Hepatocellular carcinoma (HCC) is an aggressive malignancy with poor prognosis and a lack of reliable biomarkers. In this study, we integrated weighted gene co-expression network analysis, machine-learning-based prognosticmodelling, single-cell RNA sequencing, spatial transcriptomics, and molecular docking to identify angiogenesis-related drivers in HCC. SCAF1 was identified as a hub gene and was significantly upregulated in HCC tissues compared with normal liver tissues. High SCAF1 expression was closely associated with advanced clinicopathological features and unfavourable survival outcomes.

BACKGROUND: Although both RNF214 and CAV1 are implicated in tumor progression and signaling regulation, the role of the RNF214-CAV1 axis in prostate cancer (PCa) remains unclear. This study aims to explore how RNF214 modulates CAV1 stability via ubiquitination and its influence on PCa progression and mTOR signaling.
METHODS: The expression patterns and functional impact of RNF214 were assessed using clinical PCa tissues, cultured cell lines, and mouse xenograft models. Gene silencing experiments were conducted to evaluate effects on cell proliferation, migration, and signaling activity. Prote

Glioblastoma multiforme (GBM), a highly aggressive primary brain malignancy, is characterized by its accelerated development, refractoriness to therapy, and dismal prognosis. The proteasome subunit β2 (PSMB2), a catalytic unit of the 20S proteasome, has been linked to tumorigenesis across multiple malignancies; however, its signaling and therapeutic relevance in GBM remains incompletely defined. U87 and U251 GBM cells were engineered to overexpress (OE-PSMB2) or silence (Sh-PSMB2) PSMB2. We assessed the function of PSMB2 in GBM cell proliferation, migration, and related phenotypes, and further

Accumulating evidence has revealed that upregulation of paired like homeodomain 1 (PITX1) drives the malignant progression of human cancers. Nevertheless, the role and downstream effector of PITX1 in non-small cell lung cancer (NSCLC) are poorly understood. We investigated the clinical relevance, biological function and mechanism of PITX1 in NSCLC. PITX1 was markedly upregulated in NSCLC tissues, and high PITX1 levels were correlated with the advanced stages and poorer survival of NSCLC. PITX1 overexpression facilitated the proliferative, migration and invasion capacity of NSCLC cells, while k

Gestational diabetes mellitus (GDM) is frequently linked to placental overgrowth and hypertrophy, while maternal hyperglycaemic environment consistently induces systemic vascular endothelial cell injury. Damaged endothelial cells stimulate the release of endothelial microparticles (EMPs), but their impact on placental function has not been fully characterized. In this study, we clinically observed that the levels of EMPs in the peripheral blood of GDM patients are significantly higher than those in healthy pregnant women. Furthermore, we identified that EMPs from GDM patients promote excessive

Head and neck cancer comprises a variety of malignant tumors affecting regions such as the oral cavity, pharynx, larynx, paranasal sinuses, nasal cavity and salivary glands. These tumors are highly heterogeneous, making it difficult to identify specific biomarkers and effective treatments, increasing mortality and recurrence rates. Databases analyzed to identify genes with altered expression in tumor tissue showed WDR66 able predict prognosis and survival, in HNSCC, but whose involvement in tumorigenesis is unknown. High WDR66 expression was associated with a worse prognosis, although its over

Papillary thyroid cancer (PTC) represents a prevalent endocrine neoplasm. Dysregulated tumor-associated calcium signal transducer 2 (TACSTD2) expression is functionally involved in the progression of multiple cancers. Nonetheless, the biological molecular mechanisms underlying TACSTD2 in PTC remain unclear. To verify the biological function of TACSTD2 in PTC, we conducted loss- and gain-of-function experiments, namely, cell proliferation (Cell Counting Kit-8), EdU incorporation, colony formation, and transwell migration/invasion assays, along with establishing a nude mouse xenograft tumor mode

Endothelial dysfunction plays a crucial role in the pathogenesis of vascular disease. Long noncoding RNA (lncRNA) and microRNA (miRNA) play important roles in various cellular processes and are involved in several vascular endothelial cells (VECs) biological processes, including cell growth, migration, autophagy, and apoptosis. The functions of plasmacytoma variant translocation 1 (PVT1) in VECs have been progressively investigated in recent years, mainly with regard to proliferation and migration of endothelial cells (ECs). However, the mechanism underlying the regulation of autophagy and apo

During embryonic development, 2 populations of multipotent stem cells, cranial neural crest cells (NCCs) and epibranchial placode cells (PCs), are anatomically adjacent to each other. The coordinated migration of NCCs and PCs plays a major role in the morphogenesis of craniofacial skeletons and cranial nerves. It is known that ethanol-induced dysfunction of NCCs and PCs is a key contributor to the defects of craniofacial skeletons and cranial nerves implicated in fetal alcohol spectrum disorder (FASD). However, how ethanol disrupts the coordinated interaction between NCCs and PCs was not eluci
