Actin pegs and ultrastructure of presumed sensory receptors of Beroë (Ctenophora).

We have investigated the actin content and ultrastructure of two kinds of presumed sensory projections on the lip epidermis of beroid ctenophores. Transmission electron microscopy showed that conical pegs contain a large bundle of densely packed, parallel microfilaments. Rhodamine-phalloidin brightly stained the pegs, confirming that they contain filamentous actin. Epidermal cells with actin pegs also bear a single long cilium with an onion-root structure, previously described as arising from a different type of cell. The actin peg and onion-root cilium project side-by-side, defining a polariz

Kallikrein-related peptidase 10 predicts prognosis and mediates tumor immunomodulation in colorectal cancer.

The incidence and mortality rates of colorectal cancer (CRC) have significantly increased in recent years. It has been shown that early diagnosis of CRC improves the five-year survival of patients compared to late diagnosis, as patients with stage I disease have a five-year survival rate as high as 90 %. Through bioinformatics analysis, we identified Kallikrein 10 (KLK10), a member of the Kallikrein family, as a reliable predictor of CRC progression, particularly in patients with early-stage CRC. Furthermore, single-cell analysis revealed that KLK10 was highly expressed in tumor and partial im

Pulmonary and blood dendritic cells from sarcoidosis patients more potently induce IFNγ-producing Th1 cells compared with monocytes.

Sarcoidosis is a systemic inflammatory disease mainly affecting the lungs. The hallmark of sarcoidosis are granulomas that are surrounded by activated T cells, likely targeting the disease-inducing antigen. IFNγ-producing Th1 and Th17.1 T cells are elevated in sarcoidosis and associate with disease progression. Monocytes and dendritic cells (DCs) are antigen-presenting cells (APCs) and required for T cell activation. Several subsets of monocytes and DCs with different functions were identified in sarcoidosis. However, to what extent different monocyte and DC subsets can support activation and

Bilobalide alleviates ischemia-reperfusion injury during liver transplantation by regulating macrophage polarization through inhibiting NF-κB pathway.

BACKGROUND: Hepatic ischemia-reperfusion injury (IRI) causes donor graft dysfunction and increases mortality after liver transplantation (LT). Bilobalide (BB), a natural sesquiterpene lactone extracted from Ginkgo biloba leaves, exhibits anti-inflammatory and anti-necrotic bioactivities. However, there is no study currently on the use of bilobalide in IRI therapy for liver transplant. This study aimed to investigate whether bilobalide protects grafted livers from IRI and to explore the underlying mechanisms. METHODS: Donor livers preserved in histidine-tryptophan-ketoglutarate (HTK) solution

Network mechanisms of glymphatic system dysfunction in the disruption of the "brain-lung axis".

Acute brain injury (ABI) frequently precipitates severe extracranial target-organ complications, with the lungs being the most directly and fatally affected distant organs. Traditionally, the rapid onset of pulmonary dysfunction following brain injury has been attributed to the "massive catecholamine release"-a massive, dysregulated release of catecholamines leading to intense systemic vasoconstriction, elevated pulmonary capillary hydrostatic pressure, and subsequent neurogenic pulmonary edema. However, this purely neuro-hemodynamic model falls short of explaining the delayed onset and highly

Sappanone A Promotes Motor Function Recovery in Spinal Cord Injury Mice by Inhibiting Microglial M1 Polarization via Activation of the Keap1/Nrf2 Pathway.

Spinal cord injury (SCI), a central nervous system disease resulting in significant functional impairment, neuroinflammation and oxidative stress are its core pathological features. Sappanone A (SA), a naturally occurring flavonoid compound predominantly derived from Caesalpinia sappan heartwood, demonstrates dual functionality encompassing both anti-inflammatory and antioxidant effects. The specific function of SA in SCI and its associated mechanisms constitute uncharted territory. This study examined the underlying neuroprotective mechanisms of SA in spinal cord injury using a T9 spinal cord

Jing An decoction alleviates neuroinflammation in Tourette syndrome by regulating butyrate-mediated microbiota-gut-brain axis.

BACKGROUND: Jing An decoction (JA), a traditional Chinese medicine formulation, has demonstrated notable clinical efficacy in the treatment of Tourette syndrome (TS) over the past two decades. The gut-brain axis (GBA) critically regulates neuroinflammation and neurodevelopmental processes, suggesting its potential as a therapeutic target in TS. However, the mechanisms by which JA alleviates TS via the GBA remain unclear. PURPOSE: This study investigated gut microbiota and its metabolites to explore the potential mechanisms by which JA alleviates TS, a chronic neurodevelopmental disorder. MET

The clinical impact of macrophage polarity after Kasai portoenterostomy in biliary atresia.

INTRODUCTION: Biliary atresia (BA) is a cholestatic hepatopathy caused by fibrosing destruction of intrahepatic and extrahepatic bile ducts, and its etiology has not been clearly revealed. In BA, liver fibrosis progression is often observed even after Kasai portoenterostomy (KPE), and more than half of cases require liver transplantation in their lifetime in Japan. Macrophages play an important role in liver fibrosis progression and are classically divided into proinflammatory (M1) and fibrotic macrophages (M2), whose phenotypic transformation is called "macrophage polarity." The polarity has

The Role of M1/M2 Macrophage Polarization in Rheumatoid Arthritis Synovitis.

Innate and adaptive immunity represent a harmonic counterbalanced system involved in the induction, progression, and possibly resolution of the inflammatory reaction that characterize autoimmune rheumatic diseases (ARDs), including rheumatoid arthritis (RA). Although the immunopathophysiological mechanisms of the ARDs are not fully clarified, they are often associated with an inappropriate macrophage/T-cell interaction, where classical (M1) or alternative (M2) macrophage activation may influence the occurrence of T-helper (Th)1 or Th2 responses. In RA patients, M1/Th1 activation occurs in an i

A Biphasic Calcium Phosphate Cement Enhances Dentin Regeneration by Dental Pulp Stem Cells and Promotes Macrophages M2 Phenotype In Vitro.

Calcium phosphate cement (CPC) is promising for bone and dentin repair and regeneration. However, there has been no report of biphasic CPC for inducing dentin regeneration. The aim of this study was to develop a novel biphasic CPC containing β-tricalcium phosphate (β-TCP), and investigate its effects on odontogenic differentiation of human dental pulp stem cells (hDPSCs) and macrophage polarization. New biphasic CPC was formulated with different ratios of β-TCP to an equimolar mixture of tetracalcium phosphate and dicalcium phosphate anhydrous. Mechanical properties, biocompatibility, and odon

Resolution of Pulmonary Inflammation Induced by Carbon Nanotubes and Fullerenes in Mice: Role of Macrophage Polarization.

Pulmonary exposure to certain engineered nanomaterials (ENMs) causes chronic lesions like fibrosis and cancer in animal models as a result of unresolved inflammation. Resolution of inflammation involves the time-dependent biosynthesis of lipid mediators (LMs)-in particular, specialized pro-resolving mediators (SPMs). To understand how ENM-induced pulmonary inflammation is resolved, we analyzed the inflammatory and pro-resolving responses to fibrogenic multi-walled carbon nanotubes (MWCNTs, Mitsui-7) and low-toxicity fullerenes (fullerene C60, C60F). Pharyngeal aspiration of MWCNTs at 40 μg/mou

Antivirals acting on viral envelopes via biophysical mechanisms of action.

Most antivirals target viral proteins and are specific for only one virus, or viral type. Whereas viral proteins are encoded in the plastic viral genome, virion lipids are not and their rearrangements during fusion are conserved among otherwise unrelated enveloped viruses. Antivirals that inhibit these lipid rearrangements could thus pose a high barrier to resistance and have broad-spectrum activity. Fusion occurs through a hemifusion stalk in which only the outer leaflets are fused and thus curved with a smaller radius for the polar heads than for the hydrophobic tails (negative curvature). O

Modes of the initiation of T cell-mediated immune responses.

T cell-mediated immune response is the center of adaptive immunity. Despite more than five decades of research, the knowledge of mechanisms drives the initiation of T cell-mediated immune response is still limited due to the complexity of T cell-mediated immunity. Based on the accumulating evidences of Th1-mediated immune response, a "priming-activation" model was proposed to describe the initiation of the immune response. In this model, naïve CD4 T cells undergo a priming phase in the draining lymph nodes (dLNs) to polarize to Th1 fate by signals from dendritic cells (DCs). The primed Th1 cel

Long-term cell storage of RAW 264.7 cells in a deep freezer dampens M2 marker expression.

RAW 264.7, a well-established cell line, is widely used in research; however, it is easily polarized upon subtle changes in environmental conditions or repeated passages. As the effects of cell storage conditions and duration on their phenotypic changes remain unknown, we investigated the effects of long-term storage at -80 °C on M2 marker expression in RAW 264.7 cells after stimulation with IL-4. Cells, stored in a - 80 °C deep freezer for a few years (defined as "long-stored cells") and newly purchased cells (referred to as "new cells"), were compared through flow cytometry for CD68-positive

Single-cell profiling of bronchoalveolar cells reveals a Th17 signature in neutrophilic severe equine asthma.

Severe equine asthma (SEA) is a complex respiratory condition characterized by chronic airway inflammation. It shares many clinical and pathological features with human neutrophilic asthma, making it a valuable model for studying this condition. However, the immune mechanisms driving SEA have remained elusive. Although SEA has been primarily associated with a Th2 response, there have also been reports of Th1, Th17, or mixed-mediated responses. To uncover the elusive immune mechanisms driving SEA, we performed single-cell mRNA sequencing (scRNA-seq) on cryopreserved bronchoalveolar cells from 1

Mechanistic insights into the interplays between neutrophils and other immune cells in cancer development and progression.

Cancer is considered a major public health concern worldwide and is characterized by an uncontrolled division of abnormal cells. The human immune system recognizes cancerous cells and induces innate immunity to destroy those cells. However, sustained tumors may protect themselves by developing immune escape mechanisms through multiple soluble and cellular mediators. Neutrophils are the most plenteous leukocytes in the human blood and are crucial for immune defense in infection and inflammation. Besides, neutrophils emancipate the antimicrobial contents, secrete different cytokines or chemokine

Protein Immunization Induces Memory CD4+ T Cells That Lack Th Lineage Commitment.

Acute viral infection generates lineage-committed Th1 and T follicular helper (Tfh) memory cells that recall their lineage-specific functions following secondary challenge with virus. However, the lineage commitment of effector and memory Th cells in vivo following protein vaccination is poorly understood. In this study, we analyzed effector and memory CD4+ T cell differentiation in mice (Mus musculus) following adjuvanted glycoprotein immunization compared with acute lymphocytic choriomeningitis virus infection. Glycoprotein immunization induced CXCR5- non-Tfh effector and memory CD4+ T cells

Modulation of Tetraspanin 32 (TSPAN32) Expression in T Cell-Mediated Immune Responses and in Multiple Sclerosis.

Tetraspanins are a conserved family of proteins involved in a number of biological processes including, cell-cell interactions, fertility, cancer metastasis and immune responses. It has previously been shown that TSPAN32 knockout mice have normal hemopoiesis and B-cell responses, but hyperproliferative T cells. Here, we show that TSPAN32 is expressed at higher levels in the lymphoid lineage as compared to myeloid cells. In vitro activation of T helper cells via anti-CD3/CD28 is associated with a significant downregulation of TSPAN32. Interestingly, engagement of CD3 is sufficient to modulate T

Invariant NKT cells with chimeric antigen receptor provide a novel platform for safe and effective cancer immunotherapy.

Advances in the design of chimeric antigen receptors (CARs) have improved the antitumor efficacy of redirected T cells. However, functional heterogeneity of CAR T cells limits their therapeutic potential and is associated with toxicity. We proposed that CAR expression in Vα24-invariant natural killer T (NKT) cells can build on the natural antitumor properties of these cells while their restriction by monomorphic CD1d limits toxicity. Primary human NKT cells were engineered to express a CAR against the GD2 ganglioside (CAR.GD2), which is highly expressed by neuroblastoma (NB). We compared CAR.G

Targeting hypercoagulability in ischemic stroke modulates fibrin-driven microglial polarization via JAK-STAT pathway.

Ischemic stroke is a leading cause of mortality and disability worldwide, yet its pathophysiological mechanisms remain poorly understood. In this study, we analyzed the coagulation function in 60 patients with acute ischemic stroke and found that their blood was in a hypercoagulable state. We confirmed this hematological change using a middle cerebral artery occlusion/reperfusion (MCAO/r) mouse model and improved the hypercoagulable state with the synthetic peptide Arg-Gly-Asp-Cys (RGDC), which inhibits platelet aggregation and fibrinogen binding. Interestingly, in MCAO/r mice, RGDC treatment
