Pneumocystis carinii is the most important pulmonary pathogen in patients with the acquired immunodeficiency syndrome, but host defenses against P. carinii are not well characterized. We recently reported an experimental model of P. carinii infection, in which mice selectively depleted of CD4+ lymphocytes develop pulmonary infection after inoculation with P. carinii. Lymphocytes from eight patients with adult T-cell leukemia were analyzed by two color flow cytometry. Monoclonal antibodies (Leu 3 a, Leu 8, Leu 2 a and Leu 15) labelled with fluorescein isothiocyanate or phycoerythrin were used. The purpose was to identify the subsets of the lymphocytes as helper, suppressor/inducer, suppressor or cytotoxic by the surface marker of the cells. All eight patients had antibodies for ATLA. Murine T lymphocyte clones sensitized to Listeria monocytogenes were developed to investigate specific mechanisms of T cell-mediated immunity. Splenic B and T lymphocytes metabolically labelled in vitro with [35S]-H2SO4 show differential sulphation of chondroitin sulphate, the major sulphated glycosaminoglycan in both cell-retained and secreted fractions. In B cells, 6-O-sulphation is more abundant than 4-O-sulphation, whereas in T cells sulphation occurs predominantly in the 4-O-position. The murine lymphoma lines AKTB-1b and EL-4, of B and T origin respectively, show even more marked differences. AKTB-1b follows the pattern of normal B cells, whilst in EL-4 only chondroitin 4-sulphate is synthesized.

Seven patients with selective IgM deficiency (SIgMD) were studied for cell surface immunoglobulin (SmIg), T-cell subpopulations, and immunoglobulin (Ig) synthesis in vitro by peripheral blood lymphocytes (PBL). Immunohistological techniques were used to identify activated T lymphocytes within the synovial membrane of patients with rheumatoid arthritis, using the monoclonal antibody (MoAb) RFT2, which identifies a 40-k dalton molecule preferentially expressed by T blasts or activated cells. Normal mouse serum was found to contain a factor blocking in vivo the effect of autoreactive cells accumulating in spleens of mice infected with tick-borne encephalitis (TBE), Langat, dengue type 2 (D2), and attenuated yellow fever (17D strain) viruses. The activity of the factor was manifested only, when the autoreactive lymphocytes had H-2 antigens identical with H-2 antigens of the serum donors. The hypothesis is discussed that serum factor protects the host against clones of autoreactive T lymphocytes generated in viral infections.

Two patients with disseminated mycobacterial infection presented with severe neutropenia and hematopoietic failure. Marrow cells were obtained from each patient and were cultured in methylcellulose before and after the removal of mononuclear phagocytes, T-lymphocytes, or both from the marrow cell suspension. We investigated the number of DNA-synthesizing T lymphocytes in the blood of patients with Hodgkin's disease, with infectious mononucleosis and in normal controls. T cells were characterized by their ability to form rosettes with unsensitized neuramidase-treated sheep red blood cells. Cells in DNA synthesis were evaluated autoradiographically after in vitro incubation with [3H]thymidine. Our results indicated a preferential proliferation of T lymphocytes in the blood of patients with Hodgkin's disease and infectious mononucleosis and suggested an increased turnover of these cells.
