Binge ethanol administration enhances the MDMA-induced long-term 5-HT neurotoxicity in rat brain.

RATIONALE: Ecstasy abuse commonly occurs in hot, overcrowded environments in combination with alcohol. Around 90% of ecstasy users take ethanol; over 70% of these users also often drink alcohol at hazardous levels. OBJECTIVES: We wished to examine whether binge ethanol administration enhanced the long-lasting 5-HT neurotoxicity induced by 3,4-methylenedioxymethamphetamine (MDMA) in rats maintained at high ambient temperature and the role of acetaldehyde. MATERIALS AND METHODS: Rats were treated with a 4-day ethanol regimen leading to plasma ethanol levels of around 450 mg/dl. On day 5, rats

Anxiolytic-like effect of milnacipran in the four-plate test in mice: mechanism of action.

Milnacipran is a serotonin/noradrenaline reuptake inhibitor (SNRI) which has not yet been systematically studied preclinically or clinically for the treatment of anxiety disorders. In the four-plate test (FPT) which is known to predict anxiolytic-like activity in mice, milnacipran (4, 8, 16 and 32 mg/kg) demonstrated strong anti-punishment effects following acute administration. The anxiolytic-like effect of milnacipran was not reversed by the selective GABA(A) receptor antagonist, flumazenil (2 and 4 mg/kg), the selective alpha1-adrenoceptor antagonist, prazosin (0.5 and 2 mg/kg), the selecti…

Effect of ondansetron on the contractile responses to positive inotropic agents in electrically driven left atria of rats.

We studied the interaction of ondansetron with positive inotropic agents, including serotonin (5-hydroxytryptophane, 5-HT), noradrenaline, 4-aminopyridine (4-AP), calcium chloride, or with reserpine in isolated electrically driven rat atria. Concentrations of 5-HT ranging from 1 to 64 microg/ml increased atrial contractions in a dose-dependent manner. The inotropic effect of 5-HT in the right atria appeared to be weaker than that in the left atria. Ondansetron (30 microg/ml) depressed the positive inotropic effect of lower 5-HT concentrations . This effect was thought to be due to the local an…

[Tryptophan metabolism: a key player in cardiometabolic diseases].

Tryptophan (Trp), an essential amino acid, plays a central role in metabolism, inflammation, immunity, and energy homeostasis. It is primarily metabolized through the kynurenine, indole, and serotonin pathways, generating metabolites with either protective or harmful effects depending on the physiological and tissue context. Dysregulation of these pathways contributes to obesity, diabetes, and cardiovascular diseases. Understanding these mechanisms paves the way for personalized nutritional and therapeutic strategies to prevent and manage cardiometabolic diseases.

Secondary tumoural valvulopathy in a dog.

This short report describes a case of tricuspid valvular metastasis of canine disseminated histiocytic sarcoma in a 9-year-old female Rottweiler. Immunohistochemically the malignant neoplastic cells gave a strong reaction for vimentin and lysozyme, and showed negativity for serotonin, CD3, CD79a and cytokeratin. The intratumoural microvessels were detected by immunohistochemistry using CD31 and claudin-5. This appears to be the first report of a valvular metastasis of canine malignant histiocytosis.

[Reboxetine in the treatment of bulimia nervosa].

There is a substancial body of literature demonstrating the efficacy of selective serotonin reuptake inhibitors antidepressants (SSRI) in reducing binge eating and vomiting frequency in Bulimia Nervosa. Good therapeutic response to noradrenergic agents, like desipramine and reboxetine, though not frequently reported in literature, has already been demonstrated. This case report describes the treatment of Bulimia Nervosa with reboxetine (4 to 8 mg/day) and its favorable therapeutic results.

Omental transplantation for Alzheimer's disease.

The acetylcholine, dopamine, serotonin and other neurotransmitters may be reduced in the subcommissural regions even in the early stages of Alzheimer's disease(AD), due to hypoperfusion of the anterior perforating and anterior choroidal arteries. This hypothesis was confirmed after the transplant of omental tissue on the optic chiasma, carotid crotch and anterior perforated space in a woman with moderate AD. Neurological improvement was better in the first week after the surgery than in the following months.

Catastrophic consequences secondary to psychotropic drugs, Part 1.

The use of psychotropic drugs has dramatically changed the lives of many; yet as often occurs with technological advances, negative outcomes can be encountered. Potentially fatal reactions to neuroleptic drugs and serotonin-enhancing agents (e.g., antidepressants) have been presented in Part 1, with the intent of alerting and informing psychiatric nurses. Part 2 will complete this series on catastrophic consequences of psychotropic drugs with a discussion of agranulocytosis and lithium toxicity.

Plasma biogenic amine levels in agoraphobia with panic attacks.

Eight patients with agoraphobia and panic attacks as defined by DSMIII and 11 controls were included in this study which measured the circulating biogenic amine levels. The method of assay used a high performance liquid chromatograph with electrochemical detection. Measurement of dopamine, noradrenaline and adrenaline showed no differences between the groups. Levels of serotonin (5-HT) were significantly lower in the agoraphobic group. The authors discuss the implication of this finding.

Oculocutaneous albinism, platelet storage pool disease, and progressive lupus nephritis.

Systemic lupus erythematosus (SLE) was seen in a patient with oculocutaneous albinism and platelet storage pool disease (Hermansky Pudlak syndrome). Although the patient had severely compromised platelet function, lupus nephritis developed nonetheless, which progressed to end-stage renal disease. The role of platelets in the pathogenesis of the nephritic process is discussed, and it is concluded that release of platelet granular constituents, particularly serotonin, is not required for the microvascular damage of SLE.

Further observations on the use of 5-hydroxytryptophan in a child with Lesch-Nyhan syndrome.

The effects of 5-hydroxytryptophan (5-HTP), a serotonin precursor, and carbidopa, a peripheral aromatic amino acid decarboxylase inhibitor, were studied in a boy with the Lesch-Nyhan syndrome. No effects on the self-mutilating behaviors of this disorder were noted. However, sleep patterns were normalized, with a reduction in the proportionate amounts of wakefulness and an augmentation of active sleep. A sharp reduction in waking behavioral dyskinesias was also correlated to increased drug dosage.

Radiology of carcinoid tumours of the small intestine.

Carcinoid tumors of the small intestine are particularly known in the literature for their hormonal activity and for the carcinoid syndrome caused by the serotonin they produce. The radiological symptoms are based, not only on changes caused by the tumour itself but also on changes induced by hormonal products in the intestinal wall and mesentery. The classical excentric intramural defect in the contrast column is therefore often accompanied by swollen mucosal folds, swelling of the intestinal wall and evidence of mesenteric retraction.

Translation from mice to monkeys toward human: species differences in endothelial-dependent vasorelaxation in various isolated vascular beds.

Compounds inducing effects on the blood pressure may act on arteries. The endothelium is frequently involved in these vascular effects, and … onkeys and human were evaluated using isolated organ bath or wire myography. Contraction induced by serotonin have been also investigated in various human vessels. Mice aorta, but not mesenteric arteries, was more sensitive to acetylcholine compared to rat arteries. Monkey abdominal aorta displayed a weak relaxation, whereas a potent relaxation was observed …

Tauroursodeoxycholic acid alleviates depression-like behavior induced by chronic unpredictable mild stress through amelioration of catecholamine imbalance.

Depression is thought to emerge as a result of monoamine neuromodulators' deficiency in a specific central nervous system site. Tauroursodeoxycholic acid (TUDCA) has been found to have a protective role against diseases affecting the central nervous system. The potential effects of TUDCA on brain monoamine neurotransmitters in a stress-induced depression model have not been reported. We investigated the effects of TUCDA treatment on serotonin, norepinephrine and dopamine, catecholamine biosynthesis enzyme tyrosine hydroxylase (TH) and degrading enzyme monoamine oxidase A (MAO-A), NLRP3 and pro…

[LSD revisited: Mechanisms of action and therapeutic potential in mental health].

Major depressive disorder (MDD) represents a colossal worldwide burden of health, affecting more than 350 million individuals. While traditional antidepressants with mechanisms of action at the serotonin, dopamine, or norepinephrine pathways are routinely prescribed, approximately one-third of patients fail to achieve adequate remission and develop treatment-resistant depression (TRD). This therapeutic shortfall underscores the necessity for novel approaches to treatment. Over the past two decades there has been a resurgence of scientific interest in psychedelics, such as lysergic acid diethyl…

The protective effect of DMT against neurodegeneration.

This paper explores the therapeutic potential of DMT in neuroprotective strategies, particularly concerning ischemia-reperfusion injury (IRI) and neurodegenerative disorders. Besides its potent serotonin receptor actions, DMT is also an endogenous agonist of the sigma-1 receptor (Sig-1R). Sigma receptors are a unique family of proteins with high expression in the brain and spinal cord and have been involved in the etiology, symptom course and treatment of several central nervous system disorders. Our previous theoretical and experimental work strongly suggest that targeting sigma (and serotoni…
