Rac1 Constrains Memory Consolidation.

Ras-related C3 botulinum toxin substrate 1 (Rac1) is a small GTPase that regulates actin cytoskeleton dynamics and synaptic plasticity. Rac1 has been implicated in active forgetting, but whether it also constrains the consolidation of new memories remains unclear. Here we show that systemic administration of the Rac1 inhibitor 1A-116 after training in the novel object recognition task markedly extends memory persistence in rats. A single post-training injection of 1A-116 enhanced recognition memory for at least 28 d without altering locomotor- or anxiety-related behaviors. When given after a b

[Immune Checkpoint Tim-3 Regulates Microglial Function in Alzheimer's Disease].

Microglia are resident immune cells of the central nervous system and play a central role in the pathogenesis of Alzheimer's disease (AD). Emerging evidence indicates that their functional state critically influences amyloid-β (Aβ) clearance, neuroinflammation, and disease progression. Tim-3, an immune checkpoint molecule, is highly expressed in microglia and functions as a key regulator of microglial homeostasis through the transforming growth factor-β (TGF-β)-Smad2 signaling pathway. Microglia-specific deletion of Tim-3 enhances phagocytic activity. Moreover, microglia-specific Tim-3 deficie

Emerging role of cell membrane-coated nanoparticles in targeted therapy for brain disorders.

Brain disorders, including neurodegenerative diseases such as Alzheimer's disease, Parkinson's disease, and Huntington's disease, as well as conditions like multiple sclerosis and ischemic stroke, represent a significant global health burden. These disorders are often marked by progressive neuronal degeneration, cognitive impairment, and motor dysfunction. Effective therapeutic intervention remains a challenge due to the protective but restrictive nature of blood-CNS barriers, particularly the blood-brain barrier (BBB), which limits drug delivery to the brain. Nanotechnology-based drug deliver

sEMG-Based Assessment of Engagement in VR-Integrated BALANSens Ankle Rehabilitation.

This study investigates participant engagement with various types of virtual reality (VR)-integrated passive ankle robotic rehabilitation offered by the BALANSens platform, using surface electromyography (sEMG) signals to quantitatively assess involvement. The low adherence rates in robotic rehabilitation, often due to the monotonous nature of exercises, highlight the importance of enhancing patient motivation. VR is shown to improve patient engagement by providing immersive and interactive therapy environments, ultimately increasing training time, reducing dropout rates, and enhancing motivat

Trimethyl Chitosan-Engineered Cod Skin Peptide Nanosystems Alleviate Behavioral and Cognitive Deficits in D-Galactose-Induced Alzheimer's Disease Model Mice.

Alzheimer's disease (AD) is a common neurodegenerative disorder with limited effective treatments. Cod skin collagen peptides (CSCPs) have neuroprotective potential for AD but face poor bioavailability-due to gastrointestinal enzyme cleavage and hepatic first-pass metabolism-prompting this study to develop a nanodelivery system to enhance CSCPs' efficacy. Trimethyl chitosan (TMC)-based CSCP-loaded nanoparticles (CSCPs-NPs) were synthesized via ionic gelation, characterized for physicochemical properties, and tested in a D-galactose-induced AD mouse model (six groups: normal control, model, CSC

An integrated deep learning model for early and multi-class diagnosis of Alzheimer's disease from MRI scans.

Alzheimer's disease (AD) is a progressive neurodegenerative disorder that severely affects memory, behavior, and cognitive function. Early and accurate diagnosis is crucial for effective intervention, yet detecting subtle changes in the early stages remains a challenge. In this study, we propose a hybrid deep learning-based multi-class classification system for AD using magnetic resonance imaging (MRI). The proposed approach integrates an improved DeepLabV3+ (IDeepLabV3+) model for lesion segmentation, followed by feature extraction using the LeNet-5 model. A novel feature selection method bas

Management of Alzheimer's Disease With Nanotechnological Interventions and Novel Therapeutics.

Alzheimer's disease (AD) is a progressive neurodegenerative condition marked by cognitive deterioration, β-amyloid plaque buildup, intracellular tangles, and significant neuronal loss. The increasing prevalence of AD, along with its substantial economic burden, underscores the urgent need for effective therapeutic strategies in the near future. The challenge is early diagnosis and management, hindered by the lack of reliable biomarkers. Currently, there is no definitive cure for AD. Attaining improved therapeutic outcomes necessitates delivering optimal drug concentrations to the central nervo

A novel neuroimaging based early detection framework for alzheimer disease using deep learning.

Alzheimer's disease (AD) is a progressive neurodegenerative disorder that significantly impacts cognitive function, posing a major global health challenge. Despite its rising prevalence, particularly in low and middle-income countries, early diagnosis remains inadequate, with projections estimating over 55 million affected individuals by 2022, expected to triple by 2050. Accurate early detection is critical for effective intervention. This study presents Neuroimaging-based Early Detection of Alzheimer's Disease using Deep Learning (NEDA-DL), a novel computer-aided diagnostic (CAD) framework le

Targeting Alzheimer's disease pathology: influence of nano-based drug delivery systems loaded with a combination of herbal and synthetic drugs.

INTRODUCTION: Alzheimer's Disease (AD) is a progressive neurological disorder marked by cognitive decline and memory loss. Current treatments, including acetylcholinesterase inhibitors (AChEIs) and NMDA receptor antagonists, provide only symptomatic relief due to poor Blood Brain Barrier (BBB) permeability and side effects. The integration of synthetic and natural drug combinations with nanotechnology offers a promising strategy to enhance drug delivery, efficacy, and overall therapeutic outcomes. AREAS COVERED: This review explores the integration of herbal and synthetic drugs in nano-based

Emotional prospective memory in traumatic brain injury: task characteristics matter.

This study investigated the effects of emotional valence on prospective memory (PM) performance in individuals with traumatic brain injury (TBI) compared to healthy controls. Using an emotional version of the Virtual Week paradigm, we examined how positive, negative, and neutral emotional content influenced both event-based and time-based PM tasks. Thirty-one participants with TBI and 31 matched healthy controls completed the computerised PM measure over three simulated days, and a recognition task to assess the retrospective component of PM. Participants with TBI performed less accurately tha

Vagus nerve stimulation during training fails to improve learning in healthy rats.

Learning new skills requires neuroplasticity. Vagus nerve stimulation (VNS) during sensory and motor events can increase neuroplasticity in networks related to these events and might therefore serve to facilitate learning on sensory and motor tasks. We tested if VNS could broadly improve learning on a wide variety of tasks across different skill domains in healthy, female adult rats. VNS was paired with presentation of stimuli or on successful trials during training, strategies known to facilitate plasticity and improve recovery in models of neurological disorders. VNS failed to improve either

Amphetamine Exposure during Embryogenesis Alters Expression and Function of Tyrosine Hydroxylase and the Vesicular Monoamine Transporter in Adult C. elegans.

Amphetamines (Amph) are psychostimulants broadly used as physical and cognitive enhancers. However, the long-term effects of prenatal exposure to Amph have been poorly investigated. Here, we show that continuous exposure to Amph during early development induces long-lasting changes in histone methylation at the C. elegans tyrosine hydroxylase (TH) homolog cat-2 and the vesicular monoamine transporter (VMAT) homologue cat-1 genes. These Amph-induced histone modifications are correlated with enhanced expression and function of CAT-2/TH and higher levels of dopamine, but decreased expression of C

The Effect of β-Alanine Supplementation on Performance, Cognitive Function and Resiliency in Soldiers.

β-alanine is a nonessential amino acid that combines with the amino acid histidine to form the intracellular dipeptide carnosine, an important intracellular buffer. Evidence has been well established on the ability of β-alanine supplementation to enhance anaerobic skeletal muscle performance. As a result, β-alanine has become one of the more popular supplements used by competitive athletes. These same benefits have also been reported in soldiers. Evidence accumulated over the last few years has suggested that β-alanine can result in carnosine elevations in the brain, which appears to have broa

A review of brain regions and associated post-concussion symptoms.

The human brain is an exceptionally complex organ that is comprised of billions of neurons. Therefore, when a traumatic event such as a concussion occurs, somatic, cognitive, behavioral, and sleep impairments are the common outcome. Each concussion is unique in the sense that the magnitude of biomechanical forces and the direction, rotation, and source of those forces are different for each concussive event. This helps to explain the unpredictable nature of post-concussion symptoms that can arise and resolve. The purpose of this narrative review is to connect the anatomical location, healthy f

Mindfulness-Based Cognitive Therapy in Recurrent MDD Patients With Residual Symptoms: Alterations in Resting-State Theta Oscillation Dynamics Associated With Changes in Depression and Rumination.

Many patients with major depressive disorder (MDD) suffer from residual symptoms. Rumination is a specific known risk factor for the onset, severity, prolongation, and relapse of MDD. This study aimed to examine the efficacy and EEG substrates of mindfulness-based cognitive therapy (MBCT) in alleviating depression and rumination in an MDD population with residual symptoms. We recruited 26 recurrent MDD individuals who had residual symptoms with their current antidepressants to participate in the 8-week MBCT intervention. We evaluated the efficacy and changes in the dynamics of resting-state th

Phlorizin alleviates cholinergic memory impairment and regulates gut microbiota in d-galactose induced mice.

We explored the effect of phlorizin against cholinergic memory impairment and dysbacteriosis in D-galactose induced ICR mice. The control (CON) group, D-galactose model (DGM) group, and three groups (DG-PL, DG-PM, DG-PH) treated with phlorizin at 0.01%, 0.02%, and 0.04% (w/w) in diets were raised for 12 weeks. Supplementing with phlorizin reversed the loss of organ coefficient and body weight caused by D-galactose. The functional abilities of phlorizin on hippocampal-dependent spatial learning and memory, anti-oxidation, anti-inflammation were also observed. Meanwhile, phlorizin intervention u

Mentalization-enhancing therapeutic interventions in the psychotherapy of anorexia nervosa: An analysis of use and influence on patients' mentalizing capacity.

OBJECTIVE: Improvement in patients' mentalizing capacities is considered a possible mechanism of change in psychotherapy. This improvement might take place via mentalization-enhancing interventions (MEIs) performed by psychotherapists. The study aimed to explore the use of MEIs in two evidence-based psychotherapeutic treatments for patients with anorexia nervosa (enhanced cognitive-behavior therapy, focal psychodynamic therapy) and their association with the patients' capacity to mentalize in sessions ("in-session reflective functioning" / in-session RF). Additionally, it was explored, if the
