Dysregulation of neurodevelopmental regulatory networks in Anorexia Nervosa: an integrated multi-layered omics analysis.

Anorexia nervosa (AN) is a severe metabo-psychiatric disorder with the highest mortality rate among psychiatric conditions. Its characteristic onset during adolescence suggests that disrupted neurodevelopmental processes during critical periods may contribute to disease pathophysiology. In this exploratory study, we conducted an integrated multi-layered omics analysis to identify molecular alterations affecting neurodevelopmental pathways in individuals with AN. We employed three complementary approaches: DNA methylation sequencing via Reduced Representation Bisulfite Sequencing (RRBS), microR

Vitamin D in male reproduction: molecular mechanisms and pharmacological perspectives.

Vitamin D has attracted increasing attention in male reproduction, male fertility, and sperm function, but its pharmacological relevance remains incompletely defined. This review synthesizes current evidence on the molecular mechanisms and clinical implications of vitamin D signaling in the male reproductive system. Available experimental and human data suggest that the vitamin D receptor (VDR) and vitamin D-metabolizing enzymes are present in testicular cells, the epididymis, accessory sex glands, and mature spermatozoa, supporting the possibility of both systemic and local actions. Mechanist

Dysfunction in the SOCE/EDH-mediated vasorelaxation of mesenteric arteries impairs intestinal barrier integrity in irritable bowel syndrome.

Irritable bowel syndrome (IBS) is a prevalent functional gastrointestinal disorder with an incompletely understood pathogenesis. Although dysregulation of the enteric nervous system, mucosal barrier, brain-gut axis, and immune-microbiota interactions has been extensively investigated, the contribution of intestinal microcirculatory dysfunction remains largely unclear. In this study, a water avoidance stress (WAS) model was used to induce diarrhea-predominant IBS (IBS-D) in mice, and endothelium-dependent vasorelaxation in mesenteric small resistance arteries was assessed using wire myography.

Novel Therapeutic Frontiers in Duchenne Muscular Dystrophy: Gene Therapy, Exon Skipping, and Stem Cell Approaches.

Duchenne Muscular Dystrophy (DMD) is a chronic X-linked recessive neuromuscular disorder with a prevalence of 1 in 3,500 to 5,000 live male births worldwide. Alterations in the dystrophin gene, one of the biggest known human genes with 2.5 million base pairs and 79 exons, located at Xp21.1, are responsible for the disease. The dystrophin protein, an essential structural component that links the muscle cell cytoskeleton actin to the extracellular matrix through the dystrophin-associated protein complex, is either lacking or dysfunctional due to these mutations. Dystrophin deficiency compromises

Hybrid Feature Selection-Based Machine Learning and Deep Learning Framework for Biomarker Prediction From RNA-seq Data During Dengue Fever to Severe Dengue Progression.

BACKGROUND: Severe dengue (SD) represents a life-threatening progression of dengue virus infection. Early identification of patients at risk of transitioning from dengue fever (DF) to SD remains a major clinical challenge. Unraveling the transcriptomic changes underlying this progression may aid in developing timely therapeutic interventions. METHODS: RNA-seq datasets comprising 103 samples (62 SD and 41 DF) were retrieved from the GEO repository. Following normalization using DESeq2, differentially expressed genes (DEGs) were identified between the 2 disease stages. Functional enrichment ana

Exercise as a Programmable Regulator of Mitophagy Sensitivity in Aging Muscle and Age-Related Disease.

Aging is increasingly recognized as a systems-level process marked by progressive deterioration of mitochondrial performance in tissues with high energetic demand, placing skeletal muscle at the center of systemic metabolic and functional decline. Beyond its mechanical role, skeletal muscle acts as a regulatory hub for energy homeostasis, redox balance, and inter-organ signaling, functions that depend critically on effective mitochondrial quality control. Emerging evidence indicates that age-related mitochondrial dysfunction arises not only from impaired biogenesis but also from dysregulated m

Aging of the Colon - A Mechanistic View.

The colon is one of the gastrointestinal organs most profoundly affected by aging. Recent advances in our understanding of both colonic physiology and the general mechanisms of aging have significantly expanded our knowledge of the types and underlying processes of colonic aging. In this review, we summarize current insights into the cellular and molecular mechanisms that drive physiological aging of the human colon. We examine the unique structural and functional features of key components of the colon, including the epithelium, local immune system, microbiome, enteric neurons, and smooth mus

Targeting KCNN4 channels modulates microglial activation and apoptosis in a PD-relevant inflammatory model.

Parkinson's disease (PD) is characterized by chronic neuroinflammation and progressive dopaminergic neurodegeneration, driven primarily by the activation of microglia and associated apoptotic pathways. The intermediate-conductance calcium-activated potassium channel KCNN4 has recently emerged as a potential therapeutic target, yet its role in chronic neurodegenerative conditions remains underexplored. In this study, we investigated whether pharmacological inhibition of KCNN4 using TRAM-34 can modulate both inflammatory and apoptotic responses in an LPS-induced mouse model of PD. Our in vivo fi

The Identification of Opioid Receptors and Peptide Precursors in Human DRG Neurons Expressing Pain-Signaling Molecules Confirms Their Potential as Analgesic Targets.

The presence and function of the opioidergic system in sensory dorsal root ganglia (DRG) was demonstrated in various animal models of pain. To endorse recent functional and transcriptional evidence of opioid receptors in human DRG, this study compared morphological and transcriptional evidence in human and rat DRG using immunofluorescence confocal microscopy and mRNA transcript analysis. Specifically, it examined the neuronal expression of mu (MOR), delta (DOR), and kappa (KOR) opioid receptors, opioid peptide precursors (POMC, PENK, and PDYN), and key pain-signaling molecules. The results dem

Inactivation of NMDAR and CaMKII signaling within the prelimbic cortex blocks incubated cocaine- and sucrose-craving.

The incubation of craving is a term coined to characterize the behavioral phenomenon wherein cue-elicited craving strengthens over a period of abstinence. Incubated cocaine-craving is mediated, at least in part, by increased glutamate release within the prelimbic cortex (PL). We hypothesized that this glutamate release stimulates NMDA-type glutamate receptors (NMDARs) leading to calcium-dependent activation of CaMKII signaling that drives incubated craving. To test this hypothesis, adult male and female Sprague-Dawley rats were trained to self-administer either IV cocaine or sucrose pellets (6

CXCL16 mediates nociception and inflammation in murine post-traumatic osteoarthritis.

UNLABELLED: This study investigates the role of the chemokine CXCL16 and its receptor, CXCR6, in post-traumatic osteoarthritis (PTOA) and joint nociception, highlighting the potential of targeting the CXCL16-CXCR6 axis for therapeutically managing joint inflammation and pain. Following joint injury in mice, the CXCL16-CXCR6 signaling axis is activated in synovium, driven by synovial fibroblasts and macrophages. Human OA synovium also exhibited increased CXCL16 and CXCR6 gene expression. CXCL16 stimulated a pro-inflammatory response in fibroblasts and macrophages, contrasting with an anti-infla

Notoginsenoside R1-Protocatechuic aldehyde reduces vascular inflammation and calcification through increasing the release of nitric oxide to inhibit TGFβR1-YAP/TAZ pathway in vascular smooth muscle cells.

Vascular calcification is a significant factor contributing to the rupture of vulnerable atherosclerotic plaques, ultimately leading to cardiovascular disease. However, no effective treatments are currently available to slow the progression of vascular calcification. Notoginsenoside R1 (R1) and protocatechuic aldehyde (PCAD), primary active components extracted from Panax notoginseng and Salvia miltiorrhiza Burge, have shown potential in mitigating endothelial injury and atherosclerosis. This study investigated the effects of R1-PCAD on nitric oxide (NO) production in endothelial cells (ECs) a

Comparative proteomic analysis of regenerative mechanisms in mouse retina to identify markers for neuro-regeneration in glaucoma.

The retina is part of the central nervous system (CNS). Neurons in the CNS and retinal ganglion cells lack the ability to regenerate axons spontaneously after injury. The intrinsic axonal growth regulators, their interaction and roles that enable or inhibit axon growth are still largely unknown. This study endeavored to characterize the molecular characteristics under neurodegenerative and regenerative conditions. Data-Independent Acquisition Mass Spectrometry was used to map the comprehensive proteome of the regenerative retina from 14-day-old mice (Reg-P14) and adult mice after lens injury (
