MdMYB70 transcription factor modulates malate accumulation under drought stress in apple.

Malic acid, a key component of the organoleptic quality, determines the flavor of apple (Malus domestica Borkh.) fruit. R2R3-MYB transcription factors (TFs) have important functions in stress response, but their roles in drought-mediated malate metabolism remain poorly understood. In this study, a drought-responsive gene, MdMYB70, which was relevant to the content of malate, was identified and characterized. Overexpressing MdMYB70 promoted malic acid accumulation and improved drought tolerance, while silencing MdMYB70 showed the opposite results. Moreover, MdMYB70 directly combined with the pr

Chiisanogenin Targets MLKL to Restore Autophagy and Suppress Pyroptosis in Renal Ischemia-Reperfusion Injury.

Renal ischemia-reperfusion injury (IRI) drives acute kidney injury (AKI) and its progression to chronic kidney disease (CKD), currently lacking effective treatments. This study identifies chiisanogenin, the key bioactive component from Eleutherococcus sessiliflorus fruit extract, as a potent nephroprotective agent. Mechanistically, chiisanogenin targets the mixed lineage kinase domain-like pseudokinase (MLKL)-lysosome axis. It inhibits the pathological translocation of MLKL to the lysosomal membrane, thereby preserving lysosomal integrity. This action promotes the nuclear translocation of tran

Complex phloroglucinols from glandular hairs of Mallotus philippensis and their antiviral activities.

Fourteen previously unknown phloroglucinol derivatives (1-14) and six known analogues (15-20) were extracted from Mallotus philippensis fruit glandular hairs. X-ray diffraction, NMR, ECD, OR calculations, and comprehensive spectroscopic analysis were adopted to determine their structures. Notably, compounds 1-4 represent unprecedented skeletal architectures, wherein 1 and 2 are chalcone-phloroglucinol-monoterpene adducts, 3 is a chalcone-flavonoid hybrid, and 4 is a chalcone-phloroglucinol adduct. All isolates were tested for antiviral activity against RSV and HSV-1 by determining their capabi

Scurvy mistaken for cutaneous vasculitis in an adult with obesity: A case report.

Scurvy is an increasingly recognized but frequently overlooked nutritional deficiency in developed countries. We report a 55-year-old man with obesity and type 2 diabetes mellitus who presented with progressive petechiae and ecchymoses concerning for cutaneous vasculitis. Extensive hematologic, rheumatologic, infectious, and dermatologic evaluations, including skin biopsy, were unrevealing. Symptoms were initially attributed to aspirin use without improvement following dose reduction. Subsequent nutritional assessment revealed an undetectable plasma vitamin C level (<0.1 mg/dL) in the setting

Immunoglobulin germline reference sequences enable investigations and reveal insights into bat-specific immunity.

We generated a highly contiguous, annotated genome of the Jamaican fruit bat, Artibeus jamaicensis, including annotated germline immunoglobulin heavy-chain (IGH) and light-chain lambda (IGL) loci to understand bat B cell receptor repertoires. The bat germline shares many structures and features described in human immunoglobulin loci. However, some features are unique to A. jamaicensis, including an expansion of cysteine-rich IGHV genes. To investigate the relationship between the germline IGH locus and expressed B cell receptors (BCRs), we sequenced the BCRs of wild-caught and captive A. jamai

Mitochondrial Neurodegeneration: Lessons from Drosophila melanogaster Models.

The fruit fly-i.e., Drosophila melanogaster-has proven to be a very useful model for the understanding of basic physiological processes, such as development or ageing. The availability of straightforward genetic tools that can be used to produce engineered individuals makes this model extremely interesting for the understanding of the mechanisms underlying genetic diseases in physiological models. Mitochondrial diseases are a group of yet-incurable genetic disorders characterized by the malfunction of the oxidative phosphorylation system (OXPHOS), which is the highly conserved energy transform

Latex-Free Anesthesia for Craniosynostosis Surgery Associated With Xia-Gibbs Syndrome: A Case Report.

This case report describes the anesthetic management of a one-year-old patient with Xia-Gibbs syndrome, which is a rare genetic condition caused by a mutation in the AHDC1 gene. The procedure involved calvarial vault remodeling and fronto-orbital advancement to correct a left coronal craniosynostosis. In addition, the patient had a history of seizures and latex-fruit syndrome, which necessitated careful preoperative planning and management. Despite the difficulties provided by the patient's cranial abnormalities and the paucity of literature on anesthetic experiences with the condition, the tr

"Bunch of acai berries sign": a new radiological sign in patients with CNS involvement in Chagas disease.

Chagas disease is an infection caused by Trypanosoma cruzi, a parasite endemic in Latin America. Acute involvement of the CNS by Chagas has been considered rare, but presumed reactivation of chronic disease in immunosuppressed patients has been the subject of recent reports. Our objective is to describe the clinical and imaging characteristics of four patients with Chagas disease and CNS involvement, and the patients had to have available MRI and a diagnosis confirmed by biopsy. The imaging findings were similar, highlighting the presence of focal cerebral lesions with hypointensity on T2-WI,

Model organisms for functional validation in genetic renal disease.

Functional validation is key for establishing new disease genes in human genetics. Over the years, model organisms have been utilized in a very effective manner to prove causality of genes or genetic variants for a wide variety of diseases. Also in hereditary renal disease, model organisms are very helpful for functional validation of candidate genes and variants identified by next-generation sequencing strategies and for obtaining insights into the pathophysiology. Due to high genetic conservation as well as high anatomical and physiological similarities with the human kidney, almost all gene

Cortical representation of group social communication in bats.

Social interactions occur in group settings and are mediated by communication signals that are exchanged between individuals, often using vocalizations. The neural representation of group social communication remains largely unexplored. We conducted simultaneous wireless electrophysiological recordings from the frontal cortices of groups of Egyptian fruit bats engaged in both spontaneous and task-induced vocal interactions. We found that the activity of single neurons distinguished between vocalizations produced by self and by others, as well as among specific individuals. Coordinated neural a

Multisensory interactions regulate feeding behavior in Drosophila.

The integration of two or more distinct sensory cues can help animals make more informed decisions about potential food sources, but little is known about how feeding-related multimodal sensory integration happens at the cellular and molecular levels. Here, we show that multimodal sensory integration contributes to a stereotyped feeding behavior in the model organism Drosophila melanogaster Simultaneous olfactory and mechanosensory inputs significantly influence a taste-evoked feeding behavior called the proboscis extension reflex (PER). Olfactory and mechanical information are mediated by ant

Brassinosteroids in Plants: Crosstalk with Small-Molecule Compounds.

Brassinosteroids (BRs) are known as the sixth type of plant hormone participating in various physiological and biochemical activities and play an irreplaceable role in plants. Small-molecule compounds (SMCs) such as nitric oxide (NO), ethylene, hydrogen peroxide (H2O2), and hydrogen sulfide (H2S) are involved in plant growth and development as signaling messengers. Recently, the involvement of SMCs in BR-mediated growth and stress responses is gradually being discovered in plants, including seed germination, adventitious rooting, stem elongation, fruit ripening, and stress responses. The cross

A computational overview on phylogenetic characterization, pathogenic mutations, and drug targets for Ebola virus disease.

The World Health Organization declared Ebola virus disease (EVD) as the major outbreak in the 20th century. EVD was first identified in 1976 in South Sudan and the Democratic Republic of the Congo. EVD was transmitted from infected fruit bats to humans via contact with infected animal body fluids. The Ebola virus (EBOV) has a genome size of ∼18,959 bp. It encodes seven distinct proteins: nucleoprotein (NP), glycoprotein (GP), viral proteins VP24, VP30, VP35, matrix protein VP40, and polymerase L is considered a prime target for potential antiviral strategies. The current US FDA-approved anti-E

Effect of red thyme oil (Thymus vulgaris L.) vapours on fungal decay, quality parameters and shelf-life of oranges during cold storage.

This work has been aimed at studying the effect of red thyme oil (RTO, Thymus vulgaris L.) on the shelf-life and Penicillium decay of oranges during cold storage. RTO vapours significantly reduced (P ≤ 0.05) the percentage of infected wounds, the external growth area and the production of spores in inoculated orange fruit stored for 12 days at 7 °C in a polypropylene film selected for its appropriate permeability. Among the RTO compounds, p-cymene and thymol were the most abundant in packed boxes at the end of cold storage. The RTO vapours did not affect the main quality parameters of the oran

Predictability and hierarchy in Drosophila behavior.

Even the simplest of animals exhibit behavioral sequences with complex temporal dynamics. Prominent among the proposed organizing principles for these dynamics has been the idea of a hierarchy, wherein the movements an animal makes can be understood as a set of nested subclusters. Although this type of organization holds potential advantages in terms of motion control and neural circuitry, measurements demonstrating this for an animal's entire behavioral repertoire have been limited in scope and temporal complexity. Here, we use a recently developed unsupervised technique to discover and track
