Targeting redox metabolism and low-dose radiotherapy synergistically activate cGAS-STING pathway to improve NK cell therapy efficacy in hepatocellular carcinoma.

Adoptive natural killer (NK) cell therapy (ANKCT) is a promising strategy for hepatocellular carcinoma (HCC); however, its efficacy is hampered by insufficient NK cell homing and the immunosuppressive activity of M2-polarized tumor-associated macrophages (TAMs). Activation of the cyclic guanosine monophosphate (GMP)-adenosine monophosphate synthase (AMP) synthase-stimulator of interferon genes (cGAS-STING) pathway enhances NK-cell recruitment and reprograms TAMs toward a proinflammatory M1 phenotype. Radiation therapy (RT) activates the cGAS-STING pathway by inducing reactive oxygen species (R

Bioinspired enzyme-catalytic nanoreactor enhances immunotherapy for spinal metastases by activating pyroptosis and the cGAS-STING pathway.

Bone is one of the most common sites for tumor metastasis. The "seed-and-soil" relationship renders bone tissue a favorable microenvironment for the growth of circulating tumor cells. While immunotherapies, particularly immune checkpoint blockade (ICB), have achieved breakthroughs in primary solid tumors, bone metastases often respond poorly to ICB treatment. Herein, we developed an enzyme-loaded, self-cascading nanoreactor (mCL) that integrates a CaO2 core and l-Arginine (L-Arg) with iNOS-rich macrophage membranes. This design ensures efficient tumor targeting and, upon acid-triggered decompo

Intracellular ultrastructural characterization of Azadinium and Amphidoma (Amphidomataceae, Dinophyceae), including toxigenic and non-toxigenic strains.

The marine toxigenic dinoflagellate family Amphidomataceae, comprising Amphidoma and Azadinium, has been primarily characterized by thecal morphology examined by SEM. To further clarify the detailed morphology, the intracellular ultrastructure of five Azadinium species, encompassing two toxigenic species (Azadinium poporum and Az. spinosum) and three non-toxigenic species (Az. inconspicuum, Az. dalianense, and Az. zhuanum), was examined using TEM. The ultrastructure of two Amphidoma species was also examined for comparison. All species exhibited typical dinoflagellate organelles, i.e., nucleus

Tumor Microenvironment-Responsive Biomimetic Self-Assembled Nanomedicine for Precision Combination Therapy against Triple-Negative Breast Cancer.

Although strategic combination of cuproptosis and chemotherapy is emerging as a promising strategy against triple-negative breast cancer (TNBC), current drug delivery systems remain considerable challenges in achieving co-delivery of different formulas, such as complex nanocarrier design, limited drug loading capacity, and insufficient tumor targeting. Herein, cancer cell membrane-camouflaged, self-assembled nanoparticles (DCM@CCM) were fabricated for precision combination therapy against TNBC. In the strategy, the carrier-free self-assembled nanoparticles were one-pot fabricated by co-assembl

Gadolinium impairs male steroidogenesis: In vivo and in vitro evidence.

The increasing use of gadolinium (Gd) in industrial and medical fields made it a hazardous environmental pollutant. Once ingested through water and/or food, Gd may potentially have toxic effects on all body districts. However, the effects of Gd on testicular function have been little explored. In the present study, adult male rats were allowed to drink GdCl3 or Gd2O3 (10-20-40 mg/Kg b.w.) for 4 weeks. Following Gd treatment, a significant decrease in steroidogenic-related protein (StAR, 3β-HSD, 17β-HSD, and 5α-Red) expressions, T and DHT levels, and spermatozoa concentration were observed. To

Synuclein and Mitochondrial Dysfunction: Regulating the Protein Import Complex toward PD Treatment?

Parkinson's disease (PD) is a chronic, progressive neurodegenerative disorder characterized by severe motor symptoms. While the degeneration of dopaminergic neurons in the substantia nigra plays a central role, other neurotransmitter systems also contribute to PD symptoms. α-Synuclein (αSyn), normally expressed in neurons to support synaptic function and neurotransmitter release, becomes pathologically accumulated in PD, despite not being upregulated under physiological conditions. Intracellular aggregation of αSyn into Lewy bodies is a hallmark of synucleinopathies. A vital facet of both the

Proteomic Insights into Small Molecule Modulation on Caudal Fin Regeneration in Zebrafish.

Dietary and addictive small molecules play a significant role in altering in vivo conditions. Due to their minuscule size, these molecules can seamlessly traverse tissues and cellular membranes, influencing key biological processes such as cellular growth, differentiation, and intracellular communication, which are crucial for tissue regeneration. The zebrafish serves as an excellent model for studying regenerative growth due to its remarkable ability to regrow amputated appendages. In this study, we systematically evaluated the effect of small molecules, including ethanol, glucose, and NaCl,

Dendrobine attenuates lipopolysaccharide-induced acute lung injury by modulating FAM134B-mediated endoplasmic reticulum autophagy and mitochondrial function.

BACKGROUND: Macrophage-mediated excessive inflammatory responses play a pivotal role in sepsis progression, particularly in the lungs. Endoplasmic reticulum stress (ERS) and mitochondrial dysfunction, also contribute to the development of acute lung injury (ALI). Dendrobine is a natural alkaloid known to alleviate ERS in various diseases; however, its specific role in sepsis-associated ALI is not yet fully understood. OBJECTIVE: This study investigates the protective effects of dendrobine against ALI and elucidates the underlying mechanisms. METHODS: We assessed the protective effects of den

Insights into platelet factor 4-derived peptide macrocycles; the mechanistic basis of their rapid and selective antiplasmodial actions.

The malarial parasite Plasmodium can acquire resistance to most mainstay antimalarial drugs, necessitating the development of new antiplasmodial agents with different modes of action. The innate defense protein, human platelet factor 4 (PF4), has a unique antiplasmodial action that involves selective entry into Plasmodium-infected red blood cells (RBC) and subsequent destruction of the parasite's digestive vacuole (DV). This activity is recapitulated in PF4-derived internalization peptides (PDIPs). Here, we characterized the actions of PDIP analogs and PF4 in live P. falciparum-infected human

UV-based homogeneous disinfection process for removal of antibiotic resistance genes: Efficiency, mechanisms and influencing factors.

The proliferation and dissemination of antibiotic resistance genes (ARGs) in aquatic environments pose a serious threat to global public health. Ultraviolet-driven homogeneous advanced oxidation processes (UV-AOPs) represent a prospective suite of technologies for the efficient removal of ARGs. This review critically assesses recent advances in the application of UV-AOPs, specifically UV/hydrogen peroxide (UV/H2O2), UV/peracetic acid (UV/PAA), UV/persulfate (UV/PS), and UV/chlorine (UV/Cl), for the elimination of extracellular ARGs and intracellular ARGs. The underlying mechanisms involve dire

Self-Assembling Antioxidant Nanoparticles Increase Survival and Reduce Disease Symptoms of Severe Malaria.

Severe infectious diseases trigger a rapid increase in cytokines, known as a cytokine storm, accompanied by an excessive rise in reactive oxygen species (ROS), leading to severe damage to tissues and cells. While antioxidants have been used to eliminate ROS, conventional low-molecular-weight (LMW) antioxidants fail to effectively mitigate oxidative stress in infectious diseases due to rapid metabolism, excretion, and potential disruption of the redox balance within normal cells. We developed a novel self-assembling antioxidant nanoparticle, termed a redox nanoparticle (RNP), designed to preven

Biomimetic Nanoplatform Based on the Neutrophil Membrane for Targeted Therapy of Spinal Cord Injury.

INTRODUCTION: Spinal cord injury (SCI) triggers a cascade of secondary damage, including oxidative stress and neuroinflammation, for which effective treatments remain limited. To address this, we developed a biomimetic nanoplatform, NEU@MPBNPs-HED, consisting of mesoporous Prussian blue nanoparticles (MPBNPs) loaded with hederagenin (HED) and cloaked with neutrophil membranes (NEUm) to enhance lesion targeting and immune evasion. METHODS: The physicochemical properties, drug release characteristics, and cellular uptake of NEU@MPBNPs-HED were characterized. Therapeutic efficacy was evaluated i

Antimicrobial efficacy of Punica granatum Lythraceae peel extract against pathogens belonging to the ESKAPE group.

The improper use and abuse of antibiotics have led to an increase in multidrug-resistant (MDR) bacteria resulting in a failure of standard antibiotic therapies. To date, this phenomenon represents a leading public health threat of the 21st century which requires alternative strategies to fight infections such as the identification of new molecules active against MDR strains. In the last 20 years, natural extracts with biological activities attracted scientific interest. Following the One Health Approach, natural by-products represent a sustainable and promising alternative solution. Consistent

5-HTP inhibits eosinophilia via intracellular endothelial 5-HTRs; SNPs in 5-HTRs associate with asthmatic lung function.

BACKGROUND: Previous research showed that 5-hydroxytryptophan (5HTP), a metabolic precursor of serotonin, reduces allergic lung inflammation by inhibiting eosinophil migration across endothelial monolayers. OBJECTIVE: It is unknown if serotonin receptors are involved in mediating this 5HTP function or if serotonin receptor (HTR) single nucleotide polymorphisms (SNPs) associate with lung function in humans. METHODS: Serotonin receptor subtypes were assessed by qPCR, western blot, confocal microscopy, pharmacological inhibitors and siRNA knockdown. HTR SNPs were assessed in two cohorts. RESUL

Palmitoylation-dependent regulation of cardiomyocyte Rac1 signaling activity and minor effects on cardiac hypertrophy.

S-palmitoylation is a reversible lipid modification catalyzed by 23 S-acyltransferases with a conserved zinc finger aspartate-histidine-histidine-cysteine (zDHHC) domain that facilitates targeting of proteins to specific intracellular membranes. Here we performed a gain-of-function screen in the mouse and identified the Golgi-localized enzymes zDHHC3 and zDHHC7 as regulators of cardiac hypertrophy. Cardiomyocyte-specific transgenic mice overexpressing zDHHC3 show cardiac disease, and S-acyl proteomics identified the small GTPase Rac1 as a novel substrate of zDHHC3. Notably, cardiomyopathy and …

Platelet-leukocyte crosstalk in COVID-19: How might the reciprocal links between thrombotic events and inflammatory state affect treatment strategies and disease prognosis?

Platelet-leukocyte crosstalk is commonly manifested by reciprocal links between thrombosis and inflammation. Platelet thrombus acts as a reactive matrix that recruits leukocytes to the injury site where their massive accumulation, activation and migration promote thrombotic events while triggering inflammatory responses. As a life-threatening condition with the associations between inflammation and thrombosis, COVID-19 presents diffuse alveolar damage due to exaggerated macrophage activity and cytokine storms. These events, together with direct intracellular virus invasion lead to pulmonary va

Disruption of mitochondria-associated ER membranes impairs insulin sensitivity and thermogenic function of adipocytes.

The prevalence and healthcare burden of obesity and its related metabolic disorders such as type 2 diabetes (T2D) are increasing rapidly. A better understanding of the pathogenesis of these diseases helps to find the therapeutic strategies. Mitochondria and endoplasmic reticulum (ER) are two important organelles involved in the maintenance of intracellular Ca2+ and ROS homeostasis. Their functional defects are thought to participate in the pathogenesis of insulin resistance or T2D. The proper structure and function of the mitochondria-associated ER membranes (MAMs) is required for efficient co

Pyroptosis and Its Role in Autoimmune Disease: A Potential Therapeutic Target.

Autoimmune diseases are a group of heterogeneous diseases with diverse clinical manifestations that can be divided into systemic and organ-specific. The common etiology of autoimmune diseases is the destruction of immune tolerance and the production of autoantibodies, which attack specific tissues and/or organs in the body. The pathogenesis of autoimmune diseases is complicated, and genetic, environmental, infectious, and even psychological factors work together to cause aberrant innate and adaptive immune responses. Although the exact mechanisms are unclear, recently, excessive exacerbation o
