Single-Stage Correction of Genu Recurvatum with Dual-Pillar Plating - A Case Report.

INTRODUCTION: Genu recurvatum with anterior tibial subluxation is an uncommon but functionally debilitating deformity that can cause progressive pain, instability, and altered gait mechanics in young adults. When conservative management fails, surgical correction becomes necessary to restore alignment, joint biomechanics, and functional capacity. This report describes a reproducible single-stage corrective technique for complex genu recurvatum using biplanar osteotomy, bone graft augmentation, and dual-pillar fixation. CASE REPORT: A 19-year-old male presented with a 3-month history of progre

Evaluation of osteoporosis features in cortical bone models using pattern recognition methods applied to ultrasound data.

Cortical bone plays a crucial role in bone strength in the context of osteoporosis. Thinning of the cortex and intracortical porosity developing from the endosteum are the main independent indicators of osteoporosis-related fractures risk, which can be separately quantified by advanced imaging techniques. Numerous studies have shown the sensitivity of ultrasonic guided waves to changes in the thickness and porosity of cortical bone both in models and in human subjects. The present study demonstrates feasibility of the pattern recognition approach to separate determination of factors of diagnos

Mutant activated FGFR3 impairs endochondral bone growth by preventing SOX9 downregulation in differentiating chondrocytes.

Fibroblast growth factor receptor 3 (FGFR3) plays a critical role in the control of endochondral ossification, and bone growth and mutations that cause hyperactivation of FGFR3 are responsible for a collection of developmental disorders that feature poor endochondral bone growth. FGFR3 is expressed in proliferating chondrocytes of the cartilaginous growth plate but also in chondrocytes that have exited the cell cycle and entered the prehypertrophic phase of chondrocyte differentiation. Achondroplasia disorders feature defects in chondrocyte proliferation and differentiation, and the defects in

The effect of wrist guards on bone strain in the distal forearm.

There are increasing epidemiologic and biomechanical data suggesting that wrist guards are effective in preventing wrist injuries in snowboarders and in-line skaters. However, there have been few studies designed to determine how they function. In this study we explored the load-sharing function of wrist guards at subfailure loading levels. To do so, we measured bone strain in the distal radius, distal ulna, and midshaft of the radius in cadaveric forearms with and without two types of commercially available wrist guards. We also measured construct stiffness and energy absorption during testin

[Coincidence of bone infarct and aggressive bone metastasis].

A 42-year-old male patient presented with a history of persistent pain in the right femur without trauma of 2 months, duration and an episode of bloody stools 3 months earlier with no clinical findings upon examination. X-rays and CT scan revealed a circumscribed lesion with sclerosis and periostal reaction in the right proximal femur. A three-phase bone scan showed a massive hot spot in this area. Primarily differential diagnoses included a Brodie's abscess and/or a tumor. An excisional biopsy of the area was performed and revealed the coexistence of a bone infraction and the metastasis of an

The role of macrophages in osteoclast neogenesis in vivo: quantitative analysis of osteoclasts and macrophages in endochondral bone development in rats treated with hydrocortisone or calcitonin.

Osteoclasts and macrophages in endochondral bone of rats were analyzed after long-term treatment with hydrocortisone (HC) or elcatonin (ECT). The growth plate-metaphysis junction ( GPMJ ) of the HC-treated bones showed increased numbers of osteoclasts, but the incidence of osteoclast profiles with ruffled borders was low and the cells had fewer nuclei. The number of macrophages in the GPMJ of HC-treated bone did not significantly differ from that in the control. In the metaphyseal trabecular (MT) region macrophages were numerous, but osteoclasts were depleted. Compared to controls the ECT-trea

Coupled Bone-Muscle Degeneration in Chronic Pancreatitis: A Juvenile Porcine Model of Secondary Osteosarcopenia.

Osteosarcopenia, characterized by concurrent bone loss and muscle wasting, significantly impacts mobility and quality of life. While age-related primary osteosarcopenia is well-studied, secondary osteosarcopenia (SOS) caused by chronic diseases remains poorly understood, particularly in young individuals. The present study aimed to comprehensively characterize musculoskeletal alterations associated with SOS using a juvenile porcine model of cerulein-induced chronic pancreatitis. Femoral bone analysis included densitometry, mechanical testing, histomorphometry, and serum bone turnover markers.

Artesunate, an Anti-Malaria Agent, Attenuates Experimental Osteoarthritis by Inhibiting Bone Resorption and CD31hiEmcnhi Vessel Formation in Subchondral Bone.

Osteoarthritis (OA) is a common and debilitating joint disease worldwide without interventions available to reverse its progression. Artesunate (ART), an anti-malaria agent, possesses diverse biological activities, including the inhibition of osteoclastogenesis and angiogenesis in various cells, but its role in subchondral bone during OA progression is not known. Here, we explored the curative effects of ART on the pathogenesis of OA in anterior cruciate ligament transection (ACLT) mice models. We found that ART attenuated articular cartilage degeneration, defined by lowered histologic scoring

Skeletal changes in hemochromatosis of Salers cattle.

Hemochromatosis is an inherited defect in Salers cattle, characterized by increased iron absorption and wasting in young animals. Loss of teeth and soft bones has been reported. This study details bone changes in a prospective study of an affected Salers heifer and an age-matched, half-sibling control. The heifer fell 2 weeks before euthanasia, developed non-weight bearing lameness, and then became recumbent. At necropsy, the heifer was smaller than the control and had fractures of the humerus, femur, and ribs. Distinctive circumferential laminations in the outer third of the cortex were seen

OGT mediates O-GlcNAcylation of MEIS2 and affects palatal osteogenic development.

Post-translational modifications (PTMs) have been gradually elucidated in congenital malformations such as cleft palate. Among them, O-GlcNAcylation as a dynamic PTM of proteins regulates various critical biological processes including transcription, translation, and cell fate determination. In this study, a substantial decline in O-linked β-D-N-acetylglucosamine (O-GlcNAc) levels was detected within the palatine plates of all-trans retinoic acid (atRA)-induced cleft palate mice. The role of O-GlcNAc transferase (OGT), the sole enzyme responsible for catalyzing O-GlcNAcylation, was investigate

GPR120 Inhibits RANKL-Induced Osteoclast Formation and Resorption by Attenuating Reactive Oxygen Species Production in RAW264.7 Murine Macrophages.

Osteoclasts are large, multinucleated cells that are responsible for the resorption of bone. Bone degenerative diseases, such as osteoporosis, are characterized by overactive osteoclasts. Receptor activator of nuclear factor-κB (NF-κB) ligand (RANKL) binding to its receptor on osteoclast precursors will trigger osteoclast formation and resorption. The production of reactive oxygen species (ROS) is known to play a crucial role in RANKL-induced osteoclast formation and resorption. G-protein coupled receptor 120 (GPR120) signalling has been shown to affect osteoclast formation, but the exact mech

Bone marrow mesenchymal stem cells attenuate silica-induced pulmonary fibrosis via paracrine mechanisms.

The purpose of this study was to investigate the anti-fibrotic effect and possible mechanism of bone marrow mesenchymal stem cells (BMSCs) in silica-induced lung injury and fibrosis in vivo and in vitro. In vivo, rats were exposed to 50mg/ml silica intratracheally. The rats were sacrificed on day 15 or day 30 after intravenous injection of BMSCs. Histopathological examination demonstrated that BMSCs decreased the blue areas of collagen fibers and the number of nodules. Alveolar epithelium was damaged by silica, but it was restored by BMSCs. In vitro, BMSCs co-cultured with RLE-6TN cells in 6-T
