Notch signaling sculpts the stem cell niche.

Adult stem cells depend on their niches for regulatory signaling that controls their maintenance, division, and their progeny differentiatio … e abilities to claim new tissues (e.g., bone) as their territory. Moreover, in vitro models such as organoids reveal similar Notch employment when it comes to the developing stem cell niches. Therefore, a better understanding of the processes regulating stem cell niche assembly is key for the fields of stem cell biology and regenerative medicines.

RHOA signaling defects result in impaired axon guidance in iPSC-derived neurons from patients with tuberous sclerosis complex.

Patients with Tuberous Sclerosis Complex (TSC) show aberrant wiring of neuronal connections formed during development which may contribute to symptoms of TSC, such as intellectual disabilities, autism, and epilepsy. Yet models examining the molecular basis for axonal guidance defects in developing human neurons have not been developed. Here, we generate human induced pluripotent stem cell (hiPSC) lines from a patient with TSC and genetically engineer counterparts and isogenic controls. By differentiating hiPSCs, we show that control neurons respond to canonical guidance cues as predicted. Conv

Microbial Metabolite Signaling Is Required for Systemic Iron Homeostasis.

Iron is a central micronutrient needed by all living organisms. Competition for iron in the intestinal tract is essential for the maintenance of indigenous microbial populations and for host health. How symbiotic relationships between hosts and native microbes persist during times of iron limitation is unclear. Here, we demonstrate that indigenous bacteria possess an iron-dependent mechanism that inhibits host iron transport and storage. Using a high-throughput screen of microbial metabolites, we found that gut microbiota produce metabolites that suppress hypoxia-inducible factor 2α (HIF-2α) a

Modulating the tumor immune phenotypes by radiotherapy: formulating and validating the combination therapy of radiation, PD-L1, and TIM-3 blockade in colorectal cancer.

BACKGROUND: Most colorectal cancers (CRCs) are mismatch repair-proficient (pMMR) and microsatellite stable (MSS), and they respond poorly to immune checkpoint inhibitors (ICIs). Radiotherapy (RT) can promote antitumor immunity but may also trigger adaptive immune suppression through checkpoint upregulation, providing a rationale for combination therapies. METHODS: We integrated transcriptomic analyses from The Cancer Genome Atlas and Gene Expression Omnibus cohorts-annotated for microsatellite instability (MSI)/MMR status-with single-cell RNA-sequencing data from the Human Colorectal Cancer A

The Crosstalk Mechanisms Between Ferroptosis and Pyroptosis and Their Applications in Diseases: From Molecular Networks to Clinical Strategies.

Ferroptosis and pyroptosis are two distinct forms of regulated cell death that play crucial roles in cancer, neurodegeneration, and inflammatory diseases. Ferroptosis is characterised by iron-dependent lipid peroxidation, while pyroptosis is an inflammatory cell death mediated by gasdermin proteins. Recent studies reveal extensive crosstalk between these pathways. This review establishes the first hierarchical framework coupling the autophagy bridge function (ferritinophagy-mitophagy-cyclic GMP-AMP synthase (cGAS)-stimulator of interferon genes (STING) axis) with the p53/signal transducer and

Synergistic proline-based deep eutectic solvent enables low-toxicity microdroplet cryopreservation of 3D cell spheroids.

Establishing in vitro organoid biobanks-three-dimensional (3D) self-organized microtissues derived from stem cells or patient tissues-holds great value for clinical disease modeling, drug screening, and regenerative medicine applications. However, current cryopreservation approaches are often constrained by the use of chemically harsh cryoprotectants (CPAs) and multi-step operation procedures, which may compromise biological integrity and limit practical applicability. To address these challenges, we report a low-toxicity cryopreservation strategy that integrates an L‑proline‑based deep eutect

Curcumin chemo-sensitizes intrinsic apoptosis through ROS-mediated mitochondrial hyperpolarization and DNA damage in breast cancer cells.

BACKGROUND: Curcumin (CUR), a polyphenol phyto-compound extracted from turmeric rhizome (Curcuma longa), suppresses cancer by inducing apoptosis while also limiting cell survival and proliferation. This in vitro and in silico research work focuses on the synergistic sensitization of Doxorubicin (DOXO) on regulating ROS-mediated apoptosis of the breast cancer cells (MDA-MB-231 and MCF-7) in DOXO-CUR co-treatment. We observed dose-dependent cytotoxicity, increased ROS production, and mtDNA fragmentation by reduced membrane potential. The combined molecular docking of Bcl2, Bax, and Caspase3 prot

'An NF2- wildtype malignant meningioma cell line for basic and translational science'.

BACKGROUND: Meningiomas are the most common primary nervous system neoplasm in adults. There are few good cellular models, especially of high grade/malignant meningiomas, to use to identify new therapeutic agents and treatment regimens. The widely available, partially characterized, NF2- wildtype ( NF2 wt) Grade 3 malignant meningioma cell line IOMM-Lee can help meet this need. METHODS: We generated new data to better characterize IOMM-Lee genomic and mtDNA variants, proliferation rate and colony-forming efficiency and sensitivity to ionizing radiation as a function of ATM kinase activity. Addressing Clinical Challenges of Platinum Anticancer Drugs through Rational Chemical Design.

Platinum (Pt)-based anticancer drugs have been a cornerstone of chemotherapy for decades, yet their clinical application remains constrained by dose-limiting systemic toxicity and drug resistance. In this Account, we summarize our systematic efforts to address these challenges through two complementary strategies: 1) functionalization of Pt(IV) prodrugs and 2) spatially controlled targeted delivery. The kinetic inertness and octahedral geometry of Pt(IV) complexes offer a versatile platform for axial functionalization, allowing the integration of diverse bioactive ligands that are released upo

Traditional Chinese medicine as a potential barrier-oriented sensitization strategy for immune checkpoint blockade in microsatellite-stable colorectal cancer: from resistance mechanisms to translational validation.

BACKGROUND: Microsatellite-stable colorectal cancer (MSS-CRC) is refractory to immune checkpoint inhibitors (ICIs), mainly because of low tumor immunogenicity, impaired antigen presentation, immune exclusion, and myeloid- or Treg-dominated immunosuppression. Traditional Chinese medicine (TCM), characterized by multi-component and multi-target regulation, may provide complementary approaches for modulating these resistance barriers. This review summarizes the rationale, evidence, and translational requirements for integrating TCM with ICI-based therapy in MSS-CRC. PURPOSE: To evaluate the pote

Faecalibacterium duncaniae-derived metabolites protect intestinal epithelial integrity under inflammatory conditions in dairy calves.

UNLABELLED: Preweaning diarrhea and intestinal inflammation represent leading causes of morbidity and economic loss in modern dairy calf production systems, causing impaired growth performance, increased mortality, and substantial veterinary costs that collectively constrain farm profitability. Faecalibacterium duncaniae, a key butyrate-producing commensal bacterium, is consistently depleted in diarrheic dairy calves, yet its protective effects on the ruminant intestinal epithelium remain mechanistically understood. In this study, a long-term expandable ruminant colonic organoid model was used

Polyphyllin I in malignant tumors: molecular mechanisms and advances.

BACKGROUND: Natural products derived from traditional Chinese medicine have become an important focus in anticancer drug discovery due to their favorable safety profiles and multi-target properties. Polyphyllin I (PPI), a major bioactive constituent of Rhizoma Paridis (Chong-lou), represents the key pharmacological basis for its traditional uses, including heat-clearing, detoxification, and anti-inflammatory effects. Extensive pharmacological studies have shown that PPI exhibits a variety of biological activities, such as anti-inflammatory and immunomodulatory effects. Among these, its broad-s

Modulation of mitochondrial dysfunction: Mechanisms and strategies for the use of natural products to treat stroke.

Modulations of mitochondrial dysfunction, which involve a series of dynamic processes such as mitochondrial biogenesis, mitochondrial fusion and fission, mitochondrial transport, mitochondrial autophagy, mitochondrial apoptosis, and oxidative stress, play an important role in the onset and progression of stroke. With a better understanding of the critical role of mitochondrial dysfunction modulations in post-stroke neurological injury, these modulations have emerged as a potential target for stroke prevention and treatment. Additionally, since effective treatments for stroke are extremely limi

Transcriptional responses of mouse proximal colon and colonoids during early whipworm infection.

Trichuriasis, caused by the parasitic nematode Trichuris trichiura, affects 429-508 million people worldwide. Although the early phase of whipworm infection is crucial for establishing infection, its underlying molecular mechanisms remain unclear. In this study, we examined and compared host transcriptional responses during early infection of the mouse whipworm, T. muris, using first-stage larvae (L1) in proximal colons from in vivo models (B6 and STAT6-deficient mice) and in vitro colonoid models. Differentially expressed genes and functional enrichment analysis revealed that while the "neuro

Protection of clove extract against Salmonella enteritidis-induced intestinal dysfunction in broilers through JAK2/STAT3-mediated stem cell activation.

Salmonella enteritidis(SE) is a major pathogenic bacterium causing intestinal epithelial tissue damage in broiler chickens raised without antibiotic supplementation. Clove extract, as a natural antibacterial agent, possesses activity against bacterial invasion and improves intestinal health. However, the mechanism by which clove extract alleviates SE infection remains unclear. This study aimed to elucidate the protective effects of clove extract and its underlying mechanism against SE-induced intestinal damage. Experiment 1 determined the minimum inhibitory concentration (MIC) of ten plant ext

Mitochondrial transcription factor A in RORγt+ lymphocytes regulate small intestine homeostasis and metabolism.

RORγt+ lymphocytes, including interleukin 17 (IL-17)-producing gamma delta T (γδT17) cells, T helper 17 (Th17) cells, and group 3 innate lymphoid cells (ILC3s), are important immune regulators. Compared to Th17 cells and ILC3s, γδT17 cell metabolism and its role in tissue homeostasis remains poorly understood. Here, we report that the tissue milieu shapes splenic and intestinal γδT17 cell gene signatures. Conditional deletion of mitochondrial transcription factor A (Tfam) in RORγt+ lymphocytes significantly affects systemic γδT17 cell maintenance and reduces ILC3s without affecting Th17 cells

Potential intestinal infection and faecal-oral transmission of SARS-CoV-2.

Severe acute respiratory syndrome coronavirus 2 (SARS-CoV-2) has spread to more than 200 countries and regions globally. SARS-CoV-2 is thought to spread mainly through respiratory droplets and close contact. However, reports have shown that a notable proportion of patients with coronavirus disease 2019 (COVID-19) develop gastrointestinal symptoms and nearly half of patients confirmed to have COVID-19 have shown detectable SARS-CoV-2 RNA in their faecal samples. Moreover, SARS-CoV-2 infection reportedly alters intestinal microbiota, which correlated with the expression of inflammatory factors.
