The number of elevated cytokines and chemokines in preclinical seropositive rheumatoid arthritis predicts time to diagnosis in an age-dependent manner.

OBJECTIVE: To evaluate levels of biomarkers in preclinical rheumatoid arthritis (RA) and to use elevated biomarkers to develop a model for the prediction of time to future diagnosis of seropositive RA. METHODS: Stored samples obtained from 73 military cases with seropositive RA prior to RA diagnosis and from controls (mean 2.9 samples per case; samples collected a mean of 6.6 years prior to diagnosis) were tested for rheumatoid factor (RF) isotypes, anti-cyclic citrullinated peptide (anti-CCP) antibodies, 14 cytokines and chemokines (by bead-based assay), and C-reactive protein (CRP). RESULT

Factor VIII-pulsed dendritic cells reduce anti-factor VIII antibody formation in the hemophilia A mouse model.

OBJECTIVE: Hemophilia inhibitor formation is a T-cell-dependent immune response to infused factor VIII (F.VIII). As immature dendritic cells (DCre) regulate immune response and promote tolerance, we evaluated F.VIII-pulsed DCre, propagated from bone marrow in the presence of granulocyte-macrophage colony-stimulating factor and transforming growth factor-beta, in achieving F.VIII tolerance. MATERIALS AND METHODS: The effects of intravenous F.VIII-pulsed DCre in C57BL/6 hemophilia A mice were determined by total F.VIII inhibitory antibodies, T-cell proliferation, thymidine uptake, cytokine prof

Binding sites for adeno-associated virus Rep proteins within the human genome.

The Rep proteins of adeno-associated virus type 2 (AAV) are known to bind to Rep recognition sequences (RRSs) in the AAV inverted terminal repeats (ITRs), the AAV p5 promoter, and the preferred AAV integration site in human chromosome 19, called AAVS1. Integration of the AAV genome into AAVS1 appears to be mediated by an interaction between the Rep proteins of AAV and Rep binding sites within the viral genome and the integration locus. In an attempt to identify potential alternate integration sites, we looked for recognition sites for AAV Rep proteins in the human genome by performing a BLASTN

Efficacy and Safety of Trilaciclib for the Prevention of Chemotherapy-Induced Myelosuppression in Patients With Solid Tumors: A Systematic Review and Meta-Analysis.

Trilaciclib, a cyclin-dependent kinase 4 and 6 (CDK4/6) inhibitor administered before chemotherapy, reduces chemotherapy-induced myelosuppression (CIM) in extensive-stage small cell lung cancer (ES-SCLC), but its efficacy across solid tumors remains unclear. This meta-analysis evaluated the efficacy and safety of trilaciclib in solid tumor patients receiving chemotherapy. PubMed, Embase, Cochrane Library, and ClinicalTrials.gov were searched from inception to January 21, 2026. Randomized controlled trials (RCTs) and cohort studies comparing chemotherapy with or without trilaciclib were include

Celastrol Mitigates Acute Pancreatitis Associated Inflammation by Modulating the IL-34/CSF-1R Axis and Suppressing NF-κB/ERK Signaling.

BACKGROUND: Acute pancreatitis (AP) is characterized by early acinar injury followed by rapid inflammatory amplification, yet the upstream molecular triggers linking tissue stress to cytokine escalation remain incompletely defined. Interleukin-34 (IL-34), a ligand of colony-stimulating factor-1 receptor (CSF-1R), regulates inflammatory signaling, but its role in AP has not been elucidated. Celastrol is a bioactive triterpenoid with established anti-inflammatory properties; however, whether it modulates IL-34-associated signaling in AP remains unclear. METHODS: Experimental AP was induced in r

Patient-derived decidual organoids reveal rhG-CSF-associated protective responses involving JAK2/STAT3 signaling in recurrent spontaneous abortion.

BACKGROUND: Recurrent spontaneous abortion (RSA) is a complex reproductive disorder frequently lacks effective therapeutic interventions. Recombinant human granulocyte colony-stimulating factor (rhG-CSF) has shown promise in improving pregnancy outcomes in RSA, but its underlying mechanisms remain unclear. Organoid technology offers new opportunities for disease modeling and mechanistic research in reproductive medicine. METHODS: Decidual tissue samples from RSA patients were utilized to establish decidual organoids using three-dimensional (3D) culture techniques. These organoids were charact

Loss of M1 Acetylcholine Receptor-mediated Orexinergic Activity Contributes to Immune Dysfunction in Experimental Sepsis.

BACKGROUND: Sepsis (life-threatening organ dysfunction caused by a dysregulated host response to infection) causes millions of deaths worldwide annually. Sepsis-induced changes in brain regulatory functions remain understudied. Previous work demonstrated that cecal ligation and puncture (CLP, a murine model of sepsis) affected physiologic variables and serum cytokines and hormone levels. Correction of decreased activity in the orexinergic nervous system or administration of the M1 muscarinic acetylcholine receptor (M1mAChR) agonist xanomeline reversed some of these findings. We hypothesized th

Pexidartinib Inhibits Macrophage Senescence Through Glycolysis in Periodontitis Microenvironment.

OBJECTIVES: Periodontitis, a chronic inflammatory condition, is caused by complex interactions between periodontopathic bacteria and the local innate immune response. Macrophage senescence, a pivotal contributor to immune dysfunction, has been implicated in periodontitis progression. This research was conducted to clarify how macrophage senescence and glycolysis interact within the periodontal inflammatory microenvironment. Specifically, we investigated whether pexidartinib (PLX3397), known for inhibiting the colony-stimulating factor-1 receptor (CSF-1R), could mitigate macrophage senescence b

Apalutamide-associated rash combined with severe neutropenia and eosinophilia: a case report.

BACKGROUND: Apalutamide is approved for the treatment of nonmetastatic castration-resistant prostate cancer (nmCRPC) and metastatic hormone-sensitive prostate cancer (mHSPC). Currently, the incidence of rash is higher in patients treated with apalutamide. However, the occurrence of apalutamide-associated skin rash combined with apalutamide-induced neutropenia and eosinophilia in a single patient has not been reported previously. The aim of this study is to describe a rare and severe adverse reaction induced by apalutamide, in order to improve clinical awareness and facilitate early recognition

Significantly reduced inflammatory foreign-body-response to neuroimplants and improved recording performance in young compared to adult rats.

The multicellular inflammatory encapsulation of implanted intracortical multielectrode arrays (MEA) is associated with severe deterioration of their field potentials' (FP) recording performance, which thus limits the use of brain implants in basic research and clinical applications. Therefore, extensive efforts have been made to identify the conditions in which the inflammatory foreign body response (FBR) is alleviated, or to develop methods to mitigate the formation of the inflammatory barrier. Here, for the first time, we show that (1) in young rats (74±8 gr, 4 weeks old at the onset of the

Doxorubicin-polyglycerol-nanodiamond conjugate is a cytostatic agent that evades chemoresistance and reverses cancer-induced immunosuppression in triple-negative breast cancer.

BACKGROUND: Triple negative breast cancer (TNBC) has the poorest prognosis of all breast cancer subtypes and is one of the most fatal diseases for women. Combining cytotoxic chemotherapy with immunotherapy has shown great promise for TNBC treatment. However, chemotherapy often leads to the development of chemoresistance and severe systemic toxicity compromising the immune functions that are crucial to anti-TNBC immune therapy. Tumor-induced immunosuppression also poses a great hindrance to efficacious anti-TNBC immunotherapy. Nanomedicine holds great promise to overcome these hurdles. RESULTS

A model of immunity to Burkholderia pseudomallei: unique responses following immunization and acute lethal infection.

Burkholderia pseudomallei, the etiological agent of melioidosis, causes significant mortality in endemic regions, but little is known regarding the immune mechanisms required for successful protective immunity. To establish a model of immunization that could be used to study this we screened a library of B. pseudomallei strains for immunogenicity in mice. BALB/c mice were immunized with test strains, and 2 weeks later were given a lethal challenge (LC) of virulent B. pseudomallei. Among 49 strains tested, a single strain, CL04, exhibited strong immunoprotective capacity. Interestingly, CL04 ha

The NLRP3/IL-6/EGR2/CSF-1 Axis Regulates Macrophage Differentiation and Inflammatory Resolution in Infection-Associated Wound Healing : NLRP3 Axis Controls Macrophage Fate in Wound Infection.

Chronic Pseudomonas aeruginosa (P. aeruginosa) infection impairs wound healing, yet the underlying inflammatory mechanisms remain poorly understood. This study investigates how the NLRP3 inflammasome orchestrates macrophage differentiation and inflammatory resolution during infection-associated wound healing. A murine P. aeruginosa wound infection model was established. Macrophage polarization dynamics were assessed by flow cytometry (CD11b/F4/80/Arg1/iNOS/CD206/CD86) and histology. Wound healing outcomes were compared between wild-type and NLRP3-/- mice via closure kinetics, ECM analysis (Mas

Tumor-associated M2 macrophages promote prostate cancer invasion through the M-CSF-PCLAF pathway.

BACKGROUND: Prostate cancer (PCa), particularly in its advanced and castration-resistant forms, remains a major threat to men's health, with the tumor microenvironment (TME) playing a crucial role in its progression. Tumor-associated macrophages (TAMs), especially the M2 phenotype, are key components of the TME. Our previous work identified that M2 TAMs in PCa upregulate M-CSF secretion via the MS4A6A-MYC pathway. This study aims to identify the critical downstream effector within PCa cells that mediates the tumor-promoting effects of M-CSF. METHODS: The impact of M-CSF on PCa cell (PC3 line)

Pegfilgrastim versus filgrastim for chemo-mobilized stem cell collection in multiple myeloma: A retrospective real-world study.

BACKGROUND: Autologous stem cell transplantation (ASCT) is a standard treatment for newly diagnosed multiple myeloma (MM). Achieving sufficient stem cell yield via effective mobilization promotes successful hematological reconstitution. However, clinical evidence regarding the comparative outcomes of pegfilgrastim (PEG) versus filgrastim (FIL) remains controversial, lacking regimen-specific comparisons. OBJECTIVE: To evaluate the efficacy, safety, efficiency, and costs of PEG versus FIL in MM patients, and compare the impact of cyclophosphamide-based chemo-mobilization on these outcomes. STU

The Novel Oncolytic Herpes Simplex Virus Type-1 (HSV-1) Vaccine Strain VC2 Constitutively Expressing GM-CSF Causes Increased Intratumoral T Cell Infiltration and Inhibition of Tumor Metastasis in the 4T1/Balb/c Mouse Model of Stage Four Breast Cancer.

Oncolytic virotherapy (OVT) aims to disrupt the tumor microenvironment and provide a unique therapeutic approach against solid tumors. Herpes simplex virus type-1 (HSV-1) has shown strong promise for treating various solid tumors and has been approved to treat melanoma and glioma in human patients. Previously, we reported the generation of an engineered HSV-1 vaccine strain VC2, which has shown exceptional promise as an oncolytic and immunotherapeutic virus. In the present work, we engineered VC2 to constitutively express the murine granulocyte-macrophage colony-stimulating factor (GM-CSF) gen

Dose-Dense Docetaxel and Radium-223 in Bone-Dominant Metastatic Castration-Resistant Prostate Cancer.

BACKGROUND: Disease progression in castration-resistant prostate cancer (CRPC) remains bone-dominant and docetaxel-responsive. Docetaxel and radium-223 would be a logical combination but myelosuppression is dose-limiting. Dose-dense schedules of docetaxel have comparable activity to bolus dosing with mitigated myelosuppression. We hypothesized that dose-dense docetaxel with standard radium-223 would be a feasible, safe and effective combination in bone-dominant metastatic CRPC. METHODS: Subjects had progressive bone-predominant CRPC. Design was dose escalation plus expansion with 28-day cycle
