Cytomegalovirus and the otolaryngologist.

Human cytomegalovirus (CMV) is ubiquitous and has been documented as the etiologic factor in several diseases ranging from respiratory tract infection to congenital deafness. Special clinical manifestations of CMV of interest to the otolaryngologist are presented as related to the temporal bone and audiovestibular system, the facial nerve, the aerodigestive tract, the thyroid, the hematopoietic-lymphoreticular system, and salivary gland. In addition, current concepts of CMV oncogenic potential are discussed as they relate to head and neck neoplasms. The results of a study performed on surgical

Evidence for an etiologic relation of the Epstein-Barr virus to human malignancies.

Studies on virus-induced animal tumors have provided indirect approaches to a search for viruses causing human malignancies. Epstein-Barr virus (EBV), the cause of infectious mononucleosis, served to illustrate the usefulness of these approaches in linking EBV with Burkitt's lymphoma and nasopharyngeal carcinoma. Despite its demonstrated intimate association with these tumors the role of EBV in their causation remains uncertain. While a passenger role seems excluded, it cannot be decided whether EBV is the primary or a secondary agent in the etiology of the tumors. If the primary cause, immuno

Immunological abnormalities in juvenile myelomonocytic leukaemia.

Immunological studies were performed on 24 children suffering from juvenile myelomonocytic leukaemia. Strikingly high serum immunoglobulin levels with a frequent tendency towards homogeneity and light-chain imbalance were present, together with a high incidence of antinuclear antibodies (52%) and antihuman IgG antibodies (43%). Members of the families of 19 of the children were also studied. A few similar abnormalities were found in eight families, mostly clustered in four of them. The significance of the association of these immunological aberrations with juvenile myelomonocytic leukaemia is

Drug-specific resistance mechanisms to RET inhibitors in RET fusion-positive NSCLC.

PURPOSE: Ret proto-oncogene (RET) fusions are highly heterogeneous oncogenic drivers occurring in approximately 1-2% of non-small cell lung cancer (NSCLC). While selpercatinib and pralsetinib have dramatically improved clinical outcomes of patients, acquired resistance inevitably develops and effective therapeutic strategies following resistance remain limited. Understanding RET-dependent resistance mechanisms is critical for improving clinical outcomes for this patient population. EXPERIMENTAL DESIGN: A rapid lentivirus-based mutagenesis approach, LentiMutate, was employed to identify potent

Engineered extracellular vesicles derived from sweet potato loaded with siPOLD1 for targeted therapy of glioma.

INTRODUCTION: Glioma is the most common malignant tumor of the central nervous system, with high malignancy and poor prognosis, necessitating the development of novel targeted therapies. DNA polymerase delta catalytic subunit 1 (POLD1) is implicated in multiple cancers, but its role in glioma remains unclear. Plant-derived extracellular vesicles (PDEVs) have emerged as biocompatible, targetable nanocarriers with promising applications in cancer therapy. OBJECTIVE: This study aims to elucidate the oncogenic function of POLD1 in glioma and develop a PDEVs -based delivery system for targeted the

The Dickkopf-1 (DKK1) Dichotomy in Oncology: New Insights on Tumor Progression and Immune Regulation.

Dickkopf-1 (DKK1) is a 266-amino-acid secreted glycoprotein originally identified as a high-affinity antagonist of the canonical Wnt/β-catenin signaling pathway and has emerged as a complex regulator in oncology. While historically considered as a tumor suppressor due to its ability to abrogate Wnt-driven proliferation, recent discoveries highlight a paradoxical pro-oncogenic role across various malignancies. The molecular mechanisms by which DKK1 promotes tumor progression, metastasis, and immune evasion are driven by its interaction with cell-surface receptors, specifically LRP5/6 and CKAP4.

SARS-CoV-2 Infection and Vaccination, Immune Dysregulation, and Cancer.

Severe Acute Respiratory Syndrome Coronavirus 2 (SARS-CoV-2) infection induces heterogeneous immune responses that influence both acute disease severity and long-term immune remodeling. A key question in the context of infection and vaccination is whether SARS-CoV-2 exerts direct oncogenic effects or instead acts as a transient immunological stressor capable of reinforcing tumor-permissive pathways. Current evidence does not support classical viral oncogenesis. Rather, severe infection is characterized by early interferon (IFN) imbalance followed by NF-κB-dominant inflammatory amplification, p…

Targeting the SETD2-FTO-KIT pathway as a pharmacological strategy for TKI-refractory GIST.

Tyrosine kinase inhibitors (TKIs) have significantly improved outcomes in gastrointestinal stromal tumors (GIST), but acquired resistance remains a major clinical obstacle. The underlying mechanisms that sustain oncogenic KIT signaling in TKI-refractory GIST are not fully understood. Here, we identify a previously unrecognized epigenetic-epitranscriptomic regulatory axis involving SETD2, FTO, and KIT that promotes TKI resistance by stabilizing KIT mRNA. FTO, a key N 6-methyladenosine (m6A) RNA demethylase, was found to be markedly overexpressed in high-risk and recurrent GIST samples and corre

TLR7 signature of tumour innervation reveals two distinct pathways of triple-negative breast cancer progression.

BACKGROUND: Emerging evidence indicates that tumour innervation promotes cancer progression via a non-canonical TLR7 signalling pathway. However, its impact across breast cancer subtypes, patient populations, associated molecular pathways, and oncogenic drivers remains poorly defined. METHODS: We analysed TLR7 signature scores in human breast cancer across multiple datasets and evaluated their associations with prognosis, clinical outcomes, TNBC subtypes, metastasis, molecular signatures, oncogenic signalling, and pathological complete response. RESULTS: We demonstrate that the TLR7score sig

Oncometabolites in Cancer Metabolism: Mechanistic Insights and Biomarker Potential- A Narrative Review.

BACKGROUND AND AIMS: Cancer cells undergo adaptation in hostile tumor microenvironments and go through extensive metabolic reprogramming for flourishing. Oncogenic transformation caused by mutations in some metabolic enzymes results in the generation of some metabolic intermediates called onco-metabolites. A growing body of evidence highlights the role of oncometabolites such as 2-hydroxyglutarate (2-HG), fumarate, and succinate in driving oncogenesis through epigenetic remodeling, altered redox balance, and interrupted cellular signaling. These metabolites are usually accumulated due to mutat…

Dietary Fat Content Influences PanIN Progression and Pancreatic Cancer Development in Mice.

UNLABELLED: Dietary macronutrient composition has emerged as a key modulator of pancreatic tumorigenesis, yet the impact of lipid-rich diets, particularly ketogenic diets (KD), on the earliest stages of pancreatic cancer development remains unclear. To investigate how dietary lipids shape the initiation and progression of Kras-driven neoplasia, we examined the effects of a low-fat diet (LFD), high-fat diet (HFD), and KD in the Ptf1aCreERT2;KrasG12V (AcinarKrasG12V) mouse model. KD-fed mice showed the shortest survival (median 26 ± 7 days) compared with the standard diet (SD, 87 ± 29; P = 0.02)

Multi-omics and pan-cancer analysis revealed common molecular signatures to disclose multitargeted anticancer agents through network pharmacology approach.

Cancer is characterized as a multifactorial disease due to their complex genetic and molecular mechanisms that often converge across tissue types. Shared oncogenic pathways can help us understand these functions and discover broad-spectrum therapeutics. Earlier, most studies focused on finding specific drivers for individual cancer types. However, researchers are now more interested in identifying common molecular patterns across different cancers and developing therapies that can target multiple pathways at once. This study aimed to understand the common oncogenic pathways between breast, ova

HIV-driven tumor ecosystem: from chronic immune dysfunction to cancer evolution.

Although HIV-associated malignancies have been reviewed separately through the lens of individual oncogenic viruses (KSHV, EBV, HPV) or immune dysfunction in isolation, no prior synthesis has integrated these processes into a single, systemic "tumor ecosystem" framework spanning virology, immunology, the tumor microenvironment, and clinical treatment response. Human immunodeficiency virus (HIV) infection profoundly alters the immune landscape, establishing a distinctive tumor ecosystem characterized by chronic immune dysfunction, inflammatory microenvironment remodeling, and virus-driven oncog

Distinct mutational landscapes when comparing germline and somatic cancer variants in forty tumor suppressor genes.

Germline and somatic cancer variants in tumor suppressor genes (TSGs) share loss of function mechanisms with studies of a few genes (DICER1, CEBPA) highlighting differences in variant consequence and location. To systematically assess whether TSGs display distinct mutational patterns we leveraged large public genetic databases and compared high-quality pathogenic/likely pathogenic (P/LP) germline variants in ClinVar, with oncogenic/likely oncogenic (O/LO) somatic tumor variants from cBioPortal across 40 TSGs. We obtained 32,941 P/LP germline and 12,907 O/LO somatic variants. Only 3,863 (9.2%)

Structural and energetic landscape of the human HOP-PrPC Axis: Multi-domain stabilization and peptide-mediated disruption by melittin.

The heat shock protein 70 (HSP70)-HSP90 organizing protein (HOP), also known as STIP1, is a vital co-chaperone that mediates the transfer of oncogenic client proteins between HSP70 and HSP90. While HOP's canonical role as a co-chaperone involves coordinating HSP70 and HSP90 activity to facilitate the folding, maturation, and stabilization of selected oncogenic client proteins, the interaction between HOP and the cellular prion protein (PrPC) has recently emerged as a significant driver of tumor progression, metastasis, and the maintenance of cancer stem cell characteristics. This study utilize

ALG8-Driven Metabolic Reprogramming in Polycystic Kidney Disease: A Systematic Synthesis of Evidence Linking Glycosylation Defects to Metabolic Signaling.

Endoplasmic reticulum glycosyltransferase ALG8 controls metabolic fate in autosomal dominant polycystic kidney disease (ADPKD). In this paper, we summarize human genetics, cell-based, and organ-based evidence to investigate whether ALG8 variants affect cyst initiation and metabolic states of ADPKD. Population screening showed ALG8 variant enrichment in ADPKD cohorts (OR = 9.75, P0.001); loss-of-function alleles interact with PKD1 mutations to accelerate cystogenesis. ALG8 deficiency leads to metabolic collapse by several mechanisms. Impaired polycystin-1 glycosylation disrupts ER-to-cilium tra

INTRODUCTION: Sorafenib is a first-line therapy for hepatocellular carcinoma (HCC), but its clinical benefit is limited by significant interpatient variability in response. Identifying reliable candidate biomarkers associated with sorafenib treatment is critical for optimizing treatment strategies and improving patient outcomes. METHODS: Samples in the GSE109211 dataset were divided into sorafenib-treated and control groups. Differentially expressed genes (DEGs) were screened using the limma package and filtered via LASSO and SVM-RFE algorithms to identify potential HCC biomarkers. CIBERSORT
