Chronic blockade of NO synthase activity induces a proinflammatory phenotype in the arterial wall: prevention by angiotensin II antagonism.

Chronic blockade of NO production induces hypertension and early occlusive and fibrotic end-stage organ damage owing to vascular lesions in the brain, kidney, and heart. In this study, we evaluated the inflammatory phenotypic changes induced in the arterial wall by chronic N(G)-nitro-L-arginine methyl ester (L-NAME) administration and the effect of an angiotensin II receptor (AT1) antagonist, irbesartan, on these changes. For this purpose, 2 groups of rats received L-NAME in the drinking water (50 mg x kg(-1) x d(-1)) for 2 months. One group received no other treatment and the other was treate

Incidence and morphology of endometrial angiopathies in mares in relationship to age and parity.

The morphology of endometrial blood vessels in uterine biopsy specimens from mares of varying age and reproductive status was examined by light (n = 117) and electron microscopy (n = 13), and additionally after elastase digestion (n = 86). Inflammatory vascular alterations were observed in 20.5% of the specimens. Smaller and larger arterial and venous vessels demonstrated mild to severe degenerative lesions. Unaltered vessels were detected only in maiden mares. Vessels in older maiden mares were frequently affected by angiosclerotic changes, characterized by mild to moderate perivascular and i

Platelet activating factor produced in vitro by Kaposi's sarcoma cells induces and sustains in vivo angiogenesis.

Imbalance in the network of soluble mediators may play a pivotal role in the pathogenesis of Kaposi's sarcoma (KS). In this study, we demonstrated that KS cells grown in vitro produced and in part released platelet activating factor (PAF), a powerful lipid mediator of inflammation and cell-to-cell communication. IL-1, TNF, and thrombin enhanced the synthesis of PAF. PAF receptor mRNA and specific, high affinity binding site for PAF were present in KS cells. Nanomolar concentration of PAF stimulated the chemotaxis and chemokinesis of KS cells, endothelial cells, and vascular smooth muscle cells

Study on the Role and Mechanism of TOPORS in Regulating Aortic Dissection by Mediating SUMOylation.

Aortic dissection (AD) is a fatal acute cardiovascular emergency. SUMOylation participates in cell proliferation, apoptosis, and inflammation, but its role in AD, especially via TOPORS, remains unclear. This study investigates how TOPORS regulates AD pathogenesis through SUMOylation. AD and normal aortic samples were collected to detect TOPORS expression. AD mouse and VSMCs models were constructed to assess TOPORS depletion and overexpression effects on AD progression. In AD aortic tissues, TOPORS expression was upregulated, while tripartite motif containing 27 (TRIM27) and Sentrin-specific pr

The chicken embryo model as a tool for investigating drug-induced acute kidney injury.

Nephrotoxicity remains a critical concern in drug development, with limited preclinical models that balance physiological relevance, ethical considerations, and scalability. Here, we evaluate the chicken embryo (CE) as an in vivo platform for nephrotoxicity assessment, using gentamicin-induced acute kidney injury as a proof-of-principle. Using an ex ovo culture system, we assessed renal responses in the developing CE kidney. By embryonic day 12, the CE exhibited advanced organogenesis, including a functional metanephric kidney. Immunohistochemical analyses demonstrated conservation of key stru

A neurovascular survival axis for adult sympathetic neurons.

Adult neuronal survival underlies lifelong brain function, yet its sustaining mechanisms remain unclear. We identify a neurovascular survival axis at intracranial arteries in mice, where superior cervical ganglion (SCG) sympathetic terminals form synapse-like neurosmooth muscular junctions (NsMJs) with arterial smooth muscle cells (aSMCs) that secrete netrin-4 (Ntn4). Adult aSMC-restricted Ntn4 deletion selectively reduces rostral, intracranial-projecting (cerebrospinal fluid-traced) SCG neurons; the denervation of the intracranial arterial plexus follows soma loss. Local recombinant Ntn4 deli

Fibronectin-induced overactivation of αVβ3-PI3K-PIP3-PDK1-ILK signaling drives aortic disease in Marfan syndrome.

Thoracic aortic aneurysms and dissections (TAAD), a life-threatening complication of Marfan syndrome (MFS), lack curative therapies. Our previous studies revealed that versican accumulation drives MFS aortopathy through AKT-NO pathway overactivation, but the upstream mechanisms remained unclear. Here, we show that versican-driven fibronectin (FN) accumulation activates an αVβ3-PI3K-PIP3-PDK1-ILK signaling cascade leading to AKT-NOS2 upregulation and aortic disease. FN accumulates in aortas of MFS patients and mice of both sexes and correlates with increased αVβ3 integrin and ILK expression. Di

LAMB2 p.E991K Mutation-Mediated Atherosclerosis in Rabbit.

Background: Atherosclerosis is a major pathological basis of cardiovascular disease and is influenced by both genetic and environmental factors. Although genome-wide association studies have identified the LAMB2 p.E987K variant as a susceptibility locus for atherosclerosis, its functional role remains unclear. Methods: In this study, a rabbit model carrying the homologous LAMB2 p.E991K mutation was generated using the CRISPR/Cas9-SpG system and subjected to a high-fat diet to induce atherosclerosis. Results: LAMB2 mutant rabbits exhibited increased body weight and significant lipid metabolic a

Hypertriglyceridemia Does Not Worsen Thoracic Aortic Aneurysm and Dissection in GPIHBP1 Knockout Mice-Brief Report.

BACKGROUND: Hypertriglyceridemia is an established risk factor for atherosclerotic cardiovascular disease and has recently been implicated in abdominal aortic aneurysm pathogenesis. However, its role in thoracic aortic aneurysm and dissection (TAAD) remains undefined. METHODS: We investigated the impact of hypertriglyceridemia on experimental TAAD using GPIHBP1 (glycosylphosphatidylinositol-anchored high-density lipoprotein-binding protein 1) knockout mice as a hypertriglyceridemia model. TAAD was induced in 3- to 4-week-old mice via oral β-aminopropionitrile administration for 28 days. RESU

Inflammation as a Unifying Axis: Shared Pathogenic Mechanisms in Coronary Atherosclerosis and Chronic Atrophic Gastritis.

Coronary atherosclerosis (AS) and chronic atrophic gastritis (CAG) are clinically distinct diseases but share core features of chronic, non-resolving inflammation. This review evaluates their convergent immunoinflammatory architecture and discusses how shared pathogenic nodes may inform dual-purpose therapeutic strategies. Both conditions involve barrier dysfunction driven by oxidative stress and nitric oxide dysregulation, maladaptive immune responses involving macrophage polarization and T-cell plasticity, and sustained inflammatory signaling through pathways such as TLR/NF-κB, HIF, and PI3K

Lipid overload meets S-palmitoylation: a metabolic signalling nexus driving cardiovascular and heart disease.

S-palmitoylation has emerged as a critical integrator of lipid overload and cardiovascular dysfunction. Disordered lipid metabolism inundates endothelial cells, vascular smooth muscle cells and macrophages with triglyceriderich lipoproteins, oxidized LDL and saturated fatty acids, expanding the intracellular palmitoylCoA pool and perturbing redox balance. Protein Spalmitoylation, the reversible attachment of palmitate to cysteine residues, converts excess palmitoylCoA into broad alterations in signalling and membrane dynamics. The FASN-ACSL-ZDHHC axis channel excess fatty acids into palmitoylC

Transfer RNA-derived small RNAs as novel players and biomarkers in cardiovascular disease.

An emerging field in cardiovascular research is the translational investigation of transfer RNA-derived small RNAs (tsRNAs). TsRNAs, a class of small non-coding RNA molecules, have been shown to modulate cellular functions by regulating gene expression post-transcriptionally. They are implicated in diverse pathological conditions, including cancer, cardiovascular disease (CVD), infectious disease, diabetes, neurological disease, and metabolic disorder. Accumulating evidence suggests tsRNAs as important players and biomarkers in CVD. Dysregulated tsRNAs are identified in atherosclerosis, heart

Taurolidine inhibits influenza virus infection and prevents influenza-induced cytokine storm, vasoconstriction and lung damage.

Influenza virus causes worldwide outbreaks and seasonal epidemics, posing a severe threat to public health and social development. Effective prevention and treatment of influenza infections remain major challenge for global healthcare. In this study, we observed that taurolidine effectively inhibited the proliferation of several human or animal influenza virus strains and protected mice from lethal-infection. Taurolidine treatment decreased the viral titer in the lungs of infected mice, reduced the ratio of immune cells, and alleviated lung pathology. Additionally, taurolidine treatment attenu

Anesthetic management of coronary artery reconstruction in a patient with myocardial ischemia caused by an anomalous origin of the right coronary artery running between the great vessels: a case report.

BACKGROUND: Coronary artery origin anomalies, though often incidentally detected, can lead to sudden death. Comprehensive perioperative management is essential. We report a case of an anomalous right coronary artery (RCA) arising from the left main coronary artery (LMCA) and coursing between the aorta and pulmonary artery, discovered after myocardial infarction, in which intraoperative management ensured successful coronary reconstruction. CASE PRESENTATION: A 49-year-old woman presented with chest pain and ST segment elevation. Coronary angiography revealed an anomalous RCA demonstrating com

Evolutionary Patterns of Collagen Fiber Arrangement and Calcification in Atherosclerosis.

Collagen is a major structural component of human tissues, and its spatial arrangement is critical for maintaining tissue mechanical integrity and regulating cell behavior. In atherosclerosis (AS), collagen fiber arrangement has been implicated in plaque stability and the regulation of vascular calcification, yet its evolution during disease progression and association with other pathological processes remain poorly understood. In this study, male ApoE-/- mice were fed a high-fat diet to establish a model of AS. Histological staining was performed every 5 weeks to analyze the collagen fiber al

Glutamine Protects against Mouse Abdominal Aortic Aneurysm through Modulating VSMC Apoptosis and M1 Macrophage Activation.

Glutamine (Gln), known as the most abundant free amino acid, is widely spread in human body. In this study, we demonstrated the protective effects of glutamine against mouse abdominal aortic aneurysm (AAA) induced by both angiotensin II (AngII) and calcium phosphate (Ca3(PO4)2) in vivo, which was characterized with lower incidence of mouse AAA. Moreover, histomorphological staining visually presented more intact elastic fiber and less collagen deposition in abdominal aortas of mice treated by glutamine. Further, we found glutamine inhibited the excessive production of reactive oxide species (R

Aging aggravates aortic aneurysm and dissection via miR-1204-MYLK signaling axis in mice.

The mechanism by which aging induces aortic aneurysm and dissection (AAD) remains unclear. A total of 430 participants were recruited for the screening of differentially expressed plasma microRNAs (miRNAs). We found that miR-1204 is significantly increased in both the plasma and aorta of elder patients with AAD and is positively correlated with age. Cell senescence induces the expression of miR-1204 through p53 interaction with plasmacytoma variant translocation 1, and miR-1204 induces vascular smooth muscle cell (VSMC) senescence to form a positive feedback loop. Furthermore, miR-1204 aggrava

Strategies for arterial graft optimization at the single-cell level.

Common arterial grafts used in coronary artery bypass grafting include internal thoracic artery (ITA), radial artery (RA) and right gastroepiploic artery (RGA) grafts; of these, the ITA has the best clinical outcome. Here, by analyzing the single-cell transcriptome of different arterial grafts, we suggest optimization strategies for the RA and RGA based on the ITA as a reference. Compared with the ITA, the RA had more lipid-handling-related CD36+ endothelial cells. Vascular smooth muscle cells from the RGA were more susceptible to spasm, followed by those from the RA; comparison with the ITA s

The Cell Permeant Phosphopetpide mimetic of VASP Alleviates Motor Function Deficits After Experimental Subarachnoid Hemorrhage.

Subarachnoid hemorrhage (SAH) due to the rupture of an intracranial aneurysm leads to delayed vasospasm and neuroischemia, which can result in profound neurologic deficit and death. Therapeutic options after SAH are currently limited to hemodynamic optimization and nimodipine, which have limited clinical efficacy. Experimental SAH results in cerebral vasospasm have demonstrated the downregulation of nitric oxide (NO)-protein kinase G (PKG) signaling elements. VP3 is a novel cell permeant phosphopeptide mimetic of VASP, a substrate of PKG and an actin-associated protein that modulates vasorelax

Advances and challenges in regenerative therapies for abdominal aortic aneurysm.

Abdominal aortic aneurysm (AAA) is a significant source of mortality worldwide and carries a mortality of greater than 80% after rupture. Despite extensive efforts to develop pharmacological treatments, there is currently no effective agent to prevent aneurysm growth and rupture. Current treatment paradigms only rely on the identification and surveillance of small aneurysms, prior to ultimate open surgical or endovascular repair. Recently, regenerative therapies have emerged as promising avenues to address the degenerative changes observed in AAA. This review briefly outlines current clinical
