ATP-sensitive K+ channels control the spontaneous firing of a glycinergic interneuron in the auditory brainstem.

KEY POINTS: Cartwheel neurons provide potent inhibition to fusiform neurons in the dorsal cochlear nucleus (DCN). Most cartwheel neurons fire action potentials spontaneously, but the ion channels responsible for this intrinsic activity are unknown. We investigated the ion channels responsible for the intrinsic firing of cartwheel neurons and the stable resting membrane potential found in a fraction of these neurons (quiet neurons). Among the ion channels controlling membrane potential of cartwheel neurons, the presence of open ATP-sensitive potassium channels (KATP ) is responsible for the exi

Chloride channel inhibition improves neuromuscular function under conditions mimicking neuromuscular disorders.

AIM: The skeletal muscle Cl- channels, the ClC-1 channels, stabilize the resting membrane potential and dampen muscle fibre excitability. This study explored whether ClC-1 inhibition can recover nerve-stimulated force in isolated muscle under conditions of compromised neuromuscular transmission akin to disorders of myasthenia gravis and Lambert-Eaton syndrome. METHODS: Nerve-muscle preparations were isolated from rats. Preparations were exposed to pre-or post-synaptic inhibitors (ω-agatoxin, elevated extracellular Mg2+ , α-bungarotoxin or tubocurarine). The potential of ClC-1 inhibition (9-AC

Cochlear synaptopathy: new findings in animal and human research.

In animal models, prolonged exposure (2 h) to high-level noise causes an irreparable damage to the synapses between the inner hair cells and auditory nerve fibers within the cochlea. Nevertheless, this injury does not necessarily alter the hearing threshold. Similar findings have been observed as part of typical aging in animals. This type of cochlear synaptopathy, popularly called "hidden hearing loss," has been a significant issue in neuroscience research and clinical audiology scientists. The results obtained in different investigations are inconclusive in their diagnosis and suggest new st

Autoimmune Attack of the Neuromuscular Junction in Myasthenia Gravis: Nicotinic Acetylcholine Receptors and Other Targets.

The nicotinic acetylcholine receptor (nAChR) family, the archetype member of the pentameric ligand-gated ion channels, is ubiquitously distributed in the central and peripheral nervous systems, and its members are the targets for both genetic and acquired forms of neurological disorders. In the central nervous system, nAChRs contribute to the pathological mechanisms of neurodegenerative disorders, such as Alzheimer and Parkinson diseases. In the peripheral nerve-muscle synapse, the vertebrate neuromuscular junction, "classical" myasthenia gravis (MG) and other forms of neuromuscular transmissi

Alteration of GABAergic neurotransmission in Huntington's disease.

Hereditary Huntington's disease (HD) is characterized by cell dysfunction and death in the brain, leading to progressive cognitive, psychiatric, and motor impairments. Despite molecular and cellular descriptions of the effects of the HD mutation, no effective pharmacological treatment is yet available. In addition to well-established alterations of glutamatergic and dopaminergic neurotransmitter systems, it is becoming clear that the GABAergic systems are also impaired in HD. GABA is the major inhibitory neurotransmitter in the brain, and GABAergic neurotransmission has been postulated to be m

Ovarian Hormones Organize the Maturation of Inhibitory Neurotransmission in the Frontal Cortex at Puberty Onset in Female Mice.

The frontal cortex matures late in development, showing dramatic changes after puberty onset, yet few experiments have directly tested the role of pubertal hormones in cortical maturation. One mechanism thought to play a primary role in regulating the maturation of the neocortex is an increase in inhibitory neurotransmission, which alters the balance of excitation and inhibition. We hypothesized that pubertal hormones could regulate maturation of the frontal cortex by this mechanism. Here, we report that manipulations of gonadal hormones do significantly alter the maturation of inhibitory neur

De-stabilization of the positive vago-vagal reflex in bulimia nervosa.

Bulimia nervosa is characterized by consuming large amounts of food over a defined period with a loss of control over the eating. This is followed by a compensatory behavior directed at eliminating the consumed calories, usually vomiting. Current treatments include antidepressants and/or behavioral therapies. Consensus exists that these treatments are not very effective and are associated with high relapse rates. We review evidence from literature and present original data to evaluate the hypothesis that bulimia involves alterations in vago-vagal function. Evidence in support of this include (

Anticonvulsive activity of a barbiturate derivative HB-7 in a model of allylglycine-induced epilepsy in cats.

With the aim of characterizing further the anticonvulsive activity of the newly-synthesized barbituric derivative 2-hydroxylamino-5-ethyl-5-sec. penthylbarbituric acid (HB-7), we studied its effects on allylglycine-induced epileptiform electrocorticogram (ECoG) in cats under acute conditions. Pentobarbital (PB) was used for comparison. Allylglycine (AG) was administered in a dose of 100 mg/kg i.v. to all cats. The control animals were treated with AG only. HB-7 and PB were applied in doses of 20 mg/kg i.p., about 60 min after AG. Intermittent light stimulation (ILS) at flash rate of 8/s and 25

Pharmacology of antidepressants.

Choice of an antidepressant medication is, in part, based on the side effects produced by a drug and the desire to avoid certain reactions in a particular patient. The clinician needs a reliable method of predicting which medications are most likely to produce specific untoward effects. Understanding the synaptic pharmacology of the most commonly used agents could serve as a tool for predicting possible side effects and drug-drug interactions. Antidepressant drugs alter neurotransmitter effects at nerve synapses, probably by blocking norepinephrine and serotonin reuptake, and blockade of neuro

Abnormalities of sympathetic regulation after cervical cord lesions.

Microneurographic recordings suggest that hypertensive reactions in patients with cervical spinal cord lesions usually are due, not to exaggerated sympathetic nerve reactions, but rather to the combination of more diffuse sympathetic nerve activation than normally, exaggerated vascular reactions and absence of blood pressure restraining reflexes.

Functional anatomy of the peripheral olfactory system of the African lungfish Protopterus annectens owen: development of the primary olfactory pathway during postembryonic growth.

Seven specimens of the African lungfish ranging in size from 4 to 350 gm were used for the study of changes in the olfactory nerve and bulb during postembryonic growth as expressed by the increasing number of lamellae of the olfactory organ. Serial semithin sections were used, and the data were studied mathematically. The number of axons of the olfactory nerve, the surface area of the bulb, and the number of mitral cells increase exponentially; the mitral cell density decreases exponentially; and the calculated average convergence (counted axons/counted mitral cells) of axons onto the cells of

Lambert-Eaton myasthenic syndrome.

This review of the Lambert-Eaton myasthenic syndrome (LEMS) emphasizes electrodiagnosis and includes a case report. A 50-year-old woman had become progressively weaker over 1.5 years. The suspected diagnosis was confirmed by the clinical electrophysiological findings and was made 6 months before the patient's oat-cell carcinoma was found. After treating with local radiation and chemotherapy, the myasthenic syndrome went into remission as the pulmonary lesion resolved.

Distinctive characteristics of dopamine neuron release mechanisms: Insights into Parkinson's Disease.

Parkinson's disease (PD) is the second most prevalent neurodegenerative disorder worldwide, after Alzheimer's disease, and is characterized not only by progressive motor dysfunction but also by a wide array of non-motor symptoms that frequently emerge during the prodromal phase. These early manifestations, including cognitive and neuropsychiatric impairments, autonomic dysfunction, and sleep disturbances, contribute substantially to disease burden and often precede the onset of motor impairments by years. Although degeneration of dopaminergic neurons in the substantia nigra pars compacta (SNpc
