Healthy lifestyle reduces cardiovascular risk in women with genetic predisposition to hypertensive disorders of pregnancy.

The genetic risk for hypertensive disorders of pregnancy is linked with the development of atherosclerotic cardiovascular disease. However, the effects of lifestyle and metabolic syndrome on atherosclerotic cardiovascular disease have not been evaluated. Here, we assess the long-term association between these factors and atherosclerotic cardiovascular disease in women with genetic risk for hypertensive disorders of pregnancy. We evaluate the genetic risk for hypertensive disorders of pregnancy using a genome-wide polygenic risk score derived from a large-scale GWAS. The incidence of atheroscle

Exploring the causal associations of the gut microbiota and plasma metabolites with ovarian cancer: an approach of mendelian randomization analysis combined with network pharmacology and molecular docking.

BACKGROUND: While increasing evidence suggests that alterations in the gut microbiota and metabolites are associated with ovarian cancer (OC) risk, whether these associations imply causation remains to be identified. METHODS: We conducted a two-sample Mendelian randomization (MR) study utilizing a large-scale genome-wide association study (GWAS) to explore the causal effects of the gut microbiota of 196/220 individuals and 1,400 plasma metabolites on OC and epithelial ovarian cancer (EOC) subtypes. Data on the gut microbiota were obtained from the MiBioGen consortium of 18,340 subjects and th…

Genome-wide analysis of screen behaviors among adolescents identifies novel loci and overlap with educational attainment and mental disorders.

Technological devices play a central role in adolescents' life. Despite concerns about negative effects of excessive screen time, there is little knowledge of screen behaviors' genetic architecture. Using self-reports from adolescents in the Norwegian Mother, Father, and Child Cohort Study (n = 18,490), we performed genome-wide association analysis for four screen behaviors: time spent (1) watching television; (2) gaming; (3) sitting/lying down with a screen device; and (4) using social media. The resulting summary statistics were analysed using the conditional false discovery rate (condFDR) a

Exploring the Protein-Metabolite Interplay to Discover Novel Drug Targets for Alzheimer's Disease.

BACKGROUND: The protein-metabolite interactions play a pivotal role in the pathogenesis of Alzheimer's disease (AD); however, the causal significance of these interactions in AD pathogenesis remains elusive. METHODS: We integrated datasets of protein quantitative trait loci (pQTL), metabolite quantitative trait loci (mQTL), and genome-wide association study (GWAS) for AD. Initially, we performed MR analysis to identify proteins and metabolites causally associated with AD risk. To guarantee the robustness of these findings, Bayesian colocalization analysis was employed to confirm shared geneti…

A Mendelian Randomization Study on the Association Between Sex Hormone-Binding Globulin and Membranous Nephropathy Risk.

AIM: Membranous nephropathy (MN) is the leading cause of nephrotic syndrome in adults, with 80% of cases being primary MN of unknown origin. While the influence of sex hormones on chronic kidney disease (CKD) is thoroughly established, the role of sex hormone-binding globulin (SHBG) in MN is still uncertain. METHODS: We performed a Mendelian randomization (MR) analysis to assess the causal impact of SHBG on MN, utilising data from genome-wide association studies (GWAS). The main analysis utilised the inverse variance weighted (IVW) method, while various additional and sensitivity analyses wer

Genetic insights into the relationship between immune cell traits and abnormal uterine bleeding: A Mendelian randomization study.

Abnormal uterine bleeding (AUB) is an inflammatory response involving immune cells, but the relationship between immune cell traits and AUB is highly complex and still largely unclear. This study utilized genetic data from a genome-wide association study of European participants. Mendelian randomization (MR) analysis methods, including inverse variance weighted (IVW) as the primary approach, weighted median, MR Egger regression, and Mendelian randomization pleiotropy residual sum and outlier, were employed for forward and reverse analyses. Sensitivity analyses validated the stability and relia…

Immune cell-specific gene expression and its causal role in osteoporosis and bone mineral density: Insights from single-cell eQTL and GWAS data integration.

Osteoporosis (OP) is a common metabolic bone disease, with genetic and immune system factors playing crucial roles in its pathogenesis. With the advancement of single-cell RNA sequencing (scRNA-seq), gene expression regulation at the immune cell subtype level has been more deeply explored. In this study, we integrated single-cell expression quantitative trait loci data with genome-wide association study data to systematically investigate the causal relationships between immune cell-specific gene expression and OP risk/ bone mineral density (BMD). Through summary-data-based Mendelian randomizat…

Causal relationship between sarcopenia and cognitive performance: A bidirectional Mendelian randomization study.

With the acceleration of global population aging, sarcopenia (SA) and cognitive impairment have become major public health concerns. Observational studies suggest associations between them, but such findings may be biased by confounding and reverse causation. Previous Mendelian randomization (MR) studies have yielded inconsistent results. Therefore, this study applied a bidirectional MR design to clarify their causal relationships. We obtained genetic instrumental variables for SA-related traits, including hand grip strength, lean mass, and usual walking pace, from the UK Biobank. Data on cogn

Copper and iron as unique trace elements linked to fibromyalgia risk.

Fibromyalgia (FM) is a prevalent chronic pain condition with a complex and not fully understood etiology. Abnormal metabolism of trace elements is suspected to play a role in the pathogenesis of FM, though the exact relationships have yet to be clarified. This study employed Mendelian randomization (MR) to assess potential causal relationships between 15 major trace elements and the risk of FM, focusing on the specific roles of elements that show significant associations. Genetic instrumental variables (single nucleotide polymorphisms, SNPs), related to these trace elements and FM were extract

Integrating brain proteomes and genetics to identify novel risk genes in chronic widespread musculoskeletal pain.

Chronic widespread musculoskeletal pain (CWP), a core symptom of fibromyalgia, is a complex condition with an unclear pathogenesis. Despite findings from genome-wide association studies (GWAS), translating genetic risk variants into therapeutic targets remains challenging due to limited understanding of their functional roles in CWP. In this study, we developed an integrative analytical pipeline to efficiently link genetic associations with novel risk genes for CWP. By combining high-throughput data from multiple sources, we integrated proteome-wide association studies (PWAS), transcriptome-wi

Joint analysis of genome-wide cross-trait and multi-omics reveals molecular mechanisms of inflammatory bowel disease and nominates its novel therapeutic genes.

Inflammatory bowel disease (IBD) with the two predominant endophenotypes-Crohn's disease (CD) and ulcerative colitis (UC)-represents a group of chronic gastrointestinal inflammatory conditions. Since most genetic associations with IBD are often limited to independent subtypes, we reported a genome-wide association study (GWAS) cross-trait analysis combined with CD and UC to enhance statistical power. Initially, we detected 256 association signals at 54 genomic susceptibility loci and further characterized the functionality of variants within these regions. Subsequently, we revealed tissue and …

Relationship of obesity, body fat, benign adrenal tumors and the mediating mechanism: a two-step mendelian randomization study.

BACKGROUND: Benign adrenal tumors comprise the majority of asymptomatic adrenal masses and are often associated with cortisol secretion, which increases the risk of obesity and metabolic syndrome. Hormone secretion by these tumors may confound prevailing epidemiologic findings, and the causal relationships among obesity, body fat, and benign adrenal tumors remain uncertain. Mendelian randomization (MR) uses genetic variation as an instrumental variable to simulate randomized controlled trials, thereby reducing confounding and supporting causal relationships. Therefore, we aim to use MR methods
