The environmental co-occurrence of disinfectants and antibiotics has emerged as a typical multi-pollutant issue, with river sediments serving as critical sinks and accumulation media. However, whether their combined exposure accelerate antibiotic resistance genes (ARGs) dissemination in sediments remains unclear. Here, we systematically evaluated the effects of single and combined exposures to sodium hypochlorite and representative antibiotics on the sediment resistome and microbial communities. The results demonstrated that combined exposure generally increased the relative abundances of ARGs

Microbial communities in peatlands vary with depth, but most studies have been limited in spatial extent or replication, constraining assessment of general community patterns. We characterized microbial community structure and metabolic potential across replicated depth profiles in seven boreal peatlands in Sweden. Combining 16S rRNA gene sequencing and shotgun metagenomics, we identified recurring vertical stratification across sites. Microbial richness declined with depth, and core taxa including Rice Cluster II, Terriglobales, Subgroup 2, Roseiarcus, Methylocystis, and Candidatus Solibacter

Mutations in the filaggrin (FLG) gene are recognized as major genetic predispositions for atopic dermatitis (AD), yet characterization of FLG protein expression in human subjects remains limited due to the invasive nature of skin biopsies. The present study aimed to evaluate buccal cells as a non-invasive alternative for assessing FLG protein expression and degradation profiles in AD. Buccal cells were collected from 36 patients with AD and 36 healthy controls (aged 20-40 years). FLG gene expression was evaluated using reverse transcription quantitative PCR, while FLG protein expression was as

During the COVID-19 pandemic, the need for quick public health action often conflicted with the careful methods required in phylogenetics. To explore this, we reviewed 217 SARS-CoV-2 studies published from January 2020 to March 2025 in 121 journals. We found many methodological problems that weaken the reliability and reproducibility of these studies. Key issues include missing outgroup sampling, which affects ingroup topology, tree rooting and how we interpret evolutionary changes and relationships. Another issue is the lack of gene annotations, which can cause characters from one gene to ali

Dianthus helenae Vved. is an endemic medicinal species of the Nuratau Mountains, Uzbekistan, and its genomic resources have remained largely unavailable. In this study, we sequenced, assembled, and characterized the complete chloroplast genome of D. helenae and evaluated its phylogenetic position within Dianthus. The plastome exhibited a typical circular quadripartite structure with a total length of 149,567 bp, comprising a large single-copy (LSC) region of 82,856 bp, a small single-copy (SSC) region of 17,105 bp, and a pair of inverted repeats (IRs) of 24,803 bp each. The genome contained th

RATIONALE: Thickening of the basement membrane zone (BMZ) has often been identified in airway biopsies of individuals with asthma, but the relationship of BMZ thickness to airway dysfunction and inflammation is incompletely understood. The objectives of the present study were to characterise the relationship between BMZ thickness and airway physiology, immune cell infiltration and airway gene expression.
METHODS: We used design-based stereology to measure BMZ thickness in endobronchial biopsy samples obtained from 10 healthy control subjects and 31 individuals with asthma not receiving asthma

The present study investigated the differential mRNA expression of energy metabolism and stress-response genes (ADCY10, AK1, ATP5F1B, and HSP70) in fresh and frozen-thawed Murrah buffalo spermatozoa subjected to uniform 2-h in vitro capacitation. Semen samples (n = 32 per group) from eight bulls were divided into four groups: fresh non-capacitated (G1, control), fresh capacitated (G2), frozen non-capacitated (G3), and frozen capacitated (G4). Relative expression was analyzed by RT-qPCR using GLUT5 as the reference gene. Results revealed that ADCY10 was significantly upregulated in G2, G3, and

The CCR4-NOT complex is a central regulator of post-transcriptional gene expression that controls mRNA deadenylation and decay. Although the catalytic subunits have been well characterized, the physiological roles of several non-catalytic subunits including CNOT11 in mammalian tissues remain incompletely understood. Here, we investigate the physiological role of CNOT11 using genetic mouse models. No homozygous global Cnot11 knockout mice are obtained from heterozygous intercrosses, indicating that CNOT11 is required for early development. We generate hepatocyte-specific Cnot11 knockout mice (C

Cytoplasmic male sterility (CMS) is a well-known example of mitonuclear genetic conflict over sex determination in hermaphrodite plants, where mitochondrial genes maternally inherited sterilize the male function while biparental inherited nuclear genes restore it. CMS has been recently discovered in wild animals, in the freshwater snail Physa acuta. In this species, CMS is associated with two extremely divergent mitogenomes D and K compared to the classical mitogenome N. D individuals are male-steriles, while male fertility is restored in K individuals. We hypothesized that the extreme diverge

Colistin resistance mediated by the plasmid-borne mcr-1 gene presents a significant challenge in both human and veterinary medicine. While colistin-resistant bacteria have been reported in food-producing animals and humans, mcr-1-harboring strains remain relatively underreported in companion animals, particularly in South Korea. In this study, a colistin-resistant Escherichia coli strain Z1324PEC0026 was isolated from a clinically healthy companion dog owned by a veterinary nurse, which exhibited resistance to multiple antimicrobials, including colistin, β-lactams, aminoglycosides, tetracyclin

Water scarcity impacts soybean cultivation and productivity globally. The ability of plants to withstand drought stress involves complex molecular and physiological mechanisms that facilitate the restoration and maintenance of cellular homeostasis. This study identified genes associated with carbohydrate metabolism and GABA shunt pathway that respond to water deficit in two soybean varieties. These varieties exhibited contrasting responses to water scarcity, and were subjected to two distinct cropping systems. In the drought-tolerant variety, a strategy for conferring tolerance was observed th

Background: Arboviral transmission by Aedes aegypti is exacerbated by insecticide resistance, which undermines effective vector control strategies. In the Philippines, deltamethrin resistance in several Ae. aegypti populations has been reported and prompts an investigation into the possible molecular mechanisms involved. Methods: Two Ae. aegypti populations were subjected to susceptibility assays to deltamethrin and were RNA-sequenced for transcriptome analysis. Results: The two (2) Ae. aegypti populations were resistant to deltamethrin and had LC50 values of 1.26 and 2.15 μg/mL at the zeroth

Myasthenia gravis (MG) is a T/B cell-driven autoimmune disease. The immunomodulatory mechanisms of the common immunosuppressant tacrolimus (TAC) on the immune repertoire are unclear. This study investigated TAC's immunomodulatory effects via high-throughput sequencing of peripheral blood mononuclear cells from four MG patients pre- and post-four months of TAC monotherapy, revealing dynamic T-cell receptor (TCR) and B-cell receptor (BCR) repertoire remodeling. The immune repertoire of MG patients was characterized by a skewed usage of TRBV gene families compared to healthy controls, indicating

INTRODUCTION: Hidradenitis suppurativa (HS), psoriasis (PsO), and atopic dermatitis (AD) have unique disease pathogenesis and distinct pathology, despite sharing some immunological pathways. This study aimed to evaluate the transcriptomic differences between HS, PsO, and AD lesions, which drive these unique pathogenic features.
METHODS: Bulk RNA sequencing was performed on 19 healthy controls (HC), 15 HS, 21 PsO, and 15 AD lesional skin samples.
RESULTS: PsO lesions were defined by a strong interleukin (IL)-17 signature, and AD lesions had increased T helper (Th2) signaling. HS lesions had a

Ticks, recognized as the second most significant vector of pathogens after mosquitoes, are of considerable interest in medical research. Although mitochondrial genomes are commonly employed in the phylogenetic studies of insects and arthropods, investigations into tick mitochondrial genomes are relatively scarce. The evolutionary and phylogenetic relationships at the family and genus levels remain unresolved. In this study, the mitochondrial genomes of Haemaphysalis warburtoni, Ixodes crenulatus, Rhipicephalus bursa, and Rhipicephalus pumilio were first sequenced and annotated based on the Ill

Golden pompano (Trachinotus ovatus) is an economically important marine fish in southern China, but intensive aquaculture has increased the risk of bacterial diseases, especially streptococcosis caused by Streptococcus agalactiae. As a key component of innate immunity, the NLRP3 inflammasome (NOD-like receptor family, pyrin domain containing 3 inflammasome) regulates inflammatory responses through caspase-1 (Cysteine-aspartic acid protease 1) activation and the maturation of IL-1β (Interleukin-1β) and IL-18 (Interleukin-18). In this study, five core genes associated with the NLRP3 inflammasome

The increasing incidence of vancomycin-resistant enterococci (VRE) over the past decade has reduced treatment options largely to linezolid and daptomycin. However, the emergence of resistance to both agents further complicates the management of VRE infections. While the mechanisms of linezolid resistance are relatively well understood, those underlying daptomycin resistance remain less clearly defined. In this study, we analyzed genomic changes associated with the development of linezolid and daptomycin resistance in initially susceptible isolates following treatment at a Danish university hos

Plague, caused by Yersinia pestis, has affected human societies for millennia, yet the timing, scale and biological shifts underlying its earliest spread remain unclear. Here, we screened 1,400 ancient human remains from eastern Eurasia (8,000-400 cal. BP) and recovered three prehistoric Y. pestis genomes, including one from East Asia. Integrating these data with 136 published ancient and modern Y. pestis genomes, we resolved a trans‑Eurasian polytomy dated to ~5.3 ka BP that links Scandinavia, Central Europe, and northern China. Low SNP distances and tip-dating indicate splits over century‑sc
