FAs and their metabolites as antibacterial agents: mechanisms, food applications, and translational opportunities.

Antimicrobial resistance and the demand for effective food-preservation strategies have increased interest in fatty acids (FAs) and their derivatives as potential antibacterial agents. FAs can affect bacterial membranes, cellular energetics, biofilm-associated phenotypes, and, in some organisms, specific metabolic pathways. However, these effects vary considerably with FA structure, concentration, ionization state, pH, formulation, bacterial species, and assay conditions. Additionally, microbiota-derived short-chain FAs may also contribute to host defense by influencing epithelial integrity, i

Beyond H5N1: influenza A virus infection in bovine udder organoids.

Influenza A viruses (IAVs) have recently expanded their host range to bovine species, raising critical questions about tissue tropism, viral replication capacity, and impacts on dairy production and zoonotic risk. Extensive shedding of highly pathogenic avian influenza virus (HPAIV) H5N1 in milk from naturally infected cattle in the USA highlights the bovine mammary gland as a relevant site of viral replication. To investigate these dynamics, we established and characterized bovine mammary gland organoids (MGOs) derived from primary epithelial cells and maintained in chemically defined media.

Synthetic Integration of an FCS into Coronaviruses-Hype or an Unresolved Biorisk? An Integrative Analysis of DNA Repair, Cancer Research, Drug Development, and Escape Mutant Traits.

A 19 nt fragment that spans the SARS-CoV-2 furin cleavage site (FCS) is identical to the reverse complement of a proprietary human DNA repair gene sequence. Rather than interpreting this overlap as evidence of a laboratory event, this article uses it as a theoretical springboard to explore underappreciated biorisk concerns, specifically in the context of cancer research. Although they are RNA viruses, coronaviruses are capable of hijacking host DNA damage response (DDR) pathways, exploiting nuclear functions to enhance replication and evade innate immunity. Under selective pressures (antiviral

IFI44 Functions as a Positive Regulator of Type I Interferon Signaling to Restrict Zika Virus Infection.

Zika virus (ZIKV) infection is known to cause microcephaly in newborns, and its outbreaks have previously emerged as a global public health crisis. The lack of a preventive vaccine or specific antiviral drugs underscores the urgency of investigating the detailed mechanisms of pathogenesis. We identified that interferon-induced protein 44 (IFI44) is significantly upregulated following ZIKV infection, but its role in ZIKV pathogenesis remains unclear. Using A549 and 2FTGH cells, we established ZIKV-infected cell models and employed quantitative real-time PCR and Western blotting to demonstrate t

Fungal pathogens of plants: deciphering the mechanisms of Fusarium wilt.

Fusarium oxysporum (Fo) is among the most dangerous soilborne pathogens, causing Fusarium wilt and root rots in over 100 plant species worldwide. Some pathogen strains can also infect immunocompromised animals and humans. Consequently, studying the molecular mechanisms associated with pathogen virulence and the plant immune response at different stages of disease development is of paramount importance. The process of host recognition by the pathogen and all stages of the infection process involve a wide repertoire of specific signaling molecules, effector proteins, receptor complexes, as well

The structural language of fungal cell wall sugars in WHO priority fungi: the determinant of pathogenicity and the gateway to new therapies.

Carbohydrates, far beyond their nutritional role, are fundamental biomolecules that encode biological information and mediate critical cellular interactions. In pathogenic fungi, cell wall (CW) glycans serve as both a protective armor and a dynamic interface with the host, directly influencing virulence and immune evasion. This review discusses the intricate architecture and composition of the CW in the four WHO-critical priority fungal pathogens: Aspergillus fumigatus, Candidozyma auris, Cryptococcus neoformans, and Candida albicans. We detail how distinct carbohydrate polymers, including α-

Human cytomegalovirus at the maternal-fetal interface: an overview of pathogenesis, defence, and interventions.

Human cytomegalovirus (HCMV) is the leading infectious cause of congenital disease worldwide and a major contributor to birth defects and neurodevelopmental disorders. Vertical transmission occurs when the virus breaches the maternal-fetal interface, a complex immunological environment where the placenta is in direct contact with either the uterine lining (decidua) or maternal blood covering the syncytium. The present review synthesises current understanding of HCMV pathogenesis at this critical interface, examining viral invasion mechanisms, host immune responses, and immune evasion strategie

Integrated multi-sample transcriptomic analysis of COVID-19 patients against controls using a bioinformatics pipeline.

Prior coronavirus disease 2019 (COVID-19) transcriptomic studies using diverse methods for differential gene expression (DGE) profiling of specific samples yielded inconsistent results. To validate the shared molecular patterns of COVID-19 across cell, tissue, and systemic levels, we conducted a systematic rank combination meta-analysis of differentially expressed gene (DEG) profiles sourced from various sample types using a standardised bioinformatics pipeline consisting of DESeq2, RankProd, and weighted gene correlation network analysis (WGCNA). Consistently upregulated ISGs (including key h

Tolerance Induction Strategies in Organ Transplantation: Current Status and Future Perspectives.

Achieving donor-specific immune tolerance has the potential to eliminate the need for lifelong immunosuppression in transplant recipients, but translating this goal into clinical practice remains challenging. Unlike laboratory rodents, humans are exposed to a variety of pathogens that generate memory T cells, which can interfere with tolerance induction. Establishing full donor hematopoietic chimerism, whether spontaneous or induced, can support robust immune tolerance. However, it often relies on graft-versus-host (GvH) reactivity, which carries significant risks, including graft-versus-host

Digamasellid mites (Acari: Mesostigmata) in association with saproxylic darkling beetles (Coleoptera: Tenebrionidae) and wood-decaying fungi (Polyporales) in Slovakia.

Three species of digamasellid mites adapted to life in wood-decaying polypore fungi and phoretically associated with saproxylic and fungivorous beetles of the family Tenebrionidae are described from adult and deutonymphal stages from forest habitats in Slovakia. Two of the mites are described as new species: Bulbolaelaps neomidae sp. nov., the second member of the genus, whose developmental stages were previously unknown, is specifically associated with the tinder fungus Fomes fomentarius (L.) Fr., and its deutonymphs are phoretically dispersed by the beetle Neomida haemorrhoidalis (Fabricius)

Hematological and biochemical profiles, infection and habitat quality in an urban rat population.

Host condition is key in understanding disease dynamics. In this study, we investigated the relationship between infection with Leptospira interrogans and helminths, and the hematological and hormone-biochemical stress-related profiles of urban Rattus norvegicus. Rat kidney imprints and urine were used to identify and quantify L. interrogans, while fecal samples were analyzed for helminth eggs and corticosterone metabolites. Blood samples were taken for complete blood counts and specific biochemicals in rats' sera. Principal Component Analyses were conducted to group rats according to health p

Activation and regulation of NAIP/NLRC4 and NLRP3 inflammasomes during Salmonella infection: mechanisms and therapeutic implications.

Inflammasomes constitute a critical component of host defense against Salmonella infection, among which the NAIP/NLRC4 and NLRP3 complexes serve as the two principal signaling platforms. The NAIP/NLRC4 inflammasome functions as a rapid-response sensor that specifically recognizes bacterial flagellin and T3SS components, leading to caspase-1 activation, GSDMD-mediated pyroptosis, and IL-1β/IL-18 maturation. In contrast, NLRP3 integrates broad cellular stress signals, such as K⁺ efflux, mitochondrial ROS, and lysosomal damage, to amplify and sustain inflammatory responses, particularly at later

Sulfur-oxidizing symbionts colonize the digestive tract of their lucinid hosts.

Like many marine invertebrates, marine lucinid clams have an intimate relationship with beneficial sulfur-oxidizing bacteria located within specialized gill cells known as bacteriocytes. Most previous research has focused on the symbionts in the gills of these (and other) symbiotic bivalves, often assuming that the symbionts only persistently colonize the gills, at least in the adult stage. We used 16S rRNA gene sequencing and digital polymerase chain reaction with symbiont-specific primers targeting the soxB gene on the foot, mantle, visceral mass, and gills of the lucinid clam Loripes orbicu

Quantification of the interaction forces between dengue virus and dopamine type-2 receptor using optical tweezers.

BACKGROUND: Dengue virus (DENV) causes the most significant mosquito-borne viral disease with a wide spectrum of clinical manifestation, including neurological symptoms associated with lethal dengue diseases. Dopamine receptors are expressed in central nervous system, and dopamine antagonists have been reported to exhibit antiviral activity against DENV infection in vivo and in vitro. Although identification of host-cell receptor is critical to understand dengue neuropathogenesis and neurotropism, the involvement of dopamine receptors in DENV infection remains unclear. RESULTS: We exploited t

A promising new target to control fasciolosis: Fasciola hepatica leucine aminopeptidase 2.

Parasite M17 leucine aminopeptidases (LAPs) have been associated with critical roles in different key functions such as the nutrition, migration, and invasion of the natural host. Native or recombinant LAP used as a vaccine antigen has proved effective to elicit protection against Fasciola hepatica infection in sheep, pointing to potential vaccine candidates against fascioliasis in ruminant species. Previously, the FhLAP1, abundantly secreted in vitro by the mature adult parasite was used as a vaccine antigen obtaining promising protection results against F. hepatica challenge in small ruminan

A hepatitis B virus (HBV) sequence variation graph improves sequence alignment and sample-specific consensus sequence construction for genetic analysis of HBV.

Hepatitis B virus (HBV) remains a global public health concern, with over 250 million individuals living with chronic HBV infection (CHB) and no curative therapy currently available. Viral diversity is associated with CHB pathogenesis and immunological control of infection. Improved methods to characterize the viral genome at both the population and intra-host level could aid drug development efforts. Conventionally, HBV sequencing data are aligned to a linear reference genome and only sequences capable of aligning to the reference are captured for analysis. Reference selection has additional

Memory CD8 T Cells Protect against Cytomegalovirus Disease by Formation of Nodular Inflammatory Foci Preventing Intra-Tissue Virus Spread.

Cytomegaloviruses (CMVs) are controlled by innate and adaptive immune responses in an immunocompetent host while causing multiple organ diseases in an immunocompromised host. A risk group of high clinical relevance comprises transiently immunocompromised recipients of hematopoietic cell transplantation (HCT) in the "window of risk" between eradicative therapy of hematopoietic malignancies and complete reconstitution of the immune system. Cellular immunotherapy by adoptive transfer of CMV-specific CD8 T cells is an option to prevent CMV disease by controlling a primary or reactivated infection.

A Vaccine Strategy Based on the Identification of an Annular Ganglioside Binding Motif in Monkeypox Virus Protein E8L.

The recent outbreak of Monkeypox virus requires the development of a vaccine specifically directed against this virus as quickly as possible. We propose here a new strategy based on a two-step analysis combining (i) the search for binding domains of viral proteins to gangliosides present in lipid rafts of host cells, and (ii) B epitope predictions. Based on previous studies of HIV and SARS-CoV-2 proteins, we show that the Monkeypox virus cell surface-binding protein E8L possesses a ganglioside-binding motif consisting of several subsites forming a ring structure. The binding of the E8L protein
