Telavancin: a new lipoglycopeptide for gram-positive infections.

Telavancin is a lipoglycopeptide derivative of vancomycin. Similar to vancomycin, it demonstrates activity in vitro against a variety of Gram-positive pathogens, including but not limited to methicillin-resistant Staphylococccus aureus (MRSA) and penicillin-resistant Streptococcus pneumoniae (PRSP). Modifications to vancomycin's structure expanded telavancin's spectrum of activity in vitro to include organisms such as glycopeptide-intermediate S. aureus (GISA), vancomycin-resistant S. aureus (VRSA) and vancomycin-resistant enterococci (VRE). However, the clinical implications of this are curre

Formononetin inhibits colorectal cancer via the miR-490-3p/ABCC2 axis and synergizes with 5-fluorouracil: mechanistic insights and therapeutic implications.

Colorectal cancer (CRC), one of the primary causes of gastrointestinal malignancy-related mortality worldwide, encounters significant therapeutic challenges due to multidrug resistance. Multidrug resistance-associated protein 2 (MRP2), encoded by ABCC2, plays a crucial role in the development of drug resistance during clinical CRC treatment. This study elucidates a novel mechanism by which formononetin (FMNT), a bioactive isoflavone commonly found in food and medicinal-related plants, suppresses CRC via microRNA-490-3p (miR-490-3p)-mediated regulation of ABCC2 and synergizes with 5-fluorouraci

Plasma Concentrations and Cancer-Associated Mutations in Cell-Free Circulating DNA of Treatment-Naive Follicular Lymphoma for Improved Non-Invasive Diagnosis and Prognosis.

Follicular lymphoma (FL) is the second most frequent non-Hodgkin lymphoma accounting for 10-20% of all lymphomas in western countries. As a clinically heterogeneous cancer, FL occasionally undergoes histological transformation to more aggressive B cell lymphoma types that are associated with poor prognosis. Here we evaluated the potential of circulating cell-free DNA (cfDNA) to improve the diagnosis and prognosis of follicular lymphoma patients. Twenty well-characterized FL cases (13 symptomatic and 7 asymptomatic) were prospectively included in this study. Plasma cfDNA, formalin-fixed paraffi

FGF21 serum levels are related to insulin resistance, metabolic changes and obesity in Mexican people living with HIV (PLWH).

BACKGROUND: Antiretroviral therapy has significantly improved prognosis in treatment against HIV infection, however, prolonged exposure is associated to cardiovascular diseases, lipodystrophy, type 2 diabetes, insulin resistance, metabolic alteration, as obesity which includes the accumulation of oxidative stress in adipose tissue. FGF21 is a peptide hormone that is known to regulate glucose and lipid metabolism. FGF21 is expressed and secreted primarily in the liver and adipose tissue, promoting oxidation of glucose/fatty acids and insulin sensitivity. Alterations in FGF21 may be associated w

Multi-omics Integration of Single-cell and Spatial Transcriptomics with Tissue and Urine Proteomics Identifies TRIM28 as a Translationally Relevant Candidate Regulator in Acute Kidney Injury.

Acute kidney injury (AKI) lacks disease-modifying therapies, partly because cell type-specific injury programs remain incompletely resolved and tissue mechanisms are not readily translated into clinically accessible biomarkers. We constructed a cross-scale, multi-omics atlas to prioritize candidate regulator linking renal injury circuitry with urine-detectable signals. Human single-cell, bulk, and spatial transcriptomics were integrated with mouse renal ischemia-reperfusion injury (RIRI) kidney proteomics and clinical urine proteomics. Overlap of differentially expressed genes and differential

Neuroprotective effects of platinum nanoparticle-based microreactors in a pilocarpine-induced rat model of epilepsy.

Epilepsy is a neurological disorder characterized by recurrent seizures resulting from excessive and synchronous electrical activity in the brain. Hyperexcitability leads to increased intracellular calcium, oxidative stress, and neuronal damage, promoting neurodegeneration and long-term reorganization of brain circuits. Approximately 30% of individuals diagnosed with epilepsy do not respond adequately to currently available pharmacological treatments, underscoring the need for alternative therapeutic strategies. Previous studies demonstrated that platinum nanoparticle (Pt-NP)-based microreacto

Human Derived Exosome Injections (ASCE+ vs. CELLEXOSOME) Enhance Collagen Remodeling and Angiogenesis in Intact Skin: A Comparative Experimental Study.

BACKGROUND: Exosomes derived from human cells have emerged as promising biological agents for enhancing skin quality through stimulation of collagen remodeling and angiogenesis. While their role in wound healing is well established, their effects on intact, non-injured skin remain insufficiently investigated. Their biological activity depends on their molecular cargo, including growth factors, extracellular matrix-modulating proteins, and angiogenic microRNAs. OBJECTIVE: To evaluate the impact of intradermal injection of two human-derived exosome formulations on dermal architecture and vascul

AI-integrated multi-omics platform to revolutionize anti-metastatic therapy development through circulating tumor cell profiling: SCRUM-MONSTAR-CTC.

BACKGROUND: Metastatic disease remains the leading cause of cancer-related death, yet most precision oncology strategies still emphasize profiling primary tumors and tracking cell-free tumor DNA (ctDNA). Although ctDNA has transformed genomic profiling, molecular residual disease monitoring, and early cancer detection, it cannot directly capture viable tumor cell states, phenotypic plasticity, or functional adaptations that drive metastatic spread. We propose that the next phase of precision oncology should integrate the cellular dimension of metastasis through systematic circulating tumor cel

Efficient gene transfer by pulse parameters for electrochemotherapy of cells in vitro and in muscle and melanoma tumors in mice.

BACKGROUND: In recent years, various gene therapy strategies have been developed for cancer treatment. One of these strategies is electroporation-based delivery of therapeutic transgenes - gene electrotransfer (GET). Electrochemotherapy and GET have been combined in several contemporary preclinical and veterinary studies. In most cases, two different pulse protocols are used, each for a specific treatment. The aim of our current study was to test whether the standard pulse protocol used in daily clinical practice for electrochemotherapy can also be used for effective GET. MATERIALS AND METHOD

Immune responses detected in urothelial carcinoma patients after vaccination with NY-ESO-1 protein plus BCG and GM-CSF.

Urothelial carcinoma of the bladder is a solid tumor entity for which the immunomodulatory agent Bacillus-Calmette Guerin (BCG) has been shown to have substantial efficacy with approximately 90% cure rates in select patients with superficial disease. Immune-based therapies for patients with more invasive disease do not currently exist. We previously showed that invasive urothelial carcinomas express the NY-ESO-1 tumor antigen. Here we evaluated the safety and immunogenicity of a recombinant NY-ESO-1 protein vaccine, which was administered with granulocyte macrophage colony-stimulating factor a

Stage-dependent immunity orchestrates AQP4 antibody-guided NMOSD pathology: a role for netting neutrophils with resident memory T cells in situ.

Neuromyelitis optica spectrum disorder (NMOSD) is an autoimmune disease of the CNS characterized by the production of disease-specific autoantibodies against aquaporin-4 (AQP4) water channels. Animal model studies suggest that anti-AQP4 antibodies cause a loss of AQP4-expressing astrocytes, primarily via complement-dependent cytotoxicity. Nonetheless, several aspects of the disease remain unclear, including: how anti-AQP4 antibodies cross the blood-brain barrier from the periphery to the CNS; how NMOSD expands into longitudinally extensive transverse myelitis or optic neuritis; how multiphasic

Therapeutic Extracellular Vesicles from Tonsil-Derived Mesenchymal Stem Cells for the Treatment of Retinal Degenerative Disease.

BACKGROUND: Retinal degenerative disease (RDD), one of the most common causes of blindness, is predominantly caused by the gradual death of retinal pigment epithelial cells (RPEs) and photoreceptors due to various causes. Cell-based therapies, such as stem cell implantation, have been developed for the treatment of RDD, but potential risks, including teratogenicity and immune reactions, have hampered their clinical application. Stem cell-derived extracellular vesicles (EVs) have recently emerged as a cell-free alternative therapeutic strategy; however, additional invasiveness and low yield of

Development and Validation of Novel Senescence-TIME Biomarkers for Predicting the Prognosis and Immunotherapy Responsiveness of SKCM Patients.

BACKGROUND: Increasing evidence indicates that tumor cellular senescence can impair antitumor immunity and promote skin cutaneous melanoma (SKCM) progression. However, effective methods for assessing tumor cellular senescent status and tumor immune microenvironment (TIME) status remain lacking. This study intends to establish a novel Senescence-TIME Risk Score (STIRS) based on senescence and TIME related genes to predict prognosis and immunotherapy responsiveness in SKCM patients, thereby providing new strategies for current clinical personalized treatment. METHODS: We identified distinct sen

Maxillary mesenchymal chondrosarcoma harboring HEY1::NCOA2 fusion in a 13-year-old girl: a rare case report and literature review.

BACKGROUND: Mesenchymal chondrosarcoma (MCS) is a rare and highly aggressive subtype of chondrosarcoma, accounting for less than 1% of all chondrosarcomas. It predominantly affects adolescents and young adults and frequently arises in craniofacial bones and soft tissues. Diagnosis is challenging because of significant histological overlap with other high-grade spindle cell sarcomas, particularly when the cartilaginous component is minimal or absent. The identification of the HEY1::NCOA2 gene fusion has emerged as a highly specific molecular marker for MCS, substantially improving diagnostic ac

Primary Embryonal Rhabdomyosarcoma of the Breast: A Case Report and Literature Review.

BACKGROUND: Rhabdomyosarcoma (RMS) is the most common soft-tissue sarcoma in children and adolescents, but primary RMS of the breast is exceptionally rare and diagnostically challenging. Imaging findings are nonspecific and can mimic benign adolescent lesions (e.g., fibroadenoma), making timely histopathologic confirmation crucial. Immunohistochemistry for muscle markers - particularly desmin and myogenin - supports definitive diagnosis of embryonal RMS (ERMS).
