Vesicle Nucleation Peptide Fusion Induced Extracellular Vesicles Are Distinct From Escherichia coli Outer Membrane Vesicles, and Provide an Enhanced Platform for Protein Production and Purification.

Bacterial outer membrane vesicles (OMVs) are nano-sized, spherical structures released by Gram-negative bacteria that play diverse roles in bacterial physiology, including communication, nutrient acquisition and host interactions. These vesicles bud from the bacterial outer membrane and contain lipopolysaccharides, periplasmic proteins, nucleotides and other biomolecules. The vesicle nucleating peptide (VNp) is a short peptide tag that, when fused to the amino terminus of a protein of interest, promotes the formation of bespoke recombinant extracellular vesicles (EVs) in Escherichia coli, enab

Antibody diversification in cartilaginous fishes: Mechanistic insights from the nurse shark and comparative perspectives across jawed vertebrates.

Antibody diversity in vertebrates arises through the coordinated actions of V(D)J recombination and somatic hypermutation (SHM). Cartilaginous fishes occupy a key phylogenetic position as the sister lineage to bony vertebrates and therefore provide important comparative insights into the evolution of adaptive immunity. This review focuses on the nurse shark (Ginglymostoma cirratum) as a representative model for examining antibody-diversification mechanisms in cartilaginous fishes. Shark immunoglobulin genes exhibit a multicluster organization, while immunoglobulin new antigen receptor (IgNAR),

Mechanism underlying the effects of ultrasound-assisted alkaline treatment on the structure and vitamin D₃ encapsulation-release performance of heat-induced egg yolk granule/whey protein composite gel.

This study explored the effects of ultrasound-assisted alkaline treatment between the structure and vitamin D₃ (VD₃) encapsulation efficiency, together with in vitro release behavior of heat-induced egg yolk granule/whey protein composite gels. Results showed that the alkaline environment remarkably modulated the intermolecular conformation and aggregation behavior of composite proteins. With increasing pH, protein unfolding extent augmented, while surface hydrophobicity first decreased and then increased, peaking at pH 11. Enhanced electrostatic repulsion improved system dispersibility, accom

Hyperbranched polycation-based hierarchical assembles as vaccine carrier for cascaded antigen delivery and enhanced immunity response.

Tumor vaccines have emerged as a promising strategy in cancer immunotherapy, and their therapeutic efficacy hinges on successive activation of multi-step cascade immune responses, including the uptake of antigens by dendritic cells, the presentation of antigens by mature DCs to T cells and the activation of T cells, and the recognition and killing of tumor cells by effector T cells. However, distinct or even conflicting characteristics (e.g., charge, size, stability) of vaccine delivery system are required at different stages to overcome multi-stage biological barriers remain a pivotal challen

Effect of fermentation-driven acid-mediated protein degradation on chicken and its gel properties.

This study systematically examines the impact of protein breakdown on the gel properties and texture of chicken under fermentation-induced acidic conditions. Chicken mince samples with different degrees of protein degradation were prepared by controlling fermentation time, followed by comprehensive analyses of physicochemical characteristics, protein structure, microstructure, and gel performance. Results showed that as degradation progressed, the pH value diminished from 5.86 in the control group to 4.19 at 50 days. Under acidic conditions, the surface hydrophobicity of myofibrillar protein w

Mild Heat Stimulating and Microenvironment Reprogramming Hydrogel for Accelerating Diabetic Wound Healing.

Diabetic wounds are characterized by persistent hyperglycemia, excessive ROS accumulation, sustained inflammation, and impaired angiogenesis, yet current treatments remain suboptimal. To address these challenges, we developed a mild heat stimulating and microenvironment reprogramming hydrogel (termed C-4-N) via a green synthetic strategy. L-Arginine (L-Arg) triggered the spontaneous self-polymerization of protocatechuic aldehyde (PA) into poly (protocatechuic aldehyde) (PPA) nanoparticles, onto which ginsenoside Compound K (CK) was subsequently loaded, yielding CK/L-Arg/PPA nanoparticles. Hydrogen Sulfide-Releasing Compounds Attenuate Lipopolysaccharide-Induced Inflammation in Porcine Anterior Uveal Explants, Ex Vivo.

In the present study, we investigated the pharmacological actions of both fast- and slow-releasing H2S compounds, as well as polysulfides, on acute inflammation induced by the bacterial lipopolysaccharide (LPS) in an ex vivo porcine model of anterior uveitis. Isolated porcine iris-ciliary bodies (ICB) were maintained in oxygenated Krebs-Henseleit solution and then incubated in separate wells of RPMI 1640 supplemented media with an antibiotic. ICBs were exposed to sodium hydrosulfide (NaHS), GYY4137, S-allyl-cysteine (SAC), and polysulfides, diallyl disulfide (DADS), and diallyl tetrasulfide (D

Enhancing synergistic removal of gaseous mercury by low-valence sulfur engineered V2O5-WS2/TiO2 SCR catalysts.

Elemental mercury (Hg0) is the dominant species in coal-fired flue gas, posing significant environmental hazards due to its volatility and persistence. Conventional V2O5-based selective catalytic reduction (SCR) catalysts exhibit limited Hg0 oxidation capacity (<50%) under low-temperature and SO2-rich conditions. In this study, a novel low-valence sulfur-engineered V2O5-WS2(tungsten disulfide; W = tungsten)/TiO2 catalyst is developed to enhance synergistic Hg0 removal through complementary oxidation and fixation pathways. Compared to V2O5/TiO2 and V2O5-WO3/TiO2, the V2O5-WS2/TiO2 catalyst achi

Injectable Thermal-Protective Hydrogel Enables Curative Tumor Ablation via Chemo-Immunomodulation.

Image-guided thermal ablation has been included in the National Comprehensive Cancer Network (NCCN) guidelines of multiple solid tumors. However, insufficient ablation of larger lesions and thermal injury adjacent to major organs and tissues limit its clinical application. Besides, sublethal hyperthermia at the margin of ablation can induce an immunosuppressive tumor microenvironment and increases recurrence risk. These physical and biological limitations are linked to each other. We developed an injectable hydrogel MR@CaP@HA with in situ thermal insulation and dual-responsive (pH/GSH) chemo-i

Spider peptides from Brachypelma smithi with slightly different amino acids at their C-terminal loops exert different insecticidal activities.

The insecticidal molecules of spiders persistently evolve to ensure rapid paralysis of their prey, and the best molecules are transmitted to their progeny. Here, we cloned two insecticidal peptides, Bs2 and Bs3, from the venom glands of the theraphosid Brachypelma smithi. Bs2 and Bs3 are 90.2% identical, but they exhibit interesting structural differences at their C-termini, including a connecting disulfide bond (residues Cys15-Cys36 for Bs2 and Cys15-Cys30 for Bs3). The genomic origin of Bs2 and Bs3 may be a cause for gene duplication events. Moreover, Bs2 differs in two residues from Tal1 (9

Venom gland transcriptomics and bioactivity profiling suggest bifunctional hyaluronidase activity in the venom of Mesobuthus crucittii (Scorpiones: Buthidae).

INTRODUCTION: Scorpion venom is a rich source of bioactive molecules with promising biomedical applications. Hyaluronidases are venom-associated enzymes that facilitate toxin diffusion by degrading extracellular matrix glycosaminoglycans, yet their structural diversity and substrate specificity in scorpion venoms remain insufficiently explored. This study aimed to identify and characterize a novel hyaluronidase from the venom gland of the Iranian endemic scorpion Mesobuthus crucittii and evaluate its biochemical and structural properties. METHODS: Venom gland transcriptome profiling was perfo

The new vrdg183 toxin from the Colombian spider Pamphobeteus verdolaga inhibits L-type Ca2+ currents through an allosteric mechanism.

BACKGROUND: Spider venoms are enriched in toxins that modulate mammalian voltage-gated ion channels (VGIC) with pharmacological potential. However, the inventory of toxins of American spiders with effects on voltage-gated Ca2+ channels has lagged the number of toxins interacting with other channels. Here, we aimed at identifying new Ca2+ channel-modulating toxins of the Colombian spider Pamphobeteus verdolaga. METHODS: The sequences of five novel short, positively charged, disulfide-bridged peptides, and one more short, non-bridged control peptide, were selected from the transcriptomic databa

Methyl gallate and hyaluronic acid-nanoconjugate targeting HSP90β for osteoarthritis therapy.

BACKGROUND: Osteoarthritis (OA) is a joint disease with complicated pathophysiologic process, however, the lack of effective therapeutic targets and corresponding pharmaceuticals severely limits treatment options. RESULTS: Methyl gallate (MG) could delay the pathological changes of OA by reducing cartilage inflammation and degeneration at both cellular and animal levels. Using drug affinity responsive target stability assay and mass spectrometry analysis, we identified HSP90β as a key target of MG in chondrocytes. The interaction between MG and heat shock protein 90β(HSP90β)was further valida

Scorpion Venom Peptides: From Structural Scaffolds to Therapeutic Applications-A Focus on Antioxidant Mechanisms and Translational Perspectives.

Scorpion venom peptides, with their stable disulfide backbone, compact structural framework, and highly selective regulation of ion channels, have long been regarded as important molecular probes in neuropharmacology. However, recent studies have revealed their potential for regulating oxidative stress, inflammation, and neuroprotection, making them a new research frontier. In this article, we focus on scorpion venom peptides as drugs, constructing an integrated knowledge framework from structural classification to clinical translation. First, scorpion venom peptides are systematically classif

Hierarchical ROS-scavenging hydrogel system for coordinating the modulation of RGC necroptosis and oxidative stress in an acute glaucoma model.

Glaucoma, a leading cause of irreversible blindness, is characterized by the progressive loss of retinal ganglion cells (RGCs). Beyond intraocular pressure (IOP)-dependent injury, IOP-independent pathways, specifically the self-amplifying axis between oxidative stress and the necroptotic cascade, are key drivers of disease progression. Mediated by the canonical RIPK1/RIPK3/MLKL signaling, necroptosis serves as a pivotal executioner of RGC loss. Oxidative stress functions as the critical upstream trigger that amplifies this cascade, forging a vicious cycle that demands a dual-targeted intervent

Hyaluronic acid-based reduction responsive nanoparticles for Improved anti-psoriasis effects of traditional Chinese herb monomer oleanolic acid via blocking YAP-AREG axis.

BACKGROUND: Oleanolic acid (OA), a pentacyclic triterpenoid abundantly present in various traditional Chinese herbs, exhibits promising anti-psoriatic potential owing to its broad pharmacological activities. Nevertheless, its clinical translation has been hindered by challenges including poor aqueous solubility, limited cutaneous permeation, and rapid systemic clearance, all of which compromise bioavailability when administered topically. To address these limitations, we designed and developed encapsulated and disulfide bonded OA-hyaluronic acid nanoprodrugs (OA-NPs@OA) for topical treatment o

Disrupted glutathione homeostasis in the pathogenesis of TTR-V30M amyloidosis.

BACKGROUND: Transthyretin (TTR) amyloidosis is a progressive, life-threatening disorder caused by extracellular deposition of amyloid fibrils derived from the plasma protein TTR. Inherited forms are associated with destabilizing TTR mutations; however, recent findings indicate that amyloid formation in vivo may be promoted by disulfide bond formation between TTR subunits, suggesting oxidative stress as a potential contributor to protein misfolding and disease progression. Glutathione (GSH) is a central component of the antioxidant defense system, and disruption of GSH homeostasis can lead to t

Multicolor detection of biothiols: machine learning-empowered plasmonic patterns recognition based on anti-etching of gold nanorods.

Biothiols play pivotal roles in maintaining cellular redox homeostasis, where even minor concentration fluctuations can critically affect physiological health. Their structural similarity and dynamic interconversion, however, make the accurate detection of individual species and their metabolic ratios particularly challenging. To address these challenges, we propose a robust multicolorimetric probe based on gold nanorod anti-etching, capable of rapid, sensitive, and reproducible detection and discrimination of multiple ratios of biothiols using a single signal reporter. The modulation of the a
