Multicomponent Oxicam-Metformin Salts: Toward a Strategy for Enhancing Solubility and Stability.

Drug-drug pharmaceutical multicomponent materials (PMMs) offer a promising strategy to modulate the physicochemical properties of active pharmaceutical ingredients, while enabling synergistic effects and combination therapy. Here, we report the preparation and full characterization of a new family of oxicam-metformin (MTF) salts, involving the nonsteroidal anti-inflammatory drugs piroxicam (PRX), meloxicam (MLX), and tenoxicam (TNX). Structural and computational studies revealed the role of supramolecular synthons in directing the salt formation and highlighted the relationship between molecul

A Bio-Based Chiroptical Overcrowded Alkene From Flavanone: Photoreactivity and Theoretical Investigation.

The design of light-driven overcrowded alkenes often relies on complex synthetic strategies to achieve precise control over rotational motion. In this study, we demonstrate a sustainable approach to design an overcrowded alkene and an O-heterocyclic phenanthrene derivative using a bio-based enantiopure flavanone, (-)-isolonchocarpin, as starting material. The synthesis involves a McMurry deoxygenative homocoupling reaction to generate a sterically hindered overcrowded alkene, which can further undergo photochemical (E/Z)-isomerization and oxidative 6π-electrocyclization. The oxidation product

Mesoporous peptide frameworks engineered from crystallizable collagen-mimetic peptide amphiphiles.

The rational design of porous frameworks with tunable pore dimensions and chemical functionalities is a critical step toward their implementation in diverse applications. While traditional porous materials are typically constructed from abiotic components, there is increasing interest in employing biologically derived building blocks (e.g., peptides and proteins) that offer unmatched structural and functional diversity. Here, we report the construction of crystalline mesoporous frameworks that are self-assembled from amphiphilic collagen-mimetic peptides. Comprehensive structural characterizat

Graph-Theoretical Approach for Predicting Physicochemical Properties of Stiff-Person Syndrome Drugs.

This work applies quantitative structure-property relationship analysis to examine molecular characteristics linked to stiffness person syndrome (SPS), a rare neurological condition marked by persistent muscle rigidity and spasmodic episodes. Topological indices such as Zagreb indices and their modification are employed to examine their correlations with key physicochemical properties. Linear, quadratic, exponential growth, and power curve fitting models are applied to identify the best correlations, focusing on the maximum F-statistic and the coefficient of determination R2. Furthermore, heat

Too rigid to strike: Excess desmin at Z-discs underlies a restrictive cardiomyopathy in GAN mice.

Restrictive cardiomyopathy (RCM) is characterized by myocardial stiffness and impaired diastolic filling. While human giant axonal neuropathy (GAN) is linked to RCM with desmin accumulation, the underlying mechanisms remain unclear. We combined 3D tissue staining, electron microscopy, and echocardiography to investigate RCM phenotypes in GAN mice. Phenotypic profiling revealed atrial enlargement and impaired diastolic relaxation. Ultrastructural analysis demonstrated shortened I-bands and widened, hyper-dense Z-discs, correlating with elevated desmin density at Z-discs. To elucidate the biophy

Exploring the Competition between Halogen Bonding and CH Hydrogen Bonding in Bromoarenes Using an Aryldiyne Template.

Hydrogens connected to benzene rings functionalized with electron-withdrawing groups are found to provide attractions with electron donors that are competitive with the halogen bond attraction to bromines on the same rings. An aryldiyne bridge that adequately templates this CH hydrogen bonding, along with the competing halogen bonding, provides an experimental pathway for looking at these interactions in a systematic way. Calculations using the M06-2x/6-311+G-(2d,p) density functional and basis set, along with mapping of the molecular electrostatic potentials using B3LYP/6-311++G**, support th

Bridged scaffold editing of carbocycles and heterocycles.

Bridged frameworks are widely recognized as privileged motifs in natural products and pharmaceuticals, and their distinctive three-dimensional architectures often underpin target recognition and bioactivity. However, their preparation remains a formidable challenge, with most strategies relying on either linear multi-step syntheses or structurally specialized substrates. Here we report a bridged scaffold editing strategy for heterocycles and carbocycles, which employs formaldehyde and ureas in a distinct multicomponent reaction, toward modular and efficient construction of diverse bridged poly

Chemical Oxidation of Coronene: A Crystallographic and Computational Study of Charge-Regulated π‑Stacking.

Chemical oxidation of coronene with [Et3O+SbCl6 -] in dichloromethane allowed the isolation and crystallization of a dimeric coronene radical cation in the form of [(C24H12)2 •+(SbCl6 -)]-[Et3O+SbCl6 -]2, as revealed crystallographically. The crystal structure exhibits π-stacked columns of coronene molecules, wherein alternating average interplanar contacts of 3.357(2) and 3.452(2) Å show the distinct formation of the coronene dimers. This packing is accompanied by an atom-over-atom overlap motif and core deformation often associated with a unique intermolecular interaction called pancake bond

QSPR modeling and multi-criteria ranking of antiviral drugs using degree-based topological indices, artificial neural networks, and TOPSIS.

In the present research work, graph theory has been used to represent the molecular structures of antiviral drugs corresponding to the influenza strain treatment using degree-based topological descriptors and Laplacian energy to understand their structural and physicochemical behavior. The graph theory-based descriptors are used to construct the quantitative structure property relationship and provide input to an ANN model used to predict various physicochemical properties among the selected antiviral drugs. The comparison shows that the ANN model outperforms the Linear Regression model by dem

A C-repeat binding factor-salicylic acid (CBF-SA) module links wound-induced evaporative cooling to tissue repair in plants.

Repairing damaged tissues is essential for the survival of all organisms. In plants, tissue injury rapidly triggers defense and repair programs. However, the molecular mechanisms linking early injury cues to the later stage of wound repair remain unclear. Here, we show that wounding of Arabidopsis leaves induces localized low temperature at the injury site, likely caused by evaporative cooling, which is accompanied by an activation of cold-responsive genes. Using thermal imaging combined with computer vision and deep learning, we developed a workflow to monitor the dynamics of wound healing in

On-site detection of an emerging fungicide: developing an immunochromatographic assay for thifluzamide through de novo hapten design and synthesis.

Thifluzamide (THF), a novel succinate dehydrogenase inhibitor fungicide, is widely used to control crop diseases; however, excessive residues pose potential risks to human health. To guide hapten design, a detailed analysis of the spatial structures and electrostatic potential distributions of THF and its haptens was conducted using computer-assisted molecular modeling. The challenges in synthesizing haptens that simultaneously contain both 1,3-dibromo-5-(trifluoromethoxy)benzene and 2-methyl-4-(trifluoromethyl)thiazole moieties were also addressed, ultimately leading to the development of a h

Bioimage analysis in deep visual proteomics: Advancing transparency, reproducibility, and FAIR principles.

Deep Visual Proteomics (DVP) combines high-resolution microscopy, computational instance segmentation, laser capture microdissection (LMD), and ultrasensitive mass spectrometry to enable spatially resolved molecular analysis of defined cellular populations. Within this workflow, LMD plays a central role by physically isolating microscopy-defined regions of interest for downstream proteomic profiling. However, accurate image-to-stage transfer remains challenging due to limitations in imaging resolution, tissue preparation variability, laser cutting parameters, and partially manual alignment pro

The Redesign of the Molecular Scaffold of Viral Ion Channel Blockers.

The rise of rapidly mutating viruses poses growing challenges for drug developers in the fight against antiviral resistance. Viral ion channels generally have low mutation rates and are therefore considered emerging targets for a sustainable solution to the problem of resistance. This study reports the redesign of scaffolds for drug candidates targeting viral ion channels. The redesign was powered by a combination of a computational docking protocol that accounts for water molecules (HydroDock) and a subsequent quantum mechanics-based scoring (QMH-L) of the drug candidates. Extending the antiv

Mapping the Altered Landscape of TCR Repertoire in Autoimmune Diseases.

Autoimmune diseases arise from the breakdown of immune tolerance through complex interactions between genetic predisposition, environmental exposures, and adaptive immune responses. High-throughput T-cell receptor (TCR) repertoire sequencing has transformed our ability to characterize these responses, providing unprecedented insights into clonal dynamics, antigen-driven selection, and immune history. In this review, we summarize the major alterations of TCR repertoires reported across autoimmune diseases, including changes in diversity, clonal expansion, repertoire architecture, and tissue dis

Pleural Effusion Revealing Visceral Leishmaniasis: A Diagnostic Pitfall in an Immunocompetent Patient.

Pleural involvement in visceral leishmaniasis (VL) is rare and may pose a diagnostic challenge, particularly when it mimics pleural tuberculosis. We report the case of a 39-year-old immunocompetent man who presented with a dry cough and night sweats. Initial workup revealed a small lymphocytic exudative pleural effusion, and pleural biopsy showed granulomatous inflammation with a tuberculoid appearance, leading to empirical antituberculosis treatment. Because of clinical deterioration, persistent pleural effusion, and negative molecular testing for Mycobacterium tuberculosis, further investiga
