The use of electronic-neutral penetrating peptides cyclosporin A to deliver pro-apoptotic peptide: A possibly better choice than positively charged TAT.

Cell-penetrating peptides (CPPs) are increasingly important in transporting macromolecules across cell membranes, but their use remains confined to narrow clinical applications due to the systemic toxicity induced by their positive charges. Several newly discovered electronic neutral penetrating peptides are not attracting much attention because their penetrating capacity is normally far less powerful than cationic or amphiphilic CPPs. In this study, we found the electronic neutral cyclic peptide cyclosporin A (CsA) exhibited 5.6-fold and 19.1-fold stronger penetrating capacity, respectively,

PVN infusion of GLP-1-(7-36) amide suppresses feeding but does not induce aversion or alter locomotion in rats.

Intracerebroventricular infusion of glucagon-like peptide-1-(7-36) amide (GLP-1) reduces feeding in rats, an effect that could be localized to the hypothalamic paraventricular nucleus (PVN). Intracerebroventricular GLP-1, however, may also induce conditioned taste aversion (CTA), thereby putting into question the specificity of the action of GLP-1 on feeding. The present experiments evaluated the action of PVN GLP-1 (0, 100, or 200 ng) on induction of CTA, on locomotion, and finally, on feeding and drinking in rats. PVN infusion of GLP-1 (100 or 200 ng) did not support the induction of CTA and

Modulation of peripheral inflammation by locally administered hemorphin-7.

OBJECTIVE: Sensory nerves mediate peripheral inflammation via the release of sensory peptides at the site of tissue injury. Using a blister model of inflammation, we have previously documented that endogenous opioids modulate chronic but not acute inflammation. Hemorphins are nonclassical opioid peptides found in the region of the beta-chain of hemoglobin (Hb). The heptapeptide hemorphin-7 is identical with residues 35-41 of the beta-chain of the human Hb. The aim of this study was to examine the effect of hemorphin-7 on the inflammatory response in acute and chronic injury models. METHODS: W

Construction of Naïve and Immune Human Fab Phage Display Library.

Phage display is a widely used approach for the rapid isolation of monoclonal antibodies against a wide range of target antigens. The availability of a diverse antibody library is the essential component of any phage display campaign. The generation of an antibody library is coupled with an iterative process termed as biopanning that allows the identification of monoclonal antibodies. The antibody library consists of the antibody repertoire obtained either from human peripheral blood mononuclear cell (PBMC) or directly from sorted B-cell populations. Depending on the classification of the anti

Identification of an Angiotensin-(1-7)-Producing fusion protein in the brain of transgenic rats Reveals a hypotensive effect mediated through modulation of the GABA-nNOS-NO pathway and highlighting Astrocyte-Neuron crosstalk.

This study explores the anatomical distribution of Angiotensin-(Ang)-(1-7) fusion protein within the central nervous system of the novel transgenic rat model (TG7371). The Ang-(1-7)/Mas pathway of the renin-angiotensin system (RAS) plays a key role in cardiovascular regulation and influences higher brain functions, including cognition and emotion. TG7371 expresses a transgenic Ang-(1-7)-producing fusion protein which resulted in a hypotensive phenotype. Here, we assessed the expression of Ang-(1-7) fusion mRNA and protein in primary cortical cells from neonates and identified their distributio

The brain angiotensin converting enzyme 2 and angiotensin-(1-7) as potential therapeutic targets for the central nervous system diseases.

The renin-angiotensin system (RAS) serves as a key regulator of body fluid balance and cardiovascular homeostasis, whose dysregulation is involved in the pathogenesis of various central nervous system (CNS) diseases. Beyond their canonical functions such as vascular regulation, the constituents of the RAS, particularly angiotensin (Ang) II, exert inflammatory and oxidative injury on neurons in the brain. Meanwhile, the non-classical pathway angiotensin-converting enzyme (ACE)2/Ang-(1-7) axis shows beneficial effects against CNS diseases. Activation of cerebral ACE2/Ang-(1-7) exerts regulatory

Plasma levels of an N-terminal tau fragment predict Alzheimer's and neurodegenerative disease biomarkers in autosomal dominant Alzheimer's disease.

INTRODUCTION: Tau species lacking truncation of the N-terminal region, including plasma N-terminal tau fragment 1 (NT1), have been previously associated with cognitive decline, neurodegeneration, and tau pathology in late-onset sporadic Alzheimer's disease (AD). METHODS: Here, we examined cross-sectional and longitudinal plasma NT1 as a possible predictor of cognitive, clinical, and core AD biomarker trajectories in autosomal dominant AD (ADAD). RESULTS: NT1 levels in ADAD mutation carriers (MC; n = 132) increased across the disease continuum, compared to non-carriers (NC; n = 75), becoming

Rabies Virus Glycoprotein-Decorated Liposomes in Thermosensitive Nasal Spray Gels: Facilitating Retrograde Neural Transport for Targeted Trigeminal Neuralgia Therapy.

Clinical management of trigeminal neuralgia (TN) is hindered by poor neural bioavailability and systemic toxicity of oral drugs. While the nasal route offers a direct pathway to target the trigeminal nerve, rapid mucociliary clearance and competition from systemic absorption limit its effectiveness. To address these limitations, this study aimed to develop a biomimetic nasal gel system for targeted drug delivery to the trigeminal nerve. Inspired by the neurotropism of rabies virus, we engineered a thermoresponsive nasal spray gel (OMRLP@NSG). The system utilizes rabies virus glycoprotein (RVG)

Effect of bovine beta-casomorphins on rat pancreatic beta cells (RIN-5F) under glucotoxic stress.

Beta-casomorphins (BCMs) are the bio-active peptides having opioid properties which are formed by the proteolytic digestion of β-caseins in milk. BCM-7 forms when A1 milk is digested in the small intestine due to a histidine at the 67th position in β-casein, unlike A2 milk, which has proline at this position and produces BCM-9. BCM-7 has further degraded into BCM-5 by the dipeptidyl peptidase-IV (DPP-IV) enzyme in the intestine. The opioid-like activity of BCM-7 is responsible for eliciting signaling pathways which enable a wide range of physiological effects. The aim of our study was to find

A single dose of angiotensin-(1-7) resolves eosinophilic inflammation and protects the lungs from a secondary inflammatory challenge.

OBJECTIVE: Angiotensin-(1-7) [Ang-(1-7)] is a pro-resolving mediator. It is not known whether the pro-resolving effects of Ang-(1-7) are sustained and protect the lung from a subsequent inflammatory challenge. This study sought to investigate the impact of treatment in face of a second allergic or lipopolysaccharide (LPS) challenge. METHODS: Mice, sensitized and challenged with ovalbumin (OVA), received a single Ang-(1-7) dose at the peak of eosinophilic inflammation, 24 h after the final OVA challenge. Subsequently, mice were euthanized at 48, 72, 96, and 120 h following the OVA challenge, a
