The application of the enzyme thermistor in the analysis of cyanide in standard solutions as well as in blast furnace waste water is described. The heat signal is generated in the conversion of cyanide, catalyzed by the immobilized enzymes rhodanese (E.C. 2.8.1.1) and injectase (E.C. 4.4.19). Using the combination of cyanide-metabolizing enzymes and the enzyme thermistor unit, assays down to 20 microM cyanide can be carried out. Linear relationships were obtained at 20-600 microM cyanide for injectase and 20-1000 microM for rhodanese. The stability at 27 degrees C of the heat response was init

Calciphylaxis is a rare and life-threatening condition characterized by vascular calcification and thrombosis, typically seen in patients with end-stage renal disease but also associated with long-term warfarin use in non-uremic patients. We present the case of a 78-year-old woman with a mechanical aortic valve on chronic warfarin therapy who developed progressive, painful necrotic skin ulcerations over ten months. Skin biopsy confirmed warfarin-induced non-uremic calciphylaxis. Following shared decision-making, warfarin was discontinued and replaced with low molecular weight heparin, and intr

Calciphylaxis is a rare but life-threatening complication of end-stage renal disease (ESRD), most often seen in patients undergoing hemodialysis. This condition is driven by calcium deposition in small blood vessels, leading to restricted blood flow, tissue ischemia, and often severe pain. While calciphylaxis typically affects areas with increased adiposity, such as the abdomen and proximal extremities, it can manifest on any skin surface, including rare sites like the genital region. Managing calciphylaxis is particularly challenging due to its rapid progression and limited treatment options.

Psoriasis is relatively common in clinical practice, whereas niacin deficiency is relatively rare. We describe the clinical case of a patient with plaque psoriasis for over 20 years who also had a concomitant latent tuberculosis infection. After secukinumab and anti-tuberculosis treatment for 1 year, the psoriatic rash mostly resolved, but atypical symptoms of niacin deficiency suddenly appeared. The patient's symptoms rapidly subsided after experimental treatment with niacin. After 2 weeks, the patient suddenly developed an erythroderma-like rash, manifesting as large areas of erythema and pl

Shewanella oneidensis MR-1 is a dissimilatory metal-reducing bacterium capable of reducing various metal and sulfur compounds and precipitating them in nanoparticulate form. Here, we report the synthesis of molybdenum disulfide nanomaterials at the site of S. oneidensis biofilms grown in the presence of molybdenum trioxide and sodium thiosulfate. Samples from the growth medium were imaged using scanning electron microscopy and characterized using transmission electron microscopy, energy-dispersive x-ray spectroscopy, absorbance spectroscopy, and x-ray diffraction. These methods revealed the pr

BACKGROUND: Calciphylaxis or calcific uremic arteriolopathy (CUA) is a cutaneous disease with ulcerations secondary to calcification of cutaneous and subcutaneous small arteries and arterioles. It is a rare but severe disease with significant morbidity and mortality affecting 1 to 4% of dialysis patients. The circumstances of occurrence are multiple.
CASE: We report the case of a severe bilateral lower limb calciphylaxis in a 69-year-old, obese, hemodialysis patient with a recent diagnosis of Graves' disease complicated with hypercalcemia and cardiac arrhythmia requiring the use of vitamin K

Abstract In transsulfuration reaction catalyzed by rat mercaptopyruvate sulfurtransferase (MST), a stable persulfide is formed at the catalytic site cysteine Cys(247) as a reaction intermediate. The outer sulfur atom is donated by the substrate, thiosulfate, or by mercaptopyruvate. MST serves as a thioredoxin-dependent antioxidant possessing self-regulated enzymatic activity. After oxidation of persulfurated MST by treatment with hydrogen peroxide, mass spectrometric analysis showed that the outer sulfur atom of the persulfide is oxidized to form Cys-thiosulfenate (Cys-Sγ-SO(-)), Cys-thiosulfi

Cyanide is both widely available and easily accessible throughout the world. Although the compound is not frequently encountered, it has been used as a poison and contaminant in the past and is a potential terrorist agent. Cyanide has the ability to cause significant social disruption and demands special attention to public health preparedness. It can be obtained from a variety of sources, including industrial, medical, and even common household products. Another frequently encountered source of cyanide exposure is residential fires. Exposure to high concentrations of the chemical can result i

A high-performance liquid-chromatographic method has been developed for the determination of five penicillin compounds (benzylpenicillin, phenoxymethylpenicillin, oxacillin, cloxacillin, and dicloxacillin) at trace levels in commercially available milk samples. This method comprises extraction of the lipids with ethyl acetate, clean-up and concentration on a C-18 solid-phase extraction column, and derivatization with 1,2,4-triazole and mercury(II) chloride solution, pH 8, at 65 degrees C for 10 min. The derivatized compounds are eluted from a C-2 column with a mobile phase containing acetonitr

Sulfite oxidase deficiency is characterized by severe neurologic dysfunction, dislocation of the lenses, and the accumulation and excretion of inorganic sulfite, thiosulfate, and S-sulfocysteine. We present the clinical, radiologic, and biochemical findings in two patients with this condition. In both, neurologic problems started soon after birth and progressed rapidly to profound mental retardation, microcephaly, blindness, and spastic quadriparesis. Seizures were a persistent problem throughout the course of their illness. The neurologic abnormalities were associated with progressive destruc

When grown autotrophically in a thiosulfate-mineral salts medium, cells of the facultative chemoautotrophic bacterium, Thiobacillus novellus, produced two distinct glutamate dehydrogenases, one specific for nicotinamide adenine dinucleotide phosphate (NADP) and the other specific for nicotinamide adenine dinucleotide (NAD). When glutamate was supplied exogenously as the sole carbon source, the NAD-specific glutamate dehydrogenase was fully induced. Lower levels of the enzyme were found in bacteria grown in l-arginine, l-alanine, glucose, glycerol, lactate, citrate, or succinate. Arginine, hist

UNLABELLED: Sequence comparisons showed that the sulfur oxygenase reductase (SOR) of the haloalkaliphilic bacterium Thioalkalivibrio paradoxus Arh 1 (TpSOR) is branching deeply within dendrograms of these proteins (29 to 34% identity). A synthetic gene encoding TpSOR expressed in Escherichia coli resulted in a protein 14.7 ± 0.9 nm in diameter and an apparent molecular mass of 556 kDa. Sulfite and thiosulfate were formed from elemental sulfur in a temperature range of 10 to 98°C (optimum temperature ≈ 80°C) and a pH range of 6 to 11.5 (optimum pH ≈ 9; 308 ± 78 U/mg of protein). MGI 114 (HMAF, 6-hydroxymethylacylfulvene) is a cytotoxic drug currently in phase II human clinical trials. As with other anticancer agents, inadvertent drug extravasation may result in perivascular irritation and/or necrosis. In this study the degree of soft tissue injury produced by MGI 114 after intradermal administration to rats was quantified and four potential antidotes for extravasation injuries caused by MGI 114 were evaluated. Intradermal injections of MGI 114 (0.2 ml, concentrations 0.1, 0.5 or 1.0 mg/ml) and a positive control, doxorubicin (0.2 ml, concentration 2 mg/ml) were admini

INTRODUCTION: Calciphylaxis, also known as calcific uremic arteriolopathy, is a rare, severe disease that causes diffuse skin ischemia and necrotic lesions with a high mortality rate. This disease most commonly affects patients with end-stage renal disease (ESRD). However, there are reported cases of patients developing calciphylaxis without ESRD, known as non-uremic calciphylaxis (NUC). NUC is especially rare, and little is known about its pathophysiology and treatment.
CASE PRESENTATION: We present the case of a 47-year-old male with multiple comorbidities but no history of ESRD who present

Calciphylaxis is a devastating disease characterized by painful ischemic necrotic skin lesions, primarily affecting patients with end-stage kidney disease. Its incidence has significantly increased over the past decade. Despite substantial morbidity and mortality, the pathogenesis of calciphylaxis remains poorly understood, and currently there are currently no approved treatments available. Diagnosis largely relies on clinical suspicion; skin biopsy may be useful but is not essential, particularly when biopsy risks outweigh benefits. Effective management requires a multidisciplinary approach.

Calcific uremic arteriolopathy (CUA) represents a rare but severe disease with high morbimortality. The authors present the case of a 58-year-old male patient with chronic kidney disease due to obstructive uropathy, on hemodialysis (HD). He started HD due to uremic syndrome with a severe renal dysfunction, dysregulation of calcium and phosphate metabolism, and he presented with distal penile ischemia, which was treated with surgical debridement and hyperbaric oxygen therapy. Four months later, painful distal digital necrosis of both hands was observed. Extensive arterial calcification was obse
