Recent developments in Campylobacter pathogenesis.

PURPOSE OF REVIEW: The Campylobacter species jejuni and coli are leading causes of enteritis and enterocolitis worldwide. Arthritis, Reiter syndrome, and Guillain-Barré syndrome represent post-infectious sequelae. Although the acute and chronic clinical manifestations highlight Campylobacter species as excellent models for the identification of mechanisms involved in pathogenesis, detailed investigations at the molecular level are complicated by the fastidious growth requirements of the bacteria and by the tremendous variability displayed by Campylobacter isolates. Thus, research activities in

Borrelia burgdorferi transcriptome in the central nervous system of non-human primates.

Neurological symptoms are common manifestations of Lyme disease; however, the paucibacillary nature of the spirochete in this environment has precluded a molecular analysis of the spirochete in the CNS. We have now adapted differential expression analysis by using a custom-amplified library (DECAL) in conjunction with Borrelia burgdorferi whole-genome and subgenome arrays to examine in vivo gene expression by B. burgdorferi in a non-human primate (NHP) model of neuroborreliosis. The expression profile of B. burgdorferi was examined in the CNS and heart of steroid-treated and immunocompetent NH

Proteins and proteinases in the in vitro released products (IVRP) of the tissue-invasive and lung-dwelling larvae of the pentastomid Porocephalus crotali.

Pentastomids are arthropod-like parasites which inhabit the respiratory tract of tetrapods and, despite being haematophagous, cause little or no visible pathology. Nymphs (or larvae), belonging to the advanced genus Porocephalus, encyst in the tissues of rodent intermediate hosts and migrate from this site to the lung following ingestion of the intermediate host. During migration, several moults occur and instars encounter a variety of tissue barriers and environmental cues. It was therefore relevant to define the proteins and proteinases released by the different instars during this migration

Identification of the teichoic acid phosphorylcholine esterase in Streptococcus pneumoniae.

Streptococcus pneumoniae is a major human pathogen and many interactions of this bacterium with its host appear to be mediated, directly or indirectly, by components of the bacterial cell wall, specifically by the phosphorylcholine residues which serve as anchors for surface-located choline-binding proteins and are also recognized by components of the host response, such as the human C-reactive protein, a class of myeloma proteins and PAF receptors. In the present study, we describe the identification of the pneumococcal pce gene encoding for a teichoic acid phosphorylcholine esterase (Pce), a

The lymphocytic choriomeningitis virus RING protein Z associates with eukaryotic initiation factor 4E and selectively represses translation in a RING-dependent manner.

Only a few host cell proteins that associate with arenaviruses have been identified. To date, the arenavirus Z protein associates with the promyelocytic leukemia protein PML and the ribosomal P proteins. The majority of PML is present in nuclear bodies which are translocated to the cytoplasm by infection with the arenavirus, lymphocytic choriomeningitis virus (LCMV). The Z protein is a small zinc-binding RING protein with an unknown function which is required for the viral life cycle. Here, we demonstrate an association between Z and the host cell translation factor, eukaryotic initiation fact

Studies on the role of mononuclear phagocytes in resistance to acute lymphocytic choriomeningitis virus infection.

The role of mononuclear phagocytes in various phases of the acute lymphocytic choriomeningitis virus (LCMV) infection was studied. The anti-macrophage agent carrageenan delayed virus clearance. Carrageenan was most effective when given before virus inoculation, suggesting that it interfered with early events in the host response to the virus. Correspondingly, carrageenan enhanced early virus multiplication. Pretreatment with carrageenan apparently did not inhibit induction of the T-cell response and had little or no direct effect on T-cell-dependent anti-viral activity. The LCMV-induced natura

[Hantavirus infections].

Hantaviruses are enveloped zoonotic RNA viruses hosted by rodents and responsible in the Americas for hantavirus pulmonary syndrome. In Europe, they cause hemorrhagic fever with renal syndrome and its milder form, nephropathia epidemica. The disease begins abruptly with high fever, chills, headache, back pain and abdominal pain associated with nausea and vomiting. Diagnosis is primarily made by serology. There is currently no specific medication or preventive available in Europe. Treatment is symptomatic.

The eyes have it: long-distance dispersal by an intraorbital leech parasite of birds.

A leech was found parasitizing the ocular orbit of a common redstart captured during a faunistic survey of Antikythira in the Aegean Sea during the spring migration of 2012. Morphological and molecular characterizations placed the leech in the mucous-membrane specific leech family Praobdellidae and definitively as the species Parapraobdella lineata. This is the first record of any leech parasitizing a passerine bird, Phoenicurus phoenicurus , and the first of a praobdellid leech on any avian host.

Quantification of host-microbe interactions by automated fluorescence microscopy.

We describe an automated fluorescence microscopy-based assay that quantifies the invasion of mammalian cells by intracellular pathogens. Pathogens associated with host cell surfaces, intracellular pathogens and mammalian cells are directly counted based on their specific fluorescent labeling. Such approach utilizes automated image acquisition and processing, and is thus ideally suited for high-throughput analyses. This method was validated using Listeria monocytogenes as a model intracellular pathogen.

[Structural organization and function of parasitic systems].

The structural organization and functioning of parasitic systems are considered in the context of the general theory of systems. By using the parasitic system of an epizootic process of plague as an example, the essence of the concepts of the structural organization and functioning of the systems is determined, and a scheme of a relationship of elements in the trinomial parasitic systems is presented. The mechanism of parasite-host interaction at the molecular, cellular, body's, and populational levels is considered on the basis of the theory of immune-specific wave information in an living or

Protection of mice against intestinal amoebiasis with BCG, Corynebacterium parvum and Listeria monocytogenes.

Treatment with Corynebacterium parvum or BCG, or infection with live Listeria monocytogenes was found to protect mice against subsequent infection with Entamoeba histolytica. Complete protection was obtained in mice treated with 10(7) (colony forming units) of BCG but not with 10(5). Partial protection was achieved with 10(6) of BCG. These data provide evidence that non-specific immunity plays an important role in the host defense against amoebic infection.

Noninvasive method for monitoring Pneumocystis carinii pneumonia.

The progression of Pneumocystis carinii pneumonia was temporally monitored and quantified by real-time polymerase chain reaction of P. carinii-specific DNA in oral swabs and lung homogenates from infected rats. DNA levels correlated with the number of P. carinii organisms in the rats' lungs, as enumerated by microscopic methods. This report is the first of a noninvasive, antemortem method that can be used to monitor infection in a host over time.

[Current aspects of trichinosis diagnosis].

An important approach to the study of trichinellosis is the development of sensitive diagnostic methods that allow the detection of Trichinella spiralis. Recently, ELISA assays that use surface/stichosome and/or secretion/excretion antigens from muscle larvae have been proved to be highly sensitive and specific in the diagnosis of the infection. Furthermore, the high immunodominance of carbohydrate residues on these molecules in a broad host range make them useful diagnostic markers for trichinellosis.

[Pleural effusion and Toxocara canis].

Toxocariasis is an infrequent disease which particularly affects children. Toxocara canis is an uncommon parasite of man, who is not the normal host for the nematode. Without doubt Toxocara will be recognised more and more thanks to specific serological tests, in particular the ELISA-ESA test. The authors observed the association of a pleural reaction and a generalised hypereosinophilia (blood, pleural fluid and broncho-alveolar lavage). They noted a complete regression on Mebendazole.

Studies on proliferation kinetics in liver and spleen during early graft-vs-host reaction in mice.

Autoradiographic and liquid scintillation counting studies were made during the early graft-versus-host-reaction in mice. At the height of GVHR on the 10th day after transplantation, hepatosplenomegaly correlated with the radioactive disintegrations (DPM). This coincided with a high proliferation rate of the blasts that appeared in the spleen, and of the portal infiltrations and Kupffer cells in the liver. Convenient control groups were used to show that these changes are a specific reaction in GVHR.

Beyond AMR and virulence databases: genome wide associations of closely related Vibrio alginolyticus and emerging Vibrio diabolicus provide framework for identifying novel genetic markers.

Vibrio alginolyticus is a frequently implicated species for vibriosis in humans and diverse wildlife, but it has previously been difficult to identify from the closely related and emerging Vibrio diabolicus. Comparisons of both species, including antimicrobial resistance (AMR) and virulence characterizations, are scarce and impeded by intraspecies diversity, minimal genomes, discordant classification methods, and gene databases with limited utility to understudied species. The species identities of 3,442 public domain genomes (SRA files) within the Harveyi clade were re-evaluated using genomic

Histone Acetylation Dynamics Regulate Fungal Development, Pathogenicity, and Mycotoxin Biosynthesis.

Histone acetylation, catalyzed by histone acetyltransferases (HATs) and reversed by histone deacetylases (HDACs), is a central epigenetic mechanism that regulates gene expression in eukaryotes. Identifying and characterizing these enzymes has rapidly advanced our understanding of this dynamic modification. In phytopathogenic fungi, HATs and HDACs modulate several cellular processes, including development, pathogenicity, and secondary metabolism. The filamentous fungus Fusarium graminearum causes Fusarium head blight, a destructive disease that reduces wheat yield and contaminates grain with th

Subcutaneous and intranasal immunization with Stx2B-Tir-Stx1B-Zot reduces colonization and shedding of Escherichia coli O157:H7 in mice.

The type III secretion system of Escherichia coli O157:H7 is involved in colonization of mammalian hosts by the organism. The translocated intimin receptor (Tir) is inserted into the mammalian host cell plasma membrane in a hairpin loop topology with the central loop of the molecule exposed to the host cell surface and accessible for interaction with an LEE-encoded bacterial outer membrane adhesin called intimin. Shiga toxin type 1 and 2 produced by E. coli O157:H7 are responsible for hemolytic uremic syndrome and able to promote intestinal colonization. Zonula occludens toxin (Zot) is a singl
