[The association of adherence to a Mediterranean diet during early pregnancy and the risk of gastroschisis in the offspring].

OBJECTIVES: The aim of this study was to study the association of adherence to the Mediterranean diet in early pregnancy maternal and the offspring's risk of gastroschisis. METHODS: Case-control study. We describe 11 cases of gastroschisis in the region of Murcia from 2007 to 2012 and 34 concurrent controls. At the time of diagnosis each of the cases completed a validated Food Frequency Questionnaire (FFQ) consisting of 98 items on the periconceptional diet. Confounding factors: smoking, exposure to cannabis / marihuana, age of the parents, BMI, income and educational level. We conducted a de

Diabetic embryopathy: a developmental perspective from fertilization to adulthood.

Maternal diabetes mellitus is one of the strongest human teratogens. Despite recent advances in the fields of clinical embryology, experimental teratology and preventive medicine, diabetes-related perturbations of the maternofetal unit maintain a considerable impact on the Healthcare System. Classic consequences of prenatal exposure to hyperglycemia encompass (early) spontaneous abortions, perinatal death and malformations. The spectrum of related malformations comprises some recurrent blastogenic monotopic patterns, i.e. holoprosencephaly, caudal dysgenesis and oculoauriculovertebral spectrum

Early markers of adult obesity: a review.

The purpose of this review was to evaluate factors in early childhood (≤5 years of age) that are the most significant predictors of the development of obesity in adulthood. Factors of interest included exposures/insults in the prenatal period, infancy and early childhood, as well as other socio-demographic variables such as socioeconomic status (SES) or birth place that could impact all three time periods. An extensive electronic and systematic search initially resulted in 8,880 citations, after duplicates were removed. Specific inclusion and exclusion criteria were set, and following two scre

Sperm aneuploidy in fathers of children with paternally and maternally inherited Klinefelter syndrome.

BACKGROUND: It is unclear whether frequency of sperm aneuploidy is associated with risk of fathering children with trisomy. METHODS: We recruited 36 families with a boy with Klinefelter syndrome (KS), interviewed the fathers about their exposures and medical history, received a semen sample from each father, and collected blood samples from the mother, father and child. We applied a multicolour fluorescent in-situ hybridization assay to compare the frequencies of sperm carrying XY aneuploidy and disomies X, Y and 21 in fathers of maternally and paternally inherited KS cases. RESULTS: Inherit

Maternal Body Mass Index, Gestational Weight Gain, and Cardiovascular Disease in Offspring.

IMPORTANCE: The long-term association of maternal body mass index (BMI) in early pregnancy and gestational weight gain (GWG) with offspring cardiovascular disease (CVD) risk remains unclear. OBJECTIVE: To analyze the associations of maternal BMI and GWG with offspring CVD risk. DESIGN, SETTING, AND PARTICIPANTS: This cohort study used data from national registers in Sweden for individuals born from January 1, 1982, to December 31, 2014. Participants were followed up from birth until the first diagnosis of CVD, death, emigration from Sweden, or December 31, 2023, whichever came first. Data we

Pregnancy characteristics and maternal risk of breast cancer.

CONTEXT: During pregnancy, serum levels of estrogen, progesterone, and other hormones are markedly higher than during other periods of life. Pregnancy hormones primarily are produced in the placenta, and signs of placental impairment may serve as indirect markers of hormone exposures during pregnancy. During pregnancy, these markers have been inconsistently associated with subsequent risk of breast cancer in the mother. OBJECTIVE: To examine associations between indirect markers of hormonal exposures, such as placental weight and other pregnancy characteristics, and maternal risk of developin

Ethosuximide pharmacokinetics in a pregnant patient and her newborn.

Ethosuximide concentration in serum was monitored during the last trimester of pregnancy in a patient. After delivery, the decline in serum concentration of ethosuximide was observed in the nonnursing neonate. The half-life of elimination of transplacentally acquired ethosuximide in this neonate was 41.3 hr. The ratio of breast milk to maternal serum concentration of ethosuximide was approximately 1. A total daily exposure to ethosuximide of 12.8 to 38.4 mg (3.6 to 11.0 mg/kg) as a result of nursing was predicted.

Blood methylome signatures in children exposed to maternal type 1 diabetes are linked to protection against islet autoimmunity.

Exposure to maternal type 1 diabetes (T1D) during pregnancy provides relative protection against T1D in the offspring. This protective effect may be driven by epigenetic mechanisms. Here we conducted an epigenome-wide blood analysis on 790 young children with and 962 children without a T1D-affected mother, and identified differential DNA methylation (q < 0.05) at multiple loci and regions. These included the Homeobox A gene cluster and 15 T1D susceptibility genes. The differential methylation was found in transcriptionally relevant regions associated with immune function, including sites previ

Evaluation of fetal exposure to environmental noise using a computer-generated model.

Acoustic noise can have profound effects on wellbeing, impacting the health of the pregnant mother and the development of the fetus. Mounting evidence suggests neural memory traces are formed by auditory learning in utero. A better understanding of the fetal auditory environment is therefore critical to avoid exposure to damaging noise levels. Using anatomical data from MRI scans (N = 3), we used a computational model to quantify the acoustic field inside the pregnant maternal abdomen. We obtained acoustic transfer characteristics across the human audio range and pressure maps in transverse pl

Early-onset sepsis in term infants admitted to neonatal intensive care units (2011-2016).

OBJECTIVES: Investigate characteristics of term infants culture-evaluated for early-onset sepsis (EOS) in neonatal intensive care units (NICUs), frequencies of organisms causing EOS, and factors associated with EOS. STUDY DESIGN: Using a cohort design, we identified term infants evaluated for EOS with blood, cerebrospinal fluid, or urine cultures in 326 NICUs (2011-2016). Using multivariable logistic regression, we investigated the association between EOS and demographic characteristics. RESULTS: Of 142,410 infants, 1197 (0.8%) had EOS, most commonly caused by group B Streptococcus (GBS; 40.

Increased epigenetic alterations at the promoters of transcriptional regulators following inadequate maternal gestational weight gain.

Epigenetic modifications are thought to serve as a memory of exposure to in utero environments. However, few human studies have investigated the associations between maternal nutritional conditions during pregnancy and epigenetic alterations in offspring. In this study, we report genome-wide methylation profiles for 33 postpartum placentas from pregnancies of normal and foetal growth restriction with various extents of maternal gestational weight gain. Epigenetic alterations accumulate in the placenta under adverse in utero environments, as shown by application of Smirnov-Grubbs' outlier test.

Embryo-fetal exposure and developmental outcome of thalidomide following oral and intravaginal administration to pregnant rabbits.

Studies in pregnant rabbits were conducted to evaluate if there are any differences in the uptake of thalidomide into the intrauterine compartment and developmental toxicity risk following oral and intravaginal administration. Thalidomide concentrations in maternal plasma, yolk sac cavity (YSC) fluid and embryo following intravaginal administration were 2- to 7-fold lower than their respective levels after oral administration. Ratios of thalidomide concentration in YSC fluid to maternal plasma were similar between these two routes, indicating no difference in uptake into the intrauterine compa

Placental and oral delivery of Schistosoma mansoni antigen from infected mothers to their newborns and children.

A 63-kD Schistosoma mansoni antigen was detected in 149 (86%) of 174 umbilical cord blood sera from infected women at delivery. Specific IgG antibodies to this antigen were also detected in 80% of cord blood sera. The 63-kD antigen showed the same molecular mass by Western blotting when isolated from cord blood, maternal blood, breast milk, and urine from women infected with S. mansoni. This antigen was detected in the urine of 25 infants born to infected mothers and followed for 18-24 months after delivery. It was also detected in some infants up to 21 days after parturition and then disappea

MRI during pregnancy.

The use of ultrasonography (USG) during pregnancy provides a tremendous amount of valuable information about fetal and maternal well-being. However, in some cases the image quality may be poor due to maternal fat tissue, fetal position, or some other reason. Magnetic resonance imaging (MRI) can provide clear images from multiple angles without X-ray exposure. We believe that it is important to evaluate the usefulness of MRI for diagnostic purposes during pregnancy. While MRI can provide different information to what can be obtained from USG, it is not always superior to USG. The clinical indic

Bendectin (Debendox) as a risk factor for pyloric stenosis.

The maternal use of Bendectin (Debendox) in the first trimester of pregnancy was examined in a case-control (n = 1,427 and 3,001, respectively) study of malformed infants whose mothers were interviewed between 1974 and 1976. Mothers of infants with congenital malformation showed an increased likelihood (odds ratio = 1.40) of having used Bendectin, with a stronger association of Bendectin with birth defects for mothers who also smoked (odds ratio = 2.91). A significant association was observed between the occurrence of pyloric stenosis in the infant and exposure to Bendectin in utero (odds rati

Behavioral teratologic studies using microwave radiation: is there an increased risk from exposure to cellular phones and microwave ovens?

The objective of the investigations presented in this review was to determine if there are adverse effects due to chronic prenatal microwave exposure in rats at term and/or alterations in neonatal and adult offspring psychophysiologic development and growth. Following the establishment of a nonhyperthermal power density level of microwave radiation, pregnant rats were exposed throughout pregnancy to continuous wave 915 MHz, 2450 MHz, or 6000 MHz radiation at power density levels of 10, 20, or 35 mW/cm2, respectively. Teratologic evaluation included the following parameters: maternal weight and

Exposure to Particulate Matter (PM) During Pregnancy and Non-Hodgkin Lymphomas in Children.

Background/Objectives: Lymphomas are the third most common pediatric malignancy, with non-Hodgkin lymphoma (NHL) comprising around 7% of childhood cancers. Environmental exposures have been linked to its etiology. Particulate matter (PM) prenatal exposure is increasingly scrutinized for potential effects on early-life carcinogenesis. We investigated whether exposure during pregnancy to PM2.5 and PM10 was associated with childhood NHL incidence. Methods: A population-based case-control study was conducted in Spain (2009-2016), including 3,682,038 children < 15 years. Incident NHL cases (n = 289

A systematic review and experimental study of micro/nanoplastic-induced endocrine disruption in rodents: Potential links to autism spectrum disorder.

Recent research shows that microplastic (diameter < 5 mm) and nanoplastic (diameter < 1 μm) exposures can have endocrine-disrupting effects and lead to autism spectrum disorder (ASD)-like behaviours in rodent models. We combine both a (i) systematic literature review and (ii) experimental study to synthesize the potential mechanisms underlying the link between micro-/nanoplastic (MNP) exposure and ASD, focusing on endocrine disruption and articles utilizing rodent models. First, we identify and discuss trends in the literature, outline research gaps, and suggest future directions. Most article

Exposure to gestational diabetes mellitus in utero impacts hippocampal functional connectivity in response to food cues in children.

OBJECTIVES: Intrauterine exposure to gestational diabetes mellitus (GDM) increases the risk of obesity in the offspring, but little is known about the underlying neural mechanisms. The hippocampus is crucial for food intake regulation and is vulnerable to the effects of obesity. The purpose of the study was to investigate whether GDM exposure affects hippocampal functional connectivity during exposure to food cues using functional magnetic resonance imaging (fMRI). METHODS: Participants were 90 children age 7-11 years (53 females) who underwent an fMRI-based visual food cue task in the fasted
