The effects of anti-epileptic drugs on human erythrocyte carbonic anhydrase I and II isozymes.

Carbonic anhydrase (CA) is an enzyme which plays a role in various homeostatic mechanisms, such as acid-base balance and electrolyte secretion in a various tissues. This study was aimed at determining and comparing possible alterations in activity of this enzyme caused by the use of old (Carbamazepine, Phenytoin Sodium, Sodium Valproate) and new (Levetiracetam, Pregabalin, Gabapentin, Oxcarbazepine) anti-epileptic drugs. Blood samples were collected from the volunteers. The blood samples were centrifuged to separate plasma and erythrocyte package. Hemolysate was prepared from the red cells. CA

Inhibitors of aminoglycoside resistance activated in cells.

The most common mechanism of resistance to aminoglycoside antibiotics entails bacterial expression of drug-metabolizing enzymes, such as the clinically widespread aminoglycoside N-6'-acetyltransferase (AAC(6')). Aminoglycoside-CoA bisubstrates are highly potent AAC(6') inhibitors; however, their inability to penetrate cells precludes in vivo studies. Some truncated bisubstrates are known to cross cell membranes, yet their activities against AAC(6') are in the micromolar range at best. We report here the synthesis and biological activity of aminoglycoside-pantetheine derivatives that, although

The methylerythritol phosphate (MEP) pathway for isoprenoid biosynthesis as a target for the development of new drugs against tuberculosis.

Tuberculosis remains a major infectious disease to humans. It accounts for approximately 8-9 million new cases worldwide and an estimated 1.6 million deaths annually. Effective treatments for tuberculosis consist of a combination of several drugs administered over long periods of time. Since Mycobacterium tuberculosis often acquires multiple drug resistant mechanisms, development of new drugs with innovative actions is urgently required. The 2C-methyl-D-erythritol 4-phosphate (MEP) pathway, in charge of the essential biosynthesis of isoprenoids, represents a promising and selective target for

Partial purification, characterisation and histochemical localisation of alkaline phosphatase from ascocarps of the edible desert truffle Terfezia claveryi Chatin.

In the present paper, we confirmed that alkaline phosphatase (ALP) is the main phosphatase present in ascocarps of the edible mycorrhizal fungus Terfezia claveryi. The enzyme was partially purified by precipitation with polyethylene glycol. The purification achieved from a crude extract was fivefold, with 53% of the activity recovered, and acid phosphatase, most of the lipids and phenolic compounds were eliminated. Alkaline phosphatase was kinetically characterised at pH 10.0, the optimum for this enzyme, using p-nitrophenyl phosphate as substrate. The V(max) and K(m) values were 0.3 micromol.

Transporter-mediated drug interactions: clinical implications and in vitro assessment.

Although they are less frequently compared with the reported cases of CYP-mediated drug interactions, clinically significant transporter-mediated drug interactions, which are mainly based on efflux transporter or P-glycoprotein data, have been reported. Unlike the CYP-mediated drug interactions that can be readily defined by inhibition or induction of CYP enzymes, the evidence for the so-called transporter-mediated drug interactions is often less conclusive. The difficulty in defining transporter-mediated drug interactions is due mainly to the interplay between transporters and drug-metabolizi

Biochemical and pharmacological aspects of two bradykinin-potentiating peptides obtained from tryptic hydrolysis of casein.

Peptides that display bradykinin-potentiating activity have been obtained from a number of distinct sources, such as snake venoms, fibrinogen, and casein. This paper describes the characterization of two new peptides generated by tryptic hydrolysis of casein. No homology was found with other known vasoactive or vasopotentiating peptides, especially by the lack of Ile-Pro-Pro motif. The peptides EMPFPK and YPVEPFTE, corresponding to the gamma casein sequence (108-113 and 114-121, respectively), displayed a selective potentiating activity on isolated guinea pig ileum for bradykinin. Besides, the

The role of enzyme induction and inhibition on cypermethrin hepatotoxicity.

Cypermethrin at different concentrations (100, 200, 400 and 800 ng ml(-1)) was incubated with a primary culture of rat hepatocytes. Cypermethrin was cytotoxic to rat hepatocytes at concentrations of 200 ng ml(-1)or greater. Toxicity was measured by a decrease in cell viability and leakage of ALT and AST enzymes into the culture medium. The role of cytochrome P450 in the hepatotoxicity of cypermethrin insecticide was investigated in fresh hepatocytes isolated either from phenobarbital pretreated rats or control rats and coincubated with SKF525A. Pretreatment with phenobarbital strongly protecte

Covalent binding and inhibition of cytochrome P4502E1 by trichloroethylene.

1. To investigate the effects of trichloroethylene on cytochrome P4502E1 (CYP2E1), an isozyme responsible for its metabolic activation, mice were treated with trichloroethylene and Western blot staining with both anti-dichloroacetyl and anti-CYP2E1 antisera detected a comigrating 50 kDa protein band. There was a dose-dependent increase in the intensity of the 50 kDa protein adduct stained immunochemically with anti-dichloroacetyl. 2. CYP2E1 enzyme activity was decreased from control levels in a dose-dependent manner in mice treated with 250-500 mg/kg TRI. 3. Microsomal incubations with trichlo

Effect of adenylate cyclase activators on C5a-induced human neutrophil aggregation, enzyme release and superoxide production.

The effect of adenylate cyclase activators on C5a- and f-Met-Leu-Phe-induced human neutrophil aggregation, enzyme release and superoxide production was investigated. C5a-stimulated superoxide production was markedly inhibited by adenylate cyclase activators, and the order of potency was PGE1 greater than isoproterenol greater than epinephrine greater than PGF2 alpha, which correlated with intracellular cAMP levels. However, neutrophil aggregation was inhibited by PGE1, PGE2, isoproterenol and epinephrine only at concentrations greater than 10(-6) M. Lysozyme release was inhibited only via PGEs

A trimethyl lock successfully stabilizes a nascent Michael acceptor for prodrug applications.

5-Hydroxy-γ-pyrone-based Michael acceptors have repeatedly registered as hits in screens for inhibiting key therapeutic targets, but their aqueous instability has prevented further development. Here, we report a method to stabilize such an inhibitor of STAT3 with a trimethyl lock until reduction by hNQO1, an enzyme typically overexpressed in hypoxic tumors.

Clinical use of captopril in Takayasu's disease.

Three patients with Takayasu's disease (TD), initially observed with hypertension and/or congestive heart failure, were treated with captopril, an orally active inhibitor of converting enzyme. As arteritis in this disease is diffuse and extensive, surgical correction of renovascular hypertension is often delayed or impossible. Our experience enables us to recommend captopril for conservative treatment in TD.

Genetic Interaction Analysis Reveals that Cryptococcus neoformans Utilizes Multiple Acetyl-CoA-Generating Pathways during Infection.

Cryptococcus neoformans is an important human fungal pathogen for which the external environment is its primary niche. Previous work has shown that two nonessential acetyl-CoA metabolism enzymes, ATP-citrate lyase (ACL1) and acetyl-CoA synthetase (ACS1), play important roles in C. neoformans infection. Here, we took a genetic interaction approach to studying the interplay between these two enzymes along with an enzyme initially called ACS2 but which we have found is an acetoacetyl-CoA synthetase; we have renamed the gene 2-ketobutyryl CoA synthetase 1 (KBC1) based on its biochemical activity a

Early treatment with pyridostigmine alleviates the isoprenaline-induced model of heart failure with preserved ejection fraction (HFpEF) in rats.

Acetylcholinesterase (AChE) inhibitors constitute a large and chemically diverse group of compounds that inhibit the enzyme AChE. Some of these compounds that are used to treat Alzheimer's disease have been reported to have favourable cardiovascular effects, i.e., a 35% reduction of the risk for cardiovascular disease. There has been a growing interest in AchE inhibitors as a potential therapeutic approach for cardiovascular diseases, especially heart failure (HF), since they correct the autonomic imbalance, a key component in the development of heart failure. In the present study, HF was indu

Advances in Heterocyclic Derivatives for Aromatase Inhibition in Hormone-Dependent Breast Cancer.

Breast cancer is one of the most common causes of cancer death among women in the world, and the number of tumors that depend on hormones occupies a significant percentage of cases that are diagnosed. The estrogen factor is central to the development of these cancers; hence, aromatase, the central enzyme involved in estrogen production, is a critical therapeutic target. Aromatase Inhibitors (AIs), in particular, have emerged to play an important role in endocrine therapy in postmenopausal breast cancer patients. Heterocyclic compounds, in particular, have also been receiving significant focus

Suppression of Lipopolysaccharide-Induced Cytokine Production by Chemokine Ligand-1 Short Hairpin RNA.

Objectives: Lipopolysaccharide (LPS) from oral Gram-negative bacteria induces inflammatory responses involving the chemokine CXCL1. Although CXCL1 is known to mediate inflammation, its role in disease processes remains incompletely understood. This study aimed to investigate the biological function of CXCL1, evaluate shRNAs as inhibitors, and determine whether CXCL1 suppression is associated with reduced LPS-induced cytokine production.Materials and Methods: Human CXCL1 cDNA was cloned into recombinant plasmid DNA and transfected into THP-1 monocyte-like cells. CXCL1-regulated cytokines, inclu

Molecular interaction profiling and binding dynamics of Cinnamomum zeylanicum phytochemicals with human pancreatic amylase.

Diabetes mellitus, characterized by persistent hyperglycemia, remains a critical global health challenge. Inhibition of human pancreatic alpha-amylase, a key enzyme catalyzing carbohydrate digestion, is a promising approach to manage postprandial glucose levels. Cinnamomum zeylanicum, a medicinal plant known for its therapeutic potential, harbors bioactive compounds that can act as natural alpha-amylase inhibitors, though their mechanisms remain underexplored. In this study, molecular docking and 200 ns molecular dynamics (MD) simulations were employed to evaluate the inhibitory potential of 1

Flipping the Target: Evaluating Natural LDHA Inhibitors for Selective LDHB Modulation.

Lactate dehydrogenase (LDH) catalyzes the reversible interconversion of pyruvate and lactate, coupled with the redox cycling of NADH and NAD+. While LDHA has been extensively studied as a therapeutic target, particularly in cancer, due to its role in the Warburg effect, LDHB remains underexplored, despite its involvement in the metabolic reprogramming of specific cancer types, including breast and lung cancers. Most known LDH inhibitors are designed against the LDHA isoform and act competitively at the active site. In contrast, LDHB exhibits distinct kinetic properties, substrate preferences,

New Fuels for a Failing Engine: The Impact of Novel Heart Failure Drugs on Functional Capacity.

Functional impairment is a hallmark of heart failure (HF) and a strong prognostic factor. Cardiopulmonary exercise testing (CPET) provides a robust and objective assessment of exercise capacity; however, the impact of new pharmacotherapies on CPET parameters remains largely uncharacterized systematically. This review examines the influence of contemporary HF therapies on functional capacity, with particular focus on CPET-derived metrics, such as peak oxygen uptake (VO2 peak), ventilatory efficiency (VE/VCO2 slope), and oxygen uptake efficiency slope (OUES). A critical synthesis of randomized t
