Because of the paucity of available tissue, little information has previously been available regarding the gene expression profiles of primary melanomas. The combination of ifosfamide and mesna was evaluated in a phase II trial in the treatment of metastatic prostate cancer. Two separate groups of patients were to be evaluated: patients with no prior hormonal therapy and hormonally refractory patients. Supercritical fluid chromatography (SFC) has been investigated for the separation of a pair of beta-ketonate chromium compounds and a thermally labile organochromium dimer. A limited comparison between flame ionization detection (FID) and inductively coupled plasma mass spectrometric (ICP-MS) detection of these compounds is presented. The beta-ketonate complexes were observed with both detectors, while the thermally labile dimer was not observed with ICP-MS detection. Detection limits for these compounds with ICP-MS were in the range of 0.9 to 3 pg with FID giving values between 10 and 250 pg.

A stereospecific high-performance liquid chromatographic assay for the enantiomers of phenylpropanolamine (PPA) in human plasma has been developed. The method is based on reaction of extracted PPA with the chiral derivatizing agent 2,3,4,6-tetra-O-acetyl-beta-D-glucopyranosyl isothiocyanate to form diastereomeric thiourea derivatives. These thiourea derivatives are stable for at least 24 h, readily separable by reversed-phase chromatography, and amenable to ultraviolet detection at 254 nm. The clinical significance of horizontal head-shaking nystagmus (HSN) was evaluated in 85 patients who complained of dizziness and vertigo. This was done by comparison of the horizontal head-shaking test with routine rotatory and caloric vestibular testing. A convenient and sensitive high-performance liquid chromatographic method for analysis of the enantiomers of propafenone (PPF) in human plasma was developed. Racemic propafenone and (-)-ephedrine (internal standard) were first extracted from plasma samples into a mixture of hexane-2-propanol-heptafluorobutanol (95:5:1.25, v/v). A method has been developed for the determination of tetracycline (TC), oxytetracycline (OTC), chlortetracycline (CTC) and doxycycline (DC) in animal tissues. Tissues were macerated with a buffer solution and centrifuged. The supernatant solution was purified over a Sep-Pak C18 cartridge which had previously been deactivated by silylation.
