Potential anti-inflammatory role of activin A in acute coronary syndromes.

OBJECTIVES: We sought to investigate whether activin A could be involved in the immunopathogenesis of acute coronary syndromes. BACKGROUND: Inflammatory mechanisms seem to play a pathogenic role in atherosclerosis and acute coronary syndromes, but the actual mediators have not been fully identified. Activin A, a pleiotropic member of the transforming growth factor-beta cytokine family, has recently been suggested to play a role in inflammation. METHODS: We examined the role of activin A and its endogenous inhibitor follistatin in patients with stable (n = 26) and unstable angina (n = 20) and

Immune regulation of human colonic electrolyte transport in vitro.

The role of lamina propria cells in regulating human colonic ion transport was investigated in vitro. Normal human colonic mucosae were mounted in Ussing chambers, and short circuit current changes (delta SCC) were monitored in response to immune cell activation. Anti-human immunoglobulin E (anti-IgE) and formyl-Methionyl-Leucyl-Phenylalanine (fMLP) were used to stimulate mast cells and phagocytes respectively. Anti-IgE (100 micrograms/ml) and fMLP (100 microM) evoked rapid onset, inward delta SCC (mean (SEM) max delta SCC 19.3 (2.8) and 29.4 (4.7) microA/0.63 cm2 respectively). A pharmacologi

Role of airway reactivity in pathogenesis of asthma.

Airway reactivity is taken to mean the bronchoconstrictor response of airways to non-specific (i.e. non-allergic) stimuli, most commonly to aerosols of histamine, methacholine or prostaglandin F2 alpha, to exercise, cold air or hyperventilation. Reactivity is greatly increased in subjects with asthma when compared with normal. Normal subjects may transiently acquire hyperreactivity following viral infections or exposure to ozone, although usually not in the asthmatic range. In asthma, an initiating stimulus may also be required for the development of hyperreactivity, but genetic susceptibility

AA in Hypertension and Preeclampsia.

Human essential hypertension is driven by a network of interacting nutritional, metabolic, inflammatory, and endothelial mechanisms in which AA and other PUFAs play a critical role. Beyond excess sodium intake, population data supports the role of inadequate calcium, potassium, and magnesium intake and low antioxidant vitamin status in shaping blood pressure regulation. These nutrients influence vascular smooth muscle tone and act as cofactors that support Δ6 and Δ5 desaturase activities, thereby governing tissue availability of AA, EPA, and DHA and downstream formation of vasodilator and anti

Evaluating Macrophage Polarization and Inflammation in Bitongqing-Treated Collagen-Induced Arthritis Rats.

Rheumatoid arthritis (RA) is a chronic autoimmune disease characterized by persistent synovial inflammation and immune dysregulation. Traditional Chinese medicine formulas such as Bitongqing Decoction (BTQ) are used clinically, but reproducible workflows are needed to characterize their absorbed constituents and associated immunological effects. This article presents an integrated protocol comprising herbal preparation, collagen-induced arthritis (CIA) induction, 21-day oral treatment, serum ultra-high-performance liquid chromatography coupled with quadrupole-Orbitrap mass spectrometry (UHPLC-

When the eyes and nose collide: The shared pathways of rhinoconjunctivitis.

Although allergic rhinoconjunctivitis (ARC) is highly prevalent and involves both nasal and ocular symptoms, the mechanisms proposed to connect these responses remain poorly integrated, limiting a comprehensive understanding of the disease. Multiple anatomic, neural, and immunologic pathways have been proposed to explain the nasal-ocular interactions in ARC, yet these mechanisms remain dispersed across the literature without a recent unifying analysis. This review brings together current evidence to clarify the pathways that drive the rhinoconjunctivitis response. Specifically, anatomic connec

Extracellular matrix microarchitecture modulates cellular behavior and extracellular vesicle phenotypes in biomimetic tendon models.

Tendon disease is a highly prevalent musculoskeletal disorder characterized by extracellular matrix (ECM) disorganization and fibroblast activation, contributing to fibrotic tissue healing and impaired function. While extracellular vesicles (EVs) have emerged as key mediators of intercellular communication and drivers of fibrosis in various tissues, their role in tendon pathology remains poorly understood. In this study, we developed a physiologically relevant 3D in vitro model that recapitulates biophysical features of healthy and fibrotic tendon microenvironments to investigate EV-mediated c

Inflammatory drivers of carotid atherosclerosis: signaling pathways, immune cells, and plaque stability.

BACKGROUND: Carotid atherosclerosis (CAS; herein referring to carotid atherosclerotic disease, to be distinguished from carotid artery stenting, which shares the same abbreviation) is a primary driver of ischemic stroke, with inflammation playing a central role in its pathogenesis from plaque initiation to rupture. However, the complex regulatory network governing this inflammatory process remains to be fully elucidated. PURPOSE: This review comprehensively reviews the molecular and cellular inflammatory mechanisms underlying CAS, focusing on key signaling pathways, inflammatory mediators, an

Neurovascular Signaling in the Perivascular Space: Biophysical Principles and Disruption in Hypertension.

The brain's vasculature-from pial arteries through penetrating arterioles and the capillary bed to venous drainage-is enveloped by a narrow, compositionally elaborate perivascular space that provides the primary signaling interface between neural tissue and blood vessels. Here, we advance the principle that the geometry and composition of this space are not constant but change systematically along the vascular tree, and that perivascular space composition at these locations imposes biophysical constraints that dictate the fidelity and gain of neurovascular signaling. We argue that its narrow m

Mitigating sepsis-induced vascular endothelial dysfunction through Niban phosphorylation.

INTRODUCTION: Endothelial dysfunction is a hallmark of sepsis pathophysiology, driven by inflammation, oxidative stress, and endoplasmic reticulum (ER) stress, leading to vascular abnormalities and organ failure. Niban, a multifunctional cellular stress response protein, regulates endothelial homeostasis, with its phosphorylation playing a critical role in mitigating ER stress. We hypothesized that sepsis-induced endothelial dysfunction in the aorta is driven by reduced Niban phosphorylation, which increases ER stress, but can be restored by NiPp, a cell-permeant phosphopeptide mimetic designe

Induction of activating transcription factor 3 (ATF3) in the cerebral cortex of a mouse model of blast-induced traumatic brain injury.

Traumatic brain injury (TBI) is a leading cause of long-term disabilities and mortality worldwide. Blast-induced TBI (bTBI) is the injury most commonly sustained by military personnel, but the pathomechanisms are largely unknown. Recently, accumulating evidence has suggested that neuroinflammation, characterized by the activation of microglia and astrocytes and elevated production of inflammatory mediators such as interleukins, cytokines, and chemotactic cytokines (chemokines), is a key pathological feature in bTBI. Therefore, controlling excessive neuroinflammation is critical to improve long

Dietary inflammatory index and lung cancer risk: multi-cohort evidence and plasma proteomic profiles.

BACKGROUND: Accumulating evidence links dietary inflammatory potential to cancer, yet its association with lung cancer incidence and mortality remains inconsistent. This study aimed to examine the relationship between the Dietary Inflammatory Index (DII) and lung cancer risk, and to explore the underlying molecular mechanisms. METHODS: Associations between DII scores and lung cancer incidence were evaluated using Cox proportional hazards models within the UK Biobank cohort, with external validation performed in the NHANES dataset. Mediation analyses were conducted to investigate plasma protei

Cepharanthine alleviated chronic prostatitis/chronic pelvic pain syndrome by activating Nrf2 and inhibiting the NF-κB pathway.

BACKGROUND: Chronic prostatitis/chronic pelvic pain syndrome (CP/CPPS) is a common disorder impacting the genitourinary system of adult males, primarily driven by pathogenic mechanisms involving immune and inflammatory mediators, immune cells and molecules, oxidative stress, and apoptosis. Cepharanthine (CEP), a bisbenzylisoquinoline alkaloid, exhibits properties such as immunomodulatory, antioxidant, anti-inflammatory, and antitumor activities. Nevertheless, the effects of CEP on CP/CPPS and its underlying mechanisms remain insufficiently characterized. METHODS: Two experimental models were d

Potential crosstalk between the renin-angiotensin system and Neuropilin-1 pathways in experimental rheumatoid arthritis.

Rheumatoid arthritis (RA) is a systemic autoimmune disorder characterized by persistent synovial inflammation, pathological angiogenesis, and progressive joint destruction. Emerging evidence implicates the renin-angiotensin system (RAS) and neuropilin-1 (NRP-1) as key modulators of RA pathogenesis through regulation of inflammatory signaling, vascular remodeling, and osteoclastogenesis. This study explored the mechanistic interplay between RAS and NRP-1 signaling in a rat model of RA induced by complete Freund's adjuvant. Experimental groups received valsartan (VAL), an angiotensin II type-1 r

Effect of Buccal Acupuncture with Patient-Controlled Analgesia on Pain, Inflammation, and Coagulation in Gastric Cancer Patients.

Buccal acupuncture combined with patient-controlled analgesia (PCIA) is commonly used after surgery. This study focused on the effects of acupuncture combined with PCIA on postoperative pain, inflammatory response, and coagulation function in patients with gastric cancer (GC). From March 2024 to January 2025, 142 GC patients were selected and assigned to a routine group (79 cases) and a buccal acupuncture group (63 cases). Pre- and postoperative levels of serum pain mediators (SP, PGE2, β-EP, and Glu), inflammatory markers (IL-6, TNF-α, IL-10, and CD11b), coagulation function parameters (PT, A

Curcumin attenuates uterine pain in mice through suppression of neuroinflammation in the DRG and spinal cord.

Uterine pain associated with labor, dysmenorrhea, or endometriosis is often driven by inflammation, which enhances nociceptive signaling and contributes to hyperalgesia. Although nonsteroidal anti-inflammatory drugs (NSAIDs) and glucocorticoids are commonly prescribed, their clinical application is limited by side effects, and a subset of patients exhibit inadequate or no response to NSAID therapy. Curcumin, a natural polyphenol extracted from Curcuma longa, exhibits well-documented anti-inflammatory and analgesic properties. In this study, we investigated the analgesic efficacy and underlying

The lung-muscle axis in COPD: a conceptual framework integrating evidence, mechanisms, and therapeutic perspectives.

Skeletal muscle dysfunction (SMD) is a clinically important extrapulmonary manifestation of chronic obstructive pulmonary disease (COPD) and is associated with impaired exercise capacity, reduced physical function, increased exacerbation risk, and poorer clinical outcomes. Although traditionally viewed as a consequence of pulmonary disease progression, accumulating evidence suggests that pulmonary impairment and skeletal muscle abnormalities may reflect partially overlapping processes within the systemic pathobiology of COPD. This narrative review synthesizes clinical, imaging, genetic, mechan

Mast Cells and Cardiovascular Disorders.

Mastocytosis is a neoplasm characterised by clonal proliferation of mast cells (MCs) in the skin and other organs, including the heart, and symptoms produced by MC-derived mediators. MC activation syndrome (MCAS) relates to increased and inappropriate activation of MCs without clonal proliferation. Mast cells are tissue-resident immune cells strategically located in different cardiac sites, e.g., myocardium, pericardium, aortic valve, and close to nerves, and in atherosclerotic plaques. The mediators (e.g., histamine, tryptase, chymase, cytokines, and chemokines) have roles in inflammation, an

Esculin attenuates cholestatic liver injury and fibrosis in bile duct-ligated rats by modulating oxidative stress, inflammatory responses, and fibrogenic mediators.

BACKGROUND: Cholestatic liver injury is a major contributor to hepatic fibrosis and cirrhosis, yet effective pharmacological therapies remain unavailable. Esculin, a natural coumarin glucoside, possesses potent antioxidant and anti-inflammatory activities, but its potential to mitigate cholestasis-associated fibrosis has not been fully elucidated. This study aimed to evaluate the hepatoprotective and antifibrotic effects of esculin in bile duct ligation induced cholestatic rats. METHODS: Forty male Sprague-Dawley rats were divided into four groups: control, BDL, BDL + esculin 20 mg/kg, and BD

Identification of novel gene expression programs in thrombin-stimulated hepatic stellate cells.

BACKGROUND: Evidence suggests that the coagulation factor thrombin plays a pathologic role in liver fibrosis. Studies have proposed that thrombin signaling through protease-activated receptor-1 (PAR1) promotes activation of hepatic stellate cells (HSCs), the primary collagen-producing cells in the fibrotic liver. However, a comprehensive view of the effect of thrombin on HSCs is lacking. OBJECTIVES: This study tests the hypothesis that thrombin induces expression of fibrogenic mediators in HSCs via PAR1 activation. METHODS: Immortalized human HSCs (LX-2) were cultured for 48 hours, then stim
