Exposure of Mycobacterium tuberculosis to human alveolar lining fluid shows temporal and strain-specific adaptation to the lung environment.

UNLABELLED: Upon infection, Mycobacterium tuberculosis ( M.tb ) reaches the alveolar space and comes in close contact with human alveolar lining fluid (ALF) for an uncertain period of time prior to its encounter with alveolar cells. We showed that homeostatic ALF hydrolytic enzymes modify the M.tb cell envelope, driving M.tb -host cell interactions. Still, the contribution of ALF during M.tb infection is poorly understood. Here, we exposed 4 M.tb strains with different levels of virulence, transmissibility, and drug resistance (DR) to physiological concentrations of human ALF for 15-min and 12

Optimized nonlinear robust controller along with model-parameter estimation for blood glucose regulation in type-1 diabetes.

Glucose acts as a fundamental energy source for cells and plays a pivotal role in various physiological processes, including metabolism, signaling, and cellular control. Maintaining precise regulation of blood glucose levels is crucial for overall health and equilibrium. To achieve this balance, insulin is administered either orally or through an artificial pancreas (AP) during sleep, utilizing control algorithms based on mathematical models to regulate blood glucose concentration (BGC). The extended Bergman minimal model (EBMM) is an advanced mathematical framework that incorporates a state v

Measuring Rod- and Cone-Photoreceptor-Specific Vision in Inherited Retinal Diseases Using a Commercial Perimeter.

PURPOSE: The primary pathology in most inherited retinal diseases (IRDs) is located within photoreceptors. Standard automatic perimetry (SAP) can measure photoreceptor disease severity but cannot distinguish between rods, long/middle-wavelength (L/M)-sensitive, and short-wavelength (S)-sensitive cones. Herein we developed a protocol that can provide photoreceptor-specific sensitivities. METHODS: A commercial (unmodified) perimeter was used to develop a clinical protocol that includes five profiles along the vertical meridian, utilizing different chromatic stimuli presented in the dark-adapted

Inducible expression, enzymatic activity, and origin of higher plant homologues of bacterial RelA/SpoT stress proteins in Nicotiana tabacum.

All living cells possess adaptive responses to environmental stress that are essential to ensuring cell survival. For motile organisms, this can culminate in avoidance or attractile behavior, but for sessile organisms such as plants, stress adaptation is a process of success or failure within the confines of a given environment. Nearly all bacterial species possess a highly evolved system for stress adaptation, known as the "stringent response." This ancient and ubiquitous regulatory response is mediated by production of a second messenger of general stress, the nucleotide guanosine-3',5'-(bis

Sex specific associations between common glucocorticoid receptor gene variants and hypothalamus-pituitary-adrenal axis responses to psychosocial stress.

BACKGROUND: Alterations in glucocorticoid (GC) signaling have been associated with a number of psychiatric disorders. Genetic variation of the glucocorticoid receptor (GR) might be one of the factors underlying susceptibility to stress related disease. METHODS: We investigated 206 healthy subjects and assessed associations between four common GR gene (NR3C1) polymorphisms (ER22/23EK, N363S, BclI, 9beta) and hypothalamic-pituitary-adrenal (HPA) axis responses to psychosocial stress (Trier Social Stress Test, TSST) and glucocorticoid sensitivity measured by a dexamethasone suppression test (DST

Characterization of physical health variables across fall risk appraisal groups: a cross-sectional study.

BACKGROUND: Fall risk appraisal (FRA) integrates perceived fall risk (levels of fear of falling) and physiological fall risk (static balance). Established literature has defined cut-offs for four groups (congruent, incongruent, irrational, and rational), but characterization of these groups is limited to separate studies evaluating physical activity, body composition, and fall history. This study aims to characterize physical health measures in a large sample of older adults and evaluate differences between each maladaptive group (congruent, incongruent, irrational) and the adaptive (rational)

Physiological and transcriptomic analyses reveal the critical role of MsERF15 in negatively regulating cadmium stress tolerance in Alfalfa (Medicago sativa).

Cadmium (Cd) is a highly toxic heavy metal that poses a serious threat to plant growth and the quality of agricultural and animal products. Therefore, elucidating the response and regulatory mechanisms of plants to Cd stress is essential for controlling the safety of agricultural and animal products at the source. In this study, we used a Cd-tolerant alfalfa cultivar, Medicago sativa 'Zhaodong' (ZD), and a Cd-sensitive cultivar, M. sativa 'Zhongmu No.1' (ZM), to investigate their differential responses to Cd stress in terms of morphology, photosynthetic function, and oxidative damage through c

Integrated transcriptomic and biochemical analyses reveal mechanisms of drought adaptation in strawberry (Fragaria × ananassa L.).

Drought is a major abiotic stress that limits strawberry productivity worldwide, yet the underlying physiological and molecular mechanisms of cultivar-specific tolerance remains poorly understood. This study integrated physiological, biochemical, and transcriptomic analyses to dissect drought responses of two contrasting cultivars of Fragaria × ananassa: drought-tolerant 'Kara' and drought-sensitive 'E22'. Plants were exposed to both progressive water-deficit (Def) and PEG-induced osmotic stress under controlled greenhouse conditions. Physiologically, the two cultivars diverged substantially:

[Structured data collection during neonatal anesthesia: Neo Somno Log - a self-developed digital event tracker].

INTRODUCTION: Documentation of anesthetic care in Hungary remains heterogeneous, with the scope of recorded data largely determined by local institutional protocols. Anaesthesia records are predominantly paper-based, which, with variability in data content, hinders uniform, structured, and systematic data processing. Anesthesia in newborns is associated with increased risk due to physiological immaturity and ongoing postnatal adaptation, therefore the standardized and systematic capture of perioperative data is essential to improve quality of care and enhance patient safety. OBJECTIVE: This s

Temporal multi-omics analysis of HSF-HSP and flavonoid pathways under heat stress in Iris.

Heat stress poses a significant threat to the productivity of ornamental plants. Iris tectorum (Iris) is a highly valued ornamental species known for its diverse flower colors and architectural forms. However, heat‑induced physiological damage can substantially reduce its economic value. To investigate the temporal dynamics of the heat stress response in I. tectorum 'Hot Spicy', we conducted a time-series experiment at 40 °C for 0, 1, 3, 6, 9, and 24 h, integrating physiological measurements, transcriptomics, and metabolomics. Physiologically analyses showed that heat stress induced progressiv

Less water, same buds: moderate drought as a water-saving strategy for indoor cannabis.

Drought stress (DS) can restrict plant growth while potentially enhancing secondary metabolism through adaptive physiological responses. However, its effect on cannabinoid production remains uncertain, with previous studies reporting inconsistent findings. This study investigated how DS intensity and timing affect inflorescence biomass, cannabidiol (CBD) concentration, and CBD yield in two chemotype III (CBD-dominant; high-CBD, low-THC) medicinal cannabis genotypes differing in architecture and growth habit.: Two DS intensities were imposed during flowering relative to a well-watered control h

Changes in vascular identity during vascular remodeling.

BACKGROUND: Vascular remodeling is a dynamic process characterized by changes in vascular identity that impact vessel structure and function. Molecular markers define cellular identity as arteries, veins, and lymphatic vessels: Ephrin-B2 and Notch determine arterial identity, EphB4 and COUP-TFII determine venous identity, and Prox1 determines lymphatic identity. METHODS: This is a review of experimental literature. RESULTS: These proteins determine identity during development before the first heartbeat. Hemodynamic changes in adulthood can also alter vascular identity. Changes in identity ma

Cell non-autonomous signaling through the conserved C. elegans glycoprotein hormone receptor FSHR-1 regulates cholinergic neurotransmission.

Modulation of neurotransmission is key for organismal responses to varying physiological contexts such as during infection, injury, or other stresses, as well as in learning and memory and for sensory adaptation. Roles for cell autonomous neuromodulatory mechanisms in these processes have been well described. The importance of cell non-autonomous pathways for inter-tissue signaling, such as gut-to-brain or glia-to-neuron, has emerged more recently, but the cellular mechanisms mediating such regulation remain comparatively unexplored. Glycoproteins and their G protein-coupled receptors (GPCRs)
