Zinc finger antiviral protein (ZAP) inhibits small ruminant morbillivirus replication in vitro.

Small ruminant morbillivirus (SRMV) is a highly contagious and economically important viral disease of small domestic and wild ruminants. Difficulty with its stable proliferation in ovis aries-derived cells has led to a relative lag in the study of its natural immunity and pathogenesis. Here we report the antiviral properties of ZAP against SRMV, a single-stranded negative-stranded RNA virus of the genus Morbillivirus. ZAP expression was significantly induced in sheep endometrial epithelial cells following SRMV infection. ZAP inhibited SRMV replication in cells after infection, while its overe

TBX1 is required for normal stria vascularis and semicircular canal development.

Little is known about the role of TBX1 in post-otocyst stages of inner ear development. Here, we report on mice with a missense mutation of Tbx1 that are viable with fully developed but abnormally formed inner ears. Mutant mice are deaf due to an undeveloped stria vascularis and show vestibular dysfunction associated with abnormal semicircular canal formation. We show that TBX1 is expressed in endolymph-producing strial marginal cells and vestibular dark cells of the inner ear and is an upstream regulator of Esrrb, which previously was shown to control the developmental fate of these cells. We

[HIV-negative plasmablastic lymphoma attaining sustained remission with bortezomib-combined dose-adjusted EPOCH therapy].

A 61-year-old, HIV-negative, immunocompetent woman was admitted to our hospital for significant weight loss, painful swelling of her right cheek, and rapid growth of a tumor in the right hard palate. A CT scan revealed a neoplastic lesion in the right maxillary sinus, extending to the surrounding soft bone tissue and oral cavity, as well as multiple osteolytic lesions in the skull. A pathologic examination revealed that the neoplastic cells in the oral cavity were plasmablasts tending to appear as differentiated plasma cells. The tumor cells strongly expressed CD138, Vs38c, EBER, and MYC, and

BCG vaccine mediated reduction in the MHC-II expression of macrophages and dendritic cells is reversed by activation of Toll-like receptors 7 and 9.

Tuberculosis is a major cause of death in mankind and BCG vaccine protects against childhood but not adult tuberculosis. BCG avoids lysosomal fusion in macrophages decreasing peptides required for activating CD4 T cells and Th1 immunity while suppressing the expression of MHC-II by antigen presenting cells (APCs). An in vitro model of antigen presentation showed that ligands for TLR-9, 7, 4 and 1/2 increased the ability of APCs to present antigen-85B of BCG to CD4 T cells, which correlated with an increase in MHC-II expression. TLR-activation led to a down-regulation of MARCH1 ubiquitin ligase

Retargeting Clostridium difficile Toxin B to Neuronal Cells as a Potential Vehicle for Cytosolic Delivery of Therapeutic Biomolecules to Treat Botulism.

Botulinum neurotoxins (BoNTs) deliver a protease to neurons which can cause a flaccid paralysis called botulism. Development of botulism antidotes will require neuronal delivery of agents that inhibit or destroy the BoNT protease. Here, we investigated the potential of engineering Clostridium difficile toxin B (TcdB) as a neuronal delivery vehicle by testing two recombinant TcdB chimeras. For AGT-TcdB chimera, an alkyltransferase (AGT) was appended to the N-terminal glucosyltransferase (GT) of TcdB. Recombinant AGT-TcdB had alkyltransferase activity, and the chimera was nearly as toxic to Vero

Immunology of tuberculosis.

The resurgence of tuberculosis worldwide has intensified research efforts directed at examining the host defense and pathogenic mechanisms operative in Mycobacterium tuberculosis infection. This review summarizes our current understanding of the host immune response, with emphasis on the roles of macrophages, T cells, and the cytokine/chemokine network in engendering protective immunity. Specifically, we summarize studies addressing the ability of the organism to survive within macrophages by controlling phagolysosome fusion. The recent studies on Toll-like receptors and the impact on the inna

Nuclear transport of human DDB protein induced by ultraviolet light.

Human damage-specific DNA-binding (DDB) protein can be purified as a heterodimer (p48 and p127) that binds to DNA damaged by ultraviolet light. We report here the effects of UV irradiation on the cellular localization of each DDB subunit as a function of time using green fluorescent fusion proteins in three diploid fibroblast strains: repair-proficient IMR-90 and two repair-deficient xeroderma pigmentosum group E strains (XP95TO and XP3RO). Although p48 remained in the nucleus after UV irradiation, a dynamic nuclear accumulation of p127 from the cytoplasm was found after 24 h. In IMR-90 cells,

Cyclosporin A induces prepro endothelin-1 gene transcription in human endothelial cells.

Cyclosporin A employed in treatment of organ allograft rejection, is associated with hypertension possibly due to endothelin-1. We studied transcriptional regulation of endothelin-1 by cyclosporin A in human endothelial cells using cell transfection experiments and reporter gene assays. Human umbilical vein endothelial cells were established expressing a fusion gene of the coding sequence of the firefly luciferase gene, placed under the control of the rat endothelin-1 promoter. Luciferase assays demonstrate 2.8-fold stimulation of the reporter gene by cyclosporin A (P < 0.01), and Northern blo

Copy number abnormality of acute lymphoblastic leukemia cell lines based on their genetic subtypes.

In this study, we performed genetic analysis of 83 B cell precursor acute lymphoblastic leukemia (B-ALL) cell lines. First, we performed multiplex ligation-dependent probe amplification analysis to identify copy number abnormalities (CNAs) in eight genes associated with B-ALL according to genetic subtype. In Ph+ B-ALL cell lines, the frequencies of IKZF1, CDKN2A/2B, BTG1, and PAX5 deletion were significantly higher than those in Ph- B-ALL cell lines. The frequency of CDKN2A/2B deletion in KMT2A rearranged cell lines was significantly lower than that in non-KMT2A rearranged cell lines. These fi

Ultrastructural observations on the development of triamcinolone-induced cleft palate in hamsters.

Cleft palate was induced in fetuses by administration of triamcinolone to pregnant hamsters. Twenty hours after treatment, alterations were seen at the epithelial-mesenchymal interface in the prospective fusion epithelium of the vertical shelf. The alterations included contacts between the epithelial and mesenchymal cells, and disruptions in the continuity of the basal lamina. Thirty-two hours after triamcinolone treatment, the basal epithelial cells in some of the reorienting palatal shelves were necrotic. When compared with normal cells, the drug treated necrotic cells were lighter in appear

Phytohormone Strigolactams Overcomes Oxaliplatin Resistance in Colorectal Cancer by Blocking the Late-Stage Autophagy to Tip the Metabolic Balance toward Ferroptosis.

Primary and acquired oxaliplatin (OXA)-resistance remains a major challenge in colorectal cancer (CRC) therapy. In this work, the type-II strigolactam SL 39 was identified as a promising lead compound against OXA-resistant HCT116R cells. Co-treatment of SL 39 with OXA restored its sensitivity to HCT116R cells and inhibited xenograft tumor growth. Mechanistically, SL 39-treatment specifically blocked the autophagosome-lysosome fusion in HCT116R cells without altering lysosomal pH, leading to induction of mitochondrial dysfunctions and lethal accumulation of reactive oxygen species. Additionally

Case report: Systemic presentation of ALK-positive Histiocytosis.

ALK-positive Histiocytosis (ALK-HSs) is a recently identified rare clinical entity characterized by tissue histiocytic alterations associated with ALK gene rearrangement. Clinical presentations can be solitary, multifocal, or systemic (involving multiple sites and organs). Due to limited reported cases, there is inadequate understanding of this disease. This report presents a case of ALK-HSs in a 71-year-old male patient who presented with hematuria for one week. Imaging studies conducted at an external hospital showed multiple lesions in the penis, bilateral testes, back skin, and the third l

Multivalent peptide dendrimers inhibit the fusion of viral-cellular membranes and the cellular NF-κB signaling pathway.

The binding of the influenza A virus (IAV) to host cells is multivalent interactions between the hemagglutinin (HA) trimer and sialic acid residues on the cell surface, which present a challenge for the development of efficient antiviral drugs interfering with the entry of IAV into host cells. In this study, a number of multivalent peptide dendrimers targeting the HA2 subunit of HA to block the fusion between viral-cellular membranes have been created, of which FMOC-4-KKWK showed the lowest cytotoxicity, while in the nanomolar concentration range of antiviral effects. In addition to being acti

A Peptide Inhibitor of the Human Cytomegalovirus Core Nuclear Egress Complex.

The replication of human cytomegalovirus (HCMV) involves a process termed nuclear egress, which enables translocation of newly formed viral capsids from the nucleus into the cytoplasm. The HCMV core nuclear egress complex (core NEC), a heterodimer of viral proteins pUL50 and pUL53, is therefore considered a promising target for new antiviral drugs. We have recently shown that a 29-mer peptide presenting an N-terminal alpha-helical hook-like segment of pUL53, through which pUL53 interacts with pUL50, binds to pUL50 with high affinity, and inhibits the pUL50-pUL53 interaction in vitro. Here, we

Carbon fiber amperometry in the study of ion channels and secretion.

Activation of Ca2+ channels in the plasma membrane or on internal Ca2+ stores raises cytosolic Ca2+ concentration ([Ca2+]c). Among diverse functions of Ca2+ signals, the induction of exocytosis-the process in which the contents of secretory vesicles are released by their fusion to the plasma membrane-is one of the most important. For example, in neurons and endocrine cells, it allows intercellular communication and secretion of biomolecules. Exocytosis can be detected by several physical and chemical means. By chemically oxidizing the released secretory products at a fixed electrode potential,

STAG2 mutations fine-tune the ALK-driven oncogenic program in ALK-positive anaplastic large cell lymphoma.

ALK-positive Anaplastic Large Cell Lymphoma (ALK+ ALCL) is a rare yet aggressive T-cell lymphoma, most often characterized by the NPM1::ALK fusion. Because few somatic variants have been identified in ALK+ ALCL, we conducted paired whole-exome sequencing of patient tumors with matched normal tissue. Compared with previous studies, we identified a higher frequency of mutations in cohesin complex genes, mostly STAG2 variants (6/16) predicted to impair protein function. Immunohistochemistry confirmed loss of STAG2 in additional patient tumor samples. Using an original model of NPM1::ALK-driven ly

Development of a CD3ε mAb that can identify and in vitro activate T cells in large yellow croaker (Larimichthyscrocea).

As a hallmark molecule of T cells, CD3ε forms a complex with T-cell receptor (TCR) to transduce antigen signals and drive T cell activation, playing a pivotal role in T cell-mediated immune response. However, the lack of specific monoclonal antibodies (mAbs) targeting CD3ε in fish has substantially impeded the study on adaptive immunity in these species. In this study, we constructed NIH/3T3 cells expressing CD3ε of the large yellow croaker (Larimichthys crocea) via retroviral transduction. Using these cells as immunogen to immune mice, we generated a mAb that specifically recognized a populat
