Synergistic integration of forensic transcriptome and microbiome: A robust multi-marker strategy combined with machine learning for accurate body fluid identification.

In recent years, the development of microbiome and transcriptome analyses has significantly improved the efficiency of forensic body fluid identification. However, challenging or limited biological samples in forensic practice demand highly efficient utilization of biological samples to minimize sample loss. In this study, we developed two independent assays based on a capillary electrophoresis (CE) approach: a 21-mRNA assay and a 10-bacteria system. The co-extracted RNA and DNA were amplified independently to identify five body fluids using mRNA profiling, and to specifically identify saliva

IL-8 positive cancer-associated fibroblasts drive breast cancer progression and immune evasion: insights from GWAS and single-cell transcriptomics.

BACKGROUND: Interleukin-8 (IL-8) is a key inflammatory mediator in the tumor microenvironment (TME), where cancer-associated fibroblasts (CAFs) drive breast cancer progression by secreting various cytokines. This study investigates the role of IL-8 positive CAFs in breast cancer pathogenesis and explores their underlying molecular mechanisms. METHODS: We performed Mendelian randomization (MR) analysis to evaluate the potential causal relationship between IL-8 levels and breast cancer risk using genome-wide association study (GWAS) data. Subpopulations of IL-8 + CAFs were identified through sin

Environmental and genetic risk factors for preterm birth: interplays with stressful events during pregnancy.

BACKGROUND: Preterm birth (PTB) etiology remains poorly understood. Our aim was to investigate the relation of environmental factors and specific gene polymorphisms involved in PTB in the context of stressful life events during pregnancy. METHODS: Parental sociodemographic and obstetric data as well as genetic variants of 1263 preterm newborns were analyzed. Logistic regressions were used to identify shared environmental and genetic risk factors for PTB and stressful life events. A Lasso Ridge logistic regression with cross-validation was used to select the best predictors of maternal stress.

A RE-EVALUATION OF ZYGOCOTYLE (DIGENEA, PARAMPHISTOMOIDEA) BASED ON NEW GENETIC DATA SUPPORTS ITS SYNONYMIZATION WITH WARDIUS.

The Zygocotylidae Ward, 1917 is a compact family of amphistome digeneans that until now comprised 2 genera, each represented by a single species: Zygocotyle lunata (Diesing, 1836) Stunkard, 1916 and Wardius zibethicus Barker and East, 1915 in Barker 1915. Despite highly similar morphology, these genera are separated based on the presence (Zygocotyle) or absence (Wardius) of posterolateral projections (=lappets) on the ventral sucker and esophageal bulb. In the present study, we generated partial large ribosomal subunit (28S), internal transcribed spacer 2 (ITS2) rDNA, and cytochrome c oxidase
