An improved spectrophotometric method is described for the determination of thiocyanate in plasma and urine. Thiocyanate is adsorbed on a weak anion-exchange resin with strong affinity for chaotropic ions, and eluted with perchlorate. Evidence for the existence of an NADPH oxidase-like enzyme in rainbow trout macrophages is given. Spectral evidence is presented that 0.01-0.03 mM solutions of the antiasthmatic drug doxantrazole contain the neutral or ionized form, depending on the solvent and, at least in 1 case, on concentration. In 1,4-dioxane, where only the neutral form exists, and in water, where the drug is ionized, no reaction between doxantrazole and calcium ion can be demonstrated spectroscopically. The partition coefficient (lambda) between red cell ghosts and buffer has been determined for three barbiturates over a range of pH. Experimental partition coefficients were linearly proportional to the calculated degree of association of the barbiturates. Lambda was 9.5 +/- 0.52 for phenobarbital, 12.7 +/- 0.91 for pentobarbital, and 27 +/- 4.9 for thiopental in their acid forms. Lambda for all three barbiturates in their anionic forms was zero.
