Bioengineered microbiotic levodopa therapy improves cognition and reduces pathology in a rat model of Alzheimer's disease.

BACKGROUND AND PURPOSE: Degeneration of the pontine noradrenergic and midbrain dopaminergic systems contributes to cognitive-behavioural disturbances during the prodromal stages of Alzheimer's disease (AD). We developed a genetically engineered, programmable probiotic Escherichia coli Nissle 1917 strain (EcNrha L-DOPA) capable of producing L-3,4-dihydroxyphenylalanine (L-DOPA) in a sustained and titratable manner, thus offering a novel gut-brain delivery mechanism to increase brain levels of noradrenaline and dopamine during the early stages of AD. EXPERIMENTAL APPROACH: We replicated locus c

Thymoquinone controlled obesity and invigorated cognitive and memory performance in rats consuming a high-fat diet via modulating oxidative stress, inflammation and apoptosis.

The current study investigated the mitigating effects of thymoquinone (TQ) against high-fat diet (HFD)-mediated brain injury, cognitive and memory impairment, and the underlying mechanisms. Twenty-four adult male Wistar rats were divided into four groups of six rats each. Rats were fed HFD for 12 weeks to induce obesity. On the 9th week, TQ was administered orally to obese rats for four weeks. The effects of TQ were estimated by neurobehavioral testing, biochemical analysis, DNA damage, molecular docking, and histopathological examination of brains and visceral fat. TQ reduced body weight, bod

Emotional influences on remembering and forgetting explained by frontal and parietal dynamics.

Based on item-method directed forgetting (DF) task, 60 participants were recruited to explore the influence of emotion (negative, neutral, and positive) on memory encoding processing, with all data referring to the encoding phase of the task. Behavioral results showed that participants were more successful at remembering negative pictures that needed to be forgotten, with both higher recognition rates and discrimination accuracy (Pr) compared with neutral pictures. In the brain, parietal activities reflected preferential processing during negative picture viewing through enhanced late parietal

White matter hyperintensities and the risk of vascular dementia: a systematic review and meta-analysis.

BACKGROUND: White matter hyperintensities (WMHs) are hyperintense lesions observed on magnetic resonance imaging (MRI) and are unique imaging indicators of cerebral small vessel diseases. WMH-related white matter alterations have been correlated with cognitive impairment and cerebrovascular pathology. Some studies suggest that vascular hemodynamic changes contribute to WMH development, ultimately leading to vascular dementia (VaD). However, the association between WMH burden and VaD remains inconclusive. This meta-analysis aimed to quantify the relationship between WMH volume and VaD severity

Variability in Exercise is Linked to Improved Age-related Dysfunctions, Suggesting a Potential Role for the Constrained-Disorder Principle-based Second-Generation Artificial Intelligence System.

OBJECTIVE: Regular physical activity (PA) promotes mental and physical health. Nevertheless, inactivity is a worldwide pandemic, and methods to augment exercise benefits are required. The constrained disorder principle (CDP) characterizes biological systems based on their inherent variability. Therefore, we aimed to investigate the association between intra-individual variability in PA and disability among non-athlete adults. METHODS: In this retrospective analysis of the longitudinal SHARE survey, we included non-disabled adults aged >50 with at least six visits over 14 years. Self-reported

A scoping review of financial decision-making measures in midlife and beyond: results from the advancing reliable measurement in cognitive aging and decision-making ability (ARMCADA) study.

BACKGROUND: Cognitive decline in older adults affects key functions such as memory, concentration, planning, reasoning, and decision-making (DM). This decline in cognitive abilities compromises basic DM skills, with growing evidence that DM can decline before noticeable impairment or an official cognitive impairment diagnosis, adversely impacting quality of life and leading to negative outcomes in financial management and daily activities. OBJECTIVE: This scoping review aims to identify and evaluate existing measures of financial decision-making (FDM) abilities in clinical and community-dwell

Kapβ2 Reverses Sevoflurane-Induced Hydrogel Phase Transition of hnRNPA2/B1-SG in Hypoxic Primary Rat Hippocampal Neurons.

AIMS: Sevoflurane can aggravate the progression of neurodegeneration, although the underlying mechanisms remain incompletely understood. Our previous study identified a link between heterogeneous nuclear ribonucleoprotein A2/B1 (hnRNPA2/B1) and sevoflurane-induced neurocognitive impairments. The abnormal hydrogel phase transition of stress granules (SGs) assembled via liquid-liquid phase separation (LLPS) by hnRNPA2/B1 is a crucial element in neurodegeneration. Karyopherin-β2 (Kapβ2) is known to specifically recognize hnRNPA2/B1 and reverses the hydrogel transition of SGs. This study aimed to

Post-Concussion Syndrome and Functional Neurological Disorder: Diagnostic Interfaces, Risk Mechanisms, and the Functional Overlay Model.

BACKGROUND: Post-concussion syndrome (PCS) and Functional Neurological Disorder (FND), including Functional Cognitive Disorder (FCD), are two frequently encountered but diagnostically complex conditions. While PCS is conceptualized as a sequela of mild traumatic brain injury (mTBI), FND/FCD encompasses symptoms incompatible with recognized neurological disease, often arising in the absence of structural brain damage. Yet, both conditions exhibit considerable clinical overlap-particularly in the domains of cognitive dysfunction, emotional dysregulation, and symptom persistence despite negative

Inhibition of Soluble Epoxide Hydrolase Ameliorates Cerebral Blood Flow Autoregulation and Cognition in Alzheimer's Disease and Diabetes-Related Dementia Rat Models.

Alzheimer's Disease and Alzheimer's Disease-related dementias (AD/ADRD) pose major global healthcare challenges, with diabetes mellitus (DM) being a key risk factor. Both AD and DM-related ADRD are characterized by reduced cerebral blood flow, although the exact mechanisms remain unclear. We previously identified compromised cerebral hemodynamics as early signs in TgF344-AD and type 2 DM-ADRD (T2DN) rat models. Genome-wide studies have linked AD/ADRD to SNPs in soluble epoxide hydrolase (sEH). This study explored the effects of sEH inhibition with TPPU on cerebral vascular function and cogniti

Functional neural networks stratify Parkinson's disease patients across the spectrum of cognitive impairment.

INTRODUCTION: Cognitive impairment (CI) is a significant non-motor symptoms in Parkinson's disease (PD) that often precedes the emergence of motor symptoms by several years. Patients with PD hypothetically progress from stages without CI (PD-normal cognition [NC]) to stages with Mild CI (PD-MCI) and PD dementia (PDD). CI symptoms in PD are linked to different brain regions and neural pathways, in addition to being the result of dysfunctional subcortical regions. However, it is still unknown how functional dysregulation correlates to progression during the CI. Neuroimaging techniques hold promi
