Engineering natural Saccharomyces cerevisiae isolates for enhanced one-step cellulosic ethanol production.

Engineering yeast strains for use as chassis organisms in second-generation (2G) bioethanol is a promising strategy to improve process economics. Natural isolates of Saccharomyces cerevisiae offer strain backgrounds with greater genetic diversity and enhanced robustness, with the potential for improved heterologous protein production capabilities. In this study, heterologous cellulase production using different expression strategies was evaluated in various process-relevant conditions. Enhanced cellulolytic activity was clearly demonstrated in a cell-tethered enzyme system, compared to a free

Validation of assay for measuring acetyl-coenzyme a carboxylase activity in grasses using malachite green.

Acetyl-CoA carboxylase (ACCase) is one of the most important enzymes as a herbicide target in gramineous plant species, however, assay methods for the enzyme are primarily limited to those using radioisotopes (RI). Typically, the measurement method that uses RI necessitates specialized facilities and equipment, and involves complex procedures throughout the experiment. As another method for detecting ACCase activity, the colorimetric method using malachite green (MG) is also known. However, reports on this method are limited, and information regarding the simplicity of the procedure and the sc

Evaluation of oxidative stress and mitochondrial function in a type II mucopolysaccharidosis cellular model: in vitro effects of genistein and coenzyme Q10.

Mucopolysaccharidosis type II (MPS II or Hunter Syndrome) is a lysosomal disease caused by deficient degradation of glycosaminoglycans (GAGs) heparan sulfate and dermatan sulfate due to the deficiency of the enzyme iduronate-2-sulfatase. The main treatment for MPS II is the administration of the recombinant form of the enzyme, in a process known as enzyme replacement therapy (ERT). Oxidative damage can contribute to the pathophysiology of MPS II and treatment with ERT can reduce the effects of oxidative stress. For a better understanding of pathophysiology of MPS II, we evaluated biomarkers of

Enzyme replacement therapy in a murine model of Morquio A syndrome.

Mucopolysaccharidosis IVA (MPS IVA) is an autosomal recessive disorder caused by a deficiency of N-acetylgalactosamine-6-sulfate sulfatase (GALNS), leading to accumulation of keratan sulfate (KS) and chrondroitin-6-sulfate. The pharmacokinetics and biodistributions were determined for two recombinant human GALNSs produced in CHO cell lines: native GALNS and sulfatase-modifier-factor 1 (SUMF1) modified GALNS. Preclinical studies of enzyme replacement therapy (ERT) by using two GALNS enzymes were performed on MPS IVA mice. The half-lives in blood circulation of two phosphorylated GALNS enzymes w

Antihypertensive effect of perindopril: experimental pharmacology.

This review of the pharmacological effects of perindopril based on data in the literature, consists of three approaches: i) demonstration of the antihypertensive effect; ii) characterization of converting enzyme inhibition; iii) understanding of the mechanisms of action. The antihypertensive effect of perindopril has been demonstrated in the spontaneously hypertensive rat and in renovascular hypertension (1 clip-2 kidney or 1 clip-1 kidney). Its intensity and duration are dose-dependent; they are increased in the presence of stimulation of the renin-angiotensin system (renovascular hypertensio

Discovery of Selective 17β-HSD13 Inhibitors Containing a Phenol Isostere.

Hydroxysteroid 17-β-dehydrogenase 13 is a liver-specific, lipid-droplet enzyme genetically linked to reduced risk of metabolic-associated steatotic liver disease (MASLD), making it a compelling therapeutic target for metabolic dysfunction-associated steatohepatitis. Herein, we describe optimization of a series of small-molecule 17β-HSD13 inhibitors originating from a carboxylic acid hit identified in a high-throughput screen. Advancement to a phenol-based series of interest showed potent biochemical activity but poor pharmacokinetic profiles. Discovery efforts focused on replacement of the met

Structural insight into the CUB2 domain's role in enteropeptidase-mediated trypsinogen activation.

Elucidating the structure and function of enteropeptidase (EP) is essential for advancing our understanding of its biological significance, particularly in regulating trypsinogen activation. Using cryo-EM and enzymatic activity, we uncovered the significance of the CUB2 domain in mediating the cleavage of macromolecular substrates. We identified crucial binding loops and key residues for EP's proteolytic function. The mutation E574A enhanced the proteolytic activity of EP, whereas the mutation N619A diminished its cleavage efficiency, highlighting the importance of surface-charged interactions

Multi-level mechanistic insights into phytone bioactivity: the healthy aging characteristics of C. elegans, molecular interactions and enzyme inhibition.

Phytone, an important terpene in fermented foods, has anti-aging potential. This study explored its interactions with α-amylase (αAmy), pepsin (PEP), trypsin (TPS), human serum albumin (HSA), and anti-aging effects via Caenorhabditis elegans (C. elegans). In nematodes, it prolongs lifespan, increases pharyngeal pump rate (14.98%), reduces heat stress mortality (10.10%), and preserves reproductive capacity. At the same time, it can significantly enhance the activity of SOD, CAT, and GSH-Px. Multispectral analysis and molecular simulation confirmed phytone forms stable complexes with these prote

Development of a first-in-class homolog-selective, covalent NOX4 inhibitor.

The NADPH oxidase (NOX) family in humans includes seven enzymes whose main function is to produce reactive oxygen species. Because of their role in oxidative damage, these enzymes are being explored as targets for new therapies against oxidative stress-related diseases. However, their conserved catalytic core poses selective inhibition challenging. Current inhibitors suffer from low potency, cytotoxicity, assay interference, and poor selectivity. VAS2870, a covalent NOX inhibitor targeting a cysteine in the dehydrogenase (DH) domain, does not present assay interference but still shows limited

Protein arginine methyltransferase 3: Mechanism of action in diseases and modulator discovery.

Protein arginine methyltransferase 3 (PRMT3) is a key member of the PRMT family that catalyzes the arginine methylation of proteins, thereby modulating their structure, localization, or interactions and influencing essential physiological processes, such as cell growth and signal transduction. Its dysregulation is closely associated with tumorigenesis and neurological disorders. PRMT3 is critically involved in cellular processes, such as cell growth, invasion, and apoptosis, making it a promising target for drug discovery. In this review, we summarized the structure and biological functions of

Oxime ester-linked N-phenylphthalimide derivatives as PPO inhibitors: structure-activity relationships, herbicidal activity, crop selectivity, and in silico safety assessment.

Protoporphyrinogen oxidase (PPO; EC 1.3.3.4) is an established herbicide target, yet resistance and crop-injury concerns continue to motivate new PPO-inhibiting chemotypes. Here, forty-eight oxime ester-linked N-phenylphthalimide derivatives bearing five-membered heterocycles were designed by molecular hybridization and synthesized. Greenhouse assays identified compound a6 as a lead with broad-spectrum post-emergence activity, including >90% inhibition of Ipomoea nil and Abutilon theophrasti at 37.5 g a.i. ha-1, and it provided 85% control at 14 days after treatment in a field trial at 300 g a

Recent advances in the development of small-molecule drugs based on covalent reversible inhibitors.

Reversible covalent inhibitors (RCIs) are a class of small-molecule drugs that engage target proteins through covalent bonds. Unlike irreversible covalent inhibitors, RCIs can dissociate from their target. This reversibility, combined with high selectivity, arises from a binding mechanism that requires both the spatial conformation of the target protein and the reactivity of its residues. Serving as a bridge between traditional non-covalent inhibitors and irreversible covalent inhibitors, RCIs have attracted considerable attention in drug discovery, particularly for kinases, proteases, and oth

Gram-Scale Synthesis of an Arylsulfonamide-Type Alkaline Phosphatase Inhibitor.

Alkaline phosphatases (ALP) are present in most living organisms. This family of metalloenzymes catalyzes transphosphorylation reactions and hydrolyzes phosphate monoesters. ALP enzymes have significant roles in several physiological processes and disease states. Inhibition of these enzymes makes it possible to prevent or treat certain diseases, and their role in normal physiology can also be examined. Several inhibitors with diverse chemical structures have been reported. One of them, known as SBI-425, has a low IC50 value (16 nM). An alternative, scalable method was established to prepare SB

Hydroxamate-based inhibitors reveal structural determinants of selectivity for Plasmodium falciparum aminopeptidase P.

The malarial parasite, Plasmodium falciparum (Pf), utilizes aminopeptidases in the breakdown of hemoglobin-derived oligopeptides to release amino acids for protein synthesis during growth and asexual reproduction of erythrocytic stages of the parasite. However, a N-terminal peptide bond that involves proline is difficult to hydrolyze. Aminopeptidase P (APP) is capable of cleaving peptide bonds with proline in the second position. Inhibition of PfAPP is therefore an attractive strategy for developing therapeutics for the treatment of malaria by limiting the supply of amino acids at the erythroc

Activation of protein kinase C decreases equilibrative nucleobase transporter 1-mediated substrate uptake via phosphorylation of threonine 231.

Protein kinase C (PKC) signalling has been shown to be dysregulated in various cancers including acute lymphoblastic leukemia (ALL). We have previously determined that changes in the expression levels of SLC43A3-encoded equilibrative nucleobase transporter 1 (ENBT1) can significantly alter 6-mercaptopurine (6-MP) toxicity in ALL cells. 6-MP is a common drug used in ALL chemotherapy. Furthermore, it has been reported that activation of PKC by phorbol 12-myristate 13-acetate (PMA) impacts nucleobase uptake via an ENBT1-like transporter in Lilly Laboratories Culture-Porcine Kidney 1 (LLC-PK1) cel

Design, synthesis, and anti-urease evaluations of new sulfonamide-1,2,3-triazole-acetamide derivatives.

The present study demonstrated the design and synthesis of sulfonamide-1,2,3-triazole-acetamide derivatives 11a-o and screening against urease in vitro and in silico. These compounds were designed based on reported potent urease inhibitors and optimized structurally based on substituents on acetamide moiety. In vitro studies showed that all the new compounds 11a-o (IC50 values = 0.12-4.53 µM) were more potent than stand inhibitor thiourea (IC50 value = 23.76 µM). In this regard, the most potent compounds were N-phenylacetamide derivatives 11b, 11f, and 11 h with 2-methyl, 4-methoxy, and 2-fluo
