Integrated proteomic profiling of aqueous humor reveals CDC42/RHOA-mediated pathogenic mechanisms in nAMD and PDR.

Neovascular age-related macular degeneration (nAMD) and proliferative diabetic retinopathy (PDR), though clinically distinct, are both sight-threatening ocular disorders driven by pathological neovascularization. However, their shared molecular mechanisms remain poorly characterized. In this study, we performed DIA quantitative proteomic analysis of aqueous humor (AH) from patients with nAMD, PDR, and control. A total of 3186 proteins were identified, in which 877 and 1017 differentially expressed proteins (DEPs) were detected in the nAMD and PDR groups, respectively, compared to control group

Behavioral and psychological symptoms of dementia: insights from a multivariate and network-based brain proteome-wide study.

Behavioral and psychological symptoms of dementia (BPSD) are common, profoundly troubling to patients and caregivers, and difficult to treat, yet their molecular underpinnings remain poorly understood. Here, we generated a large brain proteomic dataset with nine BPSD domains assessed in life from 376 donors from three cohorts. Protein associations with BPSD were examined using complementary approaches - domain-specific BPSD, multi-domain BPSD, and latent factor modeling - and integrated via cross-cohort meta-analysis. Four proteins (NMT1, DCAKD, DNPH1, and HIBADH) were associated with anxiety

Childhood Adversity and Cardiovascular Disease Risk: Unraveling Plasma Proteomic Pathways and Moderating Role of Genetic Predisposition.

BACKGROUND: Childhood adversity (CA) is associated with cardiovascular disease (CVD). Proteomic profiling may help elucidate underlying mechanisms. This study aimed to explore how CA and genetic risk jointly impact CVD and the potential role of proteomic biomarkers. METHODS: A total of 14 508 participants without baseline CVD and with available proteomics data from the UK Biobank were included. CA was assessed using the Childhood Trauma Screener questionnaire. Proteomic signatures were derived using least absolute shrinkage and selection operator regression. Cox proportional hazard models wer

Proteomic and Kinetic Analyses Reveal Discordant Apolipoprotein Turnover and Support a Revised Model of Human Lipoprotein(a) Metabolism.

Lipoprotein(a) [Lp(a)] is a causal risk factor for atherosclerotic cardiovascular disease composed of apolipoprotein(a) [APO(a)] covalently linked to apolipoprotein B100 (APOB). Although plasma Lp(a) concentrations are largely genetically determined, the mechanisms governing Lp(a) metabolism after particle assembly remain poorly understood. We performed a cross sectional, observational study to define the Lp(a) proteome and determine the metabolic behavior of APO(a) and APOB within circulating Lp(a) particles using integrated proteomic, kinetic, and imaging approaches. Sixteen healthy adults u

Development and Validation of a Streamlined Workflow for Proteomic Analysis of Proteins and Post-translational Modifications from Dried Blood.

It is increasingly recognized that the 'omic analysis of whole blood has applications for precision medicine and disease phenotyping. Despite this realization, whole blood is generally viewed as a challenging analytical matrix in comparison to plasma or serum. Moreover, proteomic analyses of whole blood have almost exclusively focused on (non)targeted analyses of protein abundances and much less on post-translational modifications (PTMs). Here, we developed a streamlined workflow for processing 20 microliters of venous blood collected by volumetric absorptive microsampling that incorporates se

RcAlb-PepII Perturbs the Proteomic Profile of Cryptococcus neoformans, Shutting Down Proteins Involved in Fungal Survival.

Antimicrobial peptides (AMPs) occur naturally in living organisms and play an essential role in defense against pathogens. Consequently, these peptides are significant in public health research as alternatives in addressing microbial resistance. Here, we present the proteomic profile of Cryptococcus neoformans treated with RcAlb-PepII, an AMP derived from the 2S albumin of Ricinus communis seed cake. This research is noteworthy, as C. neoformans is an emerging fungal pathogen classified by the World Health Organization (WHO) as a critical threat. Proteomic analysis revealed depletion of protei

Isoleucine absence from adult human hemoglobin is mirrored in mosquito proteomes and limits malaria parasite growth.

Amino acid composition within a proteome is conventionally viewed as a property of individual protein function. However, specific nutrient scarcity has been shown to reshape which amino acids are encoded in the genome of microbes. Mosquitoes have fed on blood for 200 million years, relying on blood amino acids for reproduction. We wondered whether this ancient dependency has left an imprint on the mosquito proteome. Here we show a proteome-wide shift to lower isoleucine usage in blood-feeding mosquitoes, mirroring the known isoleucine deficiency of adult human hemoglobin. In comparing amino ac

Long-term exposure to air pollution, proteomic aging clocks, and liver-related outcomes in Chinese adults.

Although air pollution is recognized as a contributor to accelerated biological aging, its associations with clinically significant liver-related outcomes and the potential role of proteomic aging remain unclear. We aimed to evaluate the associations of key air pollutants with liver-related outcomes and the potential mediating role of proteomic aging clocks. Among 504,006 China Kadoorie Biobank participants without liver disease or cancer, we used time-varying Cox regression to assess associations of PM2.5, PM10, NO2, and warm-season O3, and their mixtures with liver-related events (LRE) and l

Plasma Proteomic Signatures for Early Risk Stratification Across Cardiovascular-Kidney-Metabolic Stages 0-2.

AIMS: Cardiovascular-kidney-metabolic (CKM) stages 0-2 represent a potentially modifiable period before overt cardiovascular complications, yet molecular markers that identify individuals at higher downstream risk remain insufficiently defined. We investigated whether plasma proteomic signatures could characterize early CKM-related molecular risk and improve prediction of incident cardiovascular, kidney and mortality outcomes. MATERIALS AND METHODS: We analysed 43 709 UK Biobank participants with CKM stages 0-2 and measurements of 2920 plasma proteins. Multivariable Cox models evaluated prote

Proteomic insights into the combined effects of near-threshold bleaching temperature and prometryn exposure on the coral Galaxea fascicularis.

Ocean warming events increasingly co-occur with coastal agrochemical pollution, yet their joint effects on coral holobiont molecular responses remain poorly resolved. We exposed the reef-building coral Galaxea fascicularis to near-threshold warming (30 °C), prometryn (1 μg L-1), and their combination for 4 days, and quantified coral host and Symbiodiniaceae protein abundance using TMT-based proteomics to capture early-stage molecular responses. Heat was the primary driver of proteome shifts, consistent with suppressed biosynthesis and reallocation toward maintenance, whereas prometryn imposed

TurboID-based proteomic profiling reveals proxitome of the IRT1 metal transporter and new insight into IRT1 endocytosis.

Iron Regulated Transporter1 (IRT1) is the major root iron transporter responsible for broad-spectrum metal absorption in Arabidopsis thaliana root epidermal cells. Non-iron metal substrates of IRT1 were recently shown to regulate IRT1 cell surface levels by endocytosis and vacuolar degradation. TurboID-based proximity labeling was recently developed to detect protein-protein interactions and thus shed light on the intricate regulation of proteins in living cells. Although TurboID-based proximity labeling technology has been successfully established in mammals, its application in plant systems

Proteomic profiling of bone for the estimation of post-mortem interval and post-mortem submersion interval: a systematic review.

Accurate estimation of the Post-Mortem Interval (PMI) and Post-Mortem Submersion Interval (PMSI) remains a persistent challenge in forensic science, especially when traditional morphological and entomological methods fail due to advanced decomposition or in aquatic environments. Proteomic profiling of bone tissues has recently emerged as a promising approach, leveraging the predictable degradation patterns of bone proteins to estimate time since death more reliably. This systematic review, conducted in accordance with PRISMA guidelines, analyzed 24 peer-reviewed studies focusing on the applica

Rapid plastid isolation reveals the chloroplast proteome and structures of the chlororibosome large subunit and RuBisCO in Marchantia polymorpha.

Plastids house the biology of eukaryotic photosynthesis. While 1000s of plastid genomes have been sequenced, the availability of less than ten proteomes and only two species with full 70S plastid ribosomal structures limit our understanding of plant evolution. We optimized a protocol for the rapid isolation of Marchantia polymorpha plastids that provides a highly enriched and intact organelle fraction from gradient volumes as little as 2 mL. The approach was successfully applied to six other species. Focusing on M. polymorpha, we determined the proteome of the plastid fraction, identifying 133

Integrated Transcriptomic and Proteomic Analyses Reveal the Effects and Mechanisms of Glycyrrhiza Polysaccharides in Alleviating Immune Stress in Broilers.

Immune stress under intensive high density production conditions impairs broiler health and performance, highlighting the need for effective nutritional strategies. This study established an immune stress model by intraperitoneal lipopolysaccharide (LPS) injection to investigate the mechanism by which Glycyrrhiza polysaccharides (GPs) alleviate LPS-induced immune stress. A total of 240 AA broilers were randomly assigned to four groups: control (CON), GP, LPS, and GP + LPS, with six replicates per group and ten birds per replicate. During the stress phase (28 d), LPS challenge increased serum A

A Proteomic Study of the Dual Oncogenic and Tumor-Suppressive Roles of SIRT3 in Lung and Breast Cancer Cell Lines.

Mitochondria play a crucial role in metabolism and energy production by generating adenosine triphosphate (ATP) through oxidative phosphorylation. They also help maintain intracellular calcium levels, facilitate communication between the nucleus and cytoplasm, detoxify reactive oxygen species (ROS), and regulate apoptosis. Reversible acetylation of mitochondrial proteins is a key post-translational modification influencing these processes, with the NAD+-dependent deacetylase SIRT3 being a major regulator. While SIRT3 has been described as a tumor suppressor in some contexts and as a tumor prom

Proximity Mapping of the ER Proteome Reveals Metastasis-specific Candidate Biomarkers in Breast Cancer Cell Lines.

BACKGROUND/AIM: Breast cancer is the most frequently diagnosed cancer among women. While biomarkers are critical for early detection and therapy, current markers lack sufficient specificity and sensitivity. The endoplasmic reticulum (ER) plays a central role in protein folding, post-translational modification, and lipid metabolism, and its alterations are linked to tumor progression. This study aimed to map ER proteome changes associated with breast cancer invasiveness and identify novel candidate biomarkers. MATERIALS AND METHODS: We compared the ER proteomes of non-invasive MCF-7 and invasi

Physicochemical characterization of nanoparticles in highly diluted preparations and exploratory plasma proteomic correlates in an N-of-1 study.

The physicochemical properties of highly diluted homeopathic preparations remain insufficiently characterized. This study investigated particulate features of Kali carbonicum (K2CO3) at 50-millesimal potencies (LM4-LM7, ∼1:50,000 dilutions per step) and explored plasma proteomic changes in a placebo-controlled N-of-1 trial. Scanning electron microscopy showed larger particle size in Kali carbonicum (67.3 nm) than in the lactose control (47.5 nm) at LM4 in a descriptive comparison. Dynamic light scattering showed no significant differences in size, polydispersity, or zeta potential among Kali c
