Air Pollution Exposure Induces Vascular Endothelial Cell Dysfunction via Elevated Circulating F2: Multiomics Insights from an Ex Vivo Biosensor Assay.

Air pollution-induced cardiovascular disease is a leading contributor to the increasing global disease burden. Disruption of vascular endoth … tabolites predominantly enriched in energy metabolism-related pathways. Transcriptomic profiling of endothelial cells provided further corroborating evidence that acute exposure to ambient air pollutants is capable of triggering endothelial cell activation. Subsequent weighted gene coexpression network analysis identified F2 as a pivotal molecular mediator underlyi …

Evidence of endothelial anergy and T-cell exclusion in microsatellite stable colorectal liver metastases revealed through human hemi-liver perfusion and spatial transcriptomics.

BACKGROUND: Colorectal liver metastasis (CRLM) is the terminal stage of colorectal cancer and remains largely resistant to immunotherapeutics, yet the underlying immune escape mechanisms remain poorly understood. Through novel observational methods focused on human living and fixed tissue, we profiled the spatial and molecular features of microsatellite stable (MSS) CRLM and the surrounding liver microenvironment. METHODS: First, we re-purposed normothermic hepatic machine perfusion as a preclinical model of MSS CRLM, enabling delivery of labelled T-cells through physiological vascular routes

T1R3 Anchors ACE2 at the Pulmonary Endothelial Cell Surface to Attenuate Vascular Injury Following SARS-CoV-2 Spike 1 Protein Exposure In Vitro.

The development of acute lung injury in COVID-19 patients is a major cause of lethality. Activation of the pulmonary vasculature, with inflammation, oxidative stress and barrier disruption, is initiated by the SARS-CoV-2 spike protein (Sp1) at the endothelial cell surface. We previously identified the presence of a novel G-Protein Coupled Receptor (GPCR) in the lung microvasculature, T1R3. The T1R3 agonist (T1R3-A) protects the endothelium from endotoxin-induced lung injury therefore we sought to investigate the effect of T1R3-A on Sp1-induced damage to the pulmonary endothelium. Sp1 induced s

Sympathetic overactivity-induced type H endothelial cell senescence contributes to the impairment of vascularized osteogenesis by PKM2-mediated glycolysis in preclinical stages of Alzheimer's mice.

Alzheimer's disease (AD) is characterized by early-onset bone loss, yet the underlying mechanisms remain elusive. Here, we demonstrated that sympathetic hyperactivity drives vascularized osteogenesis impairment in preclinical AD through a novel PKM2-glycolysis-mediated bone vascular endothelial cell (EC) senescence pathway. Utilizing systematic transcriptome profiling complemented by in vitro functional assays and in vivo validation studies, we found that norepinephrine (NE)-induced PKM2 downregulation disrupts glycolytic flux, triggering mitochondrial dysfunction-induced senescence (MiDAS) in

Effect of temperature variation on the corneal endothelial cell during femtosecond laser-assisted cataract surgery compared with conventional phacoemulsification cataract surgery: A prospective study.

To evaluate whether the corneal endothelium experiences increased temperatures and thermal damage during femtosecond laser-assisted cataract surgery (FLACS), we performed comparative in vivo studies. These studies compared FLACS with conventional phacoemulsification cataract surgery (CPS) systems. In this study, 410 eyes with grade II or III nuclear sclerotic cataracts undergoing either CPS (n = 213) or FLACS (n = 197) were examined. All eyes received balanced salt solution at 21°C or 29°C during phacoemulsification. In FLACS, a SoftFit patient interface (PI) was used at 21°C or 29°C. The surf

Understanding the Effect of Surface-Grafted Polyethylene Glycol Chains on Scavenger Endothelial Cell Sequestration of Polymeric Nanoparticles via Quantitative Pharmacokinetic Analysis in Zebrafish Larvae.

Scavenger endothelial cells (SECs) lining the liver sinusoids play a critical role in the rapid blood clearance of nanoparticle (NP)-based drug-delivery systems. However, how these cells recognize synthetic materials is largely unknown, which hampers the establishment of NP design criteria for prolonging their blood circulation time upon systemic administration. This study investigates how the surface-grafted chain conformation on the NPs affects their uptake by SECs. Polystyrene NPs are grafted with polyethylene glycol (PEG) polymers having different molecular weights (MWs = 2, 5, 10, and 20 …

The Protective Role of Intermedin in Contrast-Induced Acute Kidney Injury: Enhancing Peritubular Capillary Endothelial Cell Adhesion and Integrity Through the cAMP/Rac1 Pathway.

Contrast-induced acute kidney injury (CIAKI) is a common complication with limited treatments. Intermedin (IMD), a peptide belonging to the calcitonin gene-related peptide family, promotes vasodilation and endothelial stability, but its role in mitigating CIAKI remains unexplored. This study investigates the protective effects of IMD in CIAKI, focusing on its mechanisms, particularly the cAMP/Rac1 signaling pathway. Human umbilical vein endothelial cells (HUVECs) were treated with iohexol to simulate kidney injury in vitro. The protective effects of IMD were assessed using CCK8 assay, flow cyt…

Inhibition of AIF-1 alleviates laser-induced macular neovascularization by inhibiting endothelial cell proliferation via restrained p44/42 MAPK signaling pathway.

Age-related macular degeneration (AMD) is a leading blinding disease worldwide, and macular neovascularization (MNV) is a common complication encountered in the advanced stages of AMD. While the underlying causes of MNV remain elusive, aberrant multiplication of choroidal endothelial cells (CECs) and increased vascular endothelial growth factor (VEGF) are thought to play significant roles in the occurrence and development of MNV. Allograft inflammatory factor-1(AIF-1) is a crucial regulatory factor of vascular tubular structure formation and growth, involving the proliferation and migration of…

Spontaneously Right-Side-Out-Orientated Coupling-Driven ROS-Sensitive Nanoparticles on Cell Membrane Inner Leaflet for Efficient Renovation in Vascular Endothelial Injury.

Biomimetic cell membrane camouflaged technology has drawn extensive attention as a feasible and efficient way to realize the biological functions of nanoparticles from the parent cells. As the burgeoning nanotherapeutic, the right-side-out orientation self-assembly and pathological dependent "on-demand" cargo release of cell membrane camouflaged nanocarriers remarkably limit further development for practical applications. In the present study, a spontaneously right-side-out-orientated coupling-driven ROS-sensitive nanotherapeutic has been constructed for target endothelial cells (ECs) repair t…

Impaired Autophagy Induced by oxLDL/β2GPI/anti-β2GPI Complex through PI3K/AKT/mTOR and eNOS Signaling Pathways Contributes to Endothelial Cell Dysfunction.

Endothelial cell dysfunction plays a fundamental role in the pathogenesis of atherosclerosis (AS), and endothelial autophagy has protective effects on the development of AS. Our previous study had shown that oxidized low-density lipoprotein/β2-glycoprotein I/anti-β2-glycoprotein I antibody (oxLDL/β2GPI/anti-β2GPI) complex could promote the expressions of inflammatory cytokines and enhance the adhesion of leukocytes to endothelial cells. In the present study, we aimed to assess the effects of oxLDL/β2GPI/anti-β2GPI complex on endothelial autophagy and explore the associated potential mechanisms…

Protective effects of phenolic acid extract from ginseng on vascular endothelial cell injury induced by palmitate via activation of PI3K/Akt/eNOS pathway.

Elevated free fatty acids may impair insulin-mediated signaling to eNOS that contributes to the pathophysiology of endothelial dysfunction. Previous studies have indicated the protective effect of ginseng and the regulatory potential of phenolic acid components from other plants on endothelial function. Therefore, this study investigated the protective effects of phenolic acid extract from ginseng (PG2) on endothelial cells against palmitate-induced damage. We found that PG2 increases cell viability, inhibits the palmitate-induced intracellular accumulation of lipids, and the overexpression of…
