The role of inner nuclear membrane protein emerin in myogenesis.

Emerin, a ubiquitously expressed inner nuclear membrane protein, plays a central role in maintaining nuclear structure and genomic organization, and in regulating gene expression and cellular signaling pathways. These functions are critical for proper myogenic differentiation and are closely linked to the pathology of Emery-Dreifuss muscular dystrophy 1 (EDMD1), a laminopathy caused by mutations in the EMD gene. Emerin, along with other nuclear lamina proteins, modulates chromatin organization, cell signaling, gene expression, and cellular mechanotransduction, processes essential for muscle de

Proteomic and metabolomic responses of priority bacterial pathogens to subinhibitory concentration of antibiotics.

This study employed a comprehensive proteomic and metabolomic analysis to characterize adaptive cellular mechanisms of priority pathogens-Escherichia coli, Klebsiella pneumoniae, Enterococcus faecium, and Staphylococcus aureus-under sub-inhibitory concentrations of antibiotics. Despite significant metabolomic perturbations, some pathogens had minimal or no significant changes in their proteome. Notably, trimethylamine metabolism was consistently altered across all species, suggesting its role in survival under antibiotic stress. Shared adaptive responses to chloramphenicol in S. aureus and E.

Role of mitogen-activated protein kinase inhibitors in Alzheimer's disease: Rouge of brain kinases.

Alzheimer's disease (AD) is the chief cause of dementia and related mortality worldwide due to progressive accumulation of amyloid peptide (Aβ) and hyperphosphorylated tau protein. These neuropathological changes lead to cognitive impairment and memory dysfunction. Notably, most Food drug Administration (FDA) approved anti-AD medications such as tacrine and donepezil are engaged with symptomatic relief of cognitive impairment but do not reverse the underlying AD neuropathology. Therefore, searching for new anti-AD is advisable. It has been shown that the inflammatory signaling pathways such as

From inhalation to systemic damage: Decoding the mechanistic cascade of silica nanoparticle-induced multi-organ toxicity.

The increasing use of silica nanoparticles (SiNPs) has raised concerns about their biotoxicity. Since respiratory exposure is the primary route of human exposure to SiNPs, this study systematically investigated their distribution and damaging effects in the lungs, heart, liver, and kidneys following tracheal drip injection. The results demonstrated that SiNPs distribute across these organs and induce mitochondrial damage, endoplasmic reticulum stress, and activate cell death pathways, including apoptosis, pyroptosis, and autophagy. The most significant damage occurred in the middle-dose group

Molecular genetics of congenital heart disease.

Congenital heart disease (CHD) is the most prevalent human birth defect and remains a leading cause of mortality in childhood. Although advancements in surgical and medical interventions have significantly reduced mortality rates among infants with critical CHDs, many survivors experience substantial cardiac and extracardiac comorbidities that affect their quality of life. The etiology of CHD is multifactorial, involving both genetic and environmental factors, yet a definitive cause remains unidentified in many cases. Recent advancements in genetic testing technologies have improved our abilit

Engineering H2S-Activatable Aptameric Binders for Tailored Cell Manipulation.

Cell surface receptors play a crucial role in mediating cellular communication and precisely regulating cellular functions. Dysregulation of these receptors is closely associated with various diseases. To enable precise and dynamic receptor control, we developed a gas-responsive aptameric binder based on a chemically masked configuration, enabling stimulus-triggered reactivation. Specifically, the receptor-targeting activity of the aptamer is sterically blocked via azide modifications at critical residues, with selective restoration of function upon exposure to hydrogen sulfide (H2S), enabling

FBXW7 regulates MYRF levels to control myelin capacity and homeostasis in the adult central nervous system.

Myelin, along with the oligodendrocytes (OLs) that produce it, is essential for proper central nervous system (CNS) function in vertebrates. Although the accurate targeting of myelin to axons and its maintenance are critical for CNS performance, the molecular pathways that regulate these processes remain poorly understood. Through a combination of zebrafish genetics, mouse models, and primary OL cultures, we find that FBXW7, a recognition subunit of an E3 ubiquitin ligase complex, is a regulator of adult myelination in the CNS. Loss of Fbxw7 in myelinating OLs results in increased myelin sheat

Bridging the gap: insights into sensorimotor deficits in NMDA receptor antibody encephalitis.

N-methyl-d-aspartate (NMDA) receptor-mediated autoimmune encephalitis (NMDAR-AE) is the most common cause of autoimmune encephalitis, especially in children and young adults. The disorder is caused by antibodies directed against the GluN1 protein, an obligatory constituent of NMDA receptors, which are key signaling molecules in brain development, learning and memory, and executive function. The manuscript by Zhou et al. offers key insights into aberrant development of cortical pathways that may underly persistent sensorimotor deficits associated with this encephalitis in a newly generated mous

Education Is Treatment: Integrating Chemosensory Dysfunction Education in Oncology Care.

Chemosensory dysfunction, defined as an altered or lost taste and smell, is a prevalent side effect of cancer treatment, with 93% of patients complaining of taste and 60% complaining of changes in smell. Despite their impact, it is an underrecognized symptom, impairing nutritional intake, quality of life, and treatment outcomes. Surprisingly, taste and smell changes are rarely assessed or addressed in oncology care. This commentary highlights the educational gap faced by clinicians and reports the results of a pilot educational intervention consisting of a 15-min podcast. The results indicate

Ototoxicity-induced c-Fos activation underlies the regenerative capacity of the vestibular sensory epithelia.

The mammalian inner ear exhibits a striking dichotomy in regenerative capacity: while vestibular hair cells retain limited ability to regenerate, cochlear hair cells permanently lose this potential after birth. Exploiting this disparity, we employed single-cell comparative transcriptomics combined with lineage tracing to identify c-Fos as a critical early responder selectively activated in the vestibular epithelium following ototoxic injury. Transcriptomic and chromatin profiling revealed that c-Fos initiates regenerative reprogramming by transcriptionally priming Atoh1, the master regulator o

The role of fibroblast-neutrophil crosstalk in the pathogenesis of inflammatory diseases: a multi-tissue perspective.

Neutrophil-fibroblast crosstalk drives inflammatory pathology across organ systems through both shared and tissue-specific mechanisms. This review synthesizes evidence from skin, lung, gut, cardiovascular, joint, sinus, and oral diseases, revealing conserved molecular pathways where fibroblasts secrete chemokines (CXCL1/8/12) to recruit neutrophils, which, in turn, release neutrophil extracellular traps (NETs), elastase, and cytokines to modulate fibroblast function. Additionally, we identify critical tissue-specific differences, including the predominance of IL-36 signaling in COPD, IL-17-car

Machine learning to improve analysis of disability in electronic health records: an untapped opportunity for health inequities research.

Electronic Health Records (EHRs) are a leading source of epidemiological data, but often lack information on a patient's disability status. This gap hampers our ability to analyse the full scope of health inequities faced by people with disabilities. Current approaches to identify disability within EHRs have limitations because of inadequate proxies for disability or issues linking data sources. Machine learning (ML) offer unprecedented opportunities to create disability markers within EHRs, such as through unsupervised learning to classify disability groups and Natural Language Processing to
