Increased Hepatocyte Growth Factor Secretion by Placenta-Derived Mesenchymal Stem Cells Improves Ovarian Function in an Ovariectomized Rat Model via Vascular Remodeling by Wnt Signaling Activation.

The vascular network contributes to the development of follicles. However, the therapeutic mechanism between vascular remodeling and ovarian functions is still unclear. Therefore, we demonstrated whether increased HGF by placenta-derived mesenchymal stem cells (PD-MSCs) improves ovarian function in an ovariectomized rat model via vascular remodeling by Wnt signaling activation. We established a half-ovariectomized rat model in which damaged ovaries were induced by ovariectomy of half of each ovary, and PD-MSCs (5 × 105 cells) were transplanted by intravenous injection. Three weeks after transp

Metabolic Changes in Focal Brain Ischemia in Rats Treated With Human Induced Pluripotent Stem Cell-Derived Neural Precursors Confirm the Beneficial Effect of Transplanted Cells.

There is currently no treatment for restoring lost neurological function after stroke. A growing number of studies have highlighted the potential of stem cells. However, the mechanisms underlying their beneficial effect have yet to be explored in sufficient detail. In this study, we transplanted human induced pluripotent stem cell-derived neural precursors (iPSC-NPs) in rat temporary middle cerebral artery occlusion (MCAO) model. Using magnetic resonance imaging (MRI) and magnetic resonance spectroscopy (MRS) we monitored the effect of cells and assessed lesion volume and metabolite changes in

Biomaterial-supported MSC transplantation enhances cell-cell communication for spinal cord injury.

The spinal cord is part of the central nervous system (CNS) and serves to connect the brain to the peripheral nervous system and peripheral tissues. The cell types that primarily comprise the spinal cord are neurons and several categories of glia, including astrocytes, oligodendrocytes, and microglia. Ependymal cells and small populations of endogenous stem cells, such as oligodendrocyte progenitor cells, also reside in the spinal cord. Neurons are interconnected in circuits; those that process cutaneous sensory input are mainly located in the dorsal spinal cord, while those involved in propri

Failure of chimerism formation and tolerance induction from Fas ligand mutant bone marrow donors after nonmyeloablative conditioning.

Formation of donor-recipient mixed chimerism after nonmyeloablative conditioning allows co-existence of donor and recipient hematopoietic stem cells, with solid organ allograft tolerance and less likeliness of graft versus host development. Using a post-transplant bronchiolitis obliterans murine model, we aimed to test the hypothesis that allograft preservation after mixed chimerism formation is dependent on the presence of a functional Fas ligand (FasL) on donor hematopoietic cells. To form mixed chimerism, two aliquots of 30 × 10(6) whole bone marrow cells (BMC) from either wild-type C57BL/6

Enhancing the safety of ovarian cortex autotransplantation: cancer cells are purged completely from human ovarian tissue fragments by pharmacological inhibition of YAP/TAZ oncoproteins.

STUDY QUESTION: Is it possible to eliminate metastasized cancer cells from ovarian cortex fragments prior to autotransplantation without compromising the ovarian tissue or follicles? SUMMARY ANSWER: Ex vivo pharmacological inhibition of YAP/TAZ by Verteporfin enabled us to efficiently eradicate experimentally induced small tumours, derived from leukaemia and rhabdomyosarcoma, from human ovarian tissue fragments. WHAT IS KNOWN ALREADY: Autotransplantation of ovarian tissue fragments that contain metastasized tumour cells may reintroduce the malignancy to the recipient. In order to enhance saf

αβ and γδ T-cell responses to Epstein-Barr Virus: insights in immunocompetence, immune failure and therapeutic augmentation in transplant patients.

Epstein-Barr Virus (EBV) is a human gamma herpes virus, which causes several diseases in immunocompetent (mononucleosis, chronic fatigue syndrome, gastric cancer, endemic Burkitt's lymphoma, head and neck cancer) and immunosuppressed (post-transplant lymphoproliferative disease, EBV-associated soft tissue tumors) patients. It elicits a complex humoral and cellular immune response with both innate and adaptive immune components. Substantial progress has been made in understanding the interplay of immune cells in EBV-associated diseases in recent years, and several therapeutic approaches have be

The Many Faces of NELL1 MN.

Neural tissue encoding protein with EGF-like repeats (NELL1) is a recently discovered target antigen in membranous nephropathy (MN). The initial study showed that most cases of NELL1 MN had no underlying disease associations, i.e. most cases of NELL1 MN were classified as primary MN. Subsequently, NELL1 MN has been found in the setting of various diseases. These include NELL1 MN associated with malignancy, drugs, infections, autoimmune disease, hematopoietic stem cell transplant, de novo MN in a kidney transplant and sarcoidosis. Thus there is marked heterogeneity in the diseases associated wi

[Allogeneic hematopoietic stem cell transplantation for inherited diseases].

Allogeneic hematopoietic stem cell transplantation (HSCT) has been applied for the treatment of inherited bone marrow failure syndromes, inherited metabolic diseases, and primary immunodeficiencies. HSCT indications for some inherited disorders are still controversial because the effects on neurocognitive function have been found to be variable, according to pre-transplant conditions including the patient's age, disease severity, disease status, and donor availability. Moreover, utmost attention must be paid to determining the preparative regimen for preventing regimen-related toxicity because

Requirements for Clinical Trials with Gene Therapy and Transplant Products in Switzerland.

This chapter aims to describe and summarize the regulation of gene and cell therapy products in Switzerland and its legal basis. Product types are briefly described, as are Swiss-specific terminologies such as the term "transplant product," which means products manufactured from cells, tissues, or even whole organs. Although some parts of this chapter may show a guideline character, they are not legally binding, but represent the current thinking of Swissmedic, the Swiss Agency for Therapeutic Products. As so far the experience with marketing approval of gene therapy and cell therapy products

[The possibilities for optimizing the surgical prevention of the progression of myopia].

Progressive myopia is a pressing problem of ophthalmology. Scleroplastic surgery is a pathogenetically validated method for treating this condition. The authors propose a new method for allotissue preservation, ensuring preservation of biological characteristics of tissues for any period without restriction, so that the material for transplantation were constantly available. A new shape of the graft is proposed: with a duplicature pouch and lateral incisions on one end, like a fur-tree. Such a design ensures a reliable position of the graft on scleral surface with an accent on the posterior po

Early United States experience with donation after circulatory death for heart retransplantation.

BACKGROUND: Donation after circulatory death (DCD) expands the heart donor pool, but outcomes after adult heart retransplantation (re-TX) are not well defined. METHODS: Using UNOS STAR data (January 1, 2019 to October 1, 2025), we studied adults undergoing orthotopic re-TX. Exposure was donor mechanism (DCD vs donation after brain death [DBD]). The primary outcome was 365-day mortality assessed with Kaplan-Meier methods and multivariable Cox regression. We also performed 2:1 propensity-score matching (PSM) using prespecified covariates. RESULTS: Among 474 re-TX recipients, 53 (11.2%) receive

Perceptions of health care providers involved in organ donation or transplantation on cardiac donation after death by circulatory criteria: a qualitative study.

PURPOSE: Cardiac donation after death determination by circulatory criteria (DCC) can be performed using either 1) direct procurement and perfusion of ex situ organs or 2) normothermic regional perfusion (NRP). Nevertheless, there are concerns regarding the acceptability and ethics of these procedures, particularly NRP in which the blood supply to the brain is surgically interrupted and circulation in the thorax and abdomen is restored prior to heart retrieval. We aimed to understand the perspectives on cardiac donation following DCC of Canadian clinicians who are involved in donation and tran

Heart donation after a fatal stab wound to the right ventricle: a case report.

Trauma victims are a significant part of the donor pool, though injuries to donor organs normally exclude them from their procurement. We present the case of a 37-year-old male with a stab wound to the chest, resulting in penetrating sharp force injury to the right ventricle of the heart. Despite the ultimately fatal injury, the heart was successfully repaired, and later donated, after brain death. Resuscitation and surgical intervention, along with the collaboration of medical and legal parties, enabled heart donation to occur without compromising the quality of forensic postmortem examinatio

Adipose derived mesenchymal stem cell-seeded regenerated silk fibroin scaffolds reverse liver fibrosis in mice.

Liver fibrosis (LF) is an important process in the progression of chronic liver disease to cirrhosis. We have previously demonstrated that a regenerated silk fibroin scaffold loaded with adipose-derived stem cells (RSF + ADSCs) can repair acute liver injury. In this study, we established a chronic LF animal model using carbon tetrachloride (CCl4) and a high-fat diet. We then investigated the liver repair capacity after transplanting RSF + ADSC scaffolds and RSF scaffolds onto the liver surface of mice. Compared with the control group, the concentrations of ALT and AST in the serum were signifi

Advances in regenerative cardiology: Stem cells versus bioartificial tissues.

BACKGROUND: Cardiovascular disease (CVD) remains the leading cause of mortality worldwide. The human myocardium has a limited regenerative capacity, prompting the development of innovative strategies to restore cardiac function. Stem cells (SCs) and bioartificial tissues (BATs) have emerged as promising tools in regenerative cardiology for myocardial repair and functional recovery. METHODS: This review analyzes current preclinical and clinical research focused on SC- and BAT-based therapies for CR. Literature was identified through comprehensive database searches. A First Experience of Distant Lung Preservation for Transplantation in the Russian Federation.

Lung transplant is an effective method of treating patients with end-stage respiratory diseases, but problems such as the imbalance between the number of donor organs and the number ofrecipients needing organs still play a leading role. From a transplant point of view, a multiorgan donor is considered of greatest efficiency, so that all organs that can potentially be used should be transplanted. The combination of the vast geographical territory of Russia, the shortage of actual donors, and the relatively small number of transplant centers has led to the need to transport donor lungs by air ov
