Regulator of G protein signaling 3 protects against cardiac hypertrophy in mice.

Regulator of G protein signaling 3 (RGS3) is a negative regulator of G protein-mediated signaling. RGS3 has previously been shown to be expressed among various cell types within the mature heart. Basic and clinical studies have reported abnormal expressions of RGS3 in hypertrophic hearts and in the failing myocardium. However, the role of RGS3 in cardiac remodeling remains unclear. In this study, we investigated the effect of cardiac overexpression of human RGS3 on cardiac hypertrophy induced by aortic banding (AB) in RGS3 transgenic mice and wild-type littermates. The extent of cardiac hypert

The primary microRNA-208b interacts with Polycomb-group protein, Ezh2, to regulate gene expression in the heart.

The Polycomb-group protein, Ezh2, is required for epigenetic gene silencing in the adult heart by unknown mechanism. We investigated the role of Ezh2 and non-coding RNAs in a mouse model of pressure overload using transverse aortic constriction (TAC) attenuated by the prototypical histone deacetylase inhibitor, trichostatin A (TSA). Chromatin immunoprecipitation of TAC and TAC+TSA hearts suggests interaction of Ezh2 and primary microRNA-208b (pri-miR-208b) in the regulation of hypertrophic gene expression. RNAi silencing of pri-miR-208b and Ezh2 validate pri-miR-208b-mediated transcriptional s

Involvement of BMPR2 in the protective effect of fluoxetine against monocrotaline-induced endothelial apoptosis in rats.

Mutations in bone morphogenetic protein (BMP) receptor II (BMPR2) are associated with the apoptosis of the pulmonary artery endothelial cells and the loss of the pulmonary small vessels. The present study was designed to investigate the involvement of BMPR2 in the protective effect of fluoxetine against monocrotaline (MCT)-induced endothelial apoptosis in rats. Models of pulmonary arterial hypertension in rats were established by a single intraperitoneal injection of MCT (60 mg/kg). Fluoxetine (2 and 10 mg/kg) was intragastrically administered once a day. After 21 days, MCT caused pulmonary hy

[Acute type A aortic dissection with partial disruption of the right coronary ostium; report of a case].

A 64-year-old man with a history of hypertension was admitted to another hospital, presenting with sudden chest pain. Although his symptoms resolved after admission, chest computed tomography (CT) revealed acute type A aortic dissection, and he was referred to our hospital for surgery. On arrival, his hemodynamic status was stable. Electrogram showed no ST-T change, and transthoracic echocardiography demonstrated pericardial effusion, mild aortic regurgitation and good left ventricular wall motion. We immediately decided on an emergency operation. Commencing a median sternotomy, the pericardia

Congenital semilunar valvulogenesis defect in mice deficient in phospholipase C epsilon.

Phospholipase Cepsilon is a novel class of phosphoinositide-specific phospholipase C, identified as a downstream effector of Ras and Rap small GTPases. We report here the first genetic analysis of its physiological function with mice whose phospholipase Cepsilon is catalytically inactivated by gene targeting. The hearts of mice homozygous for the targeted allele develop congenital malformations of both the aortic and pulmonary valves, which cause a moderate to severe degree of regurgitation with mild stenosis and result in ventricular dilation. The malformation involves marked thickening of th

Assessment of left atrial-left ventricular-arterial coupling in amateur marathon runners using three-dimensional speckle-tracking echocardiography.

BACKGROUND: Prolonged high-intensity endurance exercise has been associated with myocardial structural remodeling. This study aimed to investigate left atrial-left ventricular-arterial (LA-LV-arterial) coupling in amateur marathon runners using three-dimensional speckle-tracking echocardiography (3D-STE). By comprehensively assessing cardiac structural adaptation and arterial stiffness, this study sought to elucidate the integrated heart-vascular interaction, which may serve as a predictor for cardiovascular risk. METHODS: A total of 93 amateur marathon runners were enrolled and categorized i

Oral treatment with a zinc complex of acetylsalicylic acid prevents diabetic cardiomyopathy in a rat model of type-2 diabetes: activation of the Akt pathway.

BACKGROUND: Type-2 diabetics have an increased risk of cardiomyopathy, and heart failure is a major cause of death among these patients. Growing evidence indicates that proinflammatory cytokines may induce the development of insulin resistance, and that anti-inflammatory medications may reverse this process. We investigated the effects of the oral administration of zinc and acetylsalicylic acid, in the form of bis(aspirinato)zinc(II)-complex Zn(ASA)2, on different aspects of cardiac damage in Zucker diabetic fatty (ZDF) rats, an experimental model of type-2 diabetic cardiomyopathy. The cardiovascular risk of metabolically healthy obesity: an inflammatory perspective.

Metabolically healthy obesity (MHO) is a heterogeneous phenotype characterized by preserved traditional metabolic markers despite excess adiposity. However, accumulating evidence suggests that MHO may not be uniformly benign and may confer increased long-term cardiovascular risk through inflammatory and adipose tissue-mediated mechanisms. This narrative review synthesizes contemporary evidence on cardiovascular risk in MHO, focusing on visceral adiposity, ectopic fat, and immune metabolic pathways. Literature published between January 2010 and December 2025 was identified through PubMed/MEDLIN

When Human Tissue Meets the Driveline: Implications of Tissue Integration to Combat Driveline Infections.

Driveline infection remains a common complication of ventricular assist device (VAD) therapy. These infections often progress from the exit site through the subcutaneous tract to the deeper tunnel and, in severe cases, the pocket, underscoring the vulnerability of the driveline-tissue interface. In this review, we examine the biological and mechanical factors governing tissue-driveline interactions and their implications for tissue integration of the implanted driveline. Dysregulated inflammation, impaired vascular maturation, abnormal extracellular matrix remodeling, repetitive micromotion an

Magneto-controllable cell robots for targeted myocardial infarction repair.

Myocardial infarction (MI) triggers irreversible cardiomyocyte loss, maladaptive remodeling, and heart failure processes exacerbated by insufficient endogenous angiogenesis and unresolved inflammation. While pro-angiogenic therapies hold promise, their clinical translation is hindered by poor lesion targeting, subtherapeutic drug accumulation, and transient bioactivity. To overcome these barriers, we engineered "magnetically guided cellular robots", M2a-polarized macrophages intracellularly loaded with superparamagnetic Fe3O4 nanoparticles and genetically programmed for sustained VEGF secretio

Diabetic Atrial Cardiomyopathy: Pathogenesis, Diagnosis, Management, AI-Driven Diagnosis, and Risk Prediction.

BACKGROUND: Diabetic atrial cardiomyopathy, a component of diabetic cardiomyopathy, is increasingly recognized. However, atrial-focused mechanistic and clinical frameworks remain less developed than ventricular paradigms. MAIN BODY: This review synthesizes the pathogenesis of diabetic atrial cardiomyopathy from conventional, multiomics, and translational perspectives. We highlight how diabetes-related metabolic stress, inflammation, gut microbiota dysregulation, electrophysiological remodeling, genetic susceptibility, and epigenetic regulation converge on atrial fibrosis, conduction heterogen

Myopericarditis After Combined Idarubicin and Quizartinib in FLT3-ITD-Positive AML.

BACKGROUND: Anthracyclines are key components of induction therapy for acute myelogenous leukemia with well-understood cardiotoxicity. Quizartinib inhibits FLT3 and has been shown to potentiate apoptosis and promote maladaptive remodeling after myocardial infarction in mice; however, the impact of FLT3 inhibition on a heart already exposed to anthracyclines is poorly understood. CASE SUMMARY: A 64-year-old man with acute myelogenous leukemia developed fever, hypotension, troponin I elevation, and diffuse ST-segment elevation after treatment with cytarabine and idarubicin followed by quizartin

Adropin protects against cardiac remodeling and metabolic dysfunction in a male mouse HFpEF model.

Cardiometabolic heart failure with preserved ejection fraction (HFpEF) is a heterogenous metabolic disease, which in the heart presents as left ventricle diastolic dysfunction, ventricular stiffness, and myocardial structural remodeling. Deleterious changes in cardiac metabolism are central to HFpEF pathophysiology, and proposed treatments for the disease have focused on repairing these defects. In the present study, we used a preclinical mouse model that recapitulates cardiometabolic HFpEF to elucidate the molecular mechanisms driving cardiac dysfunction and tested whether recombinant adropin

Late-systolic to early-diastolic hemodynamic force analysis in ischemic heart disease using vector flow mapping and invasive pressure measurement.

Hemodynamic force (HDF) analysis using vector flow mapping (VFM) enables noninvasive assessment of intraventricular pressure differences (IVPDs). In this study, we evaluated the apical suction from late systole to early diastole in patients with ischemic heart disease (IHD) with or without left ventricular aneurysm (LVA), validated against simultaneous left atrial-LV pressure measurements. A total of 37 participants were evaluated: 11 healthy controls, 11 patients with IHD without LVA, and 15 with LVA. VFM-derived IVPDs were quantified as IVPD recoil (late systole) or IVPD RF (early diastole).

Role of ALDH2 in myocardial ischemia-reperfusion injury: using ALDH2 as a potential therapeutic target.

Myocardial ischemia-reperfusion (I/R) injury remains a major barrier to the full benefit of timely coronary reperfusion. This review summari … that ALDH2 deficiency aggravates infarct size, inflammatory injury, aldehyde overload, and adverse ventricular remodeling, whereas ALDH2 activation or overexpression confers protection. Clinical and genetic studies further suggest that the ALDH2 rs671/ALDH2*2 loss-of-function variant may influence myocardial infarction risk, reperfusion injury severity, and the respons …

Tachycardia-induced cardiomyopathy: evolution of the concept and contemporary insights.

Tachycardia-induced cardiomyopathy (TIC) is a reversible form of myocardial dysfunction caused by sustained or recurrent tachyarrhythmias. Despite increasing recognition, its diagnosis and management remain challenging in clinical practice. This review summarizes the historical evolution, epidemiology, pathophysiology, diagnosis, and management of TIC, and highlights current gaps in knowledge. Experimental and clinical studies have established that not only persistent tachycardia but also high arrhythmic burden and ventricular dyssynchrony contribute to myocardial dysfunction, leading to the b

Experimental animal models and patient-derived platforms to bridge preclinical discovery and translational therapeutics in pulmonary arterial hypertension.

Pulmonary arterial hypertension (PAH) is a progressive vascular disease characterized by sustained elevation of pulmonary arterial pressure, endothelial cell dysfunction, and right ventricular failure. A wide range of experimental animal models, including the monocrotaline model, Sugen combined with hypoxia, and pulmonary artery banding in large animals, have been pivotal in uncovering disease mechanisms such as vascular remodeling, metabolic dysregulation, and hypoxia-inducible signaling. More recently, human-based platforms, including induced pluripotent stem cell-derived vascular cells, org
