Human cytomegalovirus (HCMV), recognized as the largest known DNA virus in the Herpesviridae family, is widely disseminated throughout the human population and poses a substantial threat to public health. HCMV is highly infectious. A majority of HCMV-infected individuals usually exhibit asymptomatic latent infections, while the immunocompromised or immunosuppressed populations infected with HCMV have high mortality. HCMV has long stood as a hot topic of research in the international virology community, and elucidating its pathogenic mechanisms is broadly regarded as a prerequisite for the deve

Leishmaniasis, a vector-borne disease affecting millions worldwide, is caused by protozoan parasite Leishmania. In mammalian hosts, Leishmania survives in the hostile environment of macrophages by exploiting key metabolic pathways to evade their destruction and subvert the host immune responses. Understanding of how Leishmania alters host immune cell metabolism could reveal novel targets for vaccines and therapeutics for effective control of Leishmaniasis. This review focuses on Leishmania-induced modulation of host immune response, with particular focus on reprograming of key metabolic pathwa

Malaria control is challenged by parasite drug resistance and inadequate vaccine protection. Although mosquito gut microbes regulate Plasmodium development, integrated analyses of microbiota-Plasmodium-host interactions are lacking. Innate immunity imbalance is the primary driver of disease pathogenesis. This review proposes a unified "microbiota-barrier-innate immunity" axis hypothesis. Under physiological conditions, gut microbiota modulates host innate immunity via metabolites and maintains barrier integrity. However, Plasmodium infection disrupts this axis, causing microbiota imbalance, me

Leishmania infections profoundly reshape host immunity, yet their impact on central T-cell development remains underappreciated. The thymus, a key site for establishing a functional and self-tolerant T-cell repertoire, is highly sensitive to systemic inflammatory cues. Growing evidence indicates that parasitic diseases, including leishmaniasis, can perturb thymic structure and function through direct parasite presence or sustained inflammatory stress. Such disturbances may alter thymocyte maturation, leading to reduced T-cell output and the release of cells with aberrant specificity. Beyond co

Single-cell RNA sequencing (scRNA-seq) has emerged as a transformative technology in infectious disease research, enabling high-resolution characterization of cellular heterogeneity, immune dynamics, and host-pathogen interactions at the single-cell level. In the context of viral infections-including SARS-CoV-2, hepatitis B virus (HBV), human immunodeficiency virus (HIV), influenza virus, and respiratory syncytial virus (RSV)-scRNA-seq has provided critical insights into immune-cell heterogeneity, immune landscape remodeling, and the emergence of previously unrecognized cellular states. Crimean-Congo- Hemorrhagic Fever (CCHF) is the most widespread tick-borne disease in the world with a highly variable case fatality rate. It is caused by the CCHF virus (CCHFV). The disease, which has neither approved treatments nor vaccines, has long received very little attention until it was listed as a priority pathogen by WHO. Improving our understanding of mechanisms of host-virus interaction is essential for the development of effective therapeutic and prophylactic strategies. There is still much to be clarified to better understand how the virus interacts with its host and humans. Eluc

Prevotella species are an extremely common and abundant bacteria detected within the low microbial biomass of the lungs and are a core component of the oral microbiome. Clinical studies have drawn associations between Prevotella abundance and lung homeostasis, indicating potential relationships between Prevotella and lung inflammation, infection defense, and lung function. Across several studies in critically ill patients, the depletion of Prevotella and other obligate anaerobes is linked to significantly reduced survival, leading to calls for anaerobe preservation in empiric antibiotic therap

Tuberculosis is an infectious disease spread through airborne droplet nuclei. Mycobacterium tuberculosis is the etiological agent of this infection. Mycobacteria can cause active tuberculosis or asymptomatic latent infection due to its complex biology and host immunological responses. The genes of mycobacteria can change alveolar macrophages and boost their resistance to autophagosome-lysosome fusion. However, only 5%-10% of infected individuals progress to the active form. In this context, multiple factors are associated with the progression of the disease. Thus, the review aims to analyze th

Tuberculosis (TB) remains a global health challenge, driven by multiscale pathogenesis across molecular, cellular, tissue, and systemic levels. Within-host mathematical modelling provides a powerful way to explore these processes, providing mechanistic insights that are difficult to capture experimentally, including host-pathogen interactions, immune regulation, and treatment efficacy. This review synthesises the evolution of TB models from early ordinary differential equation systems to agent-based and hybrid multiscale approaches, leading to integrated pharmacokinetic/pharmacodynamic and sys

The contribution of the microbiome to sporadic colorectal cancer (CRC) has traditionally been interpreted through the detection of individual tumor-enriched taxa or isolated virulence factors. However, accumulating evidence indicates that CRC-associated microorganisms often act as structured polymicrobial consortia rather than as independent agents. This review proposes an expanded ecological framework for understanding the role of oral taxa in colorectal carcinogenesis. Oral bacteria, including genera Fusobacterium, Parvimonas, Peptostreptococcus, Porphyromonas, Prevotella, Streptococcus, and

Microfluidics has emerged as a powerful technology for precisely controlling mechanical and chemical environments across a wide range of biological systems, from single cells to complex co-cultures. In microbiology, the microscale dimensions of bacteria pose experimental challenges that microfluidic systems have effectively addressed, offering compatibility with live imaging and enabling quantitative investigations of bacterial behaviors and host-bacteria interactions. Historical microfluidic designs are now routinely used to quantify bacterial growth and lineage with unprecedented temporal-sp

Persistent colonization by gastrointestinal pathogens is an important environmental factor contributing to the development and progression of gastric and colorectal cancer. Helicobacter pylori (H. pylori), Epstein-Barr virus (EBV), Fusobacterium nucleatum (F. nucleatum), enterotoxigenic Bacteroides fragilis (ETBF), and pks-positive Escherichia coli (E. coli) can induce chronic inflammation and reshape the tumor microenvironment via virulence factors, epithelial barrier disruption, DNA damage, and immune dysregulation. Conventional views have largely focused on the tumor-promoting effects of ch

Natural product (NP) use by patients alongside their conventional cancer therapies is ubiquitous. This common, yet often hidden, practice can potentially contribute to significant patient harm, given the narrow therapeutic window of most anticancer drugs. This review takes on this challenge directly, moving past theoretical concerns to summarize current clinical evidence on interactions between widely used NPs and modern cancer treatments, including chemotherapy, targeted therapy, and immunotherapy. We break down the key pharmacokinetic (PK) mechanisms, such as the disruption of cytochrome P45

Colorectal cancer (CRC) remains one of the leading causes of cancer-related mortality worldwide, due to the limited efficacy of current therapeutic strategies in advanced stages. Necroptosis, a regulated form of necrotic cell death mediated by receptor-interacting protein kinases RIPK1 and RIPK3, and the pseudokinase MLKL, has emerged as a potential alternative pathway to induce cancer cell death. Recent studies suggest that modulation of necroptosis may enhance antitumor immunity, overcome therapeutic resistance, and improve clinical outcomes in CRC. In this review, we systematically analyzed

Traditional fermented horsemeat and horsemeat sausages in Xinjiang are rich in microbial resources, mainly including lactic acid bacteria and yeasts. Although the probiotic effects of lactic acid bacteria have been widely studied, there is relatively little research on the probiotic properties of yeasts, which limits their application in functional food development. This study isolated 44 yeast strains from traditional fermented horsemeat products in Xinjiang, and 16 strains exhibiting strong tolerance to temperature, acidic pH, simulated gastrointestinal fluids, and high bile salt solution we

Klebsiella pneumoniae (KP) is a global threat to human health due to the isolation of multidrug-resistant strains. Despite advancements in understanding KP's population structure, antibiotic resistance mechanisms, and transmission patterns, a gap remains in how KP evades defenses, allowing the pathogen to flourish in tissues despite an activated immune system. KP infection biology has been shaped by the notion that the pathogen has evolved to shield from defenses more than actively suppress them. This review describes new paradigms of how KP exploits the coevolution with the innate immune syst

In this experiment, salmon protein-fucoidan complexes (SMP-FU) were used to stabilize Pickering emulsions encapsulating astaxanthin (AXT). The hydrophobic forces and hydrogen bonding between SMP and FU were revealed as the main interaction to form the SMP-FU complex. The addition of FU led to the increase of three-phase contact angle from 51.3° to 92.1°, and the best hydrophilic-hydrophobic balance was achieved with 2 % FU. The emulsions stabilized by SMP-FU became more homogeneous and denser with the increase of FU concentration, and the smallest droplet size was obtained in the emulsion stab

Hydrophobic payloads incorporated into antibody-drug conjugates (ADCs) typically are superior to hydrophilic ones in tumor penetration and "bystander killing" upon release from ADCs. However, they are prone to aggregation and accelerated plasma clearance, which lead to reduced efficacies and increased toxicities of ADC molecules. Shielding the hydrophobicity of payloads by incorporating polyethylene glycol (PEG) elements or sugar groups into the ADC linkers has emerged as a viable alternative to directly adopting hydrophilic payloads. In this study, ADC linkers incorporating PEG or sugar group

Clostridioides difficile infection (CDI) is a significant cause of antibiotic-associated diarrhea and pseudomembranous colitis, manifesting as mild diarrhea, fulminant colitis, and even death. It is typically recognized as a healthcare-associated infection. Glucosyltransferase toxin A (TcdA) and toxin B (TcdB) are two major factors responsible for the pathogenicity of Clostridioides difficile (C. difficile). They bind to cell surface receptors and enter the cytoplasm via pH-dependent pore formation, causing cell death by inactivating GTPase. This review elucidates the pathogenic mechanisms of
