Novel role of FTO in regulation of gut-brain communication via Desulfovibrio fairfieldensis-produced hydrogen sulfide under arsenic exposure.

Fat mass and obesity-associated protein (FTO) is the key demethylase that reverses the abnormally altered N6-methyladenosine (m6A) modification in eukaryotic cells under environmental pollutants exposure. Arsenic is an environmental metalloid and can cause severe symptoms in human mainly through drinking water. However, there is no specific treatment for its toxic effects due to the uncovered mechanisms. We previously revealed that exposure to arsenic increased the level of m6A via down-regulation of FTO, which might serve as a potential target for intervention against arsenic-related disorder

Simultaneously delivery of functional gallium ions and hydrogen sulfide to endow potentiated treatment efficacy in chemo- and PARPi-resistant ovarian cancer.

Limited therapeutic options are available for patients with platinum-resistant ovarian cancer (OC). Herein, we developed gallium sulfide-embedded bovine serum albumin nanoformulations (Ga2S3-BSA NMs) with a size of ~ 11 nm via a self-assembly approach. As the nanoformulations degraded in an acidic cancer microenvironment, Ga3+ and H2S gas were simultaneously released to exert their combined anticancer effects. In A2780-CIS and SKOV3-CIS platinum-resistant OC cells, Ga3+ and H2S released from Ga2S3-BSA NMs synergistically enhanced DNA damage, which arrested the cell cycle at S and G2/M phases a

Hydrogen sulfide attenuates PM2.5-induced COPD by inhibiting cellular senescence via the Klotho/Parkin-mediated mitophagy signaling pathway.

Fine particulate matter (PM2.5), an atmospheric pollutant, plays a crucial role in mediating the occurrence of chronic obstructive pulmonary disease (COPD). Hydrogen sulfide (H2S) has shown therapeutic potential in countering COPD, but the underlying mechanisms remain elusive. This study aimed to elucidate the potential mechanisms of H2S in inhibiting cellular senescence and combating the progression of COPD. In this study, mouse and cell models were initially established by pretreating with PM, PM2.5, and NaHS. It was showed that increased expressions of p16, p21, and β-gal positive cells, al

Homogeneous dual-mode ECL-FL sensor for sensitive hydrogen sulfide detection: Mechanistic insights and applications in environmental and bioanalytical monitoring.

A dual-mode electrochemiluminescence (ECL) and fluorescence (FL) probe was developed based on functionalized metal-organic framework nanosheets (RuMOFNSs) and (3-((2,4-dinitrophenyl)thio)phenyl)methanol (DNB) for the highly sensitive detection of hydrogen sulfide (H₂S) in homogeneous systems. Compared to conventional heterogeneous detection strategies, the homogeneous nature of this probe eliminates signal loss associated with solid-phase immobilization, thereby enhancing the interaction efficiency between the probe and target molecules. The developed sensing platform exhibited exceptional sen

A novel liver-targeted and highly selective fluorescent probe for hepatic hydrogen sulfide detection in the diagnosis of drug-induced liver injury.

Drug-induced liver injury (DILI) has emerged as a significant health risk, yet reliable methods of detection at an early stage are in urgent need. Recent researches have highlighted the crucial role of hydrogen sulfide (H2S) in various pathological and physiological processes, with a correlation between its levels and the severity of DILI. We developed a novel fluorescent probe H2S-YL for the precise detection of liver H2S fluctuations in DILI, and the liver in situ accumulation capacity of H2S-YL has been significantly enhanced by incorporating a liver-targeting cholic acid moiety into its sk

Association of Kidney Graft Long-term Outcome With Recipient Cystathionine Gamma-lyase Polymorphisms and Hydrogen Sulfide Levels: A Cohort Study.

BACKGROUND: Hydrogen sulfide (H2S) produced endogenously by the CTH gene-encoded cystathionine gamma-lyase protects from renal ischemia-reperfusion injury in preclinical models. Here, we hypothesized that CTH gene polymorphisms (single nucleotide polymorphism [SNP]) and recipient H2S serum levels influence kidney graft outcomes after transplantation. METHODS: We included all consecutive recipients of a first kidney transplant in the Swiss Transplant Cohort Study and with available genotyping. In addition, 192 deceased-donor kidney transplant recipients were randomly selected to measure baseli

High-sensitivity detection of hydrogen sulfide and water content using a Triphenylamine-based fluorescent probe: Applications in food safety monitoring.

This study reports the design and synthesis of a fluorescent probe TPA-ACL based on the triphenylamine (TPA) structure for the dual detection of hydrogen sulfide (H₂S) and water content. In the presence of H₂S, TPA-ACL exhibits a pronounced "turn-on" fluorescence response, demonstrating excellent selectivity and high sensitivity with a detection limit (LOD) of 2.02 nM. Furthermore, TPA-ACL enables real-time detection of H₂S using test strips, allowing for visual monitoring of H₂S gas generated during food spoilage, indicated by a color change from pale yellow to deep yellow. The probe was also

Specific imaging of hydrogen sulfide in paw edema mice via a novel near-infrared fluorescent probe with large stokes shift.

Paw edema is a classic acute inflammation model, which is often used in clinical screening and evaluation of anti-inflammatory effects of drugs. As an endogenous gas signaling molecule, hydrogen sulfide (H2S) is closely related to the progression of paw edema inflammation. However, no fluorescent probes have been reported to visualize H2S levels in paw edema mice. Herein, a novel near-infrared (NIR) fluorescent probe (BAX-N) is developed to detect H2S. The NIR dye formed by the conjugation of benzothiazole-2-acetonitrile with xanthene serves as the fluorophore (BAX-OH), and nitrobenzoxadiazole

Role of carbon nanotubes in boosting photocatalytic hydrogen production and pollutants degradation within S-Scheme g-C3N4/ZnO nanocomposite.

In this study, the carbon nanotubes (CNTs) modified g-C3N4/ZnO photocatalytic nanocomposite was synthesized through in-situ growth of ZnO nanoparticles on the CNT-loaded graphitic carbon nitride (g-C3N4) to evaluate its hydrogen production and dye degradation capabilities. The disc-like ZnO nanoparticles were spotted alongside the CNTs on the g-C3N4 surface with a specific surface area of 140.8 m2/g. The electrostatic interactions between conjugated bonds of g-C3N4 and π bonds of CNTs resulted in a well-bonded interface and high adsorption of CNTs. CNTs significantly increased the visible ligh

The inhibition of cystathionine gamma-lyase and cystathionine beta-synthase - a strategy to reduce hydrogen sulfide emissions during livestock manure composting.

The composting of livestock manure produces a significant amount of hydrogen sulfide (H2S), which can cause environmental pollution. This has become a substantial impediment to the advancement of the composting industry. In this study, Cystathionine gamma-lyase (CSE) and Cystathionine beta-synthase (CBS) enzymes were explored from the perspective of endogenous H2S production. This study investigated aminooxyacetic acid (AOAA) and DL-propargylglycine (PAG) as enzyme inhibitors in composting to assess the feasibility and effectiveness of H2S emission reduction from chicken and swine manure. The

Effects of G protein-coupled estrogen receptor on hydrogen sulfide production and cystathionine γ-lyase expression in human endothelial cells.

Hydrogen sulfide (H2S) is an important regulator of cardiovascular homeostasis, along with nitric oxide (NO). In endothelial cells, H2S is primarily synthesized by cystathionine γ-lyase (CSE) using L-cysteine and homocysteine as substrates. However, it has not been elucidated how G protein-coupled estrogen receptor 1 (GPER) affects H2S production in endothelial cells. In this study, we investigated whether GPER activation enhances H2S production by upregulating CSE expression in endothelial cells, and identified the underlying molecular mechanism. G-1 increased the phosphorylation of cAMP resp

Haloalkane-driven dichotomous reactivity of aryl radicals as halogen and hydrogen atom transfer agents: photocatalytic olefin and alkyne functionalization cascades.

A visible-light mediated protocol for the annulative trifluoroethylation, perfluoroalkylation and di/tri-chloromethylation of olefins and alkynes tethered to aromatic rings is described. Both PIDA and diazonium salts can independently promote this transformation by simultaneously acting as both an oxidant and halogen atom transfer reagent. The reaction is applicable to multiple precursor classes, leading to the synthesis of various N- and O-containing ring systems including dihydroisoquinolinones, coumarins, fused benzimidazoles, and quinazolinone. The activation of chloroform and dichlorometh

Beyond the Tip Effect: Geometrically Symmetric Gold Nanostars Achieve Uniform Hotspots for Ultrasensitive and Reproducible SERS Monitoring of Hydrogen Sulfide.

Surface-enhanced Raman scattering (SERS) is a powerful technique for ultrasensitive molecular analysis, benefiting from exceptional sensitivity and matrix tolerance. This study moves beyond the conventional tip enhancement strategy by designing geometrically symmetric gold nanostars (S-GNSs) with finely tunable sizes and tip curvatures. A systematic investigation reveals that symmetric geometry not only promotes uniform electromagnetic field distribution but also significantly improves signal reproducibility compared to traditional asymmetric nanostars. Among the synthesized substrates, S-GNSs

Taurine ameliorates cellular senescence associated with an increased hydrogen sulfide and a decreased hepatokine, IGFBP-1, in CCl4-induced hepatotoxicity in mice.

This study investigated the protective effects of taurine against cellular senescence and hepatokine secretion in a mouse model of carbon tetrachloride (CCl4)-induced chronic liver injury. Oral taurine administration by tap water containing 3 % taurine significantly attenuated liver damage, as evidenced by reduced serum AST, ALT level and hepatic lipid peroxidation. Importantly, hepatic taurine level is reduced in CCl4-induced injury model, while taurine administration recovered it. Moreover, taurine administration decreased the numbers of p21-positive senescent cells in liver tissue of CCl4-t

Salicylic Acid, Hypersensitive Response and RBOHD-Mediated Hydrogen Peroxide Accumulation Play Key Roles in Black Rot Resistance of Crucifers.

Black rot caused by hemibiotrophic Xanthomonas campestris pv. campestris (Xcc) is a great problem in crucifer crop production. Various host responses are activated upon Xcc attack; however, their roles in black rot resistance remain ambiguous. In this study, a highly black rot resistance of host plants was achieved by applying a field-screened systemic resistance-eliciting Bacillus velezensis strain 37-1. The contributions of strain 37-1-altered host responses to Xcc resistance were then investigated in Arabidopsis. Hypersensitive response and hydrogen peroxide accumulation were demonstrated b

ReaxFF studies of surface fluorination of alumina and etching of alumina/aluminum metal heterostructures under gas-phase hydrogen fluoride exposure.

The self-limiting surface conversion reactions of alumina and alumina/aluminum metal heterostructures under HF exposure are studied at 1250 and 750 K, respectively, using a newly developed Al/O/H/F ReaxFF force field. Simulations reveal that surface termination and HF concentration significantly influence the fluorination behavior of alumina. The 100% Al-terminated α-Al2O3 (0001) exhibits the highest degree of fluorination at equilibrium, while the O-rich surface shows negligible surface conversion. Increasing HF concentration on the Al-rich surface reveals the maximum surface fluorination, re

Therapeutic potential of hydrogen-rich water in zebrafish model of Alzheimer's disease: targeting oxidative stress, inflammation, and the gut-brain axis.

Alzheimer's disease (AD) is a complex neurodegenerative disorder, with amyloid-beta (Aβ) aggregation playing a key role in its pathogenesis. Aβ-induced oxidative stress leads to neuronal damage, mitochondrial dysfunction, and apoptosis, making antioxidative strategies promising for AD treatment. This study investigates the effects of hydrogen-rich water (HRW) in a zebrafish AD model. Zebrafish were exposed to aluminum chloride to induce AD-like pathology and then treated with HRW using a nanobubble device. Behavioral assays, ELISA, Hematoxylin-eosin (H&E) staining, and reactive oxygen species

An injectable and adaptable system for the sustained release of hydrogen sulfide for targeted diabetic wound therapy by improving the microenvironment of inflammation regulation and angiogenesis.

The combined effects of persistent chronic inflammation, oxidative stress, microcirculation disorders, and dysregulated cellular energy metabolism often hinder the repair of diabetic skin wounds. Traditional treatment methods are typically insufficient in simultaneously addressing these complex factors, resulting in delayed wound healing and a high propensity for recurrence and chronic ulceration. This study developed an innovative strategy based on reactive oxygen species (ROS)-responsive nanoparticles loaded with an ultraviolet (UV)-light-responsive hydrogen sulfide (H2S) donor. The effects of heat and hydrogen peroxide treatment on the osteoinductivity of demineralized cortical bone: a potential method for preparing tendon/ligament repair scaffolds.

Recent studies have indicated that demineralized cortical bone (DCB) may be used to repair tendons and ligaments, such as the patellar tendon and anterior cruciate ligament (ACL). Hydrogen peroxide (H2O2) has been shown to reduce the osteoinductivity of DCB, and heat treatment may also decrease the osteoinductivity of DCB. The purpose of this study was (i) to determine whether heat treatment reduces the osteoinductivity of DCB and (ii) to compare the effectiveness of heat treatment and H2O2 treatment on BMP-2 inactivation. DCB was prepared by immersion in 0.6 N hydrochloric acid, and DCB-H and
