Artemisinin, a sesquiterpene lactone produced in Artemisia annua, is the most effective antimalarial compound, but its low natural abundance in A. annua remains a major bottleneck for large-scale production. Increasing its biosynthesis requires a better understanding of the regulatory networks controlling artemisinin biosynthesis genes. Several jasmonate (JA)-responsive transcription factors have been characterized, but the role of upstream kinases remains poorly understood. Here, we identify AaSnRK2.6, a SnRK2III-type protein kinase, as a novel positive regulator of artemisinin biosynthesis.

Flowering time is a critical developmental process that directly influences reproductive success, environmental adaptation, and agricultural productivity in Brassica crops. Recent climate instability, including drought, temperature fluctuations, salinity, and irregular photoperiods, has intensified the need to understand the molecular mechanisms regulating flowering adaptation. WRKY transcription factors are key integrators of flowering time. They connect environmental cues, hormone networks, and circadian rhythms to floral regulator genes. This review summarizes the structural, evolutionary,

The TKI-targeted agent lapatinib has been applied in clinical oncology for the management of multiple malignancies. Nonetheless, its therapeutic benefit is restricted by cardiotoxic effects that endanger patient survival, and the underlying molecular basis remains unclear. The GSE146096 dataset containing transcriptomic profiles of lapatinib-exposed human cardiomyocytes was analyzed to identify ferroptosis-related differentially expressed genes (DEGs). Protein expression of selected targets was subsequently confirmed by Western Blot. Reactive oxygen species (ROS) accumulation, Fe²⁺ levels, and

Atropa belladonna L. (Solanaceae) is a significant source of the pharmacologically important tropane alkaloids (TAs), particularly hyoscyamine and scopolamine. Although rolB and rolC from Rhizobium rhizogenes are known to enhance plant secondary metabolism, their functional specificities and combined actions in TAs biosynthesis remain unclear. Here, we generated transgenic A. belladonna overexpressing 35S::rolB, 35S::rolC, and 35S::rolB + rolC to systematically investigate their effects on TAs accumulation, plant morphology, and drought tolerance. Co-expression of rolB and rolC exhibited metab

The Keap1-Nrf2 pathway is a critical pathway for cellular antioxidant defense, with Keap1 acting as its central regulator. Unlike mammals, which possess only a single Keap1 gene, fish have two keap1 paralogs, keap1a and keap1b, which may have functionally diverged. In this study, we cloned and characterized keap1a and keap1b genes in grass carp (Ctenopharyngodon idella) and investigated their responses to heat stress through both in vivo and in vitro experiments. Our results showed that keap1a was downregulated and nrf2 was upregulated in both the spleen and liver during heat stress. In contra

Azole-resistant Aspergillus infections are a source of increasing concern with limited alternative therapeutic options. However, as most infections are still caused by azole-susceptible Aspergillus strains, there is a need to better understand fungal responses to azole antifungals. To this end, we discover that a long noncoding RNA, afu-182, is a major regulator of cyp51-independent sub-minimum inhibitory concentration (MIC) azole response. We observe that the loss of afu-182 leads to increased surface-attached growth and poorer disease outcomes in a murine model of invasive pulmonary aspergil

Inadequate supply of oxygen causing hypoxic cellular stress drives metabolic reprograming across diverse physiological conditions, including cancer. The activation of hypoxia-inducible factors (HIFs) to facilitate adaptation to low oxygen environments is well-characterized; however, post-transcriptional regulation by microRNAs (miRNAs) is poorly understood. Here, we investigated the mRNA and miRNA response in hypoxia-mediating reduced growth and cellular metabolism. Next-generation sequencing revealed the impact of hypoxia in cultured human hepatocellular carcinoma cells and identified over 40

Natural killer T cells (NKTs) are a promising platform for cancer immunotherapy, but few genes involved in the regulation of NKT therapeutic activity have been identified. To find regulators of NKT functional fitness, we developed a CRISPR/Cas9-based mutagenesis screen that uses a guide RNA (gRNA) library targeting 1,118 immune-related genes. Unmodified NKTs and NKTs expressing a GD2-specific chimeric antigen receptor (GD2.CAR) were transduced with the gRNA library and exposed to CD1d+ leukemia or CD1d-GD2+ neuroblastoma cells, respectively, over six challenge cycles in vitro. Neonatal bile acid metabolism is distinct from that of adults due to developmental regulation of key transporters and enzymes. The apical sodium-dependent bile acid transporter (ASBT) is transiently repressed in the intestine after birth, yet its role in neonatal bile acid homeostasis remains unclear. Here, we demonstrate that ASBT plays a crucial role in limiting fecal bile acid loss and suppressing hepatic bile acid synthesis in neonates. ASBT-deficient pups exhibited a marked decrease in serum bile acids and concomitant increase in fecal bile acids, accompanied by upregulated hepatic bile a

Iron stress adversely impacts plants' growth and development. Transcription factors (TFs) receive stress signals and modulate plant tolerance by influencing the expression of related functional genes. In the present study, we investigated the role of an apple bHLH transcription factor MxbHLH30 in the tolerance to iron stresses. The expression of MxbHLH30 was induced significantly by low-iron and high-iron treatments and MxbHLH30-overexpressed Arabidopsis plants displayed iron-stress-tolerant phenotypes. A determination of physiological and biochemical indexes associated with abiotic stress res

BACKGROUND: Malaria-associated acute respiratory distress syndrome (MA-ARDS) is a lung complication affecting 5-25% of adults with severe malaria and has a mortality rate of ~ 40% despite mechanical ventilation. Effective recovery requires the resolution of inflammation, an active process involving pathogen clearance, suppression of inflammation, and tissue repair. While antimalarial drugs eliminate the parasite, they do not adequately promote resolution.
METHODS: Using Nanostring nCounter technology, 840 inflammation- and metabolism-related genes were profiled in lung tissue from Plasmodium

Formate is a single-carbon compound that is challenging to assimilate, including when assimilation involves a CO2 intermediate that can diffuse from the cell. Mutations that overcome such challenges can be identified through adaptive laboratory evolution. We evolved the anoxygenic phototrophic bacterium Rhodopseudomonas palustris to use formate as the sole carbon source. Through gene deletions, we determined that formate is assimilated via oxidation to CO2 by formate dehydrogenase, followed by CO2 fixation by the Calvin cycle. However, this pathway had no clear link to three genes that were co

The epidermal growth factor receptor (EGFR) is a critical regulator of multiple oncogenic signaling cascades, including MAPK, PI3K/AKT/mTOR, and JAK-STAT pathways, which collectively contribute to enhanced proliferation, angiogenesis, and resistance to apoptosis in cancer. Aberrant EGFR activation has been strongly associated with tumor progression and therapeutic resistance, underscoring its importance as a molecular target for anticancer interventions. Ailanthus altissima (A. altissima), is known for its broad-spectrum anticancer potential although the underlying mechanism has not yet been c

Prenatal brain development is particularly sensitive to chemicals that can disrupt synapse formation and cause neurodevelopmental disorders. In most cases, such chemicals increase cellular oxidative stress. For example, prenatal exposure to the anti-epileptic drug valproic acid (VPA), induces oxidative stress and synaptic alterations, promoting autism spectrum disorders (ASD) in humans and autism-like behaviors in rodents. Using VPA to model chemically induced ASD, we tested whether activation of cellular mechanisms that increase antioxidant gene expression would be sufficient to prevent VPA-i

Salicylic acid (SA) plays important roles in plant local and systemic resistance. Isochorismate synthase 1 (ICS1) is a key enzyme in SA synthesis. Pathogens infection triggered the ICS1 expression and induced SA production. However, the molecular regulation mechanism of ICS1 against virus infection remains unclear. Here, we employed molecular genetics and physiobiochemical approaches to confirm a transcription factor NbNAC1 from Nicotiana benthamiana is a positive regulator of resistance against tobacco mosaic virus (TMV). The pathways NbNAC1 and NbICS1 can be triggered by TMV infection. Silen

The biological basis of the high clinical heterogeneity of T-LGL Leukemia (T-LGLL) is not completely understood and effective therapies for this disease are lacking. Through RNA-Sequencing of purified T-LGLs we reveal gene expression profiles and pathway dysregulations in the major patient subgroups, defined by CD8+ or CD4+ phenotype and STAT3/STAT5B mutational status. Overall, T-LGLL patients exhibited a marked transcriptome dysregulation compared to controls. This was more pronounced in the most symptomatic CD8 + STAT3-mutated patients, which emerged as a distinct biological entity, separate

The development of insecticide resistance in insect populations is a major challenge to sustainable agriculture and food security worldwide. Methoxyfenozide, an insect growth regulator that acts as an agonist of 20-hydroxyecdysone (20E), has severely declined in its efficacy against the rice stem borer (Chilo suppressalis), a notorious pest of rice crops in East and Southeast Asia. To date, however, the genes involved in methoxyfenozide resistance in target pests remain unclear. We conducted a long-term (seven years from 2017 to 2023) and large geographical scale (8 provinces and 45 cities in

Alopecia areata (AA) is an easy-recurring disease that presents huge challenges globally. An efficient clinical tool to predict AA onset would be valuable for timely intervention. We extracted six AA-related datasets from Gene Expression Omnibus (GEO). GO, KEGG, GSEA, GSVA and CIBERSORT algorithm were performed to elucidate the characteristics of AA. Feature genes were identified using LASSO regression and Random Forest algorithms. Five machine learning algorithms (Logistic Regression, K-nearest neighbors, Elastic Net, XGBoost and LightGBM) were employed to construct predictive models, with in
