Purification and characterization of a vasoactive intestinal polypeptide-degrading endoprotease from porcine antral mucosal membranes.

A neutral endoprotease was isolated from porcine antral mucosa and purified to homogeneity as examined by SDS-polyacrylamide gel electrophoresis (PAGE). Throughout the purification, t-butyloxycarbonyl-Arg-Val-Arg-Arg-4- methylcoumaryl-7-amide (MCA) was used as a substrate, which was found to be hydrolyzed specifically by the enzyme at the Arg-Arg bond. Unexpectedly, however, the enzyme was also found to hydrolyze vasoactive intestinal polypeptide (VIP) fairly specifically and more efficiently when various neuropeptides and related peptides were examined as substrates. It could degrade VIP by c

Drugs affecting the inner ear. A review of their clinical efficacy, mechanisms of action, toxicity, and place in therapy.

Many drugs have a site of action within the inner ear. The list includes therapeutic, diagnostic and ototoxic agents. Therapeutic agents are most useful in cases of infections, endolymphatic hydrops, vascular insufficiency, vertigo of peripheral origin, autoimmune disease, otosclerosis (otospongiosis), sudden hearing loss and tinnitus. For infections, the most widely used anti-microbial agents are the penicillins and cephalosporins. There are no antiviral agents that have been proven useful for inner ear viral infections. However, steroids have been of some value for controlling some of the se

THE SITE OF THE 5-HYDROXYTRYPTAMINE RECEPTOR ON THE INTRAMURAL NERVOUS PLEXUS OF THE GUINEA-PIG ISOLATED ILEUM.

Dose/response measurements were made on the guinea-pig isolated ileum with six agonists, acetylcholine, 5-hydroxytryptamine, nicotine, dimethylphenylpiperazinium, choline phenyl ether and histamine. The dose effects were repeated in the presence of each of twelve antagonists and one anticholinesterase. Acetylcholine and histamine were chosen because of their direct mode of action on smooth muscle, nicotine, dimethylphenylpiperazinium and choline phenyl ether were used as examples of drugs that act at the ganglionic acetylcholine receptor. 5-Hydroxytryptamine was the drug investigated. Hyoscine

Comparative impact of manual and electric toothbrushing on acid-eroded enamel.

OBJECTIVE: To evaluate and compare the enamel wear resulting from manual and electric toothbrushing following acid erosion, using natural human enamel specimens. The study aimed to quantify both surface tissue loss and subsurface prism alterations. METHODS: Twenty-four caries-free extracted human molars were decoronated and sectioned to expose natural buccal enamel surfaces. Samples were divided into two groups (manual vs electric) and subjected to four cycles of citric acid erosion (pH 2.7) followed by controlled brushing (30 strokes per cycle, total 120) using a non-fluoridated toothpaste s

Mineralization/Nano-deposition Boosts Adhesive Infiltration via Dehydration.

Preadhesion mineralization and in situ nano-deposition improving dentin bonding durability are currently explained by divergent paradigms: the former is attributed to collagen strengthening and protease fossilization, while the latter is known for the release of interface-confined water. This study challenges this dichotomy by hypothesizing that preadhesion mineralization fundamentally acts as a dehydration strategy. Two bonding strategies were formulated: a polyacrylic acid-stabilized amorphous calcium fluoride (PAA-ACF) preadhesion mineralization and in situ ACF nano-deposition. Cryo-transmi

Reducing charging polarization in quasi-solid-state rechargeable magnesium-oxygen batteries through cathode-localized 2,2,6,6-tetramethylpiperidine-1-oxyl/ionic-liquid redox mediation.

Rechargeable magnesium-oxygen (Mg-O2) batteries offer high theoretical energy density, but the electronically insulating and kinetically persistent MgO discharge product imposes severe charging polarization. Here, an aliquot of 2,2,6,6-tetramethylpiperidine-1-oxyl (TEMPO) dissolved in the ionic liquid 1-butyl-1-methylpyrrolidinium bis(trifluoromethanesulfonyl)imide (Pyr14TFSI) is localized directly at a ruthenium nanoparticle-decorated multi-walled carbon nanotube (Ru/CNT) cathode in a Mg-O2 battery employing poly(vinylidene fluoride-co-hexafluoropropylene)-based quasi-solid-state electrolyte,

An Infection-Responsive KCoF3@Fe-HA Hydrogel for Localized Co/Fe Dual Fenton-like Therapy of MRSA-Infected Wounds.

Methicillin-resistant Staphylococcus aureus (MRSA)-infected wounds remain difficult to treat because bacterial persistence, inflammation, antimicrobial resistance, and poorly controlled local toxicity can jointly impair tissue repair. Herein, we report an infection-responsive KCoF3@Fe-HA hydrogel that integrates potassium cobalt fluoride (KCoF3) with an Fe3+-crosslinked hyaluronic acid (HA) network for localized Co/Fe dual Fenton-like antibacterial therapy. Notably, the use of KCoF3 in antibacterial wound dressing systems has not yet been reported. In this design, KCoF3 serves as a confined Co

Evidence Drift and Causal Maturation Drift in Pediatric Health Research: A Dual-Drift Framework and Preliminary Appraisal Instruments for Inferential Fidelity.

Pediatric health research must translate evidence into decisions that affect children's development, safety, function, and long-term well-being. Scientific progress can still fail in two opposing ways: claims may exceed their evidentiary support, or research programs may remain productive while repeatedly refining an established signal without resolving decision-relevant uncertainty. These risks are amplified by developmental heterogeneity, ethical constraints, surrogate or short-term outcomes, long follow-up, and caregiver-mediated implementation. This concept paper formalizes these failures

Bio-based cellulose membrane from Phlomis tuberosa L. for methylene blue dye removal from wastewater and molecular interaction mechanisms.

The rapid rise in the use of synthetic dyes, particularly cationic dyes such as methylene blue (MB), has raised serious environmental and public health concerns. Effective removal of these pollutants from wastewater requires adsorbents that are both efficient and sustainable, positioning bio-based cellulose membranes as a promising alternative. In this study, cellulose was extracted from the stems of Phlomis tuberosa L. (P. tuberosa), an indigenous plant of Kazakhstan, and subsequently fabricated with polyvinylidene fluoride (PVDF). The incorporation of PVDF enhanced the mechanical strength, t

Genome-Wide Association Studies on Dental Caries: A Systematic Review.

INTRODUCTION: Dental caries is a complex, multifactorial pathological disease. Despite advances in dental care, dental caries continues to pose a significant public health challenge. In addition to the behavioural factors (e.g., sugar exposure) and environmental/oral hygiene factors (e.g., fluoride exposure in water and/or toothpaste) on the development of dental caries, potential genetic influence on dental caries susceptibility is possible, but evidence is inconclusive, especially from a candidate gene approach. Over the past 2 decades, a genome-wide association study (GWAS) approach has als

Dual-Functional Ionic Liquid Varnishes for Dental Caries Management.

The development of caries management materials has concentrated on the dual objectives of inhibiting cariogenic microorganisms and promoting remineralization. In this study, 2 dual-functional ionic liquid (IL) varnishes, exhibiting both antibacterial and remineralization capabilities, were synthesized as novel anticaries agents. The ionic liquids (ILs) were prepared by modifying 1-hexylimidazolium IL with 3-chloropropyltriethoxysilane, followed by anion exchange with F- and a coordination reaction with Sr2+, respectively. The ILs were characterized using energy-dispersive spectroscopy (EDS), i
