Personalised digital interventions for reducing hazardous and harmful alcohol consumption in community-dwelling populations.

BACKGROUND: Excessive alcohol use contributes significantly to physical and psychological illness, injury and death, and a wide array of social harm in all age groups. A proven strategy for reducing excessive alcohol consumption levels is to offer a brief conversation-based intervention in primary care settings, but more recent technological innovations have enabled people to interact directly via computer, mobile device or smartphone with digital interventions designed to address problem alcohol consumption. OBJECTIVES: To assess the effectiveness and cost-effectiveness of digital interventi

Temporal trends in abusive alcohol consumption and their projections for 2030 in Brazilian capitals.

OBJECTIVE: To analyze the temporal trend of the prevalence of abusive alcohol consumption in the Brazilian adult population residing in the country's capitals between 2006 and 2023, and to estimate projections for 2030. METHODS: Time-series study using data from the Surveillance System for Risk and Protective Factors for Chronic Diseases by Telephone Survey. Abusive alcohol consumption was defined as the intake of five or more drinks for men and four or more for women in a short period of time. The joinpoint regression model was used to identify trends, and exponential smoothing was applied t

Alcohol consumption and the risk of end-stage renal disease among Chinese men.

We examined the relationship between alcohol consumption and incidence of end-stage renal disease (ESRD) in a prospective cohort of 65 601 Chinese men aged 40 years and older. Information on the amount and type of alcohol consumed was collected at a baseline examination with follow-up evaluations conducted 8-9 years later. During the 500 876 person-years of follow-up, 176 participants initiated renal replacement therapy or died from renal failure. Compared to non-drinkers, the relative risk of ESRD was 0.67 among men consuming less than 21 drinks per week and 0.52 among men consuming this amou

Coping with stressful events: influence of parental alcoholism and race in a community sample of women.

The study explores the role of race and differences in coping among 290 white women and black women with and without alcoholic parents, addressing two questions: (1) Does coping vary by parental alcoholism or race? and (2) How is coping in adulthood affected by childhood stressors and resources and by adulthood resources? Standardized self-administered questionnaires (Coping Responses Inventory and the Children of Alcoholics Screening Test) measuring approach and avoidant coping methods were used. Collateral information was obtained from siblings who completed questionnaires focused on parenta

Persistent impairment of clomipramine demethylation in recently detoxified alcoholic patients.

The purpose of the present study was to investigate whether the metabolism of clomipramine is altered by chronic alcohol drinking. Eleven recently detoxified alcoholic patients were included (experimental group EG) and compared to a reference group of patients with no history of alcoholism (reference group RG, n = 102). Blood concentrations of clomipramine and its metabolites were measured as part of the routine drug monitoring program. Clearances were estimated from trough concentrations, according to a model developed previously. Results indicate strong inhibition of demethylation clearance …

Investigating pharmacotherapies for alcohol use disorder using Drosophila melanogaster.

BACKGROUND: Drosophila melanogaster provides a powerful whole-organism approach for the therapeutic discovery of treatments for many disorders due to the ability to perform high-throughput drug studies with detailed molecular and cellular mechanisms. While Drosophila has been critical for identifying novel molecular and neural circuit mechanisms underlying alcohol responses, few studies assess the use of Drosophila for the investigation and discovery of pharmacological targets of alcohol use disorder (AUD). Determining appropriate doses that impact alcohol consumption and memory for alcohol re

The Effects of Ethanol on the Heart: Alcoholic Cardiomyopathy.

Alcoholic-dilated Cardiomyopathy (ACM) is the most prevalent form of ethanol-induced heart damage. Ethanol induces ACM in a dose-dependent manner, independently of nutrition, vitamin, or electrolyte disturbances. It has synergistic effects with other heart risk factors. ACM produces a progressive reduction in myocardial contractility and heart chamber dilatation, leading to heart failure episodes and arrhythmias. Pathologically, ethanol induces myocytolysis, apoptosis, and necrosis of myocytes, with repair mechanisms causing hypertrophy and interstitial fibrosis. Myocyte ethanol targets includ

The circulating microbiome signature and inferred functional metagenomics in alcoholic hepatitis.

UNLABELLED: Intestinal dysbiosis is implicated in alcoholic hepatitis (AH). However, changes in the circulating microbiome, its association with the presence and severity of AH, and its functional relevance in AH is unknown. Qualitative and quantitative assessment of changes in the circulating microbiome were performed by sequencing bacterial DNA in subjects with moderate AH (MAH) (n = 18) or severe AH (SAH) (n = 19). These data were compared with heavy drinking controls (HDCs) without obvious liver disease (n = 19) and non-alcohol-consuming controls (NACs, n = 20). The data were related to en

Kudzu root: an ancient Chinese source of modern antidipsotropic agents.

Kudzu (Pueraria lobata) is one of the earliest medicinal plants used in traditional Chinese medicine. It has many profound pharmacological actions including antidipsotropic (antialcohol abuse) activity. Although both the roots and flowers of kudzu, Radix and Flos puerariae, respectively, have been used to treat alcohol abuse safely and effectively in China for more than a millennium, their true efficacy, active constituents, sites and mechanisms of action have never been critically examined. Recently, we have demonstrated that a crude extract of Radix puerariae suppresses the free-choice ethan

Criterion variability in problem-drinking research on college students.

A persistent issue confronting research on problem drinking (PD) and heavy drinking (HD) among college-age adults is the lack of a consistent, valid definition and criteria for either of the two conditions. This article presents a review and analysis of the criteria used in a selected group of studies on college students' PD and HD from 1974 to 1993. All studies within the time period that employed a random sample of students were analyzed for variations in criteria used to define either PD or HD. Sample characteristics, the specific criteria used, and data on prevalence rates found with those

A Semi-Isolation Housing Paradigm Affects Anxiety, Ethanol Intake, and the Gut Microbiota in Adult Male C57BL/6JRj Mice.

BACKGROUND: There is a complex link between environmental factors, anxiety levels, and ethanol consumption. Social conditions such as loneliness, peer influence, and housing environment shape drinking behaviors and mental health outcomes. The gut microbiota, in the context of the gut-brain axis, may act as a biomarker. OBJECTIVES: This study investigates how different housing conditions impact anxiety-like behavior, ethanol consumption, and gut microbiota in adult male mice. METHODS: Adult male C57BL/6JRj mice were housed under isolated, semi-isolated, or group-housed conditions. Anxiety-lik

Punishment of ethanol choice in group-housed male cynomolgus monkeys.

BACKGROUND: A common characteristic of individuals with alcohol use disorder (AUD) is that they continue to consume alcohol despite adverse consequences. A better understanding of the factors that influence this relative insensitivity to punishment may help identify novel treatments for AUD. In animal models, this characteristic can be studied by adding the bitter-tasting alkaloid, quinine, to an ethanol solution. In the present study, punishment of ethanol drinking was assessed in socially housed male cynomolgus monkeys to determine whether sensitivity to punishment is influenced by a monkey'

COVID-19 vaccine acceptance in three rural communes in Haiti: A cross-sectional study.

Vaccines are the most effective mechanism for ending the COVID-19 pandemic. However, reluctance to accept vaccines has hindered the efforts of health authorities to combat the virus. In Haiti, as of July 2021, less than 1% of the country's population has been fully vaccinated in part due to vaccine hesitancy. Our goal was to assess Haitian attitudes toward COVID-19 vaccination and investigate the primary reasons for Moderna vaccine hesitancy. We conducted a cross-sectional survey across three rural Haitian communities, in September 2021. The research team used electronic tablets to collect qua
