Imbalanced IgA-IgG class switching recombination: a novel mechanism of gut immune dysregulation in inflammatory bowel disease.

Inflammatory bowel disease (IBD) is a chronic intestinal inflammatory disorder with significant disease burden that markedly impairs patients' quality of life. The pathogenesis of IBD is closely associated with hyperactive immunopathological responses. Emerging evidence has established humoral immunity as a crucial contributor to IBD development. Class switch recombination (CSR), the critical process enabling B cells to produce different antibody isotypes, plays a pivotal role in humoral immunity. While healthy intestinal mucosa predominantly contains immunoglobulin A (IgA) that maintains gut

Transcriptional and epigenomic analysis of insulin sensitivity in human adipocytes.

Insulin resistance is a key feature of type 2 diabetes (T2D) and is also associated with a wide variety of other disease states. We isolated adipocytes from subjects with and without insulin resistance and subjected them to transcriptional and epigenomic profiling, allowing us to identify genes, cis-regulatory elements, and pathways that associate with insulin resistance in this critical cell type. We focused on several differentially enriched regions near the IRS1 gene that were close to, but distinct from, single-nucleotide polymorphisms associated with T2D and insulin resistance. CRISPR int

Host Tyrosine Kinase Signaling and Tyrosine Kinase Inhibitors as Candidate Host-Directed Therapies for Tuberculosis.

Tuberculosis (TB) is a major global public health issue. Although current anti-TB treatment regimens are effective, challenges such as prolonged treatment durations, high drug toxicity, and drug resistance remain, necessitating the development of new therapeutic targets and drugs. Host-directed therapy (HDT), which enhances the body's immune response to boost resistance against Mtb infection, has become a promising treatment strategy. Protein tyrosine kinases (PTKs) play a critical role in cellular signal transduction, and their aberrant activation is linked to the onset of various diseases. R

Sex differences in delirium: Neuroendocrine-immune mechanisms and implications for precision neurocritical care.

Delirium is an acute and multifactorial syndrome that remains highly prevalent in critical illness and is associated with avoidable morbidity, persistent cognitive decline, and increased mortality. Its mechanisms appear to involve a close interaction between neuroendocrine and immune pathways, including gonadal hormones, neuroactive steroids produced within the brain, and hypothalamic-pituitary-adrenal axis activity. Because these systems differ between sexes, biological sex may influence delirium risk, clinical presentation, and recovery, although this has rarely been examined in a systematic

Disruption of Signalling Pathways Induced by HBV HBx, HCV Core, and EBV LMP1 in Gastric Cancer.

Oncoviruses are a significant etiological factor in a substantial proportion of human cancers, with a particular impact on cancers of the digestive system. This review provides a comprehensive analysis of the complex molecular mechanisms through which viral oncoproteins, focusing on Hepatitis B Virus (HBV) HBx, Hepatitis C Virus (HCV) core protein, and Epstein-Barr Virus (EBV) latent membrane protein 1 (LMP1), hijack and disrupt host cell signalling networks to induce carcinogenesis. These disruptions include the inactivation of critical tumour suppressor proteins, such as p53 and the retinobl

Advances in the Molecular Mechanisms of Cholangiocarcinoma: A Comprehensive Review of Biomarkers, Regulatory Pathways and Tumor Microenvironment Reprogramming.

Cholangiocarcinoma (CCA) is a malignant tumor arising from the epithelial cells of the biliary tract. It is characterized by high heterogeneity, frequent late diagnosis, high rates of metastasis, and overall poor prognosis. Unfortunately, the development of sensitive biomarkers and effective therapeutic approaches remains challenging and an unmet need. In recent years, advances in molecular biological techniques have significantly enhanced our understanding of the molecular mechanisms underlying CCA pathogenesis. This review summarized the biomarkers identified in CCA, highlighted the critical

REAP1/AtSWAP70 integrates RAB5 and ROP signalling during sexual reproduction.

Small GTPases of the Ras superfamily are critical regulators of diverse cellular processes. While crosstalk between their signalling pathways has been documented in animals, similar mechanisms remain unexplored in plants, where small GTPases have undergone unique evolutionary diversification. Here we identify REAP1/AtSWAP70 as an effector in Arabidopsis thaliana that interacts with the active forms of both canonical RAB5 and the plant-specific RAB5, ARA6. Remarkably, REAP1 also binds to active ROP7, a plant-unique Rac-type GTPase, via its Dbl homology domain. REAP1 localizes to endosomes and f

Infectious Agents in Multiple Sclerosis: Viral Triggers, Antibody-Mediated Autoimmunity, and Parasitic Immunomodulation.

Multiple sclerosis (MS) is a chronic immune-mediated disease of the central nervous system characterized by demyelination, neuroinflammation, and progressive neurodegeneration. While there is a small component of genetic susceptibility to MS risk, environmental factors, including infectious exposures, are gaining increased recognition as playing a critical role in MS initiation and progression. Viral infections, especially by Epstein-Barr virus (EBV), have emerged as strong candidates and triggers of MS symptoms, through antibody-mediated molecular mimicry and B-cell dysregulation. In contrast

Priming Cells with Modulators of Cellular Signaling Ensures Enhanced Selectivity of HSPG-Specific Protein-Drug Conjugate Delivery.

Protein-drug conjugates (PDCs) constitute a rapidly growing group of precise anticancer agents. Receptor-mediated endocytosis is a critical step in PDC action, which ensures the delivery of PDCs into the interior of cancer cells. Here, we report TriFHS-MMAE, the first multivalent PDC specifically targeting heparan sulfate proteoglycans (HSPGs) overexpressed by pancreatic cancer cells, which induces ultrafast and highly efficient aggregation-dependent endocytosis (ADE) of HSPGs. Using high-content screening with a near-kinome-wide library of inhibitors, we identified signaling pathways that gov

Unraveling the role of miRNAs in placental function: insights into trophoblast biology and pregnancy pathology.

MicroRNAs (miRNAs) are pivotal regulators of placental development and function, orchestrating critical processes such as trophoblast proliferation, differentiation, invasion, and vascular remodeling. By modulating diverse signaling pathways, these small non-coding RNAs help ensure proper placental formation and optimal maternal-fetal nutrients and gases exchange. Dysregulated miRNA expressions can profoundly affect placental biology, leading to complications such as preeclampsia, fetal growth restriction, and recurrent pregnancy loss. Accumulating evidence implicates abnormal miRNA profiles i

Molecular Features Accompanying Richter's Transformation in Patients with Chronic Lymphocytic Leukemia.

Chronic Lymphocytic Leukemia (CLL) is a highly heterogeneous tumor. Although targeted therapies such as Bruton's Tyrosine Kinase (BTK) inhibitors and B-cell lymphoma-2 (Bcl-2) inhibitors have significantly improved patient outcomes in CLL, the disease remains incurable. A critical aspect of CLL progression is its transformation from an indolent tumor to a high-grade malignancy, a process known as Richter's Transformation (RT) or Richter Syndrome. Treatment options for RT are very limited, and patient prognosis is often poor. The molecular mechanisms driving RT are not yet fully elucidated. Thi

Partial Anterior Opercular Syndrome as Surgical Complication: Case Presentation and Brief Review of Literature.

Anterior opercular syndrome (a.k.a. Foix-Chavany-Marie syndrome) is a rare neurological condition, described as a paralysis of the mouth and tongue usually caused by a bilateral lesion of the frontal opercular area. The patient presents with speaking, chewing, and swallowing impairment, but autonomic and emotional functions-like smiling and yawning-are typically preserved. We present our patient's clinical data after critical analysis, together with a brief literature review about anterior opercular syndrome caused by unilateral opercular lesions. To our knowledge, less than 20 cases of anteri

Antibiotic Use in Livestock Farming: A Driver of Multidrug Resistance?

Antimicrobial resistance (AMR) constitutes a pressing and intensifying global health crisis, significantly exacerbated by the inappropriate utilization and excessive application of antibiotics in livestock agriculture. The excessive use of antibiotics, including prophylactic and metaphylactic administration as well as growth-promotion applications, exacerbates selective pressures, fostering the proliferation of multidrug-resistant (MDR) bacterial strains. Pathogens such as Escherichia coli, Salmonella spp., and Staphylococcus aureus can be transmitted to humans through direct contact, contamin

Metabolic regulation of immunity in the tumor microenvironment.

Metabolic-immune crosstalk in the tumor microenvironment (TME) is a critical driver of tumorigenesis, progression, and immune evasion. Tumor cells undergo profound metabolic reprogramming, causing nutrient competition, toxic metabolite accumulation, and the formation of cold niches that gradually exhaust effector immune cells. In contrast, immunosuppressive cells exhibit strong metabolic adaptability, reinforcing the suppressive milieu. Moreover, tertiary lymphoid structures provide nutrient- and oxygen-rich "moats" that sustain the functions of B and T cells. In addition, metabolic-immune int

Hypermutability bypasses genetic constraints in SCV phenotypic switching in Pseudomonas aeruginosa biofilms.

Biofilms are critical in the persistence of Pseudomonas aeruginosa infections, particularly in cystic fibrosis patients. This study explores the adaptive mechanisms behind the phenotypic switching between Small Colony Variants (SCVs) and revertant states in P. aeruginosa biofilms, emphasizing hypermutability due to Mismatch Repair System (MRS) deficiencies. Through experimental evolution and whole-genome sequencing, we show that both wild-type and mutator strains undergo parallel evolution by accumulating compensatory mutations in factors regulating intracellular c-di-GMP levels, particularly

Antibacterial Activity of GO-Based Composites Enhanced by Phosphonate-Functionalized Ionic Liquids and Silver.

The development of antibiotic-independent antimicrobial materials is critical for addressing bacterial resistance to conventional antibiotics. Currently, there is a lack of comprehensive understanding of ionic liquid-modified composites in antimicrobial applications. Here, we innovatively prepared GO-based composites modified with phosphonate ionic liquids via a series of surface functionalizations. The resulting antibacterial composites exhibit significant broad-spectrum activity against both Gram-negative and Gram-positive bacteria, including drug-resistant strains, with stronger efficacy ag

Targeting HSP70 protein-protein interactions for cancer precision Therapy: Mechanisms, structures, and inhibitor strategies.

Dysregulated protein homeostasis and aberrant signaling pathways are established hallmarks of cancer. Among key molecular players, heat shock protein 70 (HSP70) exerts critical oncogenic functions by forming stable protein-protein interactions (PPIs) with co-chaperones and client proteins, thereby sustaining malignant signaling, suppressing apoptosis, and promoting drug resistance. However, conventional ATPase inhibitors targeting HSP70 face clinical limitations, including target conservation, systemic toxicity, and induction of the heat shock response. Inhibitors targeting HSP70 PPIs demonstr

Physicians' experiences delivering provincial real-time virtual support services: A qualitative interview study.

Rural, remote, and Indigenous communities in British Columbia (BC) tend to have lower access to healthcare providers and poorer health outcomes-an inequality that the COVID-19 pandemic has exacerbated. In response, Real-Time Virtual Support (RTVS) pathways were developed to advance equitable access to care for patients and provide peer support to physicians working in underserved communities. This study aimed to describe the perspectives of Virtual Physicians (VPs) who delivered the RTVS services. Forty-five RTVS VPs engaged in 30-minute semi-structured interviews about their experiences and p

Pathogenic Fungal Sensing and Responses to Stressful Host Environments.

Despite the ubiquity of fungi in nature, only a small fraction are pathogenic to humans, and the majority of these fungi are opportunistic and affect immunocompromised individuals. In general, pathogen emergence is restricted by the ability of fungi to sense and withstand human host environmental cues and stresses. These stress responses in fungi involve immediate survival reactions as well as long-term adaptations. Additionally, some opportunistic pathogenic behavior suggests that virulence traits evolved for environmental survival, a concept known as exaptation. This review covers recent adv
