Patient-convenient long-term alopecia treatment via PLGA microsphere-loaded candlelit microneedles.

Androgenetic alopecia (AGA) is characterized by chronic and progressive hair loss, with associated psychological factors intensifying the impact on patients. Current treatments, such as oral finasteride and topical minoxidil, have low bioavailability and numerous side effects. Dissolvable microneedles (DMNs) provide a promising alternative for drug delivery. However, the presence of hair on the scalp often hinders their insertions and adhesion. Thus, candlelit microneedles (CMNs) have been developed to improve insertion and drug delivery without the use of adhesive patches. In this study, CMNs

Immunogenicity-Guided Design of an Acinetobacter baumanii Vaccine.

The development of vaccines represents a promising and safe strategy to combat multidrug-resistant (MDR) Acinetobacter baumannii (A. baumannii) infections. In this study, we designed and evaluated a dendritic cell (DC)-targeting multiepitope peptide-based biomimetic nanovaccine for its immunogenicity and protective efficacy in a murine model. Bioinformatics tools were employed to predict and screen B- and T-cell epitopes derived from the OmpW protein of A. baumannii, followed by immunological validation. The dominant epitopes were sequentially linked using 6-aminocaproic acid to synthesize a m

Rectal sunflower seed bezoar in an 11-year-old: A case report.

Sunflower seed bezoars are an indigestible mass created after consuming several unshelled sunflower seeds. We present the case of an 11-year-old who visited the emergency department with abdominal pain, constipation, and rectal bleeding. The rectal exam findings were consistent with a presumed stool ball. Symptoms persisted even after enema and polyethylene glycol administration. The patient was discharged, and after an attempted manual self-disimpaction at home, the patient's mother noted sunflower seed shell remnants in the stool. Upon further questioning, the patient endorsed consuming 10 o

Interface Design in Bimetallic PdNi Nanowires for Boosting Alcohol Oxidation Performances.

The rational design of a bimetallic nanostructure with a phase separation and interface is of great importance to enhance electrocatalytic performance. Herein, PdNi heterostructures with controlled elemental distributions were constructed via a seeded growth strategy. Partially coated Ni islands in the Pd-Ni nanowire and strained Pd branches in the Pd-NiPd nanowires are revealed, respectively. Impressively, Pd-NiPd nanowires with abundant branches exhibit a superior mass current density and cycling stability toward an ethanol oxidation reaction (EOR) and ethylene glycol oxidation reaction (EGO

Synthesis and optimization of a 2-isopropenyl-2-oxazoline based polymer for enhanced phenolic acid extraction.

Phenolic acids, widely present in plants, plant-based foods, and human metabolites, are well known for their potent antioxidant properties, which are mainly attributed to their phenolic hydroxyl groups. The number and position of these hydroxyl groups, together with methoxy and carboxylic acid groups significantly influence their antioxidant capacity. In this study, five different phenolic acids were studied: chlorogenic acid, caffeic acid, 2,3-dihydroxybenzoic acid, ferulic acid and trans-cinnamic acid. They were analyzed by solid phase extraction and high-performance liquid chromatography hy

Crowding beyond excluded volume: A tale of two dimers.

Protein-protein interactions are modulated by their environment. High macromolecular solute concentrations crowd proteins and shift equilibria between protein monomers and their assemblies. We aim to understand the mechanism of crowding by elucidating the molecular-level interactions that determine dimer stability. Using 19F-NMR spectroscopy, we studied the effects of various polyethylene glycols (PEGs) on the equilibrium thermodynamics of two protein complexes: a side-by-side and a domain-swap dimer. Analysis using our mean-field crowding model shows that, contrary to classic crowding theorie

Degradable Ureido-Polycarbonate Block Copolymers with a Complex UCST Thermoresponse.

In this work, amphiphilic block copolymers (BCs) consisting of a hydrophilic poly(ethylene glycol) methyl ether (PEG) and a degradable polycarbonate block derived from 2,2-bis(hydroxymethyl)propionic acid (bis-MPA) with pendant ureido units, along with corresponding homopolycarbonates are described. Polymers are synthesized by combining ring opening polymerization (ROP) and thiol-ene/yne functionalization to incorporate UCST-promoting ureido groups. For homopolycarbonates, increasing the ureido groups density along the polymer chain facilitates the upper critical solution temperature (UCST)-ty

Understanding how genetically encoded tags and crowding agents affect phase separation by heterochromatin protein HP1α.

The heterochromatin protein HP1α (heterochromatin protein 1 alpha) phase separates in vitro and displays properties compatible with phase separation in cells. Phase separation of HP1α in cells is typically studied using genetically encoded fluorescent tags such as green fluorescent protein (GFP). Whether such tags affect the intrinsic phase separation properties of HP1α is understudied. We assessed how tag size and linker length affect phase separation by HP1α in vitro. GFP tags inhibited phase separation by HP1α. In contrast, an UnaG tag with a 16 amino acid glycine-glycine-serine (GGS) linke

Sonochemical degradation of CCl4 in alcohol-water system.

In this study, methanol (MeOH), ethanol (EtOH), 1-propanol (PrOH), and ethylene glycol (EG) were used as alcohol-CCl4 pretreatment solutions for the first time to investigate the effect of alcohol on the sonochemical degradation of CCl4. The results showed that this pretreatment method provided the homogeneous dissolution of CCl4 in aqueous solution in a short preparation time and the enhancement of the sonochemical degradation of CCl4. In addition, the rate of the degradation of CCl4 related to the hydrophobicity of the added alcohol. It was considered that high hydrophobic alcohol molecules

Matrix directs trophoblast differentiation in a bioprinted organoid model of early placental development.

Trophoblast organoids can provide crucial insights into mechanisms of placentation, however their potential is limited by highly variable extracellular matrices unable to reflect in vivo tissues. Here, we present a bioprinted placental organoid model, generated using the first trimester trophoblast cell line, ACH-3P, and a synthetic polyethylene glycol (PEG) matrix. Bioprinted or Matrigel-embedded organoids differentiate spontaneously from cytotrophoblasts into two major subtypes: extravillous trophoblasts (EVTs) and syncytiotrophoblasts (STBs). Bioprinted organoids are driven towards EVT diff

Evaluating Cross-Linking Efficiency and Cytocompatibility of Three Commonly Used Photoinitiators across Different Cell-Compatible Hydrogel Platforms.

Biomedical hydrogels often use a photopolymerization strategy to cross-link the polymer network. There are only a few cyto-compatible photoinitiators (PIs) that are commonly used for cross-linking biomedical hydrogels, including Irgacure 2959, lithium phenyl-2,4,6-trimethylbenzoylphosphinate (LAP), and Eosin Y. Herein, we tested these PIs to optimize the cross-linking efficiency while minimizing cell death. Testing was performed on three types of hydrogels, including a synthetic material (poly(ethylene glycol)-diacrylate, PEG-DA), a semisynthetic material, PEG-fibrinogen (PF), and a modified b

Characterization of Sex-Based Differences in Integrin-Mediated Endothelial Cell Adhesion to Bioactive Hydrogels.

Endothelialization promotes thromboresistance in blood-contacting devices, but biomaterial designs often overlook sex differences in the endothelialization processes. In this study, we elucidated sex differences in endothelial cell-material interactions through an investigation of the integrin-ligand interplay with biomaterial substrates and the corollary effects on cell adhesion and spreading. First, integrin expression of human coronary artery endothelial cells (HCECs) was characterized for age-matched donors (3 male, 3 female). Sex-based differences in integrin expression were identified, w

Polyzwitterion-Based pH-Responsive Shell of Polymeric Micelle: Evaluation of Physicochemical Properties and In Vivo Functionalities.

As well-known nanocarriers for systemically injectable drug delivery systems, lipid-based polymeric micelles show potency for improving cancer treatment. Polyethylene glycol (PEG) is commonly used as a component of polymeric micelles owing to its biocompatibility but can cause immunogenic side reactions, which highlights the need for non-PEG-based systems for the delivery of therapeutic agents. To address this need, we herein synthesized a poly(N-{N'-[N″-(2-carboxyethyl)-2-aminoethyl]-2-aminoethyl}glutamide) [PGlu(DET-Car)]-lipid conjugate, prepared polymeric micelles with PGlu(DET-Car) surfac
