Enhancement of viral infection by antibodies and consequences.

SUMMARYAntibodies neutralize the infectivity of viruses, but they can also, on the contrary, enhance the infection by these agents. This harmful effect of antibodies has been described in vitro and/or in vivo and concerns both DNA and RNA viruses. The dengue virus, belonging to the Flaviviridae family, has been particularly studied, as well as members of other families of enveloped viruses but also of non-enveloped viruses, in particular viruses of the genus Enterovirus of the Picornaviridae family. Antibodies can enhance the infection of Fcγ receptor (FcγR)-bearing cells as well as cells that…

Increased public health threat of avian-origin H3N2 influenza virus caused by its evolution in dogs.

Influenza A viruses in animal reservoirs repeatedly cross species barriers to infect humans. Dogs are the closest companion animals to humans, but the role of dogs in the ecology of influenza viruses is unclear. H3N2 avian influenza viruses were transmitted to dogs around 2006 and have formed stable lineages. The long-term epidemic of avian-origin H3N2 virus in canines offers the best models to investigate the effect of dogs on the evolution of influenza viruses. Here, we carried out a systematic and comparative identification of the biological characteristics of H3N2 canine influenza viruses …

Aetiology of febrile pharyngitis in children: Potential of myxovirus resistance protein A (MxA) as a biomarker of viral infection.

OBJECTIVES: Besides group A streptococcus (GAS), microbial causes of pharyngitis in children are not well known. We aimed to document the viral and bacterial aetiology of pharyngitis and to assess the pathogenic role of viruses by determining the myxovirus resistance protein A (MxA) in the blood as a marker of interferon response. METHODS: In this prospective observational study, throat swabs and blood samples were collected from children (age 1-16 years) presenting to the emergency department with febrile pharyngitis. Microbial cause was sought by bacterial culture, polymerase chain reaction…

Molecular epidemiology of rabies epizootics in Colombia: evidence for human and dog rabies associated with bats.

Three urban rabies outbreaks have been reported in Colombia during the last two decades, one of these is occurring in the Caribbean Region (northern Colombia), while the other two occurred almost simultaneously in Arauca (eastern Colombia) and in the Central Region and ended in 1997. In order to derive phylogenetic relationships between rabies viruses isolated in these three areas, 902 nt cDNA fragments encoding the cytoplasmic domain of protein G and a fragment of protein L were obtained by RT-PCR. These amplicons contained the G-L intergenic region and were sequenced to draw phylogenetic tre…

A Global Perspective on H9N2 Avian Influenza Virus.

H9N2 avian influenza viruses have become globally widespread in poultry over the last two decades and represent a genuine threat both to the global poultry industry but also humans through their high rates of zoonotic infection and pandemic potential. H9N2 viruses are generally hyperendemic in affected countries and have been found in poultry in many new regions in recent years. In this review, we examine the current global spread of H9N2 avian influenza viruses as well as their host range, tropism, transmission routes and the risk posed by these viruses to human health.

Arbovirus surveillance in Italy.

A high number of different arboviruses have been demonstrated to be present in Italy, due to the coexistence of climates that are characteristic of continental areas in the North of the country and of subtropical areas in the South. Viruses circulating in Central Europe, such as the Tick-Borne Encephalitis (TBE) virus, are present in Central and Northern Regions, whereas viruses circulating in Mediterranean areas, such as the Sandfly fever viruses are present in Central and Southern Italy. Viruses have been isolated from mosquitoes, ticks, sandflies and vertebrates (birds, rodents, humans). Fo…

Isolation and characterization of caliciviruses from dogs with vesicular genital disease.

Two virus isolates, one from lesions of the vagina of a Bearded Collie and the other from the penis and prepuce of a Black Labrador, were partially characterized. The two viruses possessed the physicochemical properties, size and morphology of viruses belonging to the family Caliciviridae. The two isolates were shown by cross neutralization tests to be distinct from previously reported canine and feline caliciviruses. The viruses, isolated four years apart, are antigenically related. Additional studies are necessary to determine whether they are two distinct viruses or strains of another serot…

Functionalized Nanoparticles in Prevention and Targeted Therapy of Viral Diseases With Neurotropism Properties, Special Insight on COVID-19.

Neurotropic viruses have neural-invasive and neurovirulent properties to damage the central nervous system (CNS), leading to humans' fatal symptoms. Neurotropic viruses comprise a lot of viruses, such as Zika virus (ZIKV), herpes simplex virus (HSV), rabies virus (RABV), and severe acute respiratory syndrome coronavirus type 2 (SARS-CoV-2). Effective therapy is needed to prevent infection by these viruses in vivo and in vitro. However, the blood-brain barrier (BBB) usually prevents macromolecules from entering the CNS, which challenges the usage of the traditional probes, antiviral drugs, or n…

Major Shift of Influenza A Virus of Swine (IAV-S) by Human-to-Swine Spillover of the 2009 Pandemic Virus in Korea.

The 2009 influenza A H1N1 pandemic (pdm09) originated from the influenza A virus of swine (IAV-S) through multiple reassortment events with avian and human IAVs. The pdm09 reportedly reintroduced the virus to pigs, contributing to the evolution and diversity of IAV-S through frequent reassortment and drifts. Surveillance and whole-genome sequencing of IAV-S from conventional pig farms in Korea during 2021-2022 revealed that the genetic diversity of H1 and H3 IAV-S was continuously enriched after human-to-swine spillover of pdm09 viruses with long-term maintenance, persistence, and reassortment…

Influenza A virus continues to circulate among children in Linyi, northern China, after the relaxation of COVID-19 control measures.

Following the relaxation of control measures for COVID-19 in China in December 2022, there was a resurgence of influenza A among children in Linyi. This research aims to analyze the epidemiology and genetic characteristics of influenza A viruses circulating among children in Linyi from April 2022 to March 2024. Throat swab specimens were collected from children with influenza-like illness (ILI), and the types of influenza were identified. The hemagglutinin (HA) genes of influenza A viruses were amplified using reverse transcription-polymerase chain reaction (RT-PCR) and then sequenced. Subsequ…

Dissecting the Species-Specific Virome in Culicoides of Thrace.

Biting midges (Culicoides) are vectors of arboviruses of both veterinary and medical importance. The surge of emerging and reemerging vector-borne diseases and their expansion in geographical areas affected by climate change has increased the importance of understanding their capacity to contribute to novel and emerging infectious diseases. The study of Culicoides virome is the first step in the assessment of this potential. In this study, we analyzed the RNA virome of 10 Culicoides species within the geographical area of Thrace in the southeastern part of Europe, a crossing point between Asia…

Endemic and Emerging Arboviruses in Domestic Ruminants in East Asia.

Epizootic congenital abnormalities caused by Akabane, Aino, and Chuzan viruses have damaged the reproduction of domestic ruminants in East Asia for many years. In the past, large outbreaks of febrile illness related to bovine ephemeral fever and Ibaraki viruses severely affected the cattle industry in that region. In recent years, vaccines against these viruses have reduced the occurrence of diseases, although the viruses are still circulating and have occasionally caused sporadic and small-scaled epidemics. Over a long-term monitoring period, many arboviruses other than the above-mentioned vi…

ORF4 of the Temperate Archaeal Virus SNJ1 Governs the Lysis-Lysogeny Switch and Superinfection Immunity.

Recent environmental and metagenomic studies have considerably increased the repertoire of archaeal viruses and suggested that they play important roles in nutrient cycling in the biosphere. However, very little is known about how they regulate their life cycles and interact with their hosts. Here, we report that the life cycle of the temperate haloarchaeal virus SNJ1 is controlled by the product ORF4, a small protein belonging to the antitoxin MazE superfamily. We show that ORF4 controls the lysis-lysogeny switch of SNJ1 and mediates superinfection immunity by repression of genomic DNA replic…

Avian influenza virus H9N2 infections in farmed minks.

BACKGROUND: The prevalence of avian H9N2 viruses throughout Asia, along with their demonstrated ability to infect mammals, puts them high on the list of influenza viruses with pandemic potential for humans. In this study, we investigated whether H9N2 viruses could infect farmed minks. METHODS: First, we conducted a serological survey for avian influenza virus antibodies on a random sample of the field-trial population of farmed minks. Then we inoculated farmed minks with A/Chicken/Hebei/4/2008 H9N2 viruses and observed the potential pathogenicity of H9N2 virus and virus shedding in infected m…

Human Herpesviruses 6A and 6B in Brain Diseases: Association versus Causation.

Human herpesvirus 6A (HHV-6A) and human herpesvirus 6B (HHV-6B), collectively termed HHV-6A/B, are neurotropic viruses that permanently infect most humans from an early age. Although most people infected with these viruses appear to suffer no ill effects, the viruses are a well-established cause of encephalitis in immunocompromised patients. In this review, we summarize the evidence that the viruses may also be one trigger for febrile seizures (including febrile status epilepticus) in immunocompetent infants and children, mesial temporal lobe epilepsy, multiple sclerosis (MS), and, possibly, A…

[Acute viral infections in immunocompetent patients].

Viruses play a significant role in serious infections in adults and sometimes lead to the need for hospitalization and admission to intensive care units, especially in cases of severe respiratory distress or encephalopathy. Influenza and parainfluenza viruses, syncytial respiratory virus, herpes viruses and adenovirures are the most frequent causes of these severe infections. A review of the literature has been performed in order to update the epidemiology, pathogenesis and therapeutic approach of viral infections affecting immunocompetent patients. Furthermore, ventilator-associated pneumonia…

Enteric viromes of healthy farmed mink (Neovison vison) from Shandong and Liaoning provinces, China.

Farmed mink (Neovison vison) is the most common animal species used for fur farming in China. Several viruses identified in mink can be transmitted to humans, including SARS-CoV-2, suggesting that this species could play an important role in zoonotic disease emergence. Characterizing the virus diversity of farmed mink is therefore important for the prevention and mitigation of emerging infectious diseases. We utilized meta-transcriptomic sequencing to determine the enteric viromes of 109 apparently healthy farmed mink from Shandong and Liaoning provinces, China. Using this approach, we identif…

In vitro replicative fitness of early Transmitted founder HIV-1 variants and sensitivity to Interferon alpha.

Type I interferons, particularly interferon-alpha (IFN-α), play a vital role in the host's anti-viral defenses by interfering with viral replication. However, the virus rapidly evolves to exploit the IFN-α response for its replication, spread, and pathogenic function. In this study, we attempted to determine IFN-α susceptibility and productivity of infectious transmitted/founder (TF) (n = 8) and non-transmitted (NT) viruses (n = 8) derived from HIV-1 infected infants. Independent experiments were carried out to determine IFN-α resistance, replication fitness, and viral productivity in CD4+ T c…
