Advantages and research progress of three-dimensional culture systems for lung cancer drug screening (Review).

Conventional two-dimensional cell cultures predominantly grow as monolayers, lacking the complex spatial architecture of in vivo tumors. However, three-dimensional (3D) tumor culture systems can overcome these limitations by reconstituting cell-cell and cell-matrix interactions, thereby recapitulating the essential hallmarks of solid tumors, including spatial gradients of oxygen, growth factors and metabolites. 3D tumor models are essential preclinical tools in lung cancer research, providing valuable resources for studying cancer biology and developing novel anticancer drugs. The present revi

Restoring Vision to the Blind with Chemical Photoswitches.

Degenerative retinal diseases such as retinitis pigmentosa (RP) and age-related macular degeneration (AMD) affect millions of people around the world and lead to irreversible vision loss if left untreated. A number of therapeutic strategies have been developed over the years to treat these diseases or restore vision to already blind patients. In this Review, we describe the development and translational application of light-sensitive chemical photoswitches to restore visual function to the blind retina and compare the translational potential of photoswitches with other vision-restoring therapi

Strongyloides ratti and S. venezuelensis - rodent models of Strongyloides infection.

Strongyloides spp. are common parasites of vertebrates and two species, S. ratti and S. venezuelensis, parasitize rats; there are no known species that naturally infect mice. Strongyloides ratti and S. venezuelensis overlap in their geographical range and in these regions co-infections appear to be common. These species have been widely used as tractable laboratory systems in rats as well as mice. The core biology of these two species is similar, but there are clear differences in aspects of their within-host biology as well as in their free-living generation. Phylogenetic evidence suggests th

Concepts of holism in orthodox and alternative medicine.

In this essay the nature of holism in orthodox and alternative medicine will be explored and the true meaning of the words will be illustrated with a complicated case history concerning the life or death of a young pregnant mother suffering from a BRCA type breast cancer. Holism in medicine is an open-ended and exquisitely complex understanding of human biology that over time has led to spectacular improvements in the length and quality of life of patients with cancer and that this approach encourages us to consider the transcendental as much as the cell and molecular biology of the human orga

Traction force microscopy in physics and biology.

Adherent cells, crawling slugs, peeling paint, sessile liquid drops, bearings and many other living and non-living systems apply forces to solid substrates. Traction force microscopy (TFM) provides spatially-resolved measurements of interfacial forces through the quantification and analysis of the deformation of an elastic substrate. Although originally developed for adherent cells, TFM has no inherent size or force scale, and can be applied to a much broader range of mechanical systems across physics and biology. In this paper, we showcase the wide range of applicability of TFM, describe the

Unlocking the Full Potential of Spatial Omics in Plants: Practical Challenges, Solutions, and a Path Forward.

Spatial omics technologies are providing new opportunities for plant biology by enabling molecular profiling within structurally intact tissues, revealing spatially organised cell states, developmental gradients, and regulatory interactions. While spatial transcriptomics has driven early advances, the field is rapidly expanding toward integrated spatial multi-omics by combining single-cell and spatial transcriptomic, epigenomic, proteomic, and metabolomic data. These approaches offer new opportunities to study development, physiology, and plant biotic and abiotic interactions in spatially pres

An overview of the developments in 3D cancer cell models, assay techniques, and imaging modalities.

Cancer biology is a constantly evolving field of study due to the dynamic and complex nature of biological systems. The unique role of in vitro assays in cell biology has led to numerous transformations in our understanding of the functional and structural details of the cells and tissues that comprise these systems. However, the traditional monolayer assays have been reported to fall short of the in vivo physiology of cells. This has led to the development of 3D cell models, such as spheroids and organoids, that aim to recapitulate the intricate structural and functional behaviour of in vivo

Spatiotemporal precision interventions for cardiac repair and regenerative therapy.

Restoring cardiac function after myocardial infarction remains a major challenge, as current pharmacological and interventional therapies primarily mitigate symptoms and slow disease progression without addressing the irreversible loss of functional myocardium. Although a diverse range of biologically active agents has been developed to modulate inflammation, angiogenesis, fibrosis, and cardiomyocyte survival, their therapeutic impact is frequently limited by delivery strategies that fail to match the dynamic and heterogeneous nature of post-infarction healing. Advances in biomaterials, nanote

Hair Follicle Stem Cells: Emerging Physiology and Therapeutic Applications.

Hair follicle stem cells (HFSCs) have been studied for their role in developmental biology, hair cycling and transplantation. However, recent research reframes the hair follicle as a heterogeneous regenerative ecosystem capable of responding to injury cues and contributing to tissue repair beyond trichology. Despite advances, our understanding of HFSC physiology and therapeutic applications remains fragmented. Many reviews fail to bridge the gap between mechanistic biology and translatable therapeutic modalities, often overlooking critical aspects such as stemness-preserving culture systems, b

How Can Proteomics Help to Elucidate the Pathophysiological Crosstalk in Muscular Dystrophy and Associated Multi-System Dysfunction?

This perspective article is concerned with the question of how proteomics, which is a core technique of systems biology that is deeply embedded in the multi-omics field of modern bioresearch, can help us better understand the molecular pathogenesis of complex diseases. As an illustrative example of a monogenetic disorder that primarily affects the neuromuscular system but is characterized by a plethora of multi-system pathophysiological alterations, the muscle-wasting disease Duchenne muscular dystrophy was examined. Recent achievements in the field of dystrophinopathy research are described w…

Generation and Characterization of an Inducible Cx43 Overexpression System in Mouse Embryonic Stem Cells.

Connexins (Cx) are a large family of membrane proteins that can form intercellular connections, so-called gap junctions between adjacent cells. Cx43 is widely expressed in mammals and has a variety of different functions, such as the propagation of electrical conduction in the cardiac ventricle. Despite Cx43 knockout models, many questions regarding the biology of Cx43 in health and disease remain unanswered. Herein we report the establishment of a Cre-inducible Cx43 overexpression system in murine embryonic stem (ES) cells. This enables the investigation of the impact of Cx43 overexpression i

Understanding the Molecular Drivers of Disease Heterogeneity in Crohn's Disease Using Multi-omic Data Integration and Network Analysis.

Crohn's disease (CD), a form of inflammatory bowel disease (IBD), is characterized by heterogeneity along multiple clinical axes, which in turn impacts disease progression and treatment modalities. Using advanced data integration approaches and systems biology tools, we studied the contribution of CD susceptibility variants and gene expression in distinct peripheral immune cell subsets (CD14+ monocytes and CD4+ T cells) to relevant clinical traits. Our analyses revealed that most clinical traits capturing CD heterogeneity could be associated with CD14+ and CD4+ gene expression rather than dise…

Culture-independent genomics of a novel chlamydial pathogen of fish provides new insight into host-specific adaptations utilized by these intracellular bacteria.

Several Chlamydiales families are associated with epitheliocystis, a common condition of the fish gill epithelium. These families share common ancestors with the Chlamydiaceae and environmental Chlamydiae. Due to the lack of culture systems, little is known about the biology of these chlamydial fish pathogens. We investigated epitheliocystis in cultured Orange-spotted grouper (Epinephelus coioides) from North Queensland, Australia. Basophilic inclusions were present in the gills of 22/31 fish and the presence of the chlamydial pathogen in the cysts was confirmed by in situ hybridization. Giant

Aligning preclinical AML models with immunotherapy development: principles for model selection.

Advances in immunotherapy for acute myeloid leukemia (AML) have revealed critical gaps in selection of appropriate preclinical models. Conventional cytotoxic and targeted therapies could be tested in relatively straightforward systems. Immunotherapies are inherently distinct from conventional therapies, as their activity is shaped by dynamic, context-dependent interactions between leukemic cells, immune effectors, and the bone marrow microenvironment that are difficult to replicate outside the patient. Failure to recapitulate these interactions may limit the clinical translatability of promisi

Sulfolobus acidocaldarius, half a century of archaeal research.

Since its isolation from a Yellowstone hot spring in 1972, Sulfolobus acidocaldarius has become one of the most important model organisms for archaeal biology. Initially studied for its remarkable adaptation to high temperature and low pH, it has evolved into a genetically tractable system that has contributed substantially to our understanding of archaeal physiology, molecular biology, and evolution. Here, we summarize more than 5 decades of research on S. acidocaldarius, highlighting key developments in genetic and microscopy tools and their impact on our understanding of archaeal cell biolo

The fate and behavior mechanism of antibiotic resistance genes and microbial communities in flocs, aerobic granular and biofilm sludge under chloroxylenol pressure.

Chloroxylenol (PCMX), an antibacterial agent, has been widely detected in water environment and has toxic effects on biology and ecology. During 270 d, the influence of PCMX on the performance of three nitrification systems was investigated, including floc-based sequencing batch reactor (FSBR), aerobic granule-based SBR (AGSBR) and biofilm SBR (BSBR). The nitrification capability of three systems was inhibited by PCMX, but recovered after domestication, and PCMX made three systems realize partial nitrification for 10, 100 and 35 days, respectively. The extracellular polymeric substances of thr

A control theoretic primer for systems endocrinology.

Endocrine systems are characterised by complex dynamic behaviours-oscillations, transient responses, feedback regulation, and noise filtering-that are essential to physiological stability. Despite extensive molecular and clinical research, the quantitative principles governing these dynamics remain incompletely understood. In particular, the mechanistic relationship between the metabolic clearance rates of individual hormones and emergent system-level properties such as oscillatory frequency, damping, and stability has not been systematically integrated into endocrine theory. This paper introd

Remediation of wastewater by using CdS-based biohybrids: Challenges and enhancement strategies.

Co-existence of pollutants, specifically heavy metals and organic compounds, as well as multi-heavy metals, in wastewater presents a significant global environmental and public health concern. The combined presence of these pollutants can result in a synergistic increase in toxicity, making the simultaneous removal of heavy metals and organic contaminants a complex challenge. Cadmium sulfide-based photocatalyst-microbe biohybrids, which integrate the advantages of whole-cell biological catalysts and semiconducting nanomaterials, have garnered considerable interest. These biohybrids offer benef

From fandom to inquiry: what game studies reveals about biology learning at the Pokémon Fossil Museum.

Popular culture is increasingly used in biology education, but its value is often explained primarily through a narrow account of engagement: learners recognize a reference, enjoy the activity, and participate more readily. This Perspective argues that game studies offer biology education research (BER) a vocabulary for examining the deeper mechanisms that can make such engagement educationally meaningful, including fan expertise, fictional world knowledge, affect, and identity. I use the Pokémon Fossil Museum at Chicago's Field Museum as a generative case, informed by a reflective visit rathe
