Differential inhibition of Morbillivirus and Henipavirus polymerases by ERDRP-0519 and structure-guided inhibitor optimization.

ERDRP-0519 is a non-nucleoside polymerase inhibitor developed against measles virus (MeV) of the Morbillivirus genus. Here, we show that ERDRP-0519 also cross-inhibits Nipah virus (NiV) of the Henipavirus genus with reduced potency. ERDRP-0519 binds to a shared pocket within the RNA-dependent RNA polymerase (RdRp) palm domains of MeV, peste des petits ruminants virus (PPRV), and NiV polymerases. ERDRP-0519 forms more extensive interactions with Morbillivirus polymerases, whereas binding to NiV polymerase requires substantial RdRp motif rearrangements, likely incurring an energetic cost and res…

Development and validation of P113 gene specific polymerase spiral reaction assay for rapid detection of Mycoplasma ovipneumoniae in sheep and goats.

Respiratory infections caused by Mycoplasma ovipneumoniae remain a major challenge in sheep and goat production, resulting in reduced productivity and financial losses. Conventional diagnostic tools such as culture, PCR and ELISA are either expensive, labor intensive or unsuitable for on-site application in rural farming systems. At present, India lacks a rapid and affordable test for field detection of this pathogen. The objective of the present study is to develop Polymerase Spiral Reaction (PSR) assay to detect Mycoplasma ovipneumoniae in small ruminants. PSR primers specific for Mycoplasma…

Mortality associated with Dictyocaulus cervi in farmed red deer (Cervus elaphus) in Romania.

Lungworms from the genus Dictyocaulus are known to cause catarrhal bronchitis and pulmonary atelectasis in domestic and wild ruminants. In red deer (Cervus elaphus), four Dictyocaulus species can be present in the respiratory tract. This study describes the investigation of mortalities in red deer (Cervus elaphus) in a commercial farm for red deer from Romania, associated with the presence of lungworms. Postmortem examination revealed the presence of nematodes in the bronchial lumen in all cases. Morphological and molecular examination of the nematodes recovered confirmed Dictyocaulus cervi as…

Field Efficacy of Metarhizium robertsii (LCM S15) for Controlling Free-Living Stages of Gastrointestinal Nematodes in Goats.

The rise in anthelmintic resistance in small ruminants has driven the search for sustainable control alternatives. Among these, entomopathogenic fungi such as Metarhizium robertsii stand out for their potential to reduce the free-living stages of gastrointestinal nematodes (GINs). This study evaluated the field efficacy of M. robertsii (LCM S15) under climatic conditions in the Recôncavo region of Bahia, Brazil. The experiment was conducted between November 2022 and July 2023 in a completely randomized design with four groups (n = 8): aqueous control, oil control, aqueous suspension, and oil f…

First detection of Dicrocoelium dendriticum (Digenea: Dicrocoelidae) from cattle in the West Bank, Palestine.

Dicrocoelium dendriticum is a globally distributed trematode that infects the bile ducts of ruminants and causes dicrocoeliasis, leading to economic losses in livestock production. Despite favorable ecological conditions, its presence in cattle had not previously reported in Palestine. This short communication reports the first detection and characterization of D. dendriticum in cattle from the West Bank, Palestine. Adult flukes were recovered from bile ducts during routine post-mortem inspection at Al-Bireh slaughterhouse and identified based on morphological characteristics. Molecular confir…

Nationwide Distribution and Prevalence of Epizootic Haemorrhagic Disease Virus Infection in Portugal (2023-2025).

Epizootic haemorrhagic disease virus (EHDV) is an insect-borne virus that affects ruminants. It has recently emerged as a significant threat to livestock and wildlife, particularly deer and cattle. Since its introduction into Europe in late 2022, severe outbreaks have been reported across several European countries, including Italy, Spain, France, and Portugal. The first confirmed circulation of EHDV in Portugal was reported in July 2023, involving serotype 8 (EHDV-8) in cattle from the municipalities of Moura and Barrancos, in the Beja district of the Alentejo region, alongside concurrent cir…

Eastern equine encephalitis virus infection in an American pygmy goat.

Eastern equine encephalitis virus (EEEV; family Togaviridae, taxon species Alphavirus eastern) is a mosquito-borne alphavirus that is endemic to the Western Hemisphere and well established in freshwater hardwood swamps. EEEV is environmentally maintained through a mosquito-and-avian cycle, primarily by Culiseta melanura mosquitoes. Transmission to mammals, which are considered dead-end hosts, is well known and seasonal through C. melanura or other bridging mosquito vectors. EEE is well described in horses and humans, with relatively few reports in ruminants. Information regarding infection and…

An Early Treatment With BKI-1748 Exhibits Full Protection Against Abortion and Congenital Infection in Sheep Experimentally Infected With Toxoplasma gondii.

Congenital toxoplasmosis in humans and in other mammalian species, such as small ruminants, is a well-known cause of abortion and fetal malformations. The calcium-dependent protein kinase 1 (CDPK1) inhibitor BKI-1748 has shown a promising safety profile for its use in humans and a good efficacy against Toxoplasma gondii infection in vitro and in mouse models. Ten doses of BKI-1748 given every other day orally in sheep at 15 mg/kg did not show systemic or pregnancy-related toxicity. In sheep experimentally infected at 90 days of pregnancy with 1000 TgShSp1 oocysts, the BKI-1748 treatment admini…

Description of lesions in lambs intoxicated with sodium selenite included in intraruminal boluses: Lambs intoxicated with selenium.

Selenium is an essential micronutrient for ruminants, which participates in the optimal functioning of proteins and enzymes that can combat oxidative stress in the body; however, its toxicity is documented in different species. The objective of this work was to describe macroscopic and microscopic lesions in lambs intoxicated with selenium administered through intraruminal boluses. The main lesions at necropsy were pulmonary oedema; the myocardial surface presented multifocal pale areas; the thyroid and thymus glands were decreased in size, and areas of necrosis, haemorrhage and hyperkeratosis…

An LIR motif in the Rift Valley fever virus NSs protein is critical for the interaction with LC3 family members and inhibition of autophagy.

Rift Valley fever virus (RVFV) is a viral zoonosis that causes severe disease in ruminants and humans. The nonstructural small (NSs) protein is the primary virulence factor of RVFV that suppresses the host's antiviral innate immune response. Bioinformatic analysis and AlphaFold structural modeling identified four putative LC3-interacting regions (LIR) motifs (NSs 1-4) in the RVFV NSs protein, which suggest that NSs interacts with the host LC3-family proteins. Using, isothermal titration calorimetry, X-ray crystallography, co-immunoprecipitation, and co-localization experiments, the C-terminal …

Iatrogenic doramectin overdosing causes toxicity in sheep: A case report.

Macrocyclic lactones are widely used endectocides in ruminants, with a high margin of safety for labeled indications. No previous report of iatrogenic doramectin overdosing has been published. We report an outbreak in a sheep flock in Northeast Patagonia, Argentina. Toxicity signs were observed in more than 10% and 59% of ewes and lambs, respectively, particularly those with low body condition, treated with doramectin 3.5% long-acting injectable formulation, presumably at the indicated dose of 700 μg/kg. Clinical signs included lethargy, mydriasis and coma. Doramectin concentration in blood sa…

Bluetongue and epizootic haemorrhagic disease in wildlife with emphasis on the South American scenario.

Bluetongue and epizootic hemorrhagic disease affect domestic ruminants and cervids. However, other species may act as pathogen carriers in the transition of bluetongue virus (BTV) and epizootic hemorrhagic disease virus (EHDV). The wild species affected by these diseases manifest a variable range of clinical signs and lesions, and while some species appear to be extremely susceptible, showing high levels of mortality, some are resistant to these pathogens, acting as potential reservoirs of these orbiviruses.  The purpose of the following review is to describe the clinical and pathological mani…
