Yeast β-glucan alleviates alcohol-related brain injury by restoring gut-brain axis homeostasis.

This study investigated the protective effects of yeast β-glucan (YBG) against alcohol-related brain injury (ARBI), with a focus on potential mechanisms mediated by the gut-brain axis. Our findings revealed that YBG supplementation significantly alleviated alcohol-induced anxiety-like behaviors and cognitive deficits in mice. In the brain, YBG mitigated oxidative stress, reduced neuroinflammation, and ameliorated neurotransmitter dysregulation. In the gut, YBG protected the alcohol-damaged intestinal barrier, upregulating the expression of tight junction proteins (Claudin-1, Occludin, and ZO-1

A dietary sulfated polysaccharide from edible green alga Caulerpa chemnitzia modulates immune responses associated with host metabolism and gut microbiota in cyclophosphamide-induced immunosuppressed mice.

Sulfated polysaccharides from edible marine algae have attracted increasing interest as dietary components with immune-supporting potential. Caulerpa chemnitzia, an edible green alga widely distributed in Indo-Pacific regions, has long been consumed fresh as a vegetable and is rich in sulfated polysaccharides. In our previous study, a sulfated polysaccharide fraction (CRVP) isolated from the edible green alga Caulerpa chemnitzia was structurally characterized and shown to regulate macrophage immune responses in vitro. However, its in vivo immunomodulatory effects and functional relevance as a

Lifestyle Modulation of Xenobiotic Stress: Aerobic Exercise Attenuates Nanoplastic-Associated Neuroendocrine Dysfunction via a Gut-Ovary-Brain Continuum.

Xenobiotic stress can disrupt neuroendocrine function, yet whether a modifiable lifestyle factor can alter internal burden and downstream toxicity remains unclear. Here, adult female zebrafish were exposed to polystyrene nanoplastics (NPs; 80 nm; 1.0 mg/L) for 21 days, with or without moderate aerobic exercise (AE; 12 cm/s, 20 min/day). NPs exposure alone caused significant ovarian accumulation of particle-like structures (TEM), elevated oxidative stress, increased follicular apoptosis (TUNEL), and disrupted reproductive hormones (E2, FSH, LH). It also induced anxiety- and depression-like beha

Gut microbiota dysbiosis and metabolic alterations in rheumatoid arthritis: a barrier to periodontal repair.

OBJECTIVE: To investigate the impact of rheumatoid arthritis (RA) on periodontal healing and the underlying mechanisms. METHODS: Mandibular periodontal bone defect (PBD) and collagen-induced arthritis (CIA) models were established in male Sprague-Dawley rats aged 6 weeks, assigned to four groups: control (CON), CIA, PBD and CIA+PBD. Periodontal repair was evaluated at 1, 3 and 6 weeks. To examine the contribution of gut microbiota, pseudo-germ-free rats with PBD received 3-week faecal microbiota transplantation (FMT) from either healthy or CIA donors. Arthritis severity was assessed by paw th

Gut Microbial Functional Ecology and Microbiota-Derived Metabolites in Rheumatoid Arthritis Autoimmunity.

The gut microbiota is a key regulatory hub linking environmental exposure, the mucosal barrier, and joint inflammation, and plays an important role in the pathogenesis and progression of rheumatoid arthritis (RA). Mechanistic studies in this field mainly address two interrelated questions: how RA-associated gut microbiota modulate mucosal immunity and systemic autoimmunity through strain-level variation, niche competition, and metabolic remodeling; and how disease stage, host immune status, and drug exposure reciprocally reshape gut microbial structure and function. Accordingly, this review fo

Antimicrobial peptides RI8 and RW8 target bacterial membranes and genomic DNA to overcome drug resistance.

The global rise of multidrug-resistant (MDR) pathogens invalidates conventional antibiotics, while natural antimicrobial peptides (AMPs) lack proteolytic stability and precise targeting. This study reports a protease-resistant symmetric (RRYY)2P(YYRR)2 scaffold (Y = isoleucine/tryptophan) with alternating cationic-hydrophobic repeats, a central proline hinge, and palindromic topology for dual bacterial membrane-genomic DNA targeting. Two derived peptides, RI8 and RW8, were characterized: RI8 showed broad-spectrum activity against MDR pathogens (MIC: 2-16 μg/mL), high therapeutic index (39.4),

Evaluation of the immunomodulatory effects of novel phosphinic acid derivatives in vitro and in a murine colitis model.

T cells and cytokines are key contributors to the pathogenesis of inflammatory bowel disease (IBD), where dysregulated immune activation drives chronic intestinal inflammation. Phosphinic acid compounds are biologically active molecules that function as enzyme inhibitors and have been investigated for diverse pharmacological applications, including their potential therapeutics in inflammatory diseases. In this study, we investigated the immunomodulatory effects of seven novel phosphinic acid derivatives, including PAC-tryptophan (PAC-Trp), PAC-leucine (PAC-Leu), PAC-proline (PAC-Pro), PAC-phen

Microbiota-derived Indole-3-propionic acid improves FFA-induced liver injury in an in vitro model of nonalcoholic fatty liver disease by activating PINK1/parkin-mediated mitophagy.

BACKGROUND: Non-alcoholic fatty liver disease (NAFLD) is a growing global health concern, and its pathology is associated with impaired hepatic mitochondrial function and disrupted mitophagy. Emerging evidence highlights gut microbiota-derived metabolites as key regulators of NAFLD progression. Indole-3-propionic acid (IPA), a microbiota-derived tryptophan metabolite, has been linked to NAFLD development. However, its influence on mitophagy remains poorly defined. METHODS: In vitro, HepG2 cells were treated with free fatty acid (FFA) to establish a NAFLD model, followed by IPA treatment. The

Gut Microbiota as Neuroimmune Modulators in Myasthenia Gravis: Mechanistic Insights from the Gut-Brain Axis to Therapeutic Innovations.

Myasthenia gravis (MG) is a chronic autoimmune disorder characterized by an immune-mediated attack on neuromuscular junction acetylcholine receptors (AChRs), and its pathogenesis is closely linked to immune dysregulation. Emerging evidence has highlighted the pivotal role of the gut microbiota in the pathophysiology of MG through immunomodulation, microbial metabolite signaling, and gut-brain axis interactions. This review combines 16S rRNA sequencing, metagenomic, and metabolomic data to reveal distinct gut microbial signatures in patients with MG. These signatures include reduced α-diversity

Microbiota-associated metabolic networks in gut-kidney communication and renal immune regulation: mechanisms and therapeutic potential.

Kidney disease development and progression involve not only local inflammation, immune dysregulation, and fibrosis but also alterations in gut microbiota composition and metabolic function. Microbiota-associated metabolic signals connect the intestinal ecosystem, host metabolism, and the renal immune microenvironment through their associations with intestinal barrier integrity, renal tubular epithelial homeostasis, immune cell function, and inflammatory-fibrotic responses. Depending on their biological origin, receptor engagement, target-cell specificity, and disease context, these signals may

Hypoxia-Induced Gut-Brain Axis Remodeling and Insomnia: Mechanisms and Microbiota Adaptive Regulation from a Translational Perspective.

High-altitude hypoxia frequently disrupts sleep-wake cycles, causing insomnia that impairs acclimatization, performance, and health. Emerging evidence implicates the gut-brain axis (GBA) in this disorder. Prolonged or severe hypoxia can disturb intestinal epithelial homeostasis through oxidative, inflammatory, and metabolic changes, increasing barrier permeability and facilitating the translocation of lipopolysaccharide (LPS) and other bacterial products into the circulation. Concurrent gut microbial dysbiosis reduces the production of short-chain fatty acids (SCFAs) and redirects tryptophan m

Barrier restoration as a therapeutic strategy for disorders of gut-brain interaction.

Disorders of gut-brain interaction, such as irritable bowel syndrome and functional dyspepsia, are increasingly linked to defects in gut barrier function. Mucosal disruption, encompassing alterations in the epithelial and mucus layers, leads to enhanced intestinal permeability, microbial translocation, and aberrant immune and neuronal signalling, potentially contributing to symptom severity. Despite growing recognition of barrier dysfunction in disorders of gut-brain interaction, clinical interventions remain largely symptom-based, with few therapies designed to directly restore epithelial int

The role of intestinal microbial metabolites in polycystic ovary syndrome: mechanistic insights and intervention prospects.

BACKGROUND: Polycystic ovary syndrome (PCOS) is a prevalent endocrine-metabolic disorder among women of reproductive age, defined by the core features of hyperandrogenism, ovulatory dysfunction, and polycystic ovarian morphology. PCOS is frequently complicated by insulin resistance, obesity, dyslipidemia, and chronic low-grade inflammation, which substantially impair patients' reproductive health, metabolic homeostasis, and quality of life. Although the etiology of PCOS remains incompletely elucidated, accumulating evidence indicates that gut microbiota and their metabolites participate in the

Gut microbiota-derived metabolites as context-dependent modulators of Nrf2 signaling in inflammation-driven carcinogenesis.

Chronic inflammation fosters cancer development by sustaining redox imbalance, oxidative stress and persistent tissue damage. A key component to this process is the transcription factor Nrf2 (nuclear factor erythroid 2-related factor 2), which coordinates antioxidant and detoxification programs to maintain redox homeostasis. While transient Nrf2 activation protects non-malignant tissues by limiting cell and tissue damage, persistent or dysregulated Nrf2 signaling in premalignant tissue and established tumors can promote cancer cell survival, metabolic adaptation and resistance to chemotherapy.

Alterations in Trace Element and Metabolite Profiles by Graphene Nanosheets and Multi-Walled Carbon Nanotubes as Soil Contaminants in Foot Tissues of the Chinese White Jade Snails (Achatina fulica).

The increasing applications of carbon nanomaterials (NMs) raise concerns about their potential as soil contaminants, yet predictive models for NM-related soil contamination risks remain scarce. This study established the Chinese white jade snails (Achatina fulica) as a novel model to assess the toxicity of two representative carbon-based NMs, namely, graphene nanosheets (GNSs) and multi-walled carbon nanotubes (MWCNTs). Snails of 1-2 weeks old were exposed to coconut substrate containing 1% NMs for 28 days. While the exposure showed no significant effects on food consumption, weight gain, or s

Gut microbiome-derived indole-3-carboxaldehyde regulates stress vulnerability in chronic restraint stress by activating aryl hydrocarbon receptors.

Chronic stress constitutes a major precipitating factor for Major Depressive Disorder (MDD), and comprehending individual differences in stress responses is crucial for the development of effective intervention strategies for MDD. Recent studies indicate that an individual's vulnerability to chronic stress is closely associated with gut microbiota composition, but the underlying mechanisms remain unclear. This study aims to investigate whether the gut microbiota and its metabolites can serve as gut-brain signaling molecules and explores how the gut microbiota affects stress sensitivity. Here,

Quantitative analysis of neurotransmitters in fish brain: A tool to assess neurochemical effects of psychoactive pollutants.

Psychoactive drugs in fish act through mechanisms similar to those in humans because their main neurotransmitter (NT) systems are analogous. Psychoactive drugs are ubiquitous in aquatic environments, having negative effects on fish and other aquatic organisms. To understand the neurochemical changes underlying the effects, studying neurotransmitter levels is essential. The detection of NTs and their metabolites in biological samples may be challenging. A new analytical method using hydrophilic interaction liquid chromatography coupled to high-resolution mass spectrometry was developed. The met
