.

Sperm immotility is associated with epididymis metabolism disorder in mice under obstructive azoospermia.

Obstructive azoospermia (OA) accounts for approximately 40% of males who suffer from azoospermia of male infertility. Currently, available t … g mass spectrometry in epididymal fluid, we found differences in the expression of key proteins for sperm maturation, such as Angiotensinogen (AGT), rhophilin-associated tail protein 1 (ROPN1), NPC intracellular cholesterol transporter 2 (NPC2), and prominin 1 (PROM1). Furthermore, our results demonstrated that AGT, secreted by epididymal principal cells, could re …

Heat Stress and the Bull Sperm Transcriptome: Molecular Mechanisms, Fertility, and Epigenetic Inheritance.

Heat stress (HS) occurs when the body's physiological mechanisms for regulating body temperature are unable to cope with external conditions. Regarding male reproductive function, HS increases testicular temperature, leading to a reduction in sperm production and motility, and an increase in abnormal sperm, oxidative stress and DNA fragmentation. However, the molecular basis behind these induced HS changes has not been fully characterized. In bovine spermatozoa, post-transcriptional regulation of messenger RNA (mRNA) is activated during spermatogenesis, and mechanisms mediated by microRNAs (mi

Sperm-agglutinating antibodies and testicular morphology in fifty-nine men with azoospermia or cryptozoospermia.

The relationship between the state of the germinal epithelium and the type and titer of circulating sperm-agglutinating antibodies has been investigated in a series of 59 azoospermic or occasionally cryptozoospermic men. The patients were grouped according to the condition of the germinal epithelium as observed from testicular biopsy specimens, as well as to type and titer of circulating sperm-agglutinating antibodies investigated by a previously described microagglutination technique. Evidence is presented to suggest that the presence of mature spermatozoa in the testicular structures may be

Male reproductive biology was reshaped during placental mammal diversification through epididymal secretome expansion.

The evolutionary timing of the origin of secretory proteins underlying post-mating reproductive processes remains uncharacterized in vertebr … tion. These genes are predominantly expressed in the epididymis, where they protect sperm and drive sperm maturation, influencing essential post-mating reproductive processes and male reproductive outcome. In contrast, MR secretome genes that originated along other evolutionary branches are primarily associated with sperm structure, motility, egg binding, and fusi …

Somatic-Immune Cells Crosstalk In-The-Making of Testicular Immune Privilege.

Immunological infertility contributes significantly to the etiology of idiopathic male infertility. Shielding the spermatogenic cells from systemic immune responses is fundamental to secure normal production of spermatozoa. The body's immune system is tuned with the host self-components since the early postnatal period, while sperm first develops during puberty, thus rendering spermatogenic proteins as 'non-self' or 'antigenic.' Development of antibodies to these antigens elicits autoimmune responses affecting sperm motility, functions, and fertility. Therefore, the testes need to establish a

The epididymal amyloid matrix: structure and putative functions.

BACKGROUND: We previously demonstrated the normal mouse epididymal lumen contains a non-pathological amyloid matrix that surrounds spermatozoa and plays important roles in sperm maturation and protection. OBJECTIVE: The objective herein was to present a review of this work, including studies showing the amyloid structures of four members of the CRES (cystatin-related epididymal spermatogenic) subgroup are integral and essential components of the amyloid matrix. METHODS: We used conformation-dependent reagents that recognize the cross-β-sheet structure characteristic of amyloid, including thi…

Partial deletion of chromosome 8 β-defensin cluster confers sperm dysfunction and infertility in male mice.

β-defensin peptides are a family of antimicrobial peptides present at mucosal surfaces, with the main site of expression under normal condit … cellular calcium content. Thus we demonstrate in vivo that β-defensins are essential for successful sperm maturation, and their disruption leads to alteration in intracellular calcium, inappropriate spontaneous acrosome reaction and profound male infertility.

Development and Preservation of Avian Sperm.

Terminally differentiated avian sperm consist of a head which male genetic material locates and flagellum that provides the motive force to propel them towards the fertilization site. The apical end of the sperm head accommodates a secretory vesicle, called an acrosome, that undergoes acrosome reaction releasing proteolytic content to penetrate the peri-vitelline membrane of an egg. Transcriptionally and translationally inactive, sperm need to rely on these distinct compartments in which different functions are preassembled, in order to achieve the goal of "fertilization". How are these comple

Meichroacidin containing the membrane occupation and recognition nexus motif is essential for spermatozoa morphogenesis.

Meichroacidin (MCA) is a highly hydrophilic protein that contains the membrane occupation and recognition nexus motif. MCA is expressed during the stages of spermatogenesis from pachytene spermatocytes to mature sperm development and is localized in the male meiotic metaphase chromosome and sperm flagellum. MCA sequences are highly conserved in Ciona intestinalis, Cyprinus carpio, and mammals. To investigate the physiological role of MCA, we generated MCA-disrupted mutant mice; homozygous MCA mutant males were infertile, but females were not. Sperm was rarely observed in the caput epididymidis
